1.Effects of chronic heart failure on contractile/diastolic function and calcium transient in guinea pig ventricular myocytes detected simultaneously by video-based motion edge-detection system

Aim To develop the animal model of guinea pig chronic heart failure and detect effects of chronic heart failure on contractile/diastolic function and calcium transient in guinea pig ventricular myocytes by video-based motion edge-detection system simultaneously.Methods The chronic congestive heart failure(CHF) model was produced in guinea pig by a procedure that descenting aorta was constricted.The contrasting indexes in 8 weeks include: with or without dyspnea,hemodynamics,the mass ratio of left ventricle to body,the mass ratio of lung to body,and the width of left ventricle.Left ventricular myocytes were enzymatically isolated.Then the contractile and calcium transient of a single cell from both normal and failure hearts were assessed in guinea pigs by a video-based motion edge-detection system simultaneously.Results The models with dyspnea showed striking increasing in left ventricular systolic pressure and left ventricular end diastolic pressure,as well as the mass ratio of left ventricle to body,the mass ratio of lung to body and the width of left ventricular hypertrophy.The left ventricular pressure maximal rising and declining velocity significantly decreased.Compared with normal cells,the value of shorting,the contractile and diastolic velocity of myocytes decreased significantly.The diastolic calcium increased,the extent of calcium transient[FL(2K2] decreased and the time to 50% diastolic calcium lengthen.But systolic calcium,the time to peak calcium were unchanged.Conclusions The dyspnea is a important parameter for evaluating the animal CHF model of guinea pig formed by constricting the descending aortas of guinea pigs after 8 weeks.The chronic heart failure may decrease contractile and calcium transient extent in ventricular myocytes detected by video-based motion edge-detection system simultaneously,which might be helpful for the further studying the mechanism of the development of CHF.
2.Effect of TREE bleaching agent on the shear bond strength of dentin to composite resin

Objective:To compare the shear bond strength of dentin to composite resin in different post-treatment time period after tooth bleaching with the TREE dental whitening gel and observe the dentin/composite interface of each group. Methods:Eighteen intact premolars were randomly divided into 3 groups(n=6):A:The specimens were immersed into artificial saliva for 3 weeks then adhesion without bleaching; B:Adhesion immediately after bleaching for 3 weeks,during bleaching the specimens were immersed into artificial saliva; C:Before adhesion the specimens immersed in artificial saliva for further 2 weeks after bleaching for 3 weeks. Five specimens from each group were randomly submitted to shear bond strength test. The specimen remained in each group were sectioned to be observed by the scanning electron microscope.Results:(1)The shear bond strength of the group B (10.74?3.03) MPa was significantly lower than that of the group A (18.61?3.56) MPa or the group C (17.21?3.15) MPa (P0.05); (2)The resin tags present in the dentin/composite interface was scarce for the group B,and that were shorter than the group A; The resin tags of the group A and group C were similar in number,but that of the group C was shorter than that of the group A. Conclusion:The use of TREE whitening gel immediately before bonding significantly reduces bond strength,but immersion of bleached teeth in artificial saliva for 2 weeks before bonding results in a return to control bond strength.
3.The changes of Na~+,K~+-ATPase activity and ? isoforms in rat neurons suffered from global ischemia

Aim To investigate the changes of Na~+,K~+-ATPase activity and ? isoforms in rat neurons suffered from global ischemia.Methods The BCAL rat was induced by bilateral carotid artery ligation.The contents of H_2O,Na~+ and K~+ in brain tissue were detected.The method of spectrophotometry was applied to measure the activities of Na~+,K~+-ATPase.With immunohistochemical assay,the expression of the Na~+,K~+-ATPase ?_1,?_2,or ?_3 isoform was detected in neurons of rat hippocampus and cortex following ischemia.Results In the BCAL rats,the contents of H_2O and Na~+ were increased while the content of K~+ and the activity of Na~+,K~+-ATPase were decreased.The ?_1,?_2 or ?_3 isoform of Na~+,K~+-ATPase distributed in a tissue-specific fashion in neurons of hippocampus and cortex,in which ?_1 and ?_3 were abundant and ?_3 was more than ?_1,but ?_2 was much less.In BCAL rat,?_1 and ?_3 isoforms were significantly decreased in hippocampus and cortex neurons.Conclusion These results suggest that the changes of Na~+,K~+-ATPase ?_1 and ?_3 isoforms may be involved in the global cerebral ischemia
4.The application of array comparative genomic hybridization for the detection of chromosomal defects in newborns

