1.Construction and identification of eukaryotic expression vector of short hairpin RNA for IER3IP1 gene

Objective:Toconstruct the shRNA(Short Hairpin RNA)eukaryotic expression vector targetingat IER3IP1 gene and observe its silencing effect on IER3IP1 gene.Methods:Two pairs of DNA fragments targeting at IER3IP1 gene and one pair of DNA fragment of negative control were synthesized and cloned into the Eukaryotic Expression Vector pGenesil-1.These vectors were separately transfected into the L02 cells.Inhibitory effect at the time of 24,48,72 h after transfection were detected by semiquantitative RT-PCR.Results:These shRNA eukaryotic expression vectors were successfully constructed.One of them could significantly and specially inhibite the expression of IER3IP1 gene and the inhibition ratios were separately 64%,79%and 69%at the time of 24,48 and 72 h,compared with empty vector group.Conclusion:The combinated eukaryotic expression vector could inhibit the expression of IER3IP1 gene,which forms the basis for investigating IER3IP1 gene's function.

Objective: To assess the mental health status of poor medical students.Methods: 40 poor medical students and 40 ordinary medical students were studied controlly with symptom checdlist-90. Results: poor medical students than ordinary medical students had more scores in factors and all average. Conclusion: the mental health status of poor medical students is not good.The work of mental health education and consultation should be accessed according to their mental status for them.
4.Mining the relation between leukemia and genes using Weka

Objective To mine the relation between leukemia and genes using Weka. Methods The papers on leuke-mia and genes were retrieved from PubMed, their subject headings and subheadings were extracted using BICOMB to generate co-occurrence matrix and term-paper matrix. The research hotspots were found by cluster analysis of the data on co-occurrence matrix using Weka and Cobweb. The literature was verified. Results The 42 high fre-quency words were clustered into 7 classes by Weka. No high frequency words of leukemia or genes were found in classes 1, 2, 4 and 5, indicating that their clustering efficiency was poor. The clustering efficiency of the other 3 classes was good. Conclusion Cluster analysis showed that leukemia is related with myc gene, ab1 gene, p53 gene, virus gene, immunoglobulin gene and mdm gene.
5.Correlation of urine albumin and creatinine ratio and short -term prognosis in patients with acute ischemic stroke

Objective To explore the correlation of urine albumin and creatinine ratio(UACR)and short -term prognosis in patients with acute ischemic stroke (AIS).Methods 112 patients with AIS were selected as the research subjects.The admission UACR and related laboratory examination indexes were detected,the US national institutes of health stroke scale (NIHSS)scores,and 28 d after admission of the modified Rankin scale (mRS) prognostic score were recorded.The NIHSS 5 or more points and NIHSS <5 points,different levels and UACR patient observation index between the different prognosis group were compared.Results NIHSS 5 or more group of patients on admission UACR level[(15.41 ±3.21 mg/g)]was higher than the NIHSS <5 patients (9.52 ±2.04)mg/g (t =12.214,P <0.001);UACR acuity 11.60mg/g group in NIHSS of 5 or more proportion of patients (87.5%),mRS score >2 points (82.1%),the proportion of the recent death patients (23.2%)than the proportion of UACR <11.60mg/g,the proportion of the group (32.1%,25.0%,7.1%;the chi -square values 33.509,34.476,5.598,all P <0.050);MRS score >2 groups of patients UACR level[(16.10 ±3.29)mg/g]was higher than that in patients with mRS score 2 points or less (8.38 ±1.95)mg/g(t =20.132,P <0.001);Pearson correlation analysis showed that the standard of UACR and NIHSS score (r =0.412,P <0.05 ),high -sensitivity C -reactive protein (hs -CRP)(r =0.473,P <0.050),white blood cell count (WBC)(r =0.442,P <0.050),fasting plasma glucose (FPG)(r =0.375,P <0.050)were positively correlated.Multiariable Logistic regression analysis showed that UACR (OR =1.587,P <0.050)was a recent independent risk factors of poor prognosis in patients with AIS. Conclusion UACR the illness severity and recent prognosis of patients with AIS is closely related to high levels of UACR may be recent independent risk factors of poor prognosis in patients with AIS.

