1.Survey on Nurses′Job Satisfaction in Public Hospitals and Analysis of the Influential Factors-Taking the Affiliated Hospital of Weifang Medical University as an Example

Objective:This study is to investigate the job satisfaction of nursing staff in public hospital and ana-lyze the influential factors, thus to provide evidence for improving the performance of human resource management in the hospitals. Methods:A sample of 100 nurses in the affiliated hospital of Weifang Medical University was se-lected to participate in this study with cluster sampling method. Job satisfaction was investigated using self-de-signed questionnaire, and data analysis was conducted with SPSS software. Results:The total score of job satisfac-tion is(92. 52 ± 18. 17). The highest is about their working group with 3. 89 ± 0. 58, while the lowest is about their labor return with(2. 19 ± 0. 89). Authorized strength, education, working years and monthly salary all impact the job satisfaction of nurses. Conclusions:At present, nurses′job satisfaction is at a media level. The managers of public hospitals should build a scientific compensation system and humanized duty system and so on, which can meet the different needs of nursing staff and improve their job satisfaction.
2.Comparison of Brucellacapt and iELISA in serological diagnosis of brucellosis

Brucellacapt is a new serological test based on immunocapture-agglutination technique that has been used in many countries ,but there was no related article reported in China .The value of the Brucellacapt method in diagnosing human brucellosis was evaluated in this study .Among 120 suspected patients ,75 patients and 45 negatives were diagnosed by SAT and RBPT method combination with their clinical symptoms .Sera from all 120 people were tested by the method of Brucella-capt and iELISA ,and the results were ,consequently ,analyzed and compared .It showed that sensitivity ,specificity ,consis-tency rate ,Kappa value ,and area under ROC curve were found to be 82 .7% ,88 .9% ,85 .0% ,0 .69 ,and 0 .86 ,respectively for Brucellacapt ,whereas they were found to be 90 .7% ,64 .4% ,80 .8% ,0 .57 ,and 0 .78 for iELISA .In conclusion ,specific-ity ,consistency rate ,Kappa value ,and area under ROC curve were higher in Brucellacapt method than that in iELISA .How-ever ,the sensitivity of iELISA is higher .

Vasohibin-1 (VASH1),which is induced in response to angiogenic stimuli such as vascular endothelial growth factor (VEGF) and fibroblast growth factor (FGF)-2,has recently been isolated as a novel negative feedback inhibitor of angiogenesis.Several studies have demonstrated that VASH1 plays important roles in the development of various tumors and it would potentially be a biomarker and a candidate for molecular targeted therapy for patients with cancer in the future.
4.Analysis of the influence factors of central venlus catheters-related infections in postoperative patients with gastric carcinoma treated with parenteral nutrition

Objective To investigate the influence factors of central venous catheter related infections (CVC-RI) in postoperative patients with gastric carcinoma treated with parenteral nutrition,in order to provide scientific basis for prevention and treatment.Methods Using the retrospective study method,150 patients who treated with parenteral nutrition after operation of gastric carcinoma from December 2013 to March 2014 were selected.Information including age,sex,anamnesis,catheter indwelling time,infusion rate of nutrient solution,and so on were collected.Patients with central venous catheter related infections were set as infection group,patients without infection as the control group,and the group logistic regression analysis was used to analyze the collected data.Results 21 patients in infection group had CVC-RI,and the incidence was 14.00% (21/150),while 129 patients in control group had CVC-RI.Univariate analysis showed there was significant difference between infection group and control group in age,anamnesis,catheter indwelling time and infusion rate of nutrient solution (x2=17.126,5.447,12.795,7.973,P<0.05),however there was no significant difference in sex,health insurance,the success rate of catheterization and whether glutamine was in nutrient solution (P>0.05);the group logistic regression analysis showed that age (OR=15.927,95%CI 3.305-76.756,P<0.05)and catheter indwelling time (OR=3.511,95%CI 1.028-11.992,P<0.05) were independent risk factors for CVC-RI,however infusion rate of nutrient solution (OR=0.140,95%CI 0.033-0.597,P<0.05) was a protective factor.Conclusions For the postoperative patients with gastric carcinoma treated with parenteral nutrition,CVC-RI is closely related to age,catheter indwelling time and infusion rate of nutrient solution.So reducing catheter indwelling time,restoring the enteral nutrition under condition permission as so on as possible can reduce infection of elderly patients.
5.HMGB1 activates NF-κB/αvβ3 to promote A549 cells migration and invasion

