Objective:To study the changes of the NKT like cells after HIV infected. Methods:We collected peripheral blood from 47 untreated HIV infected individuals and 31 healthy controls,and analyzed the expression of Annexin-V,Ki-67,HLA-DR and other surface molecules in NKT like cells by flow cytometry. Results:The NKT like cell percentage of untreated HIV infected group was (3. 03±1. 61)%,the NKT like cell percentage of normal control group was (8. 30±7. 42)%,the percentage of NKT like cells in HIV infected individuals was significantly lower than the healthy controls ( P<0. 05 );the NKT like cell HLA-DR expression of untreated HIV infected group and normal control group were (5. 40±4. 10)% and (0. 89±0. 83)%,the NKT like cell Annexin-V expression of untreated HIV infected group and normal control group were (30. 21±13. 15)% and (5. 40±8. 05)% ,and the activation and apoptosis of NKT like cells was significantly higher after HIV infection compared with health individuals (P<0. 001,P<0. 01),the degree of activation was negatively correlated with CD4 count (r=-0. 885 7,P<0. 05);and the NKT like cell Ki-67 expression of untreated HIV infected group and normal control group were (11. 15±4. 76)% and (27. 63±18. 31)%,the proliferation ability was significantly lower after HIV infection compared with healthy controls(P<0. 05). Conclusion:HIV infection can significantly reduce the number of NKT like cells and its ability to proliferate,and increase its ability to activation and apoptosis.

Objective To explore serum IL-6 (interleukin-6) ,IL-10 (interleukin-10), the WBC (white blood cell count), CRP (C-reactive protein)and PCT ( procalcitonin ) level in early diagnosing of in children with hand, foot and mouth disease and bacterial infection. Methods 63 children in bacterial infection group and virus infection group respectively,collected 63 children in normal control group by physical examination. Enzyme-linked immunosorbent assay (ELISA) double antibody clamp method was used to determine serum IL-6 and IL-10 level. Immune turbidimetric , enzyme-linked fluorescent were used to detected serum CRP , PCT. WBC results were also compared. Resluts Serum IL-6, IL-10, CRP and PCT level were higher in normal control group compared with virus infection group , the WBC level was slightly lower than normal control grou with statistically different IL-6, IL-10, CRP level (P < 0.05);serum IL-6, IL-10, WBC, CRP and PCT levels in Bacteria infection group were higher than virus infection group and normal control group , while their serum WBC, CRP and PCT level were statistically significant compared to virus infection group (P < 0.05),IL-6 and IL-10 were no statistical different (P > 0.05). Conclusion IL-6, IL-10 levels increased significantly in the early stage of hand,foot and mouth disease, but in the aspect of combination of bacterial infection and bacterial infection, their clinical value was not so significant. WBC, CRP and PCT level had important value for early diagnosis in hand, foot and mouth disease with bacterial infection.

Objective:To better understand the changes of the NKT like cells after HIV infection and HAART treatment.Methods: Peripheral blood from HIV-infected individuals, HAART-treatment AIDS patients and healthy controls were collected, the expression of CD57 and the proliferation ability of NKT like cells before and after HAART were analyzed by flow cytometry.Results:We found that the percentage of NKT like cells before HAART was significantly lower than the healthy controls ( P<0.01 ) , and recovered after HAART treatment ( P<0.05 );the aging of NKT like cells was significantly higher before HAART compared with health individuals (P<0.01),and recovered after HAART treatment(P<0.05)the proliferation was significantly lower in vitro before HAART compared with healthy controls,and partial recovered after HAART.Conclusion: After HAART treatment,the number of NKT like cells,CD57 expression and the proliferation ability of HIV infected patients were restored.