7.Culture supernatant of bone marrow mesenchymal stem cells for treatment of bronchial asthma and its influence on lung inflammation

BACKGROUND:Bronchial asthma is considered general y to have an association with Th2 immune response disease, but there is no ideal treatment. Bone marrow mesenchymal stem cel s as a kind of adult stem cel s not only have the multipotent differentiation and proliferation capacity, but also have low immunogenicity and immunoregulation ability. OBJECTIVE:To investigate the effect of culture supernatant of bone marrow mesenchymal stem cel s on lung inflammation of bronchial asthma mice. METHODS:Twenty experimental mice were randomly divided into control and experimental groups, 10 mice in each group. At 0 and 14 days, intraperitoneal injection of ovalbumin induced sensitization in mice, and at 24-26 days, aerosolized ovalbumin solution was used for excitation. From the 24th day, in the experimental group, bone marrow mesenchymal stem cel supernatant was intraperitoneal y injected at 2 hours before excitation;meanwhile, normal saline was injected in the control group. At the last of excitation, the mice were sacrificed under anesthesia to take serum samples, bronchoalveolar lavage fluid and lung tissues. RESULTS AND CONCLUSION:(1) Mice in the control group appeared to have abnormal lung tissue structure, and there were a large amount of eosinophils and monocytes in the submucosa and muscularis. However, lung inflammation was relieved in the experimental group after bone marrow mesenchymal stem cel treatment. (2) The levels of interleukin-17 in the bronchoalveolar lavage fluid and serum were significantly lower in the experimental group than the control group (P<0.05), but there was no difference in the levels of interleukin-4 between the two groups (P>0.05). These findings indicate that the intraperitoneal injection of bone marrow mesenchymal stem cel s can ease lung inflammation and reduce levels of inflammatory markers in the bronchoalveolar lavage fluid and serum of bronchial asthma mice.

Young faculty are an essential part of a university's strength and is important for its sustainable development.And their scientific research capability is associated with the long-term development of the university.At present,there are 4 major problems with the development of young faculty's research capabilities,which include their relatively single academic knowledge structure,insufficient experience in writing proposals,insufficient opportunities for academic exchange,and inequity in resource allocation.Establishing a training mechanism may help solve some of the problems and promot their development.The research management department of a university must abandon erroneous traditional management concepts,provide research training seminars,develop a new mechanism to stimulate academic exchange,design a system for short-term laboratory rotations,and promote the participation of your faculty in major research projects.These may bring about marked improvement in the improvement of young faculty's research capabilities.

[Abstract ] Tumor, with its high morbidity and mortality, has been gained increasingly high attention from the world .Current-ly, the main treatment for solid tumor is still surgical resection , during which surgery , anesthesia and other factors may have such side effects as releasing tumor cells into blood , lymphatic , bone marrow even organs and then resulting in the formation of micrometastatic lesion, increasing the risk of tumor recurrence and metastasis and finally affecting the postoperative survival rate .Recently, studies have shown that most anesthetic agents can influence the function of immune system and the activity of tumor cells , and then impact the tumor micrometastasis .This paper will summarize the research progresses in the impacts of anesthetic agents on tumor micrometastasis during perioperative period .
10.Glucose-reducing effect of induced human bone mesenchymal stem cells on diabetic mice

Objective To study and explore Glucose-reducing effect of induced human bone mes-enchymal stem cells on diabetic mice.Methods The diabetic model of rats was duplicated by injection of STZ intraperitoneally.The induced cells were implanted into diabetic mice.Blood glucose levels were moni-tored every 3 days after implantation for 14 days.Results The mice receiving the treated Cells began to decrease their blood glucose levels after 3days.But control Animals that did not receive induced cells exhib-ited persistent hyperglycemia.Conclusions Induced cells by hBMSCs can decrease blood glucose levels on diabetic mice.