Background and purpose:High mobility group 1 (HMGB1), frequently found to be over-expressed in many human tumors, plays an important role in tumor progress and metastasis. This study aimed to investigate the mechanism of HMGB1 promoting A549 cell metastasis.Methods:A549 cells were untreated or treated with HMGB1 (200 ng/mL) in absence or presence of NF-κB inhibitors 6-amino-4-quinazoline (QNZ, 40 nmol/L) or Bortezomib (Bort, 20 nmol/L). Scratch assay and Transwell assay were performed to evaluate A549 cells migration and invasion ability. The activity of NF-κB was examined by luciferase reporter assay. NF-κBp65 and αvβ3 expressions were detected by Real-time RT-PCR or Western blot.Results:HMGB1 increased A549 cells migration and invasion ability. HMGB1 enhanced NF-κB protein level and NF-κB activity in A549 cells. Real-time RT-PCR and Western blot showed that HMGB1 up-regulated αvβ3 expression in A549 cells. NF-κB inhibitors QNZ or Bort reserved the promot-ing effects of HMGB1 on A549 cells migration and invasion, NF-κB expression and activity as well as αvβ3 expression. Conclusion:HMGB1 promotes A549 cell migration and invasion through activating NF-κB and up-regulating αvβ3.
6.A qualitative research on the survival condition of hemodialysis patients in Ningxia

Objective To explore the survival condition of hemodialysis patients in Ningxia Hui Autonomous Region. Methods Interviewed 15 patients with hemodialysis by applying Modified Grounded Theory Approach put forward by KinoSitaYasuHito,a Japanese scholar,the data of which were from qualitative inductive comparative analysis. Results As for the survival condition of hemodialysis patients in Ningxia,two relative themes were extracted, Helpless dialysis life Desire to return to society, the former contained 2 subtopics of hardship in life and susceptible mood, the latter contained 2 subtopics of cherish life and desire for care. Conclusion We should set up a social supporting system to improve the living condition of hemodialysis patients with the purpose of helping them return to the society.
7.Influence of high-fluoride on thyroid function and brain damage in rats

Objective To study the influence of high-fluoride on thyroid function and brain damage. Methods Thirty-six Wistar rats were randondy divided, according to weight and gender into 3 groups(12 rats each), i.e. control group, high fluoride group, and high fluoride plus thyroid tablet treatment group. The rats were fed with normal tap water containing no more than 5 mg/L NaF and the tap water added 100,100 mg/L NaF, respectively. After 7 months of experiment, the rats in high fluoride plus thyroid tablet treatment group were given with 0.04% thyroid tablet( 1.8 ml·kg~(-1)·d~(-1)) by gastric perfusion for three weeks. The contents of TT_3 and TT_4 in serum were detected by radio-immunological assay; the histomorphology in thyroids and brains were observed under microscopy; and the protein level of NMDAR2B subunit of glutamate receptor in the hippocampal CA1 and CA3 was measured by immunohistochemistry. Results As compared to the values of TT_3 and TT_4 in serum of rats in control group[ (0.97 ± 0.15), (84.03 ± 12.45)nmol/L], TT_3 and TT_4 in high fluoride group were obviously lower [(0.24 ± 0.07), (15.16 ± 2.08)nmol/L, all P < 0.01]; while no changes in TT_3 and TT_4 were detected in high fluoride plus thyroid tablet treatment group[ (1.02 ± 0.19), (85.63 ± 9.55)nmol/L, all P < 0.05] as compared to controls, but higher than those in high fluoride group(all P < 0.01 ). The pathological changes including partial hyperplasy, arrangement disorder, atrophy, and decreased colloid of the thyroid follicular epithelial cells in high fluoride group were observed under microscopy. In high fluoride plus thyroid tablet treatment group, the degree of the thyroid cellular hyperplasy was relatively slight as compared to high fluoride group. The swelling and disarrangement of neurons in the hippocampus were observed in high fluoride group, whereas the changes of the neurons were not so obvious in high fluoride plus thyroid tablet treatment group. The grey values of NMDAR2B positive cells in the hippocampal CA1 and CA3 in high fluoride group(167.05 ± 7.31 ) were significantly increased as compared to controls (92.53 ± 9.67 ) or high fluoride plus thyroid tablet treatment group( 101.66 ± 12.21, all P < 0.01 ). Conclusions High fluoride can induce the decreased function and changed histomorphology in thyroid and result in pathological damages in the brains of rats. However, treated with thyroid tablet to those having damages induced by high fluoride, the thyroid function and morphology can be normal, and the brain damages can be alleviated. The results indicate that hypothyroidism caused by high fluoride might be an important participating factor in brain damages caused by fluorosis.
8.Synthesis and identification of artificial antigens of paneoniflorin.

