1.Investigation of self-care ability and influencing factors of rectal cancer patients with temporary ileum stoma

Objective To describe self-care agency and its influence factors of the patients with temporary ileum stoma. Methods The descriptive statistics of non-experimental study was performed in the study. The convenience sample consisted of 144 patients from 3 hospitals in Shanghai. The self-care agency was measured by the Colostomy Self-care Agency Scale. Results 144 cases of colorectal cancer temporary ileum stoma patients′ self-care ability scored an average of 57.02 ± 7.00, including self- care willingness (37.19 ± 4.04) points, self-care knowledge 17 points, self-care skills 3 points. The multiple stepwise regression analysis showed that seven factors were statistically significant including age, gender, culture level, living area, postoperative months, presence of complications, whether to accept professional nursing training or education. Conclusions Temporary ileum stoma in patients of rectal cancer with medium self-care ability, but self-care skill level is relatively not high and should be strengthen attention in patients with rectal cancer undergoing ileum stoma. Targeted nursing intervention should be carried out to improve the self-care ability and improve the quality of life of patients with stoma.
2.FGFR1 selective inhibitor PD173074 can reduce proliferation and induce apoptosis of nasopharyngeal carcinoma.

OBJECTIVE:
To study the influence of PD173074 on proliferation and apoptosis of nasopharyngeal carcinoma.
METHOD:
With immunoblotting and RT-PCR, FGFR1 expression was detected in CNE, PONE1 and C666-1 cell lines. With MTT assay,the time-effect and dose-effect correlation between PD173074 and inhibition of CNE proliferation was evaluated. After PD173074 stimulation, the phosphorylation level of FGFR1 and AKT was detected with immunoblotting assay. Furthermore, influence of PD173074 on the activation of Caspase3 and Caspase9 was detected to study the underlying mechanism of why PD173074 could inhibit CNE proliferation.
RESULT:
FGFR1 has the highest expression in CNE cell line. Under incubation of 10 nmol/L PD173074 stimulation for 36 hours to 72 hours, the phosphorylation of FGFR1 and AKT was impaired significantly, which further reduced the proliferation of CNE. Moreover, PD173074 can activate the intrinsic apoptotic pathway by stimulating Caspase9,which activated Caspase3 and induced the apoptosis.
CONCLUSION
PD173074 could inhibit proliferation of nasopharyngeal carcinoma cell through reducing the phosphorylation of FGFR1 and AKT. Additionally, PD173074 can induce CNE apoptosis by activating intrinsic apoptotic pathway via cleaving Caspase9 and Caspase3.

3.Effect of leptin on the oxidative damage in human retinal pigment epithelial cells

Objective To investigate the effects of leptin on the oxidative damage in human retinal pigment epithelial (RPE) cells.Methods Human RPE cells (ARPE-19) were cultured in vitro,and randomly divided into control group and insulin resistance group.RPE cells were treated with 0,10,100 ng/mL leptin for 24,48,72 hours respectively.Then the levels of reactive oxygen species (ROS) expression in RPE cells were detected by 2',7'-dichlorofluorescin-diacetate (DCFH-DA),and the levels of 8-hydroxy-2′-deoxyguanosine (8-OHdG) expression in RPE cells were observed by immunocytochemistry (ICC),and the levels of human 8-oxoguanine DNA glycosylase 1 (hOGG1) expression in lysate were measured by Western blot.Results After 24,48,72 hours,the level of ROS (Control group:F=37.136,37.178,49.634; P<0.05.Insulin resistance group:F=9.822,28.881,71.150;P<0.05),8-OHdG (Control group:F =88.643,390.920,1039.276; P < 0.05.Insulin resistance group:F =273.311,299.155,82.237;P<0.05) and hOGGl (Control group:F=470.062,1073.113,295.456;P<0.05.Insulin resistance group:F =240.032,592.389,527.760 ; P<0.05) expression increased significantly with the increase of leptin concentration in control group and insulin resistance group.Under the same leptin concentration,the level of 8-OHdG has a trend that it was higher in the insulin resistance group than the control group.After 24 hours,the difference of hOGGl expression between control group and insulin resistance group was not significant (F=23.392,P>0.05).After 72 hours,the level of hOGGl expression was significantly higher in the insulin resistance group than the control group (F=129.394,P<0.05).The level of hOGGl expression was significantly higher at 48 hours than that at 24 hours and 72 hours (P< 0.05).Conclusion Leptin could induce the oxidative damage of RPE cells in normal and insulin resistance status.With the increase of leptin concentration and time extended,the degree of oxidative damage and its repair were both increased.The degree of oxidative repair increased with the increase of leptin concentration,but decreased with time extended.
4.Analysis of microsurgical repair of deep wounds of the foot near the ankle joint

Objective To evaluate simple and practical methods to repair deep wounds in the foot near the ankle joint.Methods 128cases of deep wounds near ankle jo int with exposed bone and tendons fro mJan 1990to Jan 2002were reviewed .Altogether 13types of microsurgical flaps were applied in these cases and obtained satisfactory results.Timing of sur gery:53cases of primary repair,28c ases of secondary repair,47cases of delayed repair.Results26cases of reverse skin avultion were repaired by venous anastomosis in a valsed flaps,of which19survived completely,and 7underg one partial necrosis.There were 19f ree tissue flap grafts in which 13sur vived,3failed and 3undergone partial necrosis and infection.All pedicle tissue flap grafts amounted to 83cases wi th only3partial necrosis in 58cases of sura l nerve flaps graft,and 1total necro sis in 25other types of flap grafts.Con-clusion Sural nerve flaps graft is a reliable method to repair deep wounds with exp osed bone and tendons near the ankle joint.It is easy to perform with excellent abrasion resistance and little effect on the donor limb.

