4.Enhancing effect of isoflavonoid genistein on radiosensitivity of DU145 prostate cancer cells.

OBJECTIVETo study the enhancing effect of isoflavonoid genistein in irradiation (IR) on prostate DU145 cancer cells.
METHODSProstate cancer cell line DU145 was used in this experiment. Clonogenic assay was applied to compare the survival fractions of DU145 cells after treatments with genistein alone and/or graded IR. DNA electrophoresis and TUNEL method were applied to detect cell apoptosis. Cell cycle was observed using flow cytometry and related protein expressions by immunoblotting.
RESULTClonogenic assay demonstrated that genistein, even at low to medium concentrations, enhanced the radiosensitivity of DU145 cells. After treatments with IR and/or genistein for 24 h, apoptosis was mainly seen with genistein at high concentration and was minimally dependent on IR. Apoptosis also occurred after treatments for 72 h with lower concentrations of genistein, especially when combined with IR. While IR or genistein led to a G2/M cell cycle arrest, combination of them could further increase DU145 cells at G2/M phase. This G2/M arrest was largely maintained at 72 h, and accompanied by increasing apoptosis and hyperdiploid cell populations. Cell-cycle related protein analysis disclosed biphasic changes in cyclin B1, less markedly increased cdc-2 and stably elevated p21(cip1) levels with increasing genistein concentrations.
CONCLUSIONGenistein could enhance the radiosensitivity of DU145 prostate cancer cells. The mechanisms might be involved in the increased apoptosis, prolonged cell cycle arrest and impaired damage repair induced by the combined treatment.

5.Application of Dynesys system combined with posterior lumbar interbody fusion in treating multiple lumbar degenerative disease.

OBJECTIVETo explore the clinical effects of Dynesys system combined with posterior lumbar interbody fusion (PLIF) in treating multiple lumbar degenerative disease.
METHODSThe clinical data of 46 patients with multiple lumbar degenerative diseases treated by Dynesys system combined with PLIF from September 2010 to May 2013 were retrospectively analyzed. There were 17 males and 29 females, aged from 38 to 68 years old with an average of (56.38±11.63) years. Operation section was in L2-L5 of 16 patients (6 with fusion of L4,5 and 10 with fusion of L4,5,L5S1) and in L3-S1 of 30 patients (11 with fusion of L5S1 and 19 with fusion of L4,5,L5S1). Patients were followed up for three times: postoperative 3 months, 1 year and final follow-up. Visual analogue scale (VAS) and Oswestry Disability Index (ODI) were used to assess clinical symptoms preoperatively and postoperatively. All patients underwent flexion/extension radiographs examinations before surgery and at final follow-up. Range of motion (ROM) and disc height index (DHI) were recorded.
RESULTSAll patients were followed up from 16 to 48 months with the mean of (23.23±7.34) months. At third follow-up after operation, ODI and VAS of lumbago and leg pain were significant improved than that of preoperative (P<0.01). DHI of fusion segment was significantly increased than that of preoperative (P<0.05). There was no significant difference in adjacent non-fusion segment between preoperative and postoperative (P>0.05). Postoperative ROM of fusion and non-fusion segments were obviously decreased than that of preoperative. There was no significant difference in ROM of upper adjacent non-fusion segment between 3 months and 1 year after operation (P>0.05), but at final follow-up, the ROM was increased (P<0.05).
CONCLUSIONThe preliminary clinical results of the Dynesys system combined with PLIF in the treatment of multiple lumbar degenerative diseases are satisfactory. It can be determined in fusion or non-fusion according to the individual needs and can reserve the some intervertebral motion, prevent the early degeneration of adjacent segments. However, its long-term clinical efficacy should be verified with long time.

8.Correlation of Laryngopharyngeal Reflux and Voice Disorders

Objective To study the correlation between laryngopharyngeal reflux (LPR) and voice disorders . Methods One hundred and three patients with reflux -related symptoms were recruited .The patients were evalu-ated with reflux symptom index (RSI) ,reflux finding score (RFS) evaluation and 24-hour dural-probe pH moni-toring .Eighty -nine cases with voice disorders were divided into 5 groups :vocal process granuloma (n=18) chron-ic pharyngolaryngitis (n=19) ,vocal polyps (n=15) ,vocal fold leukoplakia (n=21) and Reinke's edema (n=16) . The other 14 patients without voice disorders were the control subjects .Results According 24-hour dural -probe pH monitoring assessments ,48 .3% of the patients with voice disorders showed LPR positive .The positive rate in the vocal fold leukoplakia group (71 .4% ) and Reinke's edema group (75 .0% ) were significantly higher than the control group (35 .7% ) .RSI and RFS of the patients with Reinke's edema were both significantly higher than the control group(P=0 .020 ,P=0 .009) .RSI of the patients with chronic pharyngolaryngitis were significantly higher than the control group (P=0 .019) .The acid reflux episodes ,acid reflux time which except in the supine position of the vocal fold leukoplakia group were significantly higher than the control group .The acid reflux episodes ,acid ex-posure and acid reflux time which all except in the supine position of the Reinke's edema group were significantly higher than the control group .Conclusion The correlations between Reinke's edema ,vocal fold leukoplakia and LPR were stronger .In these two groups ,the acid reflux episodes were higher and acid reflux times were longer .
9.IMMUNOCYTOCHEMICAE LOCALIZATION OF EPIDIDYMAL SPECIFIC PROTEINS IN THE EPIDIDYMIS OF THE RABBIT

The secretion and localization of epididymal specific proteins ESP-1, ESP-2 were systematically studied by the PAGE-western blot and ABC immunocytochemical technique in the epididymis of the rabbit. The results indicated that ESP-1 and ESP-2 represented the rabbit epididymal specific proteins of 42 kd and 20 kd, respectively. They were immunoeytochemically negative in the testis, heart, liver, spleen, lung, kidney and skeletal muscle of the rabbit as well as testis and epididymis of the rat. However, in the rabbit epididymis, except the initial segment ESP-1 and ESP-2 were started to be positive from caput segment. The ESP-1 was displyed in the supranuclear area of the principle cells, while the ESP-2 was distributed evenly in the whole cytoplasm of it. Subsequently, the most heavy positive reaction was demonstrated in the corpus epididymis and the proximal segment of cauda. In contrast of the caput, the stereocilia of principle cells and lumen sperms of the cauda epididymis were positive also. However, in the distal segment of cauda the positive reaction in cytoplasm of principle cells was gradually decreased and became disappeared completely soon, while the stereocilia of it still remained heavy positive even in that of vas deferens. The epithelium of prostate gland and seminal vesicle were negative. These results indicated that both ESP-1 and ESP-2 were secretory proteins produced mainly in the principle cells of the corpus epididymis. After releasing into the lumen, they took part of epididymal fluid and might bind on the sperm to regulate sperm maturation. The possible physiological significance of ESP-1 and ESP-2 was discussed.