Oxidation method with sodium iodide was used to synthesize immunogenic antigen (PF-BSA) and coating antigen (PF-OVA) of paeoniflorin. UV spectroscopy showed that paeoniflorin was successfully conjugated with BSA and OVA. After immunized by PF-BSA, the mice can produce anti-paeoniflorin antibodies specifically. The ELISA test results showed the high titer (1:12 800) and specificity (IC50 = 0.791 mg x L(-1)) of the antiserum from mice injected with PF-BSA. Also, the antiserum showed low cross activities against nine traditional Chinese medicine (TCM) of small molecules. These artificial antigens were successfully synthesized and the anti-paeoniflorin antibody well prepared, which provides the experimental basis for the further study of ELISA and its kit.

9.Decreased hippocampal neurogenesis is involved in cognitive dysfunction induced by thiamine deficiency

Objective To investigate the influence of thiamine deficiency(TD)at early pre- pathological lesion stage on cognitive function and the correlation between cognitive dysfunction and hippocampal neurogenesis.Methods TD mouse model was prepared by feeding a thiamine-depleted diet. Learning and memory functions of TD mice were tested with Y-maze.Hippoeampal neurogenesis was studied with bromodeoxyuridine(BrdU),proliferative cell nuclear antigen(PCNA),and Doublecortin(Dcx) immunohistochemical staining on the 7th(TD7),9th,14th and TD25th day.Results TD9 mice without pathological impairment and cholinergic nerve degeneration needed more times of training(22.3?2.2)in the learning test of Y maze compared with the controls(13.5?3.5).Correspondingly,the numbers of BrdU-positive ceils and the immunoreactivity of Dcx decreased significantly in the TD9 mice(19.8?0.4, 1537.2?50.2 vs 23.9?0.3,2688.9?127.9 pixels/mm~2).Thiamine re-administration reversed the declined hippocampal neurogenesis:the number of BrdU-positive cells was 23.6?1.9 and Dcx immunoreactivity was 2052.3?269.6 pixels/mm~2:the impaired learning ability was simultaneously restored,with the number of total training trial being 16.8?0.5.Conclusion The decreased hippocampal neurogenesis contributes to retarded learning ability at early pre-pathological lesion stage of TD.
10.Laser scanning confocal microscopic imaging for Ca2 + oscillations of pancreatic acinar cells in mice.

OBJECTIVETo establish a simple but effective method of laser scanning confocal microscopic imaging for Ca2+ oscillations of pancreatic acinar cells in adult mice.
METHODSPancreatic acinar cells from adult Kunming mice were isolated acutely with collagenase, and then loaded with fluo-4-AM, a Ca2+ indicator. A laser scanning confocal microscope armed with 488 nm laser was employed to record the dynamic fluorescent signals in-time and synchronously while acetylcholine (ACh) was added in the pancreatic acinar cells.
RESULTS(1) The classic pancreatic acinar cell Ca2+ oscillations were induced by a certain concentration of ACh (100 nmol/L) successfully and steadily, which could be blocked by atropine completely. (2) Plasmic Ca2+ oscillations from different parts of one acinar cell were usually with different amplitudes and almost the same frequencies. But both of amplitudes and frequencies were different among different cells. (3) The acinar cell Ca2+ oscillations were induced by ACh in a concentration-dependent manner.
CONCLUSIONThe laser scanning confocal microscopic imaging for adult mouse pancreatic acinar cell Ca2+ oscillations was established successfully. The features of being easy to use, direct to see lively, high efficiency and good flexibility make it a popular tool for researchers to choose.
