Objective To observe the effect and safety of hemabat(H)on prevention of postpartum hemorrhage in cesarean section and after cesarean section in high risk pregnant women.Methods Four hundred and sixty-nine pregnant women with high hemorrhagic risk factors including twin pregnancy, polyhydramnios,fetal macrosomia,placenta previa were planned cesarean section.A total of 457 pregnant women were divided into 3 groups by operation indications.There were 239 cases of fetal macrosomia,145 cases of twin pregnancy and polyhydramnios,and 73 cases of placenta previa.Three kinds of hysterotonics were used randomly in each group.Group oxytocin(O):20 U oxytocin injected into the uterine plus 20 U oxytocin intravascularly,152 women;Group oxytocin+hemabate(O+H):20 U oxytocin and 250?g hemabat injected into the uterine,192 women;group H:250 p,g hemabat,injected into the uterine,125 women.The amount of bleeding during the operation and within 2-hour after delivery were measured.The side effect of each group was observed.Results The amount of bleeding during cesarean section in group O was(445?262)ml,in group O+H(33?218)ml,and in group H(375?265)ml.There was an extremely significant difference between group O and group O+H(P

The clinical data of 8 patients with the fourth-degree laceration of perineum, who were admitted in Beijing Obstetrics and Gynecology Hospital, Capital Medical University from January 2004 to December 2010, were reviewed retrospectively. The causes and clinical strategies were analyzed. The fourth-degree laceration of perineum was mainly caused by improper skills of midwifery, too faster labor,insufficient estimation of fetal-weight and perineal condition and obstetric forceps in this group of patients.Primary suture was performed and all cases achieved complete healing. The improvement of midwifery skill,carefully monitoring the uterine contraction, accurate evaluation of condition of perineum and the fetal-weight and good communication between patients and midwives are the key points to prevent the fourth-degree laceration of perineum.

Porphyromonas gingivalis is a specific causative agent of human chronic periodontitis. This anaerobe produce collagenase PrtC encoded by prtC. To constructed the prokaryotic expression system for the prtC gene from P.gingivalis so as to be used to explore antigenicity and immunoreactivity of prtC gene product as well as the relationship between the local level of PrtC and the periodontitis damage, the entire length of prtC genes fragment from the ATCC-33277 and 47A-1 strains of P.gingivalis was amplified by PCR. After T-A cloning and sequencing, the prokaryotic expression system for prtC was constructed by using pET32a plasmid and E.coli BL21DE3 strain. The expression of the target recombinant PrtC protein was induce with different concentrations of IPTG. Western blot assay was used to detect the antigenicity and immunoreactivity of PrtC protein, and ELISA assay was used to detect the PrtC level in the subgingival plaque samples of patients with chronic periodontitis. The experimental results showed that the nucleotide sequences of the prtC genes from ATCC-33277 and 47A-1 strains of P.gingivalis were entirely identical, and the sequence similarities of nucleotides and amino acids were 98.46% and 99.07% respectively. Under the induction of different concentration of IPTG, the output of recombinant expressed product PrtC may reach up to 50% of the total bacterial proteins. It was also proved that the recombinant PrtC could bind with antibody against the whole cell of P.gingivalis, and could induce the production of specific antibodies in rabbit. In 91.39% of the subgingival plaque samples, the PrtC could be detectable, in which the level of positive rates of detection was higher in severe cases of chronic periodontitis than that in the mild cases. So far, a prokaryotic expression system for the prtC gene from P.gingivalis with high expression efficiency was successfully constructed in the present study, and the expressed product PrtC possesses well antigenicity and immunoreactivity, suggesting the possibility to be used as the candidate antigen for developing the serological kit and P.gingivalis vaccine.

Two lipase active fractions Lip1 and Lip2 were purified from the cell extract of Rhizopus chinensis CCTCCM201021.Both gave a single band on SDS-PAGE after using ammonium sulfate precipitation、Phenyl-Sepharose FF、DEAE-Sepharose FF and Sephadex G100 gel filtration chromatographies.The molecular masses of two lipases were 59.2kD and 39.4kD respectively.Lip1 and Lip2 showed optimal pH at 8.0 and 8.5 and their optimal temperatures were 40℃ and 35℃ respectively.The substrate specificity of the two lipases was obviously different.Lip1 was more specific to long chain fatty acid of p-nitrophenyl esters while Lip2 had a preference for the hydrolysis of short chain fatty acid of p-nitrophenyl esters.Lip1 had 1,3-position specificity for triacylglycerols hydrolysis while Lip2 had nonspecific position.Both lipases were stimulated by Ca 2+、Mg 2+ while SDS had strong inhibition on their activities.Lip1 and Lip2 had good stability in cyclohexane、hexane、heptane and isooctane(30% V/V).

Objective To analyze the growth and development condition of the children with cerebral palsy,and to investigate the influence of puberty on their adult height.Methods 56 children with cerebral palsy were selected as research subjects.34 healthy children were selected as control group.Their height and weight were measured,the development condition of their sexual character was checked,and then standard deviation of height and weight,height age and bone age were calculated,adult height and target height were predicted and analyzed.Results The standard deviation of height and weight in cerebral palsy group was (-1.29 ± 1.39) and (-0.77 ±1.20) respectively,which was lower than that of the normal control group[(0.40 ±0.95),(0.38 ± 1.01)] (t =-6.270,-4.6 7 6,all P ＜ 0.0 5).In preadolescent cerebral palsy group,the chronobiological age was bigger than their height age and bone age,the difference was significant (t =6.381,7.939,all P ＜ 0.05),but there was no significant difference between height age and bone age (P ＞ 0.05),there existed no significant difference between predicted adult height and target height(P ＞ 0.05).In adolescem cerebral palsy group,the chronobiological age and bone age were larger than their height age,which indicated significant difference (t =3.438,-3.759,all P ＜ 0.05),but there was no significant difference between the chronobiological age and bone age (P ＞ 0.05),the predicted adult height was lower than target height,the difference was significant (t =-5.204,P ＜ 0.05).Conclusion The growth and development of children with cerebral palsy would usually fall behind the normal children,but showed similarity in terms of starting age and process of puberty.After puberty,their bone age would increase dramatically,their predicted adult height would fall behind target height distinctly.
