1.Quantitative study on loss of dopaminergic neurons in SNpc of C57/BL6 mice induced by MPTP

Objective To observe the distribution of dopaminergic neurons in SNpc and to establish standard curve in normal mice so as to measure the changes of dopaminergic neurons in number in SNpc of the mice toxicated with MPTP. Methods Ten male C57BL6 mice aged 8-12 weeks, weight 20-22 g, were randomized to receive 20 mg/kg MPTP or physical saline every 3 h for 4 times, then killed 7 d later. The mouse mesocerebrum was taken out and fixed, frozen, sectioned. All sections containing SNpc were observed under the guide of mouse brain atlas. Every other sections were chosen to stain tyrosine hydroxylase (TH) to show dopaminergic neurons immunohistochemically. The TH positive cells in SNpc were counted in each section and the standard curve was established. Results The standard curve of SNpc compact part position and TH positive cells was established. By comparing the standard curves for the MPTP intoxicated mice and the saline mice, TH positive cells in SNpc from MPTP toxicated mice decreased significantly, which confirmed the validity and feasibility of the standard curve. Conclusion The establishment of standard curve greatly facilitates the comparison of specimen from different groups and makes the assessment of dopaminergic neuron loss more accurate and efficient. The standard curve can serve as an excellent reference curve for the assessment of dopaminergic neurons in SNpc in normal C57BL6 mice.
2.Serum epidemiological analysis of community patients with hepatitis B

Objective To discuss and analyze the serum epidemiological characteristics of hepatitis B among people in Jinan in order to formulate applicable community nursing measures to control hepatitis B. Methods The serum marker of hepatitis B was tested by enzyme-linked immunosorbent assay in 26756 serum samples by strict operation according to the kit introduction. The data were analyzed by SPSS 13.0 software. Results The positive rate of HBsAg was 7.39%, among which the positive rate of male was 8.81% and female 5.54%, which was lower than that of the male (P<0.01). The positive rate of people lower than 20 years old was lower than those of above 20 years old. The positive rate of male from 21 to 30 years old was lower than those from 41 to 50 years old (P<0.05). No difference was seen between people with different professionals. Conclusions Vaccine inoculation of hepatitis B for susceptible population combined with community health education about hepatitis B related knowledge and comprehensive nursing intervention proved to be the pivotal measures for the control and prevention of hepatitis B.
3.Construction and Practice of Teaching Evaluation System of PBL Mode in Clinical Pharmacology

OBJECTIVE:To establish an overall teaching evaluation system for clinical pharmacology with PBL(problem-based learning) mode. METHODS:The teaching evaluation system was established by“Delphi”method,and it has been applied to 270 students at China Medical University in 2013-2014. RESULTS:According to the result of test score,PBL method can improve abil-ity of analyzing medical records,while formative evaluation indicates most of the students are satisfied with class response.And PBL method also can enhance student’s skill of clinical practice,broaden the knowledge an so on. CONCLUSIONS:The evaluation sys-tem was comprehensive with contents,systematic and objective,and it also can provide an effective feedback in teaching,which can accelerate the optimization of the teaching mode and improve the quality of education.
4.Relationship between expression of Syk,VEGF-C and lymph node metastasis in breast carcinoma

Purpose To investigate the relationship between the expression of Syk,VEGF-C and lymph node metastasis in breast carcinoma.Methods Immunohistochemical EnVision and SP methods were used to detect the expression of Syk,NFκB(p65)and VEGF-C in 55 cases of breast carcinoma.Results The positive rates of Syk, VEGF-C and NFκB(p65)in 55 cases of breast carcinoma were 50.9%,56.4% and 81.8%,respectively.Lower positive rate of Syk was obtained in the group of positive lymph-node metastasis than that in the group of non-lymph-node metastasis (P<0.05);Higher positive rate of VEGF-C was obtained in the group of positive lymph-node metastasis than that in the group of non-lymph-node metastasis (P<0.05);But no significant differences was found in p65(P>0.75).Higher positive rate of p65 nuclear expression was obtained in the group of positive lymph-node metastasis than that in the group of non-lymph-node metastasis (P<0.025).The expression of Syk was negatively associated with VEGF-C (r=-0.620,P=0.000);the nuclear expression of p65 was associated with the low expression of Syk(r=0.448,P=0.002)and the high expression of VEGF-C (r=0.310,P=0.036).Conclusions In breast carcinoma, Syk may downregulate the expression of VEGF-C by inhibiting the activation of NFκB, which suppresses the lymph node metastasis of the cancer.
5.Protective effects of sulforaphane on the oxidative damage of kidney mitochondria complex in obese rats induced by high-fat diet.

