1.Expression of cyclooxygenase-1 in cervical carcinoma and its significance

Objective To explore the expression of cyclooxygenase-1 (COX-1) in cervical carcinoma and its significance. Methods The pathological specimens were collected from 62 female patients, who were admitted to 307 Hospital of PLA from Jan. 1999 to Mar. 2005, including 31 cases of cervical carcinomas, 15 cases of cervical intraepithelial neoplasia and 16 cases of normal cervix. Surgery or biopsy was performed. Expression of COX-1 was detected by immunohistochemistry, and the relationship between COX-1 and clinicopathological feature was analyzed. Results The major sites of COX-1 expression were localized in cytoplasm, and next in cell membrane. Strongly positive expression of COX-1 was observed in cervical carcinomas, and weakly positive expression of COX-1 in cervical intra-epithelial neoplasia, with positive rates of 81% and 13%, respectively. There was no COX-1 expression in normal cervix. A significant difference was observed among these specimens. No obvious correlation was found between COX-1 expression and patient's age, tumor differentiation degree snd clinical stages. Conclusion Expression of COX-1 may serve as an auxiliary parameter for diagnosis, therapeutic scheme option, and prognosis of patients with cervical carcinoma.
2.Effects of rehabilitation on patients dependent on mechanical ventilation

Objective To explore weaning dependent patients off mechanical ventilation through rehabilitative treatment. Methods Eighty-four patients dependent on mechanical ventilation were randomly divided into a rehabilitation group (n =44) and a control group (n =40). All were treated with routine medication, but in addition patients in the rehabilitation program followed a rehabilitation program. Three weeks later, the success rates and the time of weaning off mechanical ventilation were observed in the two groups. Results In the rehabilitation group,88.6% of the patients were successfully weaned off mechanical ventilation, versus 60% of the control group. The respective average weaning times were 6.8 ± 3.8 days and 14.3 ± 5.2 days. Both differences were statistically significant. Conclusions Rehabilitative treatment improves the weaning success rate and reduces the time needed for weaning mechanical ventilation-dependent patients.
3.Expression of cyclooxygenase-1 in ovarian cancer and its significance

Objective To explore the expression of cyclooxygenase-1 in ovarian cancer and its significance. Methods Expression of COX-1 and CA125 was detected by immunohistochemistry in 37 cases of ovarian cancer and 31 cases of ovarian cyst. The serum level of COX-1 and CA125 was tested in 40 cases of ovarian cancer and 31 cases of ovarian cyst and 60 healthy volunteers by ELISA. Results The positive expression of COX-1 and CA125 was 78% and 57% in ovarian cancer and 3% and 7% in ovarian cyst,respectively. The expression of COX-1 and CA125 was 65% and 93% at the serum level. Conclusion Expression of COX-1 could act as an auxiliary diagnostic criterion in ovarian carcinoma.
4.Faciliated primary culture and amplification of breast cancer cells and their biological properties

Objective To efficiently builds up and expand breast cancer cells from cancer tissue and to identify their biological properties , provide abundant materials for research and personalized medicine .Methods Feeder cell layer and ROCK inhibitor Y-27632 were employed to faciliate the breast cancer cells;CCK-8 was used to determine the proliferation of the breast cancer cells; Cell cycle distribution was analyzed by flow cytometry; Histochemistry ( FH) assay to show the expression level of CK .The mRNA expression of HER-2, ER, PR and the breast cancer stem cell associated molecules (such as CD44, CD24, etc.) were detected by RT-PCR;STR assay was used for identifying verification of the cells .Results The use of feeder cells and Y-27632 facilitates rapid expand of the original breast cancer cells , and the cells have kept the original features of the tumor .Conclusions To use the method could obtain a large number of cells within a short time , which can promptly be used for the research of per-sonalized medicine .
5.Prevalence Study of GJB2,SLC26A4 and mtDNA 12S rRNA1555A>G Mutations in Hui Ethnic Group Patients with Non-Syndromic Hearing Loss from Northwest China

Objective To investigate the prevalence of GJB2 ,SLC26A4 and mitochondrial DNA 12S rRNA m .1555A>G(mtDNA 1555A>G) mutations in Hui ethic group patients with nonsyndromic hearing loss (NSHL) from Northwest China .Methods A total of 420 peripheral blood samples were collected from unrelated Hui ethic group probands with NSHL in Northwest China .Amplified the target gene by polymerase chain reaction (PCR) af-ter extracting genomic DNA from whole blood .The mtDNA 1555A>G mutation was detected by PCR -Alw26I di-gestion ,then direct sequencing was used to the positive samples of mtDNA 1555A> G ,the coding region of GJB2 gene ,exon 8 and 19 of SLC26A4 gene .Results There were 11(2 .62% ) cases caused by mtDNA 1555A>G homo-zygous mutation in 420 patients with NSHL .There were 41(9 .76% ) cases including homozygote and compound het-erozygote ,caused by GJB2 gene mutation ,which was the most frequent deafness -related gene .The allel frequency of c .235delC accounted for 6 .90% ,as well as the most frequent(51 .33% ) mutational pattern in GJB2 gene .There were 20 patients(4 .76% ) were found carring two allel mutations in SLC26A4 gene .The allel frequency of c .919 -2A>G was 5 .0% ,accounting for a total of 68 .85% in all base alterations of SLC26A4 gene ,which was the major mutant form of SLC26A4 gene .Conclusion GJB2 gene is the most common deafness -gene in Hui ethnic group pa-tients with NSHL from Northwest China ,while c .235delC is the main mutant form ,and c .919-2A>G is the hot-spot mutation of SLC26A4 gene .Through this study we can provide the molecular epidemiology basis for Hui ethnic group patients with NSHL from Northwest China in genetic diagnosis ,genetic counseling and therapy by associated testing of three frequent hearing loss genes .
6.Second trimester maternal serum screening for Down's syndrome in mainland China :a multi-center prospective study

