1.Clinical significance of changes of coagulation, fibrinolytic function and platelet parameters before and after chemotherapy in lung cancer patients

Objective To discuss the clinical significance of changes of coagulation,fibrinolytic function and platelet parameters before and after chemotherapy in lung cancer patients.Methods Sixty-five cases patients with lung cancer were selected as research object and 65 health persons were selected as the control group.The prothrombin time (PT),thrombin time (TT),activated partial clotting enzyme live time (APTT),fibrinogen (FIB),D-dimer (D-D),the original plasma protein C (PC),fibrinolytic enzyme activator inhibitor-Ⅰ (PAI-Ⅰ),the platelet count (PLT),platelet deposited (PCT),platelet distribution width (PDW) and mean platelet volume(MPV) of the control group,lung cancer patients before and after chemotherapy were tested and compared.Results The levels of PT,TT,APTT,PC,FIB,D-D,PAI-T,PLT,PCT,MPV and PDW of the control group,lung cancer group of before and after chemotherapy were (11.24±0.36) s,(11.56±0.64) sand (10.42±0.64) s,(13.52±1.23) s,(13.56±1.04) sand (12.43±1.26) s,(29.74±2.53) s,(30.05 ± ±2.31) sand (28.53±2.25) s,(4.52±0.52) mg/L,(3.67±0.42) mg/L and (3.25±0.35) mg/L,(3.24±0.53) g/L,(4.26±0.63) g/L and (3.72± 0.25) g/L,(143.53± 34.73) μg/L,(745.63 ± 253.543) μg/L and (435.67±164.73) μg/L,(26.53±7.35) μg/L,(46.63±12.65) μg/L and (85.67±32.63) μg/L,(221.74± ± 53.62) × 109/L,(284.24± 58.25) × 109/L and (204.73 ± 47.1) × 109/L,(0.22 ± 0.06) %,(0.26 ± 0.12) % and (0.21±0.04)%,(10.03±0.64)fl,(10.64±1.052) fl and (9.64±0.84) fl,(15.59±1.53) fl,(16.15±2.53) fland (15.22±2.18) fl respectively.There were no statistically significant difference about PT,TT,APTT and PDW between lung cancer group of before chemotherapy and control group (t =-0.773,-0.142,-0.241 and-0.844;P>0.05).The PC level of lung cancer group of before chemotherapy was obviously lowerthan that of control group(t=5.354,P<0.05),and levels of FIB,D-D,PAI-T,PLT,PCT and MPV obviouslyhigher than that of control group(t=-4.246,-7.563,-6.533,-5.243,-4.956 and-5.354;P<0.05).After chemotherapy,the levels of PT,TI,APTI,FIB,PC,D-D,PLT,PCT and MPV of lung cancer group decreased obviously while the level of PAI-T was obviously increased,the differences were statistically significant (t =4.366,5.104,4.963,5.453,6.452,7.343,5.452,6.043,5.452 and-5.934;P<0.05).Conclusion The coagulation,fibrinolytic function and platelet of patients with lung cancer and platelet function obvious anomalies and show a high condensation and secondary fibrinolytic state of hyperthyroidism.Chemotherapy can increase the disfunction of coagulation.
2.Influence of Retn gene expression on glucose uptake by 3T3-L1 cells and the related mechanism

Objective:To investigate the influence of high expression of Retn gene on glucose uptake by 3T3-L1 cells and to study its mechanism in inducing insulin resistance.Methods:(1)Radioimmunoassay was used to determine the glucose uptake by 3T3-L1 cells with low-,normal-and high-level Retn expression under basal and insulin-stimulated states.(2)RT-PCR and real-time RT-PCR were used to determine the mRNA levels of several glucose transport proteins in 3T3-L1 cells with different expression of Retn,including insulin receptor substrate-1(IRS-1),phosphatidylinositol 3-kinase(PI-3K),AKT-2,glucose transporter-4(GLUT-4),p38 mitogen-activated protein kinase(p38MAPK),and glycogen synthase kinase-3?(GSK-3?).(Results:) The uptake of glucose decreased with the increase of Retn expression under basal and insulin-stimulated conditions.The mRNA expression of 2 signal protein PI-3K and AKT-2 decreased with the increase of Retn expression;and the expression of GSK-3? and p38MAPK increased with the increase of Retn expression.Conclusion: Resistin protein can induce insulin resistance in adipocytes,which might be related to the expression changes of some proteins in PI-3K and Ras pathways.
3.Relationship of Vitamin D and preeclampsia