OBJECTIVETo realize the oxidative damage of kidney mitochondrial complex in obese rats induced by high-fat diet and investigate the protective effects of sulforaphane against the damage.
METHODSEighty-eight adult male SD rats were used, after 1 week adaptability feeding, 8 rats were selected as control group and given low-fat diet. The other 80 rats were given high-fat diet. After 2 weeks, the 32 diet-induced obesity models were choosen whose weight gain was higher than 40%. The 32 rats were randomly divided into 4 groups, i.e. high fat group, high fat+sulforaphane low dose group, high fat+sulforaphane middle dose group and high fat+sulforaphane high dose group. The rats in the sulforaphane low, middle and high dose groups were orally administered with sulforaphane 5, 10 and 20 mg/kg, all the 4 groups were kept feeding high-fat diet for 5 weeks. All rats were sacrificed and their kidneys were removed to assay the index of oxidative damages.
RESULTSThe content of ROS (0.26 ± 0.04) and MDA((0.87 ± 0.05) U/mg) in the hight-fat group were significantly higher than those in the control group((0.20 ± 0.02),(0.57 ± 0.08) U/mg)(t values were -3.02 and -4.72, P < 0.05). The activity of T-AOC((0.43 ± 0.04) U/mg) and MMP (12.09 ± 1.56) were lower than the control group ((0.48 ± 0.04 U/mg, (16.08 ± 3.12) )(t values were 2.06 and 2.28, P < 0.05). Gavage intervention with sulforaphane, the MDA amount ((0.67 ± 0.05), (0.55 ± 0.05), (0.56 ± 0.07) U/mg) in the sulforaphane low, middle and high dose groups were lower than the hight-fat group ((0.87 ± 0.05) U/mg (t values were 3.65, 5.71 and 5.60. P < 0.05). The activity of T-AOC ((0.49 ± 0.05), (0.55 ± 0.05), (0.54 ± 0.04) U/mg), T-SOD ((61.07 ± 2.79), (55.95 ± 2.39), (60.26 ± 6.02) U/mg) and the level of MMP ((17.17 ± 2.52), (18.24 ± 2.54), (18.21 ± 3.65)) were higher than in the high-fat group ((0.43 ± 0.04) U/mg,(47.22 ± 2.43) U/mg,(12.09 ± 1.56)) (tT-AOC values were -2.36, -4.83 and -4.30; tT-SOD values were -6.37, -4.71 and -5.99; tMMP values were -2.90, -3.52 and -3.50, P < 0.05). The activity of GSH-Px in the sulforaphane low and middle dose groups ((69.12 ± 8.63), (64.43 ± 6.58) U/mg) were higher than those in the high-fat group((53.03 ± 5.70) U/mg)(t values were -3.82 and -2.71, P < 0.05). But there were no significant difference between the high dose group ((60.02 ± 7.05) U/mg) and the high-fat group (t = -1.66, P > 0.05).
CONCLUSIONHigh-fat diet can induce the mitochondrial oxidative dysfunction in kidney, and sulforaphane shows protective effect on the kidney mitochondrial complex from oxidative damage in obese rats induced by high-fat diet.