Objective: To explore the values of fecal mircoRNA (miRNA, miR) in diagnosis and prognostic evaluation of colorectal cancer. Methods: Fifty colorectal cancer patients and fifty healthy controls were included in this study. The contents of fecal miR-141, miR-17-3p and miR-92a-3p in all subjects were detected by real-time fluorescent quantitative PCR before operation. The differences of the three mircoRNA levels between the two groups were analyzed. Then the diagnostic efficiency of the three miRNAs for colorectal cancer was assessed by using receiver operating characteristic (ROC) curve. Furthermore, the differences of fecal miR-141 and miR-92a-3p levels in colorectal cancer patients between preoperation and postoperation (6 months after operation) were analyzed. The recurrence of colorectal cancer patients in 12 months after operation was followed up, then the differences between postoperative and preoperative fecal miR-141 contents in patients with and without recurrence were analyzed. Results: The levels of fecal miR-141 and miR-92a-3p in colorectal cancer group were significantly higher than those in the control group (both P < 0.01). There was no significant difference in fecal miR-17-3p level between the two groups (P = 0.30). ROC curve analysis showed that the areas under the curve (AUC) of fecal miR-141 and miR-92a-3p were 0.874 [95% confidence interval (CI): 0.803-0.946] and 0.789 (95% CI : 0.703-0.876), respectively. The level of fecal miR-141 in colorectal cancer patients 6 months after operation was significantly lower than that before operation (P < 0.01), while there was no significantly difference between postoperative and preoperative fecal miR-92a-3p levels (P = 0.08). The difference between postoperative and preoperative fecal miR-141 contents in 4 patients with recurrence was significantly less than that in 46 patients without recurrence (P < 0.05). Conclusion: The fecal miR-141 and miR-92a-3p were highly expressed in patients with colorectal cancer, so that they may be used as the non-invasive biomarkers for the auxiliary diagnosis of colorectal cancer. In particular, the fecal miR-141 may have a certain value in evaluating the surgical outcomes and prognosis of colorectal cancer, as well as monitoring the postoperative recurrence.

To probe into the characteristics of traditional Chinese medicine syndromes and their element distributions in sub-health status.

Objective To compare the fatigue characteristics between people with fatigue-predominant sub-health and patients with ulcerative colitis.Methods Fatigue Self-assessment Scale (FSAS) was applied in people with fatigue-predominant sub-health and patients with ulcerative colitis.By using the multivariate statistical analysis,the fatigue characteristics of them were analyzed.Results The fatigue characteristics of two groups of people were different.The scores of physical fatigue,mental fatigue,fatigue consequences,total fatigue,reaction of fatigue after sleep and rest,situation of fatigue in people with fatigue-predominant sub-health were 31.06 ± 17.08,24.63 ± 14.50,24.38±13.39,26.36 ± 11.97,30.63 ± 22.70,42.89 ± 21.47,and in patients with ulcerative colitis were 25.60 ± 22.98,17.80 ± 18.44,20.54 ± 18.53,21.20 ± 17.15,18.18 ± 25.47,41.21 ± 25.70.There were statistical differences on the two groups (P ＜ 0.05 ).There were statistical differences between the percents of four degrees ( unconspicuous,mild,moderate,heavy) of physical fatigue,mental fatigue,fatigue consequences between in people with fatigue-predominant sub-health and in patients with ulcerative colitis (P ＜ 0.05 ).There were statistical differences on the percents of five degrees ( no obvious,slightly,less obvious,obvious,very obvious) of unrelieved fatigue after sleep and rest,situation of fatigue between in people with fatigue-predominant sub-health and in patients with ulcerative colitis (P ＜ 0.05 ).Conclusion The type,severity of fatigue,and characteristics ( such as unrelieved fatigue after sleep and rest,situation of fatigue) are different between people with fatigue-predominant sub-health and patients with ulcerative colitis.

