The factor of antiapoptosis in cancer is recently identified as survivin. Survivin is prominently overexpressed in embryonic and fetal tissues, and various human cancers. It is potentially involved in both inhibition of apoptosis and control of cell division. Recently, survivin is found to be correlated with the genesis, progress, and prognosis of urological cancer. Manipulation of the antiapoptotic pathway maintained by survivin may be beneficial for cancer therapy. Progressive research of survivin in urological cancer.
2.Diagnosis and surgical treatment of adult congenital choledochal cyst

Objective To evaluate the diagnosis and surgical treatment of adult congenital choledochal cyst(ACCC). Methods Clinical data of 345 ACCC cases from 1974 to 2007 were analyzed retrospectively. Results There were only 110 patients (31.9%) with typical triad (abdominal pain, palpable mass and jaundice). B-ultrasound, CT, ERCP, PTC and MRCP were methods for diagnosis. Cancer arising from choledochal cysts was found in 19 cases who had undergone internal drainage and its incidence of cholangiocarcinoma is much higher than in the general population. All cases underwent surgical treatment, in which total excision of choledochal cyst and Roux-en-Y hepaticojejunostomy was main operation, 337 cases recovered well and 8 (2. 3%) died. C..ondusions Accurate diagnosis of ACCC was difficult depending on clinical symptoms only. B-ultrasound was the choice of atraumatic diagnosis. All patients with an established diagnosis of ACCC should undergo exploratory laparotomy. Total excision of eholedochal cyst and Roux-en-Y hepaticojejunostomy is most effective treatment for ACCC.
3.Effects of anti-viral therapy in combination with transcatheter arteria chemoembolization on recurrence of HBVrelated hepatocellular carcinoma after operation

Objective To investigate the preventive effects of anti-viral therapy in combination with transcatheter arteria chemoembolization (TACE) on recurrence of HBV-related hepatocellular carcinoma (HCC) after operation. Methods According to selection standards, 49 patients treated in this hospital from January 2003 to June 2009 were enrolled in this study and divided into a control group and combined group.The cumulative tumor-free survival rate, cumulative survival rate, the 1-, 2- and 3-year recurrence rates and cumulative recurrence rates were determined in all the patients and compared between the 2 groups. Results The cumulative tumor-free survival rate and cumulative survival rate were significantly higher in the combined group than in the control group (P=0. 019, 0. 008). The 1-, 2-, and 3-recurrence rates were 24% vs. 38.9%, 28% vs. 33.3%, and 28% vs. 22.2% in the combined group and control group, respectively. The 1-, 2-, and 3-year cumulative recurrence rates were 24% vs. 38. 9%, 52% vs. 72. 2%, and 80% vs.94. 4% in the combined group and control group, respectively. Conclusion Anti-viral therapy in combination with TACE can exert significant preventive effects on recurrence of HBV-related HCC after operation.
4.Influence of sirtinol in cell cycle of prostate cancer DU145 cells and its mechanism

Objective To observe the influence of sirtinol,a silent information regulator 1(SIRT1)inhibitor,in the cell proliferation, cell cycle progression and the expression levels of positive regulator proteins of the cell cycle including Cyclin D1,CDK4 and pRb in prostate cancer DU145 cells,and to explore the possible mechanism of SIRT1 in occurrence of prostagte cancer.Methods The DU145 cells at logarithmic growth phase were cultured in vitro and divided into control group(DMSO)and different doses (10,25,50μmol·L-1 )of sirtinol groups.The inhibitory rate of growth of DU145 cells was detected with MTT method,the SIRT1 mRNA and protein expression levels were determined by RT-PCR and Western blotting method, and the cell cycle was measured by flow cytometry.The Cyclin D1,CDK4 and pRb protein expression levels were examined by Western blotting method. Results Compared with control group, the inhibitory rates of growth of the DU145 cells in different doses of sirtinol groups were increased markedly in a dose-dependent manner(P<0.01),and the flow cytometry analysis result showed the DU145 cells at G1 phase were increased (P<0.01 ). Compared with control group, the expression levels of SIRT1 mRNA and protein in DU145 cells in different doses of sirtinol groups were decreased significantly(P<0.01);the expression levels of Cyclin D1 and pRb proteins were decreased(P<0.01),whereas the expression levels of CDK4 had no change(P>0.05).Conclusion SIRT1 inhibition by sirtinol can inhibit the cell growth of prostate cancer DU145 cells in a dose-dependent manner and arrest the cell cycle progression,and its mechanism may be related to decreasing the CyclinD1 and pRb protein expressions.
5.Damage of endothelial cells induced by OX-LDL and its influence in expressions of TLR-4 and EpCAM

Objective To study the damage effect of oxidized low density lipoprotein (OX-LDL)on the endothelial cells and its influence in the expressions of Toll like receptor 4 (TLR-4)and epithelial cell adhesion molecule (EpCAM).Methods The human endothelial cells ECV-304 were cultured in vitro and treated by different concentrations of OX-LDL for 24 h,and the cell vitality was measured by Taipan blue rejection test.The reactive oxygen species (ROS)level,mitochondrial membrane potential (MMP),cell cycle and apoptosis were detected by flow cytometry (FCM).The expression of TLR-4 and EpCAM were also analyzed by FCM.Results Compared with control group,the ROS levels in ECV-304 cells in 25,50,100,and 200 mg·L-1 OX-LDL were increased, but the cell vitalities and MMP were decreased (P < 0.05).Compared with control group,the percentages of S phase cells in 25 and 50 mg·L-1 OX-LDL groups were increased,the G0 G1 phase cells were slightly reduced,and the SubG1 phase cells (apoptotic cells)were increased with the increasing of OX-LDL concentration.Compared with control group,the TLR-4 and EpCAM expression levels in ECV-304 cells in 50 mg·L-1 OX-LDL group at 24 h were significantly increased (P < 0.05 ). Conclusion OX-LDL could increase the ROS level and induce the apoptosis in vascular endothelial cells,while increase the cell vitality and MMP.During the damage process,the expresion levels of TLR-4 and EpCAM are up-regulated.
6.Preparation of Irisquinone freeze-drying Injection and its stability

