1.Expression of miR-134 in breast cancer and its relationship with epithelial-mesenchymal tran-sition

Purpose To investigate the expression of miR-134 in breast carcinoma and its c1inica1 significance,and to ana1yze the as-sociation between its expression with bio-markers of epithe1ia1-mesenchyma1 transition( EMT). Methods The expression of miR-134 in 97 breast cancer samp1es was detected by in situ hybridization and the expression of E-cadherin,N-cadherin,and vimentin was de-tected by MaxVision two-step method of immunohistochemistry,to ana1yze the re1ationship between their expression and c1inicopatho1og-ica1 characteristics. Results The expression of miR-134 in breast cancer was negative1y associated with higher histo1ogica1 grade, HER-2 overexpression,1ow expression of E-cadherin as we11 as high expression of N-cadherin and vimentin. Both 1ow-expression of E-cadherin and up-expression of N-cadherin and vimentin in breast cancer were positive1y corre1ated with higher histo1ogica1 grade,1ymph node invo1vement and HER-2 overexpression,whi1e negative1y associated with the expression of ER and PR. Conclusion The expres-sion of miR-134 in breast cancer tissue is negative1y re1ated to both the ma1ignant progression and EMT,indicating that it might be a potentia1 bio-marker for breast cancer.
2.The relationship between resilience and cognitive function of older adults

Objective To investigate the role of resilience on cognitive function in older adults.MethodsBy random sampling,a total of 321 older adults participated the present study and were investigated by Connor-Davidson Resilience Scale,Mini Mental State Examination and self-designed general questionnaire.ResultsThe score of resilience in older adults (82.75±15.13) was significantly higher than that of Chinese community people (65.40±13.90) (t=20.68,P<0.01).Older adults with poor resilience performed significantly lower on all dimensions of cognitive abilities than the other two groups (middle & higher resilience group) (all P<0.05).With both age and educational level controlled,tenacity and strength were of positive prediction for the cognitive abilities of older adults (β=0.25,0.17,P<0.05).ConclusionThere is a close relationship between resilience and cognitive function of older adults.The training of tenacity,strength and optimism will help delay the decline of cognitive function.

Abtract: Acute coronary syndrome is a common clinical and frequently occurring disease, belonging to the chest discomfort, heartache, and true heart pain of TCM category. The clinical observation shows that the heat and blood stasis, and poison damage heart nutrient is one of the important mechanisms of triggering coronary syndrome. Therefore, the method of promoting blood circulation to remove meridian obstruction and the Qing Ying detoxication was established as the basic treatment, and Danshen Tongluo Jiedu Decoction is applied as the main formula. According to the different types of disease and syndrome differentiation, flexible modification can achieve good efficacy. It is expected that these will provide new ideas and methods for clinical treatment of acute coronary syndrome.
4.Establishment of Ethical Assessment System in the Admittance of the Limitative Medical Technologies

Objective to establish an ethical assessment system in the admittance of the limitative medical technologies.Method:literature review and Delphi method are employed to adjust and set the structure,index numbers,and index weights of ethical assessment system of the limitative medical technologies.Result:An ethical assessment system in the admittance of the limitative medical technologies was established,which included 3 primary indices,11 secondary indices and 35 tertiary indices.There were 4 qualitative levels with different weights in tertiary indices,based on which the global grade and proposed-admittance criterion were obtained.Conclusion:Establishment of ethical assessment system in the admittance of the limitative medical technologies provides an ethical evidence for the assessment of clinical admittance of the limitative medical technologies with qualitative and quantitative approaches and class-setting evidence.
5.Effects of Intervention of Soothing Liver and Activating Blood Chinese Medicine on Cardiac Function and Myocardial Pathologic Morphology of BMSCs Transplanting Myocardial Ischemia Reperfusion Injury Rats