Objective To explore the expression and diagnostic value of serum microRNA-574-3p (miR-574-3p) in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC).Methods A total of 90 patients with HBV-related HCC,40 patients with HBV-related liver cirrhosis (LC) and 50 healthy controls were recruited.The expression levels of serum miR-574-3p,α-fetoprotein (AFP) and α-Lfucosidase (AFU) of all subjects were determined.The difference of serum miR-574-3p level between groups was compared.The relation between serum miR-574-3p level of HCC patients and its clinical pathological characteristics was analyzed.The t-test was performed.The relationship between serum miR-574-3p level of HCC patients and AFP,AFU was analyzed by Spearman correlation analysis.The diagnostic efficacy of them as diagnostic markers was evaluated by receiver operating characteristic curves (ROC) and the area under the curve (AUC)(95％ CI (confidence interval)).Results The relative quantity expression of serum miR-574-3p of HCC group,LC group and healthy group was 2.152(1.654,3.061),1.292 (0.984,1.666) and 1.018 (0.750,1.726),respectively.The expression level of serum miR-574-3p of HCC group was significantly higher than those of LC group and healthy control group,and the differences were statistically significant (t=2.726 and 2.845,both P＜0.01).The expression level of serum miR-574-3p of HCC patients was significantly different in tumors with different degree of differentiation (t=2.262,P=0.039),different stage (t=2.354,P=0.025) and different HBV DNA concentrations (t=2.771,P＜0.01).There was no correlation between serum miR-574-3p level and AFP (r2 =0.076,P=0.505),AFU (r2 =0.082,P=0.422) in HCC patients.When compared HCC group with LC group,AUC of serum miR-574-3p of was 0.823 and 95％ CI was 0.750 to 0.897.When compared HCC group with healthy control group,AUC of serum miR-574-3p was 0.840 and 95％CI was 0.769 to 0.910.Conclusions The expression level of serum miR-574-3p of HCC patients is significantly higher than those of LC patients and healthy controls.Serum miR-574-3p may be an important biomarker in the diagnosis of HCC.

Objective To develop a specific trans-splicing intron ribozyme type Ⅰ-mediated dual reporter gene system (Rib53-Fluc-tk) for targeting CEA.Methods The novel CEA-targeting trans-splicing ribozyme with the downstream reporter system (Rib53-Fluc-tk) was constructed by genetic engineering technology.The trans-splicing reaction product was evaluated using the 131I-5-iodo-2'-fluro-l-beta-D-arabinofuranosy-luracil (FIAU) cellular uptake rates and the bioluminescence.Two-sample t test,the analysis of variance and the least significant difference (LSD) t test was performed for data analysis.Results The sequence of Rib53-Fluc-tk was proved by gene-sequencing test.Human MCF-7 breast cancer cells showed a high ratio of firefly luciferase/renilla luciferase (0.64±0.10,n =4).A 520 bp band of product existed,which matched with the predicted size using RNA from cells transfected with Rib53-Fluc-tk in MCF-7.Signals were detected by bioluminescence in human embryonic kidney 293T cells co-transfected with Rib53-Fluc-tk and pCDNA3.1-CEA.The labelling rate of 131I-FIAU was (64.02±4.79)％ (n =3).The radiochemical purity was (95.96± 1.07)％ (n=3),and the stability of the radiocompound remained high in human serum at least for 24 h.The uptake of 131I-FIAU in 293T cells transfected with Rib53-Fluc-tk was (0.31±0.01)％ (n=4),while it increased with the incubation time in 293T cells co-transfected with pCDNA3.1-CEA and Rib53-Fluc-tk and reached (1.40±0.06)％ at 4.5 h (F=1 007.29,t=136.34,both P＜0.01).Conelusions A novel and specific reporter gene in the cellular level was established.Taking advantage of trans-splicing reaction of the ribozyme,it could improve the specificity of the reporter gene imaging.

Objective:To systematically review parents ′ experience of caring for children with chronic kidney disease (CKD) to fully understand care needs and improve the psychological state and caregiving quality of parents. Methods:The qualitative studies on parents ′ experience of caring for children with CKD were retrieved from following databases, including PubMed, Web of Science, CNKI, VIP, CBM, and WanFang Data from inception to March 2020. The quality of included researches was evaluated according to the JBI Critical Appraisal Tool for qualitative studies in Australia. The results were integrated by pooled integration methods. Results:A total of 14 studies were included. 69 results were summarized and integrated to form 10 categories. These categories extracted 4 integrated results: parents' physical and mental condition are affected, relationship between parents and their support system has changed, reconstruction of parents ′ life, unmet care needs and problems. Conclusions:Parents are crucial to the disease management of children with CKD, so clinical medical staff should not only provide medical services for children, but also pay more attention to the psychological status and needs of parents, so as to provide guidance and support to promote parents to better implement care and disease management for children.