AIM: To prepare the irisquinone freeze-drying injection and investigate its stability. METHODS: The degradation pattern of irisquinone and its effects were measured. At the same time, the stability of freeze-(drying) injection by classic thermostatical test. RESULTS: Irisquinone solution was unstable to heat and its content decreased remarkably in acid and alkaline solutions. The result showed that oil/water partition coeffecient of irisquinone was very large. The irisquinone-hydropropyl1-?-cyclodextrin inclusion complex freeze-drying injection was very effective to enhance the drug stability. CONCLUSION: Irisquinone inclusion complex injection has character of good stability.
7.Solubilization of hydropropyl-?-cyclodextrin to paclitaxel and its relative molecular inclusion mechanism

Objective To investigate the solubilitization effect of macromolecular paclitaxel with cyclodextrin and mechanism between inclusion complex and drugs. Methods The influence effects on paclitaxel solubility were studied with ?-cyclodextrin (?-CD) and hydroxypropyl-?-cyclodextrin (HP-?-CD) as inclusion carriers. The chemical shifts of hydrogen atom in host and guest molecules were also investigated with 1H-NMR. Results The form of inclusion complex was responsible by host molecules and the enhancement solubility of paclitaxel in HP-?-CD was larger than that in ?-CD and increased with the ratio increasing. The benzene ring in the molecular of HP-?-CD and paclitaxel showed the most significant effect. Conclusion The solubility of diossolved paclitaxel is improved by hydroxypropyl-?-cyclodextrin.
8.Identification of irisquinone hydroxypropyl-?-cyclodextrin inclusion compoundand its thermodynamic stability

Objective To parperare and identify the irisquinone-hydroxypropyl-?-cyclodextrin (irisquinone-HP-?-CD) inclusion compound. The inclusion mechanism and mol ratio of irisquinone and HP-?-CD were studied simultaneously. Methods The irisquinone-HP-?-CD was prepared with lyophilization technique. The mol ratio between host and guest moleculars was also researched by molecular gradient and continuing variational methods in inclusion processing. At the same time, the inclusion compound was identified by X-ray diffraction (XRD) and differential scanning calorimetry (DSC) methods, respectively. Results The above-mentioned systems showed the mol ratio of HP-?-CD-irisquinone (2 : 1) and the inclusion compound enhanced remarkably the most solubilization and more combined constant of irisquinone in 25, 35, and 45℃. The lyophilized powder had been formed inclusion compound by identifying. Conclusion It is advantageous to increase the solubilization and to strengthen the stability of irisquinone by preparing inclusion compound.
9.Pharmacokinetics of Paclitaxel Lyophilized Injection in rat

AIM: To study the pharmacokinetics of Paclitaxel Lyophilized Injection and Taxol in rats. METHODS: Paclitaxel inclusion lyophilized Injection was prepared with hydroxylpropyl ? inclusion as the inclusion vehicle. At the same time, Paclitaxel Lyophilized Injection pharmacokinetic pattern and parametes had been modeled with 3p87 program and statistic rules comparing with Taxol. RESULTS: Paclitaxel Lyophilized Inject and Taxol abided by two department model. The parametes T 1/2? (3.40?0.27)h and AUC (34.46?4.34) mg?L -1 ?h,MRT 3.50 h to Lyophilized Injection and T 1/2? (3.50?0.62)h, AUC (25.73?6.42) mg?L -1 ?h,MRT 2.52 h to Taxol. CONCLUSION: The results indicate that distribution rate of the two preparations are in coincidence with in rats and Paclitaxle Lyophilized extends the retention time of paclitaxel in rats.
10.Treatment of 7 patients with medial meniscal cyst under arthroscopy.

OBJECTIVETo investigate clinical effects of arthroscopy in the treatment of medial meniscal cyst.
METHODSFrom June 2011 to January 2013, 7 patients with medial meniscal cyst were treated with arthroscopy. There were 3 males and 4 females,ranging in age from 27 to 63 years old,with a mean age of (43.93±2.10) years old. The cysts have been discovered for 3 to 30 months,with a mean time of (10.6±1.3) months. All the patients complained of knee pain,especially in the medial joint gap. The Pisani sign, Caklin sign and medial McMurray sign were all positive. Preoperative MRI examination confirmed the diagnosis. Lysholm score changes and clinical efficacy were observed through a six-month follow-up.
RESULTSThe postoperative Lysholm scores were all significantly higher than the preoperative scores. According to Sarimo standard, 6 patients got an excellent result, and 1 good.
CONCLUSIONArthroscopic treatment of medial meniscal cyst has replaced the traditional method, which could retain the normal meniscus as much as possible and repair the meniscus injury simultaneously, as well as get a good curative effect and a good recovery of knee function. This method is worthy of clinical application.
