Objective To investigate the protective effect of carnosine on Glutamate induced apoptosis in SH-SY5Y cells and its mechanism. Methods The injury model was established by treating SH-SY5Y cells with glutamate in vitro.Inverted microscope was used to observe cell morphology. The cell viability was detected by MTT assay, and changes in nucleus were detected by Hoechst33258 staining under fluorescence microscopy. The cell apoptosis rates were measured by flow cytometry. The reactive oxygen species ( ROS ) level in cells was detected by fluorescent probe CDFH-DA. Results Compared with normal control group, the cell viability in glutamate group decreased (P<0.01), the morphology changes of cell apoptosis such as karyopyknosis and split were observed by Hoechst33258 staining, while the apoptosis rate and the level of ROS were increased (P<0.01). Compared with glutamate group, the cell viability of carnosine 0.6,3,15 mmol/L groups obviously increased(P<0.01), while the morphology changes of cell apoptosis significantly improved, the apoptosis rate and the level of ROS were obviously reduced ( P<0.01 ) .Compared with normal control group, the cell viability, the morphology changes and the apoptosis rate did not significantly change in carnosine group. The level of ROS was reduced, but there was no statistically significant difference between two groups. Conclusion Carnosine has apparent protective effect on apoptosis of SH-SY5Y cells injury induced by glutamate. The mechanism may be related to carnosine prevent the occurrence of oxidative stress.

Traditional processes are mostly adopted in traditional Chinese medicine (TCM) preparation production and the quality of products is mostly controlled by terminal. Potential problems of the production in the process are unpredictable and is relied on experience in most cases. Therefore, it is hard to find the key points affecting the preparation process and quality control. A pattern of research and development of traditional Chinese medicine preparation process based on the idea of Quality by Design (QbD) was proposed after introducing the latest research achievement. Basic theories of micromeritics and rheology were used to characterize the physical property of TCM raw material. TCM preparation process was designed in a more scientific and rational way by studying the correlation among enhancing physical property of raw material, preparation process and product quality of preparation. So factors affecting the quality of TCM production would be found out and problems that might occur in the pilot process could be predicted. It would be a foundation for the R&D and production of TCM preparation as well as support for the "process control" of TCMIs gradually realized in the future.

Objective To investigate the effect of transduction of heme oxygenase-1 (HO-1) protein on oxygen-glucose deprivation and restoration (OGD/R)-induced injury to hippocampal neurons in rats. Methods Plasmid 11R-HO-1 was constructed using plasmid pET-21a(+)-p53-11R (plasmid 11R) and 11R-HO-1 fusion protein was identified and collected.Hippocampal neurons obtained from newborn Wistar rats (< 48 h) were cultured for 7 days in vitro and then the neurons were randomly divided into 5 groups (n =171 each) using a random number table:OGD/R group, normal saline group (group NS),plasmid 11R group (11R group),300 nmol/L 11R-HO-1 group (H1 group),and 1 500 nmol/L 11R-HO-1 group (H2 group). In NS,11R,H1 and H2 groups, the neurons were incubated for 2 h with 300 nmol/L normal saline,300 nmol/L plasmid 11R,300 nmol/L 11R-HO-1 fusion protein, and 1 500 nmol/L 11R-HO-1 fusion protein, respectively, and then OGD/R was performed. The neurons were incubated in deoxygenated glucose-free DMEM medium and sealed under 5 % CO2-95 % N2 in an anaerobic chamber equilibrated to 37 ℃ for 45 min.OGD was terminated by replacement of the medium with high glucose DMEM medium and by returning the cultures to a standard incubator maintained at 37 ℃ in 5 % CO2-95 % air and the neurons were then incubated for 24 h.Immediately after OGD/R was established, the cell survival rate (by MTT assay),apoptosis rate (using TUNEL),and expression of HO-1 and caspase-3 protein (by using Western blot) were measured. Results Compared with group OGD/R,the cell survival rate was significantly increased, the apoptosis rate was decreased, the caspase-3 expression was down-regulated,HO-1 protein expression was up-regulated in H1 and H2 groups (P < 0.05),and no significant change was found in the parameters mentioned above in NS and 11R groups (P > 0.05).Compared with group H1,the cell survival rate was significantly increased, the apoptosis rate was decreased, the caspase-3 expression was down-regulated,and HO-1 protein expression was up-regulated in group H2 (P < 0.05).Conclusion Transduction of HO-1 protein can reduce OGD/R-induced injury to hippocampal neurons of rats.

Objective To investigate the influence of hyperbaric oxygen treatment (HBOT) on blood pressure (BP) and the incidence of rebleeding in patients with hypertensive intracranial hemorrhage (HICH)Methods One hundred and twenty patients with HICH were treated for 60 min with 100% oxygen at 2.0 absolute atmospheres (ATA) daily when the condition was stable and BP was controlled ideally. Blood pressure was measured before the patients were sent into the HBO chamber and remeasured following completion of each HBOT session and 1 hour later. Rebleeding was monitored during and after each HBO session . Results HBOT caused a significant elevation of systolic BP in 25.83% (31/120) patients and a significant decrease in 16.67% (20/120) patients (P <0. 05 ),whereas the rest 57.5% (69/120) patients had no significant changes, when the BP was measured right after the HBOT session. The mean diastolic BP increased in 69 (57. 50% ) patients and decreased in 4. 17%(5/120) patients (P < 0. 05 ), whereas we found no significant changes in the rest 46 (38. 33% ) patients. No differences were found in the comparison of BP before and 1 hour after the HBOT session and no one suffered from rebleeding during and after HBOT session. Conclusions HBOT may cause temporal blood pressure changes in most patients with HICH, however, it will not cause an increasing incidence of rebleeding if the patient's condition is stable and the blood pressure has been well controlled.
