1.Research on the induction of T cell subset response via oral vaccination with liposome-mediated recombinant pCDNA3.1~+/Ag85A plasmid

Objective:To prepare recombinant pCDNA3.1+/Ag85A DNA vaccine encoding tuberculosis Ag85A gene mediated with LipofectamineTM 2000 and to study the effect on T cell subpopulation via oral vaccination.Methods:Recombinant plasmid containing Ag85A using liposome as a vector was constructed and administered to C57BL/6 mice via oral route.Determination of the contents of IFN-r,IL-4 in serum of C57BL/6 mice was performed by double antibody sandwich ELISA.Furthermore,the ability of splenocytes to secret IFN-? and IL-4 was tested by ELISPOT method.When the mice were vaccinated with recombinant eukaryotic expressing vector 5 weeks later,titers of serum antibody against Ag85A were detected by ELISA.The percentage of the CD4+ and CD8+ T cell subsets in the splenocytes was determined by flow cytometry.Results:Lymphocytes obtained from the spleen of liposomal pcDNA3.1+/Ag85A vaccine-immunized mice exhibited lower IFN-? production and higher IL-4 production than those of pcDNA3.1+ vector immunized mice.The number of spleen MNC secreting IFN-? stimulated by Ag85A protein in vitro was significantly lower than that of plasmid vector group.Liposomal pcDNA3.1+/Ag85A vaccine immunized mice elicited higher Ag85A-specific antibodcy titres.The percentage of the CD4+ and CD8+ T cell subsets in the splenocytes was decreased.The subset both were shown the profile of Th2 responses.Conclusion:The oral recombinant plasmid pCDNA3.1+/Ag85A mediated with LipofectamineTM 2000 It shows down-regulation effect on the subsets of CD4+ T cells and CD8+ T cells.The regimen has good immunogenecity and could induce Th2 type humoral response in C57BL/6 mice.The immunization suppresses secretion of IFN-?.It can greatly enhance the titres of Ag85A-specific antibodies.LipofectamineTM 2000 can act as an adjuvant through comparation with negative control group.

4.Chinese medicine induces differentiation of bone marrow mesenchymal stem cells into cardiomyocytes

BACKGROUND:Traditional Chinese medicine shares wide application in clinical treatment because of its high security. At present, a lot of studies have been reported, in which, traditional Chinese medicine or monomers are used for in vitro differentiation of bone marrow mesenchymal stem cells into cardiomyocyte-like cells.
OBJECTIVE:To summarize and analyze the research progress of Chinese medicine to induce the differentiation of bone marrow mesenchymal stem cells into cardiomyocyte-like cells and to looking prospect for its application.
METHODS:CNKI database was searched for the articles about Chinese medicine-induced differentiation of bone marrow mesenchymal stem cells into cardiomyocytes, published from January 2001 to January 2013. The key words were“Chinese medicine, bone marrow mesenchymal stem cells, cardiomyocyte”in Chinese and“bone marrow mesenchymal stem cells, myocardial cells”in English. Obsolete or repetitive articles were excluded. Final y, 36 articles were included in result analysis.
RESULTS AND CONCLUSION:Different Chinese medicine formulations were used as inducers to induce differentiation of bone marrow mesenchymal stem cells into cardiomyocytes, including Panax notoginseng saponins, salvianolic acid B, Jiawei Danshen Yin, icari n, astragaloside. Then, the induced cells were examined by immunohistochemistry and reverse transcription-PCR. The results showed that the induced cells were positive for Desmin, cardiac troponin I and major histocompatibility complex. Positive cells were fusiform and fibroblast-like morphology, which indicates a role in promoting proliferation and differentiation. These findings show that Chinese medicine-induced differentiation of bone marrow mesenchymal stem cells into cardiomyocytes has become a worldwide research hotspot, providing a theoretical basis for clinical treatment of ischemic heart diseases.

Saliva and samples containing saliva were regarded as a kind of potential biological trace evidencefor individual identification in forensic science practice. In this paper, the authors have studied theDNA analysis of saliva and saliva containing samples. The results demonstrated that DNA can be extracted from saliva and saliva-containing samples. RFLPs analysis by DNA fingerprinting and PCRbased typing can be performed according to the amount of DNA extracted from these samples.
6.Optimization of Bacteriocin Produced by Lactobacillus pentosus 31-1

To optimize bacteriocin production,researching was done on the respect of incubation condition and the media components. And the optimum incubation time was 24h, the optimum temperature was 30℃, the optimum broth initial pH was 6.5. The optimum media component was: lactose 30g, trypone 15g, soybean peptone 20g, meat extract 30g, peptonethe 20g, Tween 80 1mL, K_(2)HPO_(4 )2g, NaAc 5g, Tri-Ammonium citrate 2g, (MgSO_(4)) 0.58g, MnSO_(4) 0.25g, H_(2)O 1000mL. Production of bactericoin under the optimum broth was 640AU/mL, 8 times than MRS media.
7.The clinical efficacy of hydroxyfasudil in senile heart failure caused by coronary heart disease

