OBJECTIVE:To investigate the protective effects of diphenyl ethylene glycoside(TSG)on hippocampal neuron cells ischemic injury model rats.METHODS:Hippocampal nerve cells ischemic injury rats’model was established with sodium dithionite,antagonism of TSG on ischemic cerebral injury was determined by methyl thiazolyl tetrazolium(MTT)microcol-orimetry and lactate dehydrogenase(LDH)efflux assay.RESULTS:Pseudo-ischemic cerebral injury could cause significant injury to neurons with elevated cell death rate,significantly decreased MTT value and increased LDH efflux rate.The appli-cation of TSG could lead in significantly decreased LDH efflux rate,elevated MTT value and reduced cell death rate.CON-CLUSION:TSG has a significant protective effect on nerve cells ischemic injury model rats.

The results showed that Con A-LAK cells could kill NK sensitive YAC-1, TNF sensitive L929 and H-2 low/non expression HCa-F target cells; The cytotoxicitity of Con A-LAK cells was performed by direct contact, the non-secretary pathway mediated by interaction of surface ligands of effector cells and receptors of target cells play an important role; Con A-LAK cells expressed mTNF-?, which participated in the killing of TNF sensitive tumor cells and might be one of the important effector molecules in MHC non-restricted, antigen non-specific cvtotoxicitv.

Objective To investigate the effects of anti-sense hTERT on the expression of VEGF and the receptor in SGC7901 cells, and study the influence in angiogenesis and progression in gastric carcinoma. Methods SGC7901 cell line was transfected by the recombinant virus containing sense and anti-sense hTERT cDNA. Then the expression of VEGF-C and Flt-4 was observed with RT-PCR, distribution of cell cycles was determined with flow cytometry. The expression of VEGF-C and Flt-4 protein was assessed with immunohistochemistry. Result The distribution of cell cycle of antisense hTERT transfected SGC7901 cells was changed, and the mRNA and protein expression of VEGF-C and Flt-4 was significantly down-regulated. Conclusion Anti-sense hTERT can act as an agent for inhibiting VEGF-C and Flt-4 mRNA and protein level, and it blocks tumor angiogenesis and lymphogenous metastasis.

BACKGROUND:Early diagnosis and accurate treatment of ligament injuries are the premise of avoiding chronic instability of knee joint and promoting functional recovery of knee joint.
OBJECTIVE:To investigate the clinical efficacy of limited incision with autologous tendon graft for knee ligament injury.
METHODS:Eighty patients with knee ligament damage were randomly divided into two groups. The cases of control group were given orthosis treatment, and the cases of observation group were treated with the limited incision plus autologous tendon graft.
RESULTS AND CONCLUSION:At 6 months post-treatment, the excellent efficacy rate and postoperative quality of life score were increased in the observation group, compared with the control group (P<0.05). The active and passive ranges of motion of knee joint in the observation group were significantly lower than that in the control group (P<0.05). At 3 months post-treatment, Lysholmp scores of the observation group was significantly lower than that in the control group (P<0.05). Limited incision plus autologous tendon graft treatment can significantly improve knee function after knee ligament injury.

Objective To investigate the inhibitory effect of siRNA human telomerase transcriptase (hTERT) on activity of telomerase and proliferation of lung carcinoma cell A549. Methods A plasmid including U6 promoter and siRNA of hTERT was designed and constructed. The plasmid was transfected into A549 cell line. The telomerase activity was tested by telomerase repeat amplification protocol ELISA (TRAP-ELISA). MTT assay was used to assay the cell proliferation activity,and hTERT expression was assessed by Western blot. Result The U6 expression plasmid that was constructed for hTERT gene 745 showed obvious interfering effect. hTERT-siRNA could down-regulate the expression of hTERT protein,inhibit telomerase activity and proliferation of A549 cells. Conclusion siRNA of hTERT can inhibit the expression of human telomerase and proliferation of A549 cells. It may open a new approach to the use of siRNA as a new tool to study gene function in cancer cell lines,and may be developed to be a new gene therapeutic agent for cancer.

