Objective:To investigate the relationship of urinary 8-oxoguanosine(8-oxoGsn)with muscle mass, muscle strength, advanced glycation end products(AGEs), trace elements, heavy metals and other health-related indictors in different age groups of the Beijing area.Methods:A cross-sectional research was conducted.Healthy adults aged 25 to 93 years who sought health examination in the Health Examination Center of Beijing Hospital were recruited.Participants were divided into the young and middle-aged group and the elderly group with age 60 as the cutoff.Urinary 8-oxoGsn levels were detected by mass spectrometry and adjusted using urine creatinine values.Body composition was measured by multifrequency bioelectrical impedance analysis(BIA). Grip strength, 6-meter walking speed and 5-times sit to stand test were conducted by experienced team members.Skin autofluorescence was used to detect skin AGEs.A portable optical scanner was used to detect heavy metals and trace elements using reference points of the palm.Levels of fasting blood glucose, glycosylated hemoglobin, high-density lipoprotein and other common blood biochemical indicators were measured.Results:A total of 106 subjects were enrolled, including 68 in the young and middle-aged group and 38 in the elderly group.The proportion of patients with hypertension(14 ases or 36.8% vs.7 ases or 10.3%), systolic blood pressure[130(120, 140) vs.120(110, 126)mmHg], fasting blood glucose[5.7(5.2, 5.9)mmol/L vs.5.2(4.9, 5.5)mmol/L], glycosylated hemoglobin[6.0(5.7, 6.2)% vs.5.7(5.4, 5.9)%], 8-oxoGsn/Cre[1.9(1.4, 2.6) vs.1.3(1.0, 1.6)], AGEs(2.44±0.46 vs.2.01±0.29), 5-times sit to stand test scores[7.8(6.9, 9.8)s vs.6.0(5.0, 6.8)s], magnesium(31.4±7.2 vs.27.7±6.4), mercury(0.013±0.003 vs.0.008±0.003)and silver[0.011(0.010, 0.012) vs.0.010(0.009, 0.011)]were higher in the elderly group than in the young and middle-aged group, while grip strength[28.0(22.0, 35.1)kg vs.36.6(28.5, 49.1)kg], fat-free mass[44.9(37.5, 51.1)kg vs.53.3(42.4, 58.5)kg], trunk muscle mass[21.0(17.5, 23.9)kg vs.25(19.8, 27.4)kg], appendicular skeletal muscle mass[20.9(17.6, 23.9)kg vs.24.9(19.8, 27.3)kg], calcium[273.3(219.1, 480.0) vs.457.8(428.5, 489.1)], cobalt[0.029(0.027, 0.031) vs.0.031(0.028, 0.034)], selenium[1.44(0.93, 1.71) vs.1.61(1.53, 1.68)]and nickel[3.5(3.3, 4.0)*10 -3vs.3.8(3.6, 4.1)*10 -3]were lower in the elderly group than in the young and middle-aged group( P<0.05). Urinary 8-oxoGsn/Cre levels were positively correlated with age, time of 5-times sit to stand test, AGEs, fasting blood glucose, mercury and aluminum( rs=0.443, 0.292, 0.357, 0.205, 0.316 and 0.214, P<0.05), and negatively correlated with trunk muscle mass, appendicular skeletal muscle mass, fat-free mass, grip strength, silicon and manganese( rs=-0.334, -0.333, -0.332, -0.366, -0.246 and -0.234, P<0.05), with statistical significance. Conclusions:Increased urinary 8-oxoGsn/Cre levels are correlated with decreased muscle mass, poor physical function, accumulation of AGEs, decreased trace element levels and increased heavy metal levels.Therefore, 8-oxoGsn has the potential to be a broadly representative and sensitive indicator for health assessment.