Objective To study the relationship of Vitamin D and preeclampsia.Methods Four hundred and forty-two pregnant women were enrolled in this study,they were divided into two groups,420women without preeclampsia (control group) and 22 women with preeclampsia (preeclampsia group).Blood serum was collected during the 10th-14th and 24th-28th week of gestation,25-hydroxyvitamin D [25(OH) D]was detected.The level of 25 (OH) D was compared between two groups and Logistic regression analysis was used to investigate the association of 25 (OH) D and preeclampsia.Results The level of 25 (OH) D at 24th-28th week in control group [ (56.3 ± 18.8) nmol/L] was significantly higher than that in preeclampsia group [(46.2 ± 17.4) nmol/L] (P<0.01).The level of 25 (OH) D had significant relationship with preeclampsia at 24th-28th week (OR =0.67,P =0.01),regression coefficient was -0.3910,Wald was 12.3174.Conclusion The level of 25 (OH) D is associated with preeclampsia at 24th-28th week of gestation.
4.Comparative Study of CT and X-ray Plain Film in Diagnosis of Giant Pulmonary Bullae

Objective To investigate the disgnostic value of thoracic CT and X-ray plain film in giant bulla and its differential diagnosis with pneumothorax.Methods 6 patients with surgical proved giant pulmonary bulla were analyzed retrospectively,all patient underwent chest radiography and CT scan.Results The lesions occupied 30%~80% of thoracic cavity,4 cases lied left thoracic cavity and 2 cases lied right thoracic cavity.3 cases were misdiagnosed as pneumothorax by X-ray plain film.CT could show the wall and internal condition of bulla as well as compression of the normal pulmonary tissue.Conclusion Giant pulmonary bulla is easily misdiagnosed,thoracic CT is of significant value in diagnosis of giant bulla.
5.The application of case management model on the stable patients with chronic obstructive pulmonary disease

Objective To evaluate the effect of case management model on the stable patients with chronic obstructive pulmonary disease (COPD),containing health knowledge level,the number of acute exacerbation and the quality of life.Methods Fifty patients were divided into the experimental group and the control group with 25 cases each by random digits table method.The experimental group received the case management model for 3 months.The control group received routine care for 3 months.The COPD health Knowledge Questionnaire(CKQ) scores and St George's Respiratory Questionnaire(SGRQ) scores were measured for 4 times (the beginning of the intervention and 1 months,2 months,3 months later),and the numbers of acute exacerbation were measured at 3 months later.Results The CKQ scores at 1,2,3 months after intervention were (28.04±1.43),(28.20±1.00),(28.40±1.04) points in the experimental group,and (18.08±2.30),(18.50±1.87),(18.33±1.66) points in the control group,and there were significant differences (t=18.109,22.556,5.320,all P < 0.01).The scores of SGRQ and symptom at 2 months and 3 months after intervention were (50.56±8.68),(49.40±8.05),(41.52±10.86),(37.16±8.90) points in the experimental group,and (57.42±10.22),(59.46±9.06),(54.50±8.67),(56.71 ±10.03) points in the control group,and there were significant differences (t=-7.222-2.536,P < 0.05 or 0.01).The influence scores at 1,2,3 months after intervention were (46.28±8.74),(44.40±8.11),(42.04±8.22) points in the experimental group,and (51.50±7.77),(51.79±8.06),(54.08±6.96) points in the control group,and there were significant differences(t=-2.206,-3.197,-5.523,P < 0.05 or 0.01).There was no statistically significant difference about activity scores between two groups before and after intervention (P > 0.05).The numbers of acute exacerbation during 3 months with 3,22 cases in 1 and 0 time in the experimental group and 9,15 cases in the control group,and there was statistically difference between two groups(x2=4.306,P< 0.05).Conclusions The case management model can promote the stable COPD patients to know more disease-related health knowledge,and improve the health knowledge level.The case management model can reduce the symptoms of the stable COPD patients,reduce the frequency of acute exacerbation,reduce the influence of disease on daily life,prevent further decline in activity endurance,and improve the quality of life.
6.The expression level of carcinoembryonic antigen under hypoxic condition in breast cancer ;microenvironment and its prognostic value of breast cancer