Objective To study the application of the array comparative genomic hybridization (Array-CGH) for the detection of chromosomal disorders in newborns.Method The Array-CGH technique was used to analyze the whole genome of the patients who were suspected of chromosomal disease in neonatal ward of our hospital from January to December in 2014,and further verification in genomic unbalanced ectopia was carried out by FISH (fluorescence in situ hybridization,FISH).Result Among 514 patients,104 were found carrying chromosomal abnormalities with a detection rate of 20.2%.The most common chromosomal disease is the Down syndrome syndrome (24 cases),followed by the chubby Willy and Angel syndrome(17 cases),while the Wolf-Hirschhorn syndrome in 5 cases,Williams syndrome in 5 cases and the Criduchat syndrone in 5 cases.The results of FISH were consistent with Array-CGH.Conclusion The technique of Array-CGH can be used to scan the whole genome of children with unknown disease.As a high-throughput and rapid research method,this technique has important clinical significance in the screening of chromosomal diseases.
5.A Cognition Survey of Basic Doctors on Essential Drugs in Lu-oping County, Yunnan Province

Objective To investigate the cognition of basic doctors on essential drugs in Luo County. Methods The random sampling was used to investigate the basic doctors in Luoping County, and 200 copies questionnaire were distributed. Results The questionnaire response rate was 98%. The survey results indicated that the cognition on essential drugs among basic doctors was poor, but most basic doctors have positive attitudes and behaviors in essential drugs. Conclusion It is necessary to improve the awareness of essential drugs among basic doctors in Luo County.
6.Clinical values of using platelet rich plasma with autologous fat injection in facial rejuvenation

Objective To explore the clinical value of autologous fat injection with platelet rich plasma in facial rejuvenation.Methods 35 female beauty seekers were collected in this study.They were divided into two groups:sample autologous fat group (FAT group,n=20) and combined autologous fat and platelet-rich plasma group (PRP group,n =15).The average level of injection was 34 ml.The fat was collected at the abdominal or thigh sites using 18 G needle connecting with negative pressure suction-pump.The fat was centrifugated 2 min to get rid of the lower layer of water and the upper layer of oil.Weigao PRP extract kit was used to obtain the PRP.The ratio of PRP to fat was 1:5.The injection technique was according to Colemen method.The survival rate,satisfaction and complication were compared between the 2 groups.Results There were no infection,hematoma,fat liquefaction necrosis occurred in the 2 groups.Compared with the FAT group,the skin texture and facial contour of the PRP group was improved.Conclusions The platelet rich plasma can improve the survival rate of fat and has a stable effect,which is one of the safe and ideal methods of facial rejuvenation.
7.Effects of renovascular hypertension on contractile/ diastolic function,calcium transient and calcium sensitivity in rat ventricular myocytes

Aim To observe effects of hypertension on contractile/diastolic function and calcium transient in rats ventricular myocytes. Methods The model of one-kidney-one-clip (1k1c) hypertensive rat was pre-pared by partially ligating the left renal artery and removing the right kidney. Left ventricular myocytes were enzymatically isolated. Then the contraction and calcium transient of a single cell from both normal and renovascular hypertensive rats were observed by a video-based motion edge-detection system simultaneously. Effects of calcium in various concentrations on contractile/diastolic function and calcium transient in ventricular myocytes from renovascular hypertensive rats were assessed in the same way. Results Compared with normal cardiac myocytes, the shorting amplitude and the contractile and diastolic velocity were increased significantly in 1k1c hypertensive rat cardiac myocytes. However their intracellular calcium in contractile and diastolic periods, the extent of calcium transient and the parameters of intracellular calcium dynamics were unchanged. But the extracellular calcium of different concentrations could shift the Fura-2 fluorescence ratio-cell shorting amplitude curve from hypertension rat myocytes to the left compared with that from normal rats. Conclusions The hypertension increases the contractility of rat cardiac myocytes, which is due to raising their sensitivity to calcium.
8.Correlation study on the serum levels of ProGRP and NSE with different treatments in limited disease small cell lung cancer

