1.Analysis of Drug Use Normalization in Inpatients with Cancer Pain Based on the Latest Cancer Pain Treat-ment Guidelines

Objective:To investigate the therapeutic medication for inpatients with cancer pain, and compare with the latest cancer treatment guidelines to improve the rational drug use. Methods:By a retrospective study, the medical record analysis for the inpatients with cancer from October to December in 2014 was performed, and the incidence of pain and the therapeutic medication were investiga-ted and the rationality of drug use was evaluated based on the latest cancer pain treatment guidelines. Results:The incidence of cancer pain was 25. 46% of 774 cases with new cancer, and 76. 58% of the patients with cancer pain were treated with analgesic drugs. In ac-cordance with the guidelines for the treatment scheme, there was no significant difference between the Opioid-Naive group ( guide group 1) and the Opioids-Tolerant group (guide group 2) in the combination therapy, while there was significant difference in the drug selec-tion and the time of administration. In the guide group 1 the proportion of oxywdone controlled-release tablets was signiticantly higher than that in the guide group 2, the ratio of taking the medicine on time in the guide group 1 was also higher than that in the guide group 2. Both groups showed difference from the guidelines in the pain assessment and titration and specific analgesic drug use. Conclusion:The findings indicate the difference between the pain treatment and the guidelines in therapeutic medication of cancer pain, and the propaganda and education of relevant knowledge should be strengthened to promote the treatment standard.
2.Use of a vacuum-assisted closure device in the repair of defect after enlarged excision of dermatofibrosarcoma protuberans

Objective To develop a new method to repair the defect after enlarged excision of dermatofibrosarcoma protuberans.Methods This study included 8 patients with dermatofibrosarcoma protuberans measuring 2.5 to 5.5 cm in diameter.All the patients underwent enlarged excision of the affected skin and subcutaneous tissue.The defect measured 12.5 to 17.5 cm in diameter.Sieve skin flaps secured with a vacuum-assisted closure device were used to repair the huge surface defects.Results All the patients experienced the survival of sieve skin flaps at stage Ⅰ after operation,with no infection,effusion or necrosis.No relapse was observed during the 3 to 40 months of follow up.A satisfactory recovery was achieved in skin appearance and function with the formation of a flat scar,and no obvious proliferation occurred.Conclusion The vacuum-assisted closure device offers a safe and simple method for securing skin grafts to the defect after enlarged excision of dermatofibrosarcoma protuberans.
3.Microanatomic study on arrangement and distribution of the fascicular groups in ulnar nerve trunk to the intrinsic muscles

Objective To observe the arrangement and distribution of the fascicular groups in the ulnar nerve trunk to the intrinsic muscles so as to provide anatomical evidence for selective repair in the clinic.Methods 12 cadaveric upper extremities recently embalmed with formalin were obtained. The ulnar nerve and its branches were exposed. Microanatomy and longitudinal contradissection of each functional fascicular group were performed by tracing their terminal branches, and the arrangement and distribution were observed and recorded. The fascicular groups to the intrinsic muscles of hand in the nerve trunk were observed to see how they crossed and mixed with other fascicular groups. The diameter of the nerve trunk and the fascicles to the intrinsic muscles were measured, and the area and the distribution range of the fascicles calculated. Results The shape and the diameter of the ulnar nerve trunk varied at different positions. The fibers of functional fascicular groups had the definite differentiation at distal while interfused and diffused at proximal. The fascicular groups to the intrinsic muscles of hand were the largest and their fibers relatively concentrated (the distribution range occupied 40% ~ 80% of the ulnar nerve), and had the definite localization at the forearm and distal to the 3/8 segment of upper arm. Conclusion Since the fascicles to the intrinsic muscles of the hand are the most important in the ulnar nerve, selective repair by using epineural and perineural techniques at the forearm and upper arm will reduce the loss of nerve fibers and will contribute to the functional recovery.
4.Prediction and bioinformatic analysis of hsa-miRNA-451 target genes

Objective To predict as well as bioinforrmatically analyze the target genes of has-miR-451.Methods miRBase, miRanda, TargetScan and PicTar were used to predict the target genes of hsa-miRNA-451.The functions of the target genes were demonstrated by Gene Ontology and pathway enrichment analysis.P<0.05 was set as statistically significant.Results 18 target spots of hsa-miRNA-451 were predicted by 3 databases or prediction software at least.The functions of the target genes were enriched in proliferation and development of epithelial cells and regulation of kinase activity (P< 0.05).Pathway analysis showed that transforming growth factor-beta signaling pathway, mitogen-activated protein kinase signaling pathway, epidermal growth factor signaling pathway, Wnt signaling pathway and mammalian target of rapamycin signaling pathway were significantly enriched (P<0.05).Conclusion hsa-miRNA-451 might be involved in various signaling pathways related to proliferation and development of epithelial cells.
5.Change of Cytoskeleton and Variance of Ca~(2+) in Cultured Cells During the Invasion of Toxoplasma gondii