Objective To observe the clinical efficacy of hydroxyfasudil in senile heart failure caused by coronary heart disease (CHD). Methods 110 Patients with heart failure caused by CHD were randomly divided into two groups. 55 patients in control group were given conventional treatment, whereas the remaining patients were assigned into hydroxyfasudil group in the base of conventional treatment. After 6 months, we observed and compared the changes in cardiac function of the two groups. Results The effective rate of treatment group was 96.36% and the control group was 85.45%. There was statistical difference between the two groups (P < 0.05). Then we compared the degree of improvement of left ventricular diastolic function using ultrasonic cardiogram and Tei index. Left ventricular ejection fraction of treatment group improved significantly but biochemical indicators of two groups had no statistical difference. Conclusion In patients with heart failure caused by CHD, supplementation of hydroxyfasudil based on conventional therapy could improve diastolic function , potentiate left ventricular remodeling and enhance the life quality. Furthermore, it′s more sensitivity and reasonable to assess cardiac function using Tei index as compared with using ultrasonic cardiogram.
8.Inhibitory effect of joint gene expression of GM-SCF, IL-21, and Rae-1 in treatment of liver cancer model mouse

Objective To observe recombinant plasmids were constructed with the macrophage colony-stimulating factor ( GM-SCF) , interleukin -21 (IL-21) and retinoic acid early transcription factor-1 (Rae-1), and observe the inhibitory effects in subcutaneous liver cancer model in mice with the recombinant plasmids.Methods The recombinant plasmids of GM-SCF, IL-21 and Rae-1 were constructed with RT-PCR method, mouse model was constructed, the model mice were randomly divided into six groups including control, IRES/GFP, IRES/IL21, IRES/GM-SCF, IRES/GM-SCF-IL21 and IRES/combination with 10 mice included in each group, each groups (15 mice) were treated with the corresponding gene therapy.The survival rate were observed after 60 days.The blood levels of interferon -γ(IFN-γ) and interleukin -2 (IL-2) were detected in each group.Results The pGM-CSF-GFP-IRES-Rae-1-IL-21 has been successfully constructed.All mice had demised 14 and 16 days after treatment in the control and IRES/GFP groups, respectively.There were 2, 1, 11 mice remaining after 60 days of treatment in the IRES/GM-SCF, IRES/IL21 and IRES/GM-SCF-IL21 groups respectively.The survival rate of mice at 60 days of treatment was 73.33%, 13.33%, and 6.67% for groups IRES/GM-SCF-IL21, IRES/GM-SCF and IRES/IL21, respectively.The survival rate of the mice was significantly higher in IRES/GM-SCF-IL21 than the other groups.The levels of IL-2 and INF-γof mice 1-6 days after treatment gradually increased in the IRES/combination groups, including IRES/GM-SCF-IL21, IRES/GM-SCF and IRES/IL21.They were highest in the IRES/combination group and lowest (P<0.01) in the IRES/GM-SCF and IRES/IL21 groups, with the IRES/GM-SCF-IL21 group showing intermediate levels.By 6-10 days after treatment, IL-2 and INF-γlevels had stably increased in the IRES/combination groups, but had gradually decreased in the IRES/GM-SCF-IL21, IRES/GM-SCF and IRES/IL21 groups.At the end of treatment, IL-2 and INF-γlevels were significantly (P<0.01) higher in the IRES/GM-SCF-IL21 than were found in either the IRES/GM-SCF group or IRES/IL21 group, which were also significantly (P<0.01) higher than either the IRES/GFP or control groups.The levels of IL-2 and INF-γwere highest in the IRES/combination group ( P<0.01) and not significantly different among the IRES/GM-SCF, IRES/IL21, IRES/GFP, and control groups.Conclusion The inhibitory effects in subcutaneous liver cancer model in mice were obvious significantly, and its mechanism maybe be related to the activation of the body's immune.
9.Nursing of skin flaps after 72 hours' transplantation of femoral skin flap for repairing soft tissue defect in lower extremity

Objective To summarize the strategies for nursing of skin flaps after 72 hours' transplantation of femoral skin flap for repairing soft tissue defect in lower extremity.Method One hundred and sixty trauma patients with lower extremity soft tissue defect underwent transplantation of femoral skin flap for repairing.The skin flaps were nursed within 72h after transplantation.Results The skins flaps in 160 patients receiving to repair tissue defect of lower limbs all survived.Within 72 hours after operation, 12 patients suffered from vascular crisis, which were cured after pertinent treatment.Conclusions For the patients receiving to repair tissue defect, the skin flaps should be carefully nursed. Meanwhile, preoperative psychological nursing, postoperative pain nursing and health education are critical for the survival of skin flaps.
10.Analysis of platelet-associated antibodies among cancer patients

Objectives To analyze the mechanisms and implications involved in the generation of platelet associated antibodies in cancer patients.Methods HLA IgG antibody from 116 cancer patients who had received repeated platelet transfusion (more than 3 times) was screened by ELISA and the GTI QUICKSCREEN kit. The positive samples from the initial screening were confirmed for HLA and HPA antibodies using the PAKPLUS reagent. Patients with antibodies were observed for symptoms and clinical effectiveness of platelet transfusions. Results A total of 44 cases of HLA IgG antibody were screened out of 116 samples, with an antibody rate of 37.93%. Among those 44 samples, 18 were confirmed to contain anti-HLA-Ⅰantibody, 8 with anti-HPA antibody, 10 with both anti-HPA and anti-HLA antibodies, and 8 were not identified.