Objective To investigate the correlationbetween thecarcinoembryonic antigen ( CEA) level and hypoxic status of breast cancer, and itspotential of being a prognostic factor in breast cancer. Methods Breast cancer cell line MDA-MB-231 was cultured under hypoxic or normoxic condition to determine the protein level of CEA using Western blotting.30 breast cancer tissue sections were collected in Tianjin Medical University Cancer Institute and Hospital from April 2013 to June 2015.and were detected the expression of CEA, hypoxia-inducible factor-1α( HIF-1α) and carbonic anhydrase-9 ( CA-9 ) by immunohistochemistry.Serum CEA were detected by immunofluorescence assay of 166 breast cancer patients collected from April 2009 to June 2011.The correlation between the expression levels of CEA and breast cancer patients'clinical data was analyzed by Logistic univariate analysis.Kaplan-Meier method was employed to calculate the progression-free survival of patients with breast cancer. Log-rank test was conducted to compare the difference between groups.Results CEA levels were significantly higher in breast cancer cells under hypoxic condition in a time-dependent manner, compared to normoxiccontrol. Immunohistochemical staining indicated that CEA was coexpressed with HIF-1αand CA-9 in breast cancer and linear regression analysis revealed that CEA expression correlated with HIF-1α( P =0.0096 );Preoperative serum levels of CEA were closely related with tumor size ( P =0.015 ) andLymph node metastasis(P=0.032); CEA positive patients achieved a progression-free survival of 29.85months(versus 39.08 months in CEA negative patients, P=0.003).Conclusions Hypoxia upregulates CEA expression in breast cancer and the levels of preoperative serum CEA is a potential prognostic factor ofbreast cancer.
7.Research advances in role of myeloid-derived suppressor cells in tumorigenesis

The role of myeloid-derived suppressor cells (MDSCs)in promoting inflammatory response and immune escape of tumor cells and the mechanism by which MDSCs promote the development and progression of tumors are reviewed.This could provide a new way for the investigation of immunological mechanism of liver fibrosis and liver cancer.Besides,MDSCs may be an effective therapeutic target for liver cirrhosis and liver cancer.
8.EFFECT OF ANTISENSE VEGF-(165) cDNA TRANSFECTION ON THE GROWTH OF HUMAN NEUROBLASTOMA SH-SY5Y CELLS IN VITRO AND IN NUDE MICE

Objective The present study was designed to explore the inhibitory effect of antisense VEGF-(165) cDNA on angiogenesis,growth rate of human neuroblastoma.Methods The eukaryotic expression vectors bearing antisense VEGF-(165) cDNA or sense VEGF-(165) cDNA were constructed.The stable cell lines transfected with the sense or antisense VEGF-(165) cDNA were established by using the selective medium containing 400?mg/L of G418.These cell lines were further studied for the exogenous antisense VEGF mRNA expression by RT-PCR and inhibition of expression of endogenous VEGF protein by immunocytochemical staining and ELISA.The proliferation of the transfected cells was analyzed by MTT method.Transfected cells were subcutaneously transplanted into nude mice and the growth of tumor masses was observed and weighted.Results The antisense VEGF was only detected in SH-SY5Y/AsVEGF cells by RT-PCR.The expression of VEGF protein was dramatically declined in the SH-SY5Y/AsVEGF cells compared with the original cells and empty vector transfected cells by immunocytochemical staining and ELISA.There was no effect of antisense VEGF transfection on cell proliferation in vitro.The growth of tumor mass with the cells transfected with antisense VEGF was significantly decreased in nude mice.Conclusion Antisense VEGF cDNA transfection to SH-SY5Y cells can effectively inhibit the expression of endogenous VEGF protein and tumor growth in nude mice.