Objective To study the correlation of the serum levels of pro-gastrin-releasing peptide (ProGRP) and neuro-specific enolase (NSE) with different treatments in limited-disease small cell lung cancer (L-SCLC) patients.Methods 150 L-SCLC patients were randomly divided into three groups including concurrent chemo-radiotherapy group,sequential chemo-radiotherapy group,and chemotherapy group.The serum levels of ProGRP and NSE were detected by enzyme-linked immunosorbent assay (ELISA) and electrochemiluminescence immunoassay before and after different treatments.The follow-up phase was 12 months.Results The serum levels of ProGRP and NSE were significantly decreased in all 3 groups after treatment (318.96,250.77,226.18 pg/ml and 31.72,23.95,17.89 μg/L),and the lowest level was observed in concurrent chemo-radiotherapy group,then the sequential chemo-radiotherapy group and chemotherapy group.The short term therapeutic effects were in the same sequence,and there was statistical significance between concurrent chemoradiotherapy group and chemotherapy (P < 0.05).The decrease extent of ProGRP and NSE in effective cases (CR+PR) was significantly higher than that in failure cases in all 3 groups.The serum levels of ProGRP and NSE were increased with disease progress (P < 0.05).Conclusion The serum levels of ProGRP and NSE can be used to reflect the patient' s condition and evaluate therapeutic effect in L-SCLC.The concurrent chemo-radiotherapy is more effective than sequential chemo-radiotherapy and chemotherapy only in L-SCLC.
9.Effect of adding time of human milk fortifier on growth and incidence of complications of very low birth weight premature infants

Objective To investigate the effects of human milk fortifier(HMF)addition at different time points on the growth,development and the incidence of complications in very low birth weight(VLBW)infants.Methods A total of 93 VLBW infants admitted into Neonatal Intensive Care Unit of Tianjin Central Hospital of Obste-trics and Gynecology from January to September 2015 with more than 80%of total milk intake during hospitalization,excluding those who had severe asphyxia or abandoned treatment and died,were collected.The included cases were divided into 2 groups by using completely randomized grouping method,early fortification group(n=48)and delayed fortification group(n=45)adding HMF with the enteral intake of 50 mL/(kg·d)and 100 mL/(kg·d),respectively.The outcomes included growth development and the incidence of complications during hospitalization.Then,t test and chi-square test of independent samples were used for statistical analysis.Results There was significant difference in the weight growth rate between the 2 groups,and the growth rate of early fortification group and delayed fortification group were(15.4±2.4)g/(kg·d)and(13.6±2.3)g/(kg·d),respectively(t=3.043,P=0.004).There was no significant difference in height growth rate,head circumference growth rate,weight at 34 weeks postmenstrual age,time of recovering birth weight and parenteral nutrition,hospitalization duration,body weight,body length,head circumference at discharge and the incidence of extrauterine growth retardation between the 2 groups(all P>0.05).There was no statistical difference in incidence of feeding intolerance,necrotizing enterocolitis,nosocomial infection,retinopathy of prematurity,bronchopulmonary dysplasia between the 2 groups(all P>0.05).Conclusions HMF with enteral intake of 50 mL/(kg·d)contributes to weight gain rate in VLBW infants during hospitalization,but not to the increase in the incidence of complications.
10.COX-2 involved in the antitumor effect of valdecoxib on Lewis tumor

Objective To study the inhibitory effect of valdecoxib on Lewis tumor and the potential relationship with cyclooxygenase-2(COX-2).Methods HE staining was used to observe lymphocyte infiltration in tumor tissue and the cell structure of the stomach and colon.Western blotting was used to detect the expression of COX-2 in tumor tissue.PGE_2 ELISA kit was used to detect the content of PGE_2 in tumor tissue.Results ① Valdecoxib inhibited the growth of the tumor,and the survival rate was increased.②There was lymphocyte infiltration in treatment group and the content of PGE_2 was decreased.③Valdecoxib did not affect cell structure of stomach and colon,bleeding and clotting time.Conclusion Valdecoxib inhibits the growth of the Lewis tumor and enhances the survival rate.The effect of valdecoxib is related to the inhibition of COX-2.