6.Analysis of influencing factors of cardiac rehabilitation information needs of patients after PCI

Objective:To investigate the information demand level and its influencing factors of cardiac rehabilitation in patients after PCI, in order to provide scientific basis for doctors and nurses to formulate cardiac rehabilitation intervention strategies.Methods:A total of 146 patients after PCI were investigated with general data questionnaire and Cardiac Rehabilitation Inventory in the First Affiliated Hospital of Dalian Medical University from December 2019 to February 2020, and multiple linear regression was used to analyze the influencing factors of cardiac rehabilitation information demand.Results:The average scores of each dimension of Cardiac Rehabilitation Inventory after PCI were autonomy, process anxiety and result anxiety from high to low. Multiple linear regression showed that occupational type was a significant predictor of autonomy of patients after PCI, which could explain 4.6% of the variation of autonomy, and autonomy of in-service patients after PCI was higher than that of retirees ( t=2.81, P<0.05). Sex and age were significant predictors of process anxiety of patients after PCI, which could explain the variation of 8.6% of process anxiety. The occupational type, the medical insurance type and whether they had received professional rehabilitation guidance during hospitalization were significant predictors of anxiety after PCI, which could explain the 15.2% variation of anxiety, and the anxiety of in-service patients after PCI was lower than that of retirees ( t=-3.76, P<0.05). Conclusions:The level of cardiac rehabilitation information needs of patients after PCI is worthy of attention. Medical staff can give targeted, personalized and different forms of cardiac rehabilitation in the process of cardiac rehabilitation, and emphasize the implementation of health education and guidance for their cardiac rehabilitation in order to meet their cardiac rehabilitation information needs, improve their autonomy and alleviate their anxiety.
7.Meta-analysis of the diagnostic value of the Simple Five item Scoring Scale for Sarcopenia in elderly patients with sarcopenia

Objective:To evaluate the diagnostic accuracy and clinical applicability of the Simple Five item Scoring Scale for Sarcopenia (SARC-F) by systematic review and Meta-analysis.Methods:PubMed, The Cochrane Library, Wed of science, EMBASE, MEDLINE, Ovid, China HowNet database, China biomedical data, VIP database and Wanfang database were searched by computer, and all the researches related to SARC-F questionnaire's diagnosis of sarcopenia from the establishment of the database to March 1, 2020 were collected. Two researchers independently selected literature, extracted data, and evaluated the bias risk of the study. Revman 5.3 software, Stata16.0software were used to comprehensively evaluate the diagnostic value of SARC-F questionnaire for sarcopenia.Results:A total of 13 studies involving 8 860 patients were included. The results of meta-analysis showed that the sensitivity and specificity of the diagnostic criteria established by the European Working Group of Sarcopenia, the Asian Working Group for Sarcopenia and the International Working Group for Sarcopenia were used as the gold standard. The area under the receiver operating characteristic curve was 0.71, 0.69 and 0.67 respectively, and the Q-index was 0.659 3, 0.650 9 and 0.631 6.Conclusion:SARC-F questionnaire has a certain value in the screening and diagnosis of sarcopenia. In the future research, it is suggested to add the simple measurement of muscle quality on the basis of this scale to improve the accuracy of the questionnaire screening
8.Study on early integrated Chinese and western medicine cluster intervention strategy in preventing intensive care unit acquired weakness

Objective:To investigate the effect of early integrated Chinese and western medicine cluster intervention strategy on the prevention of intensive care unit acquired weakness(ICUAW).Methods:A total of 70 patients admitted to the ICU were divided into the intervention group and the control group by random digits table method with 35 cases each. The control group was given limb pressure treatment and limb rehabilitation exercise according to ICU nursing routine, and the intervention group was given early integrated Chinese and western medicine cluster intervention based on the control group, after intervention, the incidence of ICUAW, muscle strength, Barthel index score, mechanical ventilation time, length of stay in ICU and total length of stay between the two groups were compared.Results:The Medical Research Council score of the intervention group and control group were respectively (53.23±5.94) and (43.94±12.58), the Barthel index score were (79.57±7.80) and (66.14±19.25), the incidence of ICUAW were 17%(6/35) and 46%(16/35), the mechanical ventilation time were (162.26±72.27) and (254.06±126.22) h, the length of stay in ICU were (12.20±4.98) and (15.57±6.04) d, the total length of stay were (18.83±6.76) and (23.09±7.88) d, and the differences were statistically significant ( t values were -3.82-3.69, χ2 value was 6.63, P<0.05 or 0.01) . Conclusions:Early integrated Chinese and western medicine cluster intervention strategycan reduce the incidence of ICUAW, improve the muscle strength and daily life activities of patients, shorten the mechanical ventilation time, ICU length of stay and total length of stay in critically ill patients.
9.Relationship of CD62E+microparticles level with degree of severity, prognosis and vascular risk factors in patients with acute cerebral infarction