Objective To observe the effects of intervention of soothing liver and activating blood Chinese medicine on cardiac function and myocardial pathologic morphology of BMSCs transplanting on myocardial IRI of rats, and investigate its myocardial protection mechanism. Methods Model of myocardial IRI was established by coronary artery ligation in rats. SD rats were randomly divided into sham operation group, IRI group, BMSCs group and combined group. Rats in combined group were filled the stomach with soothing liver and activating blood Chinese medicine, and rats in other groups were filled the stomach with the same dose of normal saline. After 4 weeks, myocardial pathologic morphology was observed with light microscope. Cardiac function was detected with ultrasonic cardiogram.Results Compared with BMSCs group, heart function of the combined group improved, with significant statistical difference (P<0.05,P<0.01). Pathological observation showed that myocardial structure and pathological morphology were obviously promoted in the combined group.Conclusion Soothing liver and activating blood Chinese medicine could improve heart function and myocardial pathological morphology of IRI rats with BMSCs transplantation.
6.The correlation study of mandibular second molar root growth and bone age in male teenagers

Objective To explore the relationship between cervical bone age and male juvenile mandibular second molar (Msm) root growth ,and to provide evidence for the development of bone and teeth in patients with orthodontic treatment . Methods The CBCT and lateral cephalometric radiographs of 114 cases of 9 .5 to 15 .5 years old children in Chongqing city and ad‐olescents cervical vertebral bone and MSM root length were measured ,using statistical analysis to explore the relationship between cervical vertebral bone ,tooth root length change of age and Msm .Results By Pearson correlation analysis ,Msm root length chan‐ges and cervical vertebral bone were positively correlated(r= 0 .737 ,P< 0 .01) .Msm root length changes showed a positive correla‐tion with age(r= 0 .681 ,P< 0 .01) .Msm root length changes was positively related with dental age(r = 0 .795 ,P< 0 .01) .Conclu‐sion The root growth of the mandibular second molar was highly correlated with cervical vertebral bone in male children .When the growth and development of patients were judged by orthodontic treatment ,we can adopt the mandibular second molar root length forecast growth level of male adolescents .
7.Effects ofDanshen Tongluo Detoxication Decoction on Inflammatory Response of Rats with Bone Marrow Stem Cell Transplantation in Acute Myocardial Infarction

Objective To explore the effects ofDanshen Tongluo Detoxication Decoction on the inflammatory response of rats with bone marrow stem cell (BMSC) transplantation in acute myocardial infarction (AMI); To discuss its mechanism of action.Methods The whole bone marrow adherent method was adopted for BMSCs separation and culture. The AMI model was established by closing the left anterior descending coronary artery of SD rats. After the modeling, SD rats were randomly divided into sham-operation group, model group and BMSCs group (BMSCs transplantation group),Danshen Tongluo Detoxication Decoction group, Danshen Tongluo Detoxication Decoction + BMSCs group, 10 rats in each group. BMSCs cell suspension was injected directly into the edge of the myocardial tissue infarction area; Chinese medicine or normal saline were administered for gavage. 4 weeks later, the contents of MCP-1 and sICAM-1 in each group were detected by ELISA. TLR-4 expression was measured by Western blot method, and HE staining was used to observe the myocardial tissue pathological changes.Results Compared with the model group, the contents of MCP-1 and sICAM-1 and the protein expression of TLR-4 in BMSCs group,Danshen Tongluo Detoxication Decoction group, andDanshen Tongluo Detoxication Decoction + BMSCs group decreased, Danshen Tongluo Detoxication Decoction group was better than BMSCs group (P<0.05,P<0.01), andDanshen Tongluo Detoxication Decoction + BMSCs group was better than BMSCs group andDanshen Tongluo Detoxication Decoction group(P<0.05,P<0.01).Conclusion BMSCs transplantation combined withDanshen Tongluo Detoxication Decoction can restrain the inflammatory response of AMI model rats and repair ischemic myocardium issue, which mechanism may be related to regulating TLR-4 induced inflammatory response.
8.Association of NF-κB and its downstream pathway with acute radiation-induced myocardial fibrosis in rats