Methods: From March to December 2009, 37 386 eligible residents from four cities in northern China (Tianjin, Shenyang, Taiyuan, and Rizhao) were enrolled in a follow-up study by using the random cluster sampling method. Demographic characteristics, lifestyle, history of diseases, and self-report situation of hypertension were collected by using questionnaire. Based on the average annual concentration of NO₂ during the period from the cohort to the onset of hypertension as an estimate of exposure, the effect of NO₂ exposure on hypertension was analyzed by employing Cox proportional hazards model. The interactions between NO₂ exposure and different characteristics (age, sex, body mass index, smoking, alcohol consumption, education, economy, exercise, and fruit intake) were also examined. Results: The baseline age of residents was (43.74±13.78) years, and the body mass index (BMI) was (22.56±2.92) kg/m². During an average follow-up time of 11.40 years, 2 619 (7.0%) new cases of hypertension were reported. The overall mean environmental pollution levels during the study period for the entire cohort was (40.74±17.07) μg/m³. After adjusting for age, sex, BMI, family history of hypertension, socio-economic information, and lifestyle, the hazard ratio (HR) of incident hypertension with a 10 μg/m³ increase of NO₂ was 1.21 (95%CI: 1.18-1.25). Compared with residents aged 60 years and over (HR=1.19, 95%CI: 1.14-1.26), former and current smoking (HR=1.20, 95%CI: 1.14-1.25), and high-frequency fruit consumption (HR=1.17, 95%CI: 1.13-1.21), residents younger than 60 years (HR=1.28, 95%CI: 1.25-1.32), non-smoker (HR=1.23, 95%CI: 1.19-1.27), and low-frequency fruit consumption (HR=1.27, 95%CI: 1.20-1.35) had stronger interaction effect with NO₂ (all P values for interaction<0.05). Conclusion NO₂ exposure may lead to the onset of hypertension, which has a stronger effect on people younger than 60 years old, without smoking history and with low-frequency fruit consumption.

Objective To evaluate the effects of extracts of Semen Coicis (ESC) on a BALB/c mouse model of atopic dermatitis (AD),and to explore its potential mechanism.Methods Forty specific pathogen-free (SPF) female BABL/c mice were randomly divided into blank group (8 mice,receiving no treatment) and AD model group (32 mice).The mice in the model group were topically treated with 2,4-dinitrochlorobenzene (DNCB) in acetone/olive oil to establish the mouse model of AD.After modeling,8 mice in the blank group and 8 in the model group were sacrificed immediately.The other 24 mice in the model group were randomly and equally divided into 3 groups:model control group receiving no treatment,ESC group and ESC vehicle group topically treated with ESC and ESC vehicle respectively once every day on the back and aural region of the mice for 28 consecutive days.Changes in skin lesions were observed by naked eyes every day.A thickness tester was used to measure the thickness of skin lesions on the left ear before modeling,at completion of modeling and 12 hours after the final treatment.At 12 hours after the final treatment,the mice in the above 3 groups were sacrificed,and the eyeballs were removed for collecting blood.Then,the sera were isolated,and skin tissue specimens were obtained from the skin lesions on the back.These tissue sections were subjected to hematoxylin and eosin (HE) staining and toluidine blue staining for observing the infiltration of inflammatory cells in skin lesions.An immunohistochemical study was performed to determine the expression of aquaporin 3 (AQP3),Toll-like receptor 2 (TLR2) and TLR4,and enzyme-linked immunosorbent assay (ELISA) to detect the serum levels of IgE,interleukin-4 (IL-4) and interferon-/ (IFN-γ).Results After 28-day treatment,skin lesions were improved in the ESC group.Compared with the model control group,the ESC group showed a significantly lower clinical symptom score (1.50 ± 0.58 vs.2.50 ± 0.58,P ＜ 0.05),decreased lesional thickness on the left ear ([0.31 ± 0.01] mm vs.[0.33 ± 0.01] mm,P ＜ 0.05),and lower number of infiltrating mast cells per high-power field (15.18 ± 1.64 vs.28.94 ± 1.28,P ＜ 0.05).Immunohistochemical findings indicated that the ESC group showed significantly lower expression of AQP3,TLR2 and TLR4 compared with the model control group,and decreased AQP3 expression in the spinous layer.Compared with the model control group,the ESC group showed significantly lower total serum IgE and IL-4 levels,but higher IFN-γ levels (all P ＜ 0.05).Conclusion Topical ESC is effective for the treatment of skin lesions in mouse models of AD,likely by regulating serum levels of IgE,IL-4 and IFN-γ and affecting the expression of AQP3,TLR2 and TLR4.