Objective To explore the change of cytoskeleton and the variance of Ca2+ in cultured cells during the invasion of Toxoplasma gondii. Methods Tachyzoites suspensions were gathered by routine method and used to infect phagocytic cells(J774A.1)and non-phagocytic cells (HUVEC). The ability of T. gondii invading into the cells and the influence of cytoskeleton inhibitor,colchicine and cytochalasin D,were observed by microscopy. The rearrangement of cytoskeleton of cells was observed by fluoromicroscopy. By using laser scanning confocal microscope,the variance of Ca2+ in J774A.1 and HUVEC was detected. Results Ca2+ increased greatly in J774A.1 during the invasion of T.gondii(P0.05). The microfilaments of J774A.1 were agglomerated during the invasion of T. gondii. Cytoskeleton inhibitor,cytochalasin D(P0.05)during the invasion and cytoskeleton was not changed. Cytochalasin D and colchicine showed little effect on the infection rate of HUVEC. Conclusion The concentration of Ca2+ increases greatly and cytoskeleton(mainly the microfilament)has been rearranged in phagocytic cell during the invasion of T. gondii,while both of them show no significant change in non-phagocytic cell.
6.Three-dimensional imaging reconstruction of the ulnar nerve functional fascicular groups

Objective To reconstruct on the basis of histological study the computerized 3D ima ges of the fascicular groups of ulnar nerve,which can be used to guide the microsurg ical repair of injuries of brachial plexus and ulnar nerve and to provide morpholog ical evidence for the functional rec overy of intrinsic muscles of hand after root avulsion of brachial plexus.Methods The successive AchE-staining histo logical cross-sections were made.T he photos of the sections were transformed into digital information and sto red in the computer.Different functional fas-cicular groups were distinguished a nd marked on each section.The topographic database was analyzed by image pro-cessing software,with the support o f OpenGL three dimensional graphic l ibrary.3D images of the fascicular g roups of ulnar nerve were reconstructed usin g the contour-based surface constru ction method in the program written w ith VC language.Results The 3D images of the individual external surface of the reconstructive fa scicular groups were displayed and the human-computer in teraction was realized by performin g the arbitrary rotation and the rand om cross-section.By selecting the int erested object,the spatial structu re of any fascicular group could be di splayed in the same space simultaneously.Compare d with the result of microanatomy,th e 3D shape could truly reflect the str ucture,adjacent relationship and arrangem ent of fascicular groups in the ulnar nerve trunk.Conclusions Since the re-constructive 3D images can truly reflect the arrangement and distribution of the functional fascicular grou ps,they will have positive effects on understanding the internal structure of ulnar nerve,designing operative procedur e and adjusting operative method for surgeons.[
7.Advances in the Isolation Methods of Fungal Polyketide Synthase Genes

Fungal polyketide synthases are responsible for the biosynthesis of secondary metabolites such as pigments, mycotoxins, and they are very important in pharmacology , food science and agriculture. The recent advances in the methods for the isolation and manipulation of multiple classes of polyketide synthase genes from fungi were introduced. It is useful for discovery of novel fungal polyketide synthase gene clusters. These methods can also be useful for revealing the genetic potential of fungi to produce multiple types of bioactive polyketide.
8.The advantages and research progress of T cells of the chimeric antigen receptor in the treatment of primary tumors

Chimeric antigen receptor (Car) T cells, not only have the characteristics of strong specific recognition of tu-mor antigens, but also have destruction and high affinity advantages, thus receiving more attention. Although it has played a lot of advantages in anti-tumor, it still has some shortcomings, which needs to be further optimized to improve the safety of its clinical application. In this study, The cell structure and biological function, treatment process, application development and application risk of Car T cells are reviewed, which provide references for further clinical immunotherapy of Car T.
9.Construction of GST/HIF-1α Fusion Protein Expression Vectors and the Expressions in Escherichia coil

Objective To construct GST/HIF1α fusion protein expression vector and induce its expression in Escherichia coli (E.coli). Methods The coding sequence of hypoxia inducible factor-la (HIF-la) and its deletion fragments were amplified from the plasmid pcD-NA3.1-HIF-la by PCR and inserted into pGEX-4T-2 by BamHI and Not I. The positive recombinanls were identified by restriction endonu-clease digestion and DNA sequencing. Then they were transformed into E.coli BL21, induced by IPTG and identified by SDS-PAGE and Western blot. Results The prokaryotic expression plasmid pGEX-4T-2-HIF-lα and its deletion mutants were successfully constructed and confirmed by enzyme digestion and sequencing. The desired CST/HIF-1α fusion proteins were expressed and confirmed by Western blot. Conclusion The prokaryotic expression plasmid of HIF-la and its deletion mutants were successfully constructed and the expression of fu-sion proteins was confirmed. This study provides the basis for the further research on purifying HIF-la protein and the biological function of HIF-lα.