Objective To evaluate the performance characteristics of the second trimester double-marker test for the detection of fetal Down's syndrome in mainland China. Methods This prospective national multi-centered study used alpha-fetoprotein (AFP) and free β-subunit of human chorionic gonadotrophin( free β-hCG)as the serum markers. From May 2004 to September 2006, 11 centers participated in the collection and analysis of maternal serum AFP and free β-hCG between 14 and 20+6 weeks of pregnancy. The screening results were calculated using the standard algorithm based on the standard database provided with the analytic software. Patients with an increased risk of Down's syndrome pregnancy (≥1/270) were offered genetic anmiocentesis. Outcomes of all pregnancies were obtained.Results A total of 66 132 singleton pregnancies were included in the study. The median maternal age was 27 years. At a cut-eft of 1 in 270, the detection rate (DR) based on a Caucasian database was 72% corresponding to a false positive rate (FPR) of 5%, and the DR based on the Chinese database was raised to 76% corresponding to an FPR of 5%. Conclusion The double-marker test using AFP and free β-hCG is an effective screen strategy for second-trimester detection of fetal Down's syndrome in mainland China. Ethnic variance exists between the Caucasian and Chinese populations. The accuracy of screening is increased by the use of race-specific medians.
7.Effects of butylphthalide combined with shuxuening on cognitive function and activity of daily living of patients with vascular dementia

Objective To investigate the effects of butylphthalide combined with shuxuening on cognitive function and activity of daily living (ADL) of patients with vascular dementia (VD).Methods A prospective research method was conducted, one hundred and one light and moderate VD patients admitted to Harrison International Peace Hospital from July 2014 to November 2016 were enrolled, and they were divided into a control group (49 cases) and an observation group (52 cases) according to random number tables. The two groups were given routine basic treatment, the control group received Shuxuening injection once a day for 2 courses (2 weeks as a course), while the observation group was additionally treated by butylphthalide capsule 0.2 g on the basis of the control group treatment. Before the treatment and 1 and 2 courses after treatment, the cognitive function was evaluated by mini-mental state examination (MMSE) and Hastgawa Dementia Scale (HDS), ADL was evaluated by ADL scale; the efficacy and adverse reactions were observed.Results The MMSE, HDS and ADL scores were increased gradually after treatment in the two groups, reached the highest degree after 2 courses of treatment, and the degrees of increase in the above indicators in observation group were more significant than those in the control group (MMSE score: 27.92±3.51 vs. 23.88±3.31, HDS score: 20.13±2.33 vs. 17.68±2.42, ADL score: 82.14±6.68 vs. 68.17±5.79, allP < 0.05). The total effective rate of observation group was significantly higher than that in the control group [86.5% (45/52) vs. 75.5% (37/49), P < 0.05], and the incidence of adverse reactions was lower than that in the control group [1.9% (1/52) vs. 4.1% (2/49)], being no significant difference between the two groups in adverse reaction (P > 0.05).Conclusions Polybutylene phthalate capsule combined with shuxuening injection can significantly improve the cognitive function and quality of life of patients with vascular dementia,and the clinical efficacy is safe and reliable.
8.Study on Anti-inflammatory and Detumescent Pharmacodynamic Material Basis of Jingyaokang Capsule

OBJECTIVE: To study the anti-inflammatory and detumescent pharmacodynamic material basis of Jingyaokang capsule, and to provide reference for secondary development, the establishment of quality control method and technological upgrading of the preparations. METHODS: The constituents of Jingyaokang capsule were extracted and separated with different solvents and macroporous adsorption resin to obtain constituent A (overall enrichment part), constituent B (chloroform extraction part), constituent C (water-course part) and constituent D (elution part of 60% ethanol). Using dexamethasone acetate as positive control, the anti-inflammatory and detumescent effects of Jingyaokang capsule and different extraction parts (constituents A, B, C, D) were investigated by mice ear edema and rat paw edema tests to screen the active fraction. UPLC-Q-TOF-MS method was used to analyze active constituent, identify compounds and attribute medicinal material. RESULTS: Anti-inflammatory and detumescent effects of constituent B (chloroform extraction part)+constituent D (elution part of 60% ethanol) were similar to those of Jingyaokang capsule in rats or mice, indicating both had synergistic anti-inflammatory and detumescent effects and were active constituents of Jingyaokang capsule. UPLC-Q-TOF-MS detection and identification showed that constituent B contained 13 compounds as strychnine, phellodendrine, periplogenin, tetraketone alcohol, 11-carbonyl-β-mastic acid, attributing to Strychnos nux-vomica, Stephania tetrandra, Periploca sepium, Lycopodium japonicum, Boswellia carterii, etc. Constituent D contained 7 compounds as adenine, hydroxysafflower yellow A, phellodendrine, neoeriocitrin, zingibroside R1, attributing to rainworm, Carthamus tinctorius, Stephania tetrandra, Davallia mariesii, Achyranthes bidentata, etc. CONCLUSIONS: Jingyaokang capsule shows the significant anti-inflammation and detumescent effects. The chloroform extraction part is synergistic with 60% ethanol elution part, which are the active constituents of anti-inflammation and detumescence,mainly including alkaloids, flavonoids and boswellic acids.