Objective To evaluate the effects of interleukin-10 and dexamethasone on expression of TNF-? mRNA, IL-1? mRNA, and IL-1ra mRNA in acute lung injury (ALI). Methods A rat model of ALI was reproduced by intratracheal instillation of endotoxin (LPS) with a dose of 10mg/kg. 72 male S-D rats were randomly divided into control group, LPS group, LPS+IL-10 group, and LPS+Dex group, 18 rats in each group (6 rats at 2h, 6h, and 24h respectively). RT-PCR method was used to determine the expression of TNF-? mRNA, IL-1? mRNA, and IL-1ra mRNA in lung tissue. Results In LPS group TNF-? mRNA expression peaked at 2h, then decreased sharply; IL-1? mRNA expression increased markedly at 2h, peaked at 6h, then decreased; IL-1ra mRNA expression increased and peaked at 6h, and remained higher than control group at 24h. Both IL-10 and Dex inhibited TNF-? mRNA and IL-1? mRNA expression, but with no effect on IL-1ra mRNA expression. Pathological examination revealed that IL-10 or Dex attenuated injury to lung parenchyma. Conclusions The up-regulation of expression of both TNF-? mRNA and IL-1? mRNA was much earlier than that of the expression of IL-1ra mRNA, suggesting that there was an imbalance of inflammatory and anti-inflammatory mediators in the early phase of ALI. Both IL-10 and Dex could inhibit the expression of inflammatory mediators and exert no effect on the expression of anti-inflammatory mediators, thus contributing a balance effect between them and relieving ALI in rats.

A total of 128 patients with coronary heart disease/angina pectoris who underwent coronary arterial stent implantation were collected in this study between January and December 2002.① At 6-18 months of follow up,coronary artery angiography (CAG) showed that diameter stenosis rate ＞I 50% was considered the restenosis.While,36 out of 128 lesions had restenosis,and the rate was 28.1%.Serum lipoprotein (a) level in the restenosis group was significantly higher than non-restenosis group (P ＜ 0.01).②All patients were divided into high lipoprotein (a) group (the concentration ≥ 230 mg/L) and low lipoprotein (a) group (the concentration ＜ 230 rag/L).Restenosis rate in the high lipoprotein (a) group was significantly higher than low lipoprotein (a) group (P ＜ 0.05).Intravascular ultrasound showed that area and volume of newborn endomembrane in the high lipoprotein (a) group were significantly higher than slow lipoprotein (a) group after coronary arterial stent implantation (P ＜ 0.01).③ Correlation analysis suggested that.lipoprotein (a) was positively correlated with area and volume of newborn plaque (P ＜ 0.05).

ObjectivesTo study the effect of community interventions and management on asthma in children and its impact on knowledge, attitude, and practice (KAP)of parents and the home environment.MethodsAn asthma health management profile of 0 to 14-year-old asthmatic children (n =93 ) in Sanlitun and Liulitun communities in Chaoyang District in Beijing was established.The children were randomly divided into a management group ( n =49) and control group ( n =44 ) based on community.Community-integrated management,such as regular follow-up,condition monitoring and health education,was implemented in the asthmatic children in the management group but not in the control group.The parents' KAP and the household environment in the two groups were compared after 1 year based on the changes shown in the health management profile.ResultsThe asthma relapse rate decreased to 27.9％ (12/43) in the management group.Compared with the control group,the rates of hospitalization (x2 =8.174,P =0.004) and school absences ( x2 =4.962,P =0.026) significantly decreased.The KAP level of parents increased to 67.4％ ( 29/43 ) in the management group and 20.4％ (9/44) in the control group,and the difference was statistically significant (x2 =19.517,P ＜0.01 ).Knowledge improved the most and showed a significant difference from the control group ( x2 =19.517,P ＜0.01 ).Home environment in the management group improved to 76.7％ (33/43).The number of indoor pets ( x2 =3.906,P =0.048) and indoor cockroaches ( x2 =4.962,P =0.026 ) reduced and showed significant differences between the two groups.In addition,children's allergy-related symptoms decreased to 30.2％ ( 13/43 ) in the management group compared with 9.1％ (4/44) in the control group,which was a significant difference ( x2 =6.183,P =0.013).ConclusionsParents' knowledge of asthma,compliance behaviors,and home environmentwere effectively improved through community-integrated management.This management technique can reduce the allergy-related symptoms of asthmatic children,improve asthma severity,and reduce the influence of asthma on children's daily lives.