Objective To examine the pathological changes in the myocardial tissues such as inflammatory response and fibrosis in a rat model of acute radiation-induced heart damage (RIHD),and to explore whether NF-κB and its downstream pathway are associated with acute radiation-induced myocardial fibrosis.Methods Fourteen nale adult Sprague-Dawley rats were randomly divided into control group and radiation group.Local heart irradiation was delivered to the precordial region of rats to establish an RIHD model in a single fraction with a dose of 20 Gy generated by a 6 MV linear accelerator.At 14 days after irradiation,the histopathological changes in myocardial and interstitial tissues were examined by HE staining;the distribution of collagen fibers was observed by Masson staining,and collagen volume fraction (CVF) was used as a semi-quantitative evaluation for myocardial collagen deposition,which was defined as the percentage of collagen area occupied in total area,and was compared using the independent-samples t test.The protein and mRNA expression levels of the NF-κB members p50 and p65 and the downstream pathway members hypoxia-inducible factor 1α(HIF-1o),connective tissue growth factor (CTGF),and type I (COL-1) were quantitatively analyzed by Western blot and qPCR,respectively.Results At 14 days after local heart irradiation,the radiation group showed significant myocardial edema and derangement,rupturc of some myocardial ceils,mild nuclear pyknosis,darkened nuclear staining,a small number of irregular nuclei,and myocardial interstitial inflammatory cell infiltration accompanied by increased fibroblast,as compared with the control group.The Masson staining showed that the collagen fibers in radiation group were widely distributed at the interstitial tissue and increased significantly compared with those in the control group;normal myocardial cells were in disordered array and had a tendency to be replaced by collagen fibers.The semi-quantitative analysis showed that radiation induced a significant increase in CVF (22.05% vs.3.76%,P =0.003).Western blot and qPCR revealed that the protein and mRNA expression of p50,p65,HIF-1 α,CTGF,and COL-1 was significantly higher in the radiation group than in the control group (all P < 0.05).Conclusions The pathological features of acute RIHD include significant myocardial edema and myocardial interstitial inflammatory cell infiltration accompanied by increased fibroblasts and collagen fibers.Radiation exposure can activate NF-κB and cause the upregulation of HIF-1α and CTGF at both protein and mRNA levels,which may play an important role in the progression of radiation-induced myocardial inflammation to fibrosis.
9.The expression and significance of smac, XIAP, caspase-3 in nonnasal inverted papilloma.

OBJECTIVE:
To explore the expression and significance of second mitochondria derived activator of caspase (Smac), X-linked inhibitor of apoptosis protein (XIAP)and cysteine containing aspartate specific protease 3 (caspase-3) in the growth, development and carcinogenesis of the nonnasal inverted papilloma (NIP).
METHOD:
Immunohistochemical method was used to detect the expression of Smac, XIAP, caspase-3 in 10 cases of nasal cavity mucosae (NM) and 45 cases of NIP, the group of NIP including 25 cases of NIP without dysplasia, 11 cases of NIP with dysplasia, and 9 cases of NIP with malignant transformation to squamous cell carcinoma (SCC).
RESULT:
The intensity of the positive expression of Smac, Caspase-3 in NIP were lower than NM, the intensity of the positive expression decreased with the decreasing degree of histological differentiation. There was a significant difference between NIP without dysplasia and SCC. It was presented with a progressive tendency for the expression of XIAP in the group of NM and NIP. The lower degree of histological differentiation, the higher intensity of the positive expression. The expression between NIP without dysplasia and SCC had a significant difference. Smac negatively correlated with XIAP (r(s) = -0.323, P < 0.05), XIAP negatively correlated with caspase-3 (r(s) = -0.408, P < 0.01), Smac positively correlated with caspase 3 (r(s) = 0.424, P < 0.01).
CONCLUSION
Smac, XIAP, caspase 3 might be associated with the growth and carcinogenesis of NIP.

10.Rapid and high throughput measurement of lipase thermo-stability through ANS fluorescence signal assay.

We have developed a rapid and high throughput lipase-ANS (8-Anilino-l-naphthalenesulfonic acid) assay to evaluate the thermo-stability of lipases based on the ANS fluorescence signal's increasing and shifting when this small fluorescence probes binds to lipase. The testing lipase samples were incubated at a temperature range of 25 degrees C to 65 degrees C for 30 min before mixed with ANS solution (0.20 mg/mL lipase and 0.05 mmol/L ANS in the buffer of 20 mmol/L Tris-HCl, 100 mmol/L NaCl, pH 7.2) in a cuvette or microplate. Fluorescence signals of the samples were measured at EX 378 nm, EM 465 nm with a fluorescence photometer or a plate reader, and Tm was calculated with the software of GraphPad Prism5.0. The Tm values of several mutants of Penicillium expansum lipase (PEL) were measured with this ANS assay and conventional method simultaneously and the results show that Tm values are comparative and consistent between these methods, suggesting that the lipase-ANS assay is a reliable, rapid and high throughput method for lipase thermo-stability measurement.