Objective To explore the level of CD62E+ endothelial microparticles in acute cerebral infarction patients and its relationship with vascular risk factors,degree of severity and prognosis.Methods Seventy patients with acute cerebral infarction and 70 non-cerebral infarction patients (controls),admitted to our hospital from January 2013 to September 2013,were selected in our study; U.S.national institutes of health stroke scale (NIHSS) was performed to evaluate the degree of severity; Barthel index (BI) was used to evaluate the prognosis; the levels of blood lipids,blood glucose,uric acid,fibrinogen (FIB),homocysteine (Hcy),C-reactive protein (CRP) and lipoprotein-associated phospholipase A2 (Lp-PLA2) were routinely measured; CD62E+ levels were detected by flow cytometry,and the relationships of CD62E+ level with vascular risk factors,degree of disease and prognosis were analyzed.Results The blood CD62E+ level of patients with acute cerebral infarction ([1.55 ±0.67]microparticle/μL) was significantly higher than that in the control group ([1.01 ±0.66] microparticle/μL,P<0.05); CD62E+ level in patients with NIHSS scores>5 ([2.03±0.61] microparticle/μL) was significantly higher than that in patients with NIHSS scores ≤ 5 ([1.25±0.37] microparticle/μL,P<0.05).Spearman correlation analysis showed that CD62E+ level was significantly correlated with the stroke severity (NHISS scores,r=0.537,P=0.000) and lipoprotein-associated phospholipase A2 (Lp-PLA2) level (r=0.327,P=0.006),and negatively correlated with prognosis indicator (r=-0.634,P=0.000); other factors showed no significant correlation.Conclusion CD62E+ microparticle can reflect the severity and prognosis of acute brain infarction,and it may be an important biological marker for clinical application.Besides,the correlation between Lp-PLA2 and CD62E+ means that the level ofCD62E+ micro particles is associated with the stability of athemsclerotic plaques.
10.Hippocampal corticotropin releasing hormone mRNA expression and neuroprotective mechanism of adrenocorticotropic hormore in immature rats after N-methyl-D-aspartate induced spasm seizures

Objective To study the hippocampal corticotropin releasing hormone (CRH) mRNA expression and neuroprotective mechanism of adrenocorticotropic hormore (ACTH) in immature rats after N-methyl-D-aspartate (NMDA)-induced spasm seizures.Methods Sixty 10-day-old Wistar rats were randomly divided into blank control group,NMDA-induced seizure group and ACTH treatment group (n=20).Rats in the blank control group did not give any treatment;rat models of infantile spasm in the NMDA-induced seizure group and ACTH treatment group were induced by intraperitoneal injection of NMDA (7 mg/kg) for a consecutive 7 d;3 h after NMDA injection,intraperitoneal injection of ACTH 0.5 mg/(kg· d) was performed in the rats of ACTH treatment group,and NMDA-induced seizure group was given an equal volume of saline.By in situ hybridization (ISH),the mean optical density of CRH mRNA-positive neurons in the hippocampus was measured.Results In the ACTH treatment group,the latencies of epileptic seizures one week after treatment were significantly prolonged as compared with those before treatment,and the scores of epileptic seizures one week after treatment were significantly decreased as compared with those before treatment (P<0.05);the latencies of epileptic seizures were significantly prolonged and the scores of epileptic seizures were significantly decreased in the ACTH treatment group as compared with those in the NMDA-induced seizure group (P<0.05).The CRH mRNA expression in NMDA-induced seizure group was significantly increased as compared with that in the blank control group (P<0.05),and the CRH mRNA expression in the ACTH treatment group was significantly decreased as compared with that in the NMDA-induced seizure group (P<0.05).Conclusion Systemic ACTH has neuroprotective effect via down-regulating the hippocampal CRH mRNA expression.