Objective To investigate the association of cytotoxic T lymphocyte associated antigen 4 (CTLA 4) gene polymorphism with type 1diabetesmellitusandautoimmunethyroiddiseases. Objective To study the effect of Gabapentin(GBP)on the transient receptor potential vanilloid l(TRPV1)in dorsal root ganglion (DRG)of rats with chronic constriction injury(CCI). Methods SD male rats were used to establish CCI model and randomly divided into 3 groups,the control group,the CCI group,and the CCI+GBP group. Mechanical allodynia and thermal hyperalgesia were measured to evaluate the es?tablishment of CCI model and the analgesic effect of GBP. At the postoperative 8 days,all rats were sacrificed and dorsal root ganglions were collect?ed for further RT?PCR and Western?blotting analysis to detect expression of TRVP1 in DRG. Results The mRNA and protein expression of TRPV1 were both significantly increased in DRG after CCI operation and GBP could decrease mRNA expression of TRPV1 gene in DRG after CCI. Conclusion The expression of TRPV1 was increased in DRG after CCI operation which was decreased after administration of GBP.
8.The Development of Endogenous CO and its Mechanism in Asthma

Objective To explore the development of endogenous CO and its mechanisms in asthma.Methods 50 guinea pigs were equally separated 5 groups.After the models of asthmatic guinea pigs were established in 4 groups,3 groups of them were treated with dexamethasone,the specific stimulator(hemin)and inhibitor(Sn-PP)of HO-1 respectively.The rest of groups were asthmatic one and normal control one(NCG).From all tested animals,the COHb in blood and lung HO-1 activity were examined,the lung tissues were pathologically observed and immunohistochemically stained.Results In asthmatic and hemin stimulated groups,the levels of COHb with enhanced expression of lung HO-1 and their activity were significantly increased compared with NCG(t=4.43~9.97,P<0.01).For example,the lung HO-1 activities in asthmatic group were (881±361)pmol/(mg*pro*h),COHb (4.94±2.15)% respectively.In dexamethason prevented and Sn-PP inhibited groups,however,all determined levels were markedly reduced compared with asthmatic group(P<0.01).Conclusion The increased HO-1 had resulted in the growth of endogenous CO in asthma.
9.Regulatory effect of tumor necrosis factor-α induced protein 8 like-2 expression on immunosuppressive activity of CD4 + CD25 + regulatory T cells in mice

Objective To observe the expression of tumor necrosis factor-α induced protein 8 like-2 (TIPE2) in CD4 + CD25 + regulatory T cells ( CD4 + CD25 + Tregs) and analyze its potential effect on immunosuppressive activity of CD4 + CD25 + Tregs. Methods CD4 + CD25 + Tregs were purified from spleen of the BALB/c mice by using magnetic cell sorting system.The expressions of TIPE2 mRNA and protein in CD4 + CD25 + Tregs were detected by using RT-PCR and Western blot.CD4 + CD25 +Tregs were further infected with the recombinant lentiviruses that carried small interference RNA(siRNA)to knock down the TIPE2 expression.Based on the expressions of cell surface molecules including cytotoxic T-lymphocyte-associated antigen (CTLA)-4 and forkhead/winged helix transcription factor p3 (Foxp3) detected by flow cytometry and the levels of cytokines including interleukin (IL)-10 and transforming growth factor (TGF)-3 examined by ELISA in CD4 + CD25 + Tregs,the functional role of TIPE2 in controlling suppressive activity of CD4 + CD25 + Tregs was analyzed.In the meantime,the proliferation activities of T effector cells were assayed by MTT test. Results A 147 bp TTPE2 gene band and a clear TIPE2 band with a molecular mass of approximately 21 000 in CD4 + CD25 + Tregs were detected by using Westem blot.Cell surface molecule as well as cytokine expressions were significantly down-regulated when the CD4 + CD25 + Treg stimulated and activated by anti-CD3/CD28 was cultured for 24 hours after the siRNA silenced CD4 + CD25 + Treg cells ( P < 0.01 ).Meanwhile,the suppression role of CD4 +CD25 + Tregs on the proliferation activity of T effector cells was weakened obviously ( P < 0.05 ). Conclusions As an important immunoregulatory molecule,TIPE2 not only expresses in the CD4 + CD25 +Tregs,but also affects the immunosuppressive function of CD4 + CD25 + Tregs.
10.Transconjunctival approach to the fractures of the orbital floor and infraorbital rim

Objective:To study the surgical approach to the orbital floor and infraorbital rim.Method:15 cases of fractures of the orbital floor and infraorbital rim were treated with the transconjunctival approach.Result:All fractures of the orbital floor and infraorbital rim were repositioned and fixed through transconjunctival approach. Except for 1 case with mild canthal malposition, no other complication was observed.Conclusion:The transconjunctival approach can provide adequate exposure of the infraorbital rim and orbital floor without scar formation in the face.
