4.Investigation in the levels of Th1 and Th2 type cytokines in patients with cervical cancer
Cancer Research and Clinic 1999;0(05):-
Objective To detect the levels of Th1 and Th2 type cytokines of peripheral blood CD+4 T lymphocytes in the patients with cervical cancer in order to find out the laboratory evidence of tumor immunotherapy. Methods The peripheral blood lymphocytes were stimulated by stimulator in order to enhance expression of cytokines. Then specific fluorochrome-conjugated monoclonal antibody was added to be combined with its specific antigen. Results The level of IL-2 produced by Th1 cells were lower in patients of cervical cancer than those of normal controls, there were significant difference between them. But the level of IFN-? produced by Th1 cells had no significant change in patients of cervical cancer in comparison with normal controls. The levels of Th2 type cytokines such as interleukin-4(IL-4), interleukin-6(IL-6), interleukin-10(IL-10) were higher in patients of cervical cancer than those of normal controls, there were significant difference between them. Conclusion Th1/Th2 is imbalance in cervical cancer .Th1 type cytokines are inhibited, and Th2 type cytokines are relatively enhanced, so Th1/Th2 shifts to Th2. It may be the mechanism of tumor arising and transferring by immune escape from immunosurveillance.

This paper summarized the application of LC and mass spectrometry technology in inactivated vaccines, vaccines of virus-like particles(VLPs), polysaccharide vaccines, and polysaccharide conjugate vaccines.
6.Effect of peroxisome proliferator activated receptor γ agonist on the proliferation and differentiation of rat osteoblasts in vitro
Chinese Journal of Geriatrics 2011;30(3):241-244
Objective To study the effect of rosiglitazone (RSG), the agonist of peroxisome proliferator activated receptor γ (PPARγ), on the proliferation and differentiation of rat osteoblasts and the related mechanisms. Methods The identification of rat primary osteoblasts was performed by alkaline phosphatase (ALP) staining and mineralized nodules. The 3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-di-phenytetrazoliumromide (MTT) assay and p-nitrophenyl phosphate (PNPP) assay were used to observe the effects of different concentrations of RSG on proliferation and differentiation of the osteoblasts. The reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of connective tissue growth factor (CTGF) mRNA. The effects of the different concentrations (0,1,2,5,10 and 20 μmol/L) of RSG on TGF-β1-induced CTGF mRNA expression in osteoblasts were detected. Results (1)Different concentrations of RSG could not change the proliferation of osteoblasts (P>0. 05). (2)Compared with control group, all different concentrations of RSG could suppress ALP activity in osteoblasts (P<0. 01 ). (3) RSG suppressed the osteoblats CTGF mRNA expression induced by TGF-β1 in a dose-dependent manner (P<0. 01). Conclusions In vitro, RSG can inhibit the TGF-β1 induced rat osteoblasts CTGF mRNA expression. RSG may play a potential role in preventing the differentiation of the rat osteoblasts.
7.The relationship of placental expression of leptin and disease development in patients with pre-eclampsia
Chinese Journal of Postgraduates of Medicine 2010;33(12):11-13
Objective To study the relationship of placental expression of leptin and disease development in patients with pre-eclampsia.Methods The placental expression of leptin gene was determined in gestational hypertension(10 cases),mild pre-eclampsia(10 cases),Severe pre-eclampsia(10cases)and normal pregnant women(15 cases, control group)by reverse txanscription/polymerase chain reaction(RT/PCR).Meanwhile,24 hour urine protein and mean arterial blood pressure(MAP)were detected in patients with pre-eclampsia.Results The expression of leptin mRNA in placenta were significantly higher in severe and mild pre-eclarnpsia than that in normal pregnant women(0.507 ±0.036,0.476±0.023vs 0.441 ±0.030)(P<0.01),meanwhile,in the severe pre-eclampsia was higher than that in the mild preeclampsia(P<0.05).The difference was not significant between the gestafional hypertension(0.463±0.024)and the normal pregnant women(P>0.05).There was a positive correlation between the placental leptin mRNA expression levels in pre-eclampsia and the 24 hour urine protein and MAP (P<0.05).Conclusions The placental leptin mRNA expression levels in pre-eclampsia increase. The pre-eclampsia women placental hptin mRNA expression levels correlate with pathogenetie condition. The placental leptin mRNA expression levels in pre-eclampsia correlate, with disease development.
8.Roles of calreticulin on anti-tumor effection
Journal of International Oncology 2009;36(12):900-902
Calreticulin (CRT) participates in the progression and apoptosis of many kinds of tumor cells. Recent studies indicate expression of CRT on tumor cells acts as an "eat me" signal for dendritic cells, allowing them to phagocytose tumor cells. It can induce immunogenic tumor cell death by adsorbing recombinant calreticulin to tumor cells or by enforcing the translocation of endogenous calreticulin to the cell surface. In animal experiments,CRT related tumor vaccine has good therapeutic effects in some tumor bearing mice, which can be used in tumor immunotherapy and targeted therapy.
