Objective To investigate the role of nitric oxide/cyclic guanosine monophosphate (NO/ cGMP) signaling pathway in dexmedetomidine-induced reduction of neuropathic pain (NP) in the rats. Methods Forty-two healthy male Sprague-Dawley rats, weighing 200-250 g,were randomly divided into 7 groups (n =6 each) using a random number table:normal control group (C group);NP group;dexmedetomidine group (D group);dimethyl sulfoxide (DMSO) group;a non-selective nitric oxide synthase inhibitor N-nitro-L-arginine methyl ester (L-NAME) group (L-NAME group);inactive enantiomer D-NAME group (D-NAME group);a soluble guanylyl cyclase inhibitor 1H-[1,2,4] oxidazole[4,3-a] quinoxalin-1-one (ODQ) group (ODQ group).NP was induced by chronic constriction injury to the sciatic nerve in anesthetized rats.On day 7 after chronic constriction injury, dexmedetomidine 1.5 μg/kg was injected intrathecally in group D,and in DMSO,L-NAME,D-NAME and ODQ groups,DMSO l0 μl,L-NAME 100 μg,D-NAME 100 μg and ODQ 10 μg were injected intrathecally, respectively, and 25 min later dexmedetomidine 1.5 μg/kg was injected intrathecally. The thermal paw withdrawal latency was measured immediately after intrathecal administration and at 30,60,90,120,150,180 and 210 min after intrathecal administration, and the area under the curve of thermal paw withdrawal latency was calculated to reflect the thermal pain threshold. Results Compared with group C,the thermal pain threshold was significantly decreased in the other six groups (P<0.05).The thermal pain threshold was significantly higher in D,DMSO,L-NAME,ODQ and D-NAME groups than in group NP (P<0.05).Compared with group D,the thermal pain threshold was significantly decreased in L-NAME and ODQ groups (P<0.05),and no significant change was found in the thermal pain threshold in D-NAME and DMSO groups (P>0.05).Conclusion The mechanism by which dexmedetomidine reduces NP is related to activation of NO/cGMP signaling pathway in the rats.

Objective: To study the mechanism of Tuina in the treatment of skeletal muscle injury. Methods: Rabbits were heavily beaten at gastrocnemius muscle to make acute contusion model and then treated respectively by early Tuina and routine Tuina. The number of satellite cells of skeletal muscles was observed. Results: The number of the satellite cells continued to grow in both groups, and it began to increase significantly 3-5 days after Tuina treatment. Early Tuina treatment produces larger number of satellite cells than routine Tuina treatment. Conclusion: Early Tuina treatment is helpful to the marked recovery of skeletal muscles by increasing the number of satellite cell.

Journal of Jilin University(Medicine Edition) 2000;26(2):142-143
Objective:To observe the living cells on the surface of intraocular lenses and to compare the cellular numbers and morphology of prestaining to that of poststaining. Methods:Twenty adult pigment rabbit eyes were given posterior chamber intraocular lens implantation.Lenses were extracted and put into 1640 cell cultural liquid immediately on the 1 st,3 rd,7 th,14 th and 28 th day respectively after operatrion. The samples were observed under inverted phase contrast microscope subsequently. Results:We found cells deposited on the surface of lenses from 1 st to 28 th day after operation. These cells contracted and lost three-demensional appearance while its number was less than that of prestaining obviously. Conclusion:A part of cells remove from the surface of lenses during staining;however,we could count the number but could not affirm cellular types which would make error in cellular classification and counting. The fact suggests that we should discover new accurate and reliable methods to avoid mistakes during experiment.

Chinese Journal of Digestion 2011;31(9):613-618
Objective To observe the protective effect of probiotics VSL # 3 on the rats of trinitrobenzene sulphonic acid (TNBS) induced acute colitis, and the effect on the expression of tight junction proteins claudins (claudinl, 2, and 3) in colon mucosa. Methods Rat of colitis model was induced by TNBS one time enema. Since the second day after the model established, 2ml 0.9% sodium chloride was intragastric administrated into 8 rats each day (TNBS group), 8 rats with 100 mg VSL # 3 (VSL # 3 group ), another 8 rats with 5-aminosalicylic acid (5-ASA) intragastric administration (5-ASA group), and 0.9% sodium chloride was intragastric administrated daily into 5 no modeling rats as control group. All were treated for 7 days and then sacrificed. Fecal samples were collected for bacterial culturing and detecting the changes of colon bacteria colonies. Disease activity index (DAI was calculated according to diarrhea, hemafecia and weight. The colon tissue was taken for HE staining and the pathological changes were observed under microscope. The changes of myeloperoxidase (MPO) level in colon tissue were examined by enzyme-linked immunosorbent assay (ELISA). The expression and distribution of claudins 1, 2, and 3 in colon tissue was detected by a novel quantum dots immunofluorescence labeling technique. Results Compared with TNBS group, both VSL # 3 and 5-ASA treatment could improve colitis, as well as decreas DAI and colonic MPO level, and lower the inflammatory score of colitis. Compared with control group, the bacteria counting in the colon of colitis rats changed, the expression of Claudins 1, 2, and 3 expression was significantly increased (average density: 66.200±5.737, 71.780±6.670 and 61.300±5. 199,t=17.237, 27.909and 21.788, all P<0.01). Compared with TNBS group or 5-ASA group, VSL# 3 could adjust the balance of intestinal bacteria and increase the expression of Claudin 1 and 3 (Claudin 1 : 75. 550 ±8.717, 66.200±5.737 and 67.080±5.401;t=9.348, 8. 469; all P<0.05; Claudin-3: 68. 820±7.443, 61.300±5. 199 and 59. 830±5. 930;t=7. 519, 8. 988, all P<0.05), however claudin 2 expression was down-regulated (58. 740±6. 457, 71. 780±6. 670 and 66. 870±5. 791; t= 13. 033,P<0.01, t=8. 123, P<0. 05). Conclusion Probiotics VSL# 3 may enhance the intestinal mucosal barrier function through changing the claudins expression, and play a role in the relief of inflammatory reaction in acute colitis.