Objective To investigate the influence of an anti-integrin α6 monoclonal antibody (GoH3) on the in vitro infection of a human keratinocyte cell line HaCaT with HPV6/11 virus particles (VP). Methods HaCaT cells were infected in vitro with 4 different concentrations of HPV6/11 VP alone, HPV6/11 VP of 106 copies/ml after incubation with 6 different dilutions of GoH3, or 8 clinical isolates of HPV6/11 VP of 106 copies/ml before or after the incubation with 1∶ 100 dilution of GoH3. After additional culture, the infected HaCaT cells were collected and fluorescence quantitative (FQ)-PCR was performed to detect the HPV DNA load in cells. The inhibition rate of CoH3 on the infection was calculated. Results The viral load was different among the HaCaT cells infected with different concentrations of HPV6/11 VP (P < 0.01). The inhibition rate on the infection positively correlated with the concentration of CoH3 when the dilution was more than 1∶ 100; however, when the dilution was less than 1∶ 100, the increase in CoH3 concentration had no influence on the inhibition rate. The average viral load in HaCaT cells infected with clinical isolates of HPV6/11 VP was (5.81 ± 2.51) × 104 copies/ml in the absence of GoH3, (3.02 ± 1.21) × 104 copies/ml with the presence of CoH3, and the average inhibition rate of GoH3 was (46.9 ± 4.7)%. Conclusions GoH3 could partially suppress the adhesion of HPV6/11 VP to HaCaT cells, hinting that integrin a6 is an important HPV6/11 VP receptor on host cells.

Objective To evaluate the therapeutic effects of magnetically labeled mononuclear stem cells (MR-MNC) and mesenchymal stem cells (MR-MSC) transplantation in a swine acute myocardial infarction (AMI) model by MR imaging.Methods AMI model was established in swines by balloon occlusion of the left anterior descending coronary artery,107 autologous MR-MSC (n=7),MR-MNC (n=6) or PBS(n=6) were delivered via intracoronary infusion within 1 week after AMI [(4.8±1.3) days].Changes of infarct size and cardiac function were assessed with the use of 3.0T MR scanner before AMI,at 1 and 8 weeks post AMI.Results Magnetically labeled stem cells could be identified in the region of AMI by cardiac MR imaging.Eight weeks post transplantation,infarct size was significantly reduced in MR-MSC transplantation group(8.5%±0.5% vs.24.7%±3.1%,P＜0.05) and in MR-MNC transplantation (12.3%±1.5% vs.26.1%±1.5%,P＜0.05) while infaret size remained unchanged in PBS group (P＞0.05) compared to values at 1 week post AMI,left yentricular ejection fraction (LVEF) was also significantly higher in MR-MSC transplantation group (56.9%±1.3% vs.40.7%±2.0%,P＜0.05) and MR-MNC transplantation group (52.8%±1.4% vs.41.9%±3.3%,P＜0.06) compared to LVEF at 1 week post AML LVEF increase was more significant in swines received MR-MSC transplantation than MRMNC transplantation(16.2%±1.2% vs.10.9%±3.0%,P＜0.05).Prussian blue staining identified stem cells in corresponding myocardial regions with as by MRI.Western blot analysis demonstrated that cardiac expressions of myosin heavy chain (MHC) in MR-MSC group (100.3±5.5) and in MR-MNCs group (95.5±4.2) were significantly higher than that in PBS group (75.7±5.7,P＜0.05),myocardial troponin T (cTNT) expression in MR-MSC group (124.0±5.8) and MR-MNC group (118.4±4.4) were also significantly higher than in PBS group (93.3±3.9,P＜0.05) while MMP2/TIMP1 ratios in MR-MSC group (0.6±0.1) and MR-MNC group (0.6±0.1) were significandy lower than that in PBS group (4.2±0.2,P＜0.05).Conclusions Magnetically labeled MR-MSC and MR-MNC homed to heart post myocardial infarction and reduced infarct size,improved cardiac function.MR-MSC is superior to MR-MNC on improving cardiac function.
