1.Bone marrow mesenchymal stem cell transplantation alleviates myocardial injury due to systolic hypertension

BACKGROUND:Mesenchymal stem cel s can be used to treat a variety of injuries, but little is known about their effect on systolic hypertension. OBJECTIVE:To study the therapeutic effect of bone marrow mesenchymal stem cel s on systolic hypertension rats. METHODS:Twenty-four rats were randomized equal y into control, model and cel transplantation groups. A rat model of systolic hypertension was made by injection of vitamin K and warfarin sodium in the model and cel transplantation groups. Rats in the cel transplantation group were given injection of bone marrow mesenchymal stem cel s into the left ventricle, while those in the other two groups given normal saline. RESULTS AND CONCLUSION:Compared with the model group, bone marrow mesenchymal stem cel s could reduce left ventricular mass index, increase cardiac tissue type I col agen volume fraction, col agen volume fraction and perivascular col agen volume fraction, but the ratio of end-diastolic volume/body weight did not change significantly. These findings indicate that bone marrow mesenchymal stem cel transplantation can reduce myocardial hypertrophy and cardiac fibrosis in systolic hypertension rats.
2.Uterine arterial embolization for uterine fibroid:correlation between uterine fibroid and ovarian blood supply

Objective To retrospectively analyze the correlation between uterine fibroid and ovarian blood supply, and to discuss the visualization of ovarian artery and uterine ovarian branch in uterine arterial embolization. Methods A total of 363 patients with clinically-confirmed uterine fibroid were enrolled in this study. Uterine arterial embolization was carried out in all patients. The visualization of ovarian artery and uterine ovarian branch during angiography was recorded. After the procedure the changes of uterine fibroid and ovarian functions were followed up. The results were statistically analyzed. Results The blood supply of uterine fibroid could be divided into three types: unilateral artery as the main source of blood supply (34.16%), bilateral balanced blood supply (43.80%) and unilateral uterine artery (22.04%). The visualization rate of uterine ovarian branch in bilateral balanced blood supply type was significantly higher than that of other two types (P<0.05). The visualization rate of ovarian artery at the side which was lack of blood vessels was higher than that at the opposite side (P< 0.05). Transient ovary functional impairment was observed in patients with visualization of uterine ovarian branch, which restored to normal in three months. No significant difference in the reduction of the tumor mass existed between each other among the different blood supply types. Conclusion The blood supply types of uterine fibroids have a parallel relationship with the ovarian blood supply from ovarian artery and/or uterine ovarian branch. Special attention should be paid to the uterine arterial embolization of the side that is lack of blood vessels, and it is needed to carefully observe the embolization extent, the flow velocity, etc. during the performance of embolization so as to avoid ovarian function impairment.
3.The effect of crocetin on Akt/GSK-3β/eNOS signaling pathway in myocardial ischemia-reperfusion injury of isolated rat hearts

Objective To study the protective effects of crocetin on myocardial ischemia-reperfusion injury, and their correlation with the signaling pathway of serine/threonine protein kinase (Akt)/glycogen synthase kinase (GSK)-3β/nitric ox?ide synthase (eNOS). Methods Forty healthy SD rats were divided into normal group (N), ischemia reperfusion group (IR) and 5, 10 and 15 mg/L of crocetin groups (CRO1, CRO2 and CRO3) by random number table method. The values of heart rate (HR), coronary flow (CF) and left ventricular pressure measurement (LVDP, LV+dp/dtmax, LV-dp/dtmax) 30 minutes after reperfusion were compared between five groups. TTC staining was used to detect the infarct volume. Spectrophotometric method was used to determinate the expression of lactate dehydrogenase (LDH) and creatine kinase (CK-MB). The levels of Akt, the phosphorylation of Akt (p-Akt), GSK-3β, phosphorylation of GSK-3β(p-GSK-3β), eNOS and phosphorylation of eNOS (p-NOS) were detected by Western blot assay. Results The HR, CF, LVDP, LV+dp/dtmax and LV-dp/dtmax were significantly lower 30 min after reperfusion in IR group than those of N group and crocetin groups (P<0.05). The myocardial infarction area was bigger in IR group than that of crocetin groups. The expression levels of LDH and CK-MB were signifi?cantly higher in IR group than those of N group and crocetin groups (P<0.05). The reperfusion index was higher in CRO3 group than that of CRO1 group. The infarction area, LDH and CK-MB expressions were significantly decreased in CRO3 group than those of CRO1 group (P<0.05). There were no significant differences in expressions of Akt, GSK-3βand eNOS between IR group, N group and crocetin groups. But p-Akt, p-GSK-3βand p-NOS were significantly decreased in IR group than those of N group and crocetin groups. The p-Akt, p-GSK-3βand p-NOS were significantly increased in CRO3 group than those of CRO1 group (P<0.05).Conclusion Crocetin has protective effects on myocrdial ischemia reperfusion injury in rats, which may be involved in the enhancing the phosphorylation of signalling pathway of Akt/GSK-3β/eNOS.
4.The effects of sonicated extract of Porphyromonas endodontalis on the cell cycle and apoptosis of osteo-blasts

Objective:To observe the effects of sonicated extract of Porphyromonas endodontalis(P.endodontalis)on the cell cy-cle and apoptosis of human osteoblastic hFOB 1 .1 9 cells.Methods:hFOB 1 .1 9 cells were treated with sonicated extract of P.end-odontalis at 0 (the control),1 ,1 0 and 1 00 μg/ml for 1 2,24 and 48 h respectively.MTT assay was used to examine cell prolifera-tion.The cell cycle distribution and apoptosis were examined by flow cytometry.Results:The sonicated exact of P.endodontalis in-hibited the proliferation of hFOB 1 .1 9 cells in a time-and dose-dependent manner.24 h treatment of 1 0 μg/ml and 1 00 μg/ml ex-act arrested the cell cycle of the cells in G1 phase,48 h treatment induced apoptosis in a dose-dependent manner.Conclusion:Sonicated exact of P.endodontalis may arrest hFOB 1 .1 9 cells in G1 phase and induce cell apoptosis,leading to inhibition of the cell proliferation.
5.Clinical Observations on the Treatment of Ramsay Hunt Syndrome by Blood-letting Puncture and Cupping as Main Therapy

Purpose: To investigate the clinical efficacy of blood-letting puncture and cupping in cooperation with acupuncture and moxibustion for treating Ramsay Hunt syndrome. Atreatment group of 32 cases was treated by blood-letting puncture with plum-blossom needle and cupping in cooperation with acupuncture and moxibustion and a control group of 32 cases,with acupuncture and moxibustion, and andaciclovir. Results and conclusion: The cure and marked effectiveness rate was 81.2% in the treatment group and 56.2% in the control group after 4 courses of treatment. It was higher in the treatment group than in the control group (P<0.05). The short-term otalgia-relieving rate was also significantly higher in the treatment group than in the control group (P<0.01).
6.Effect of metformin on connexin43 expression in H9c2 cells cultured with high glucose

Aim To investigate the effect of metformin (Met) on connexin43(Cx43) expression in H9c2 cells cultured with high glucose and the relevant mecha-nisms.Methods Rat myocardial cells H9c2 were cul-tured with high glucose , then combined with 3μmol· L-1 and 5 μmol · L-1 of Met for 24 h respectively . The viability and toxicity of H9c2 cells were detected by MTT and LDH , respectively .The expression and distribution of Cx43 were detected by immunofluores-cence staining .The intracellular oxygen species reac-tive ( ROS) level was detected by fluorescencemicrosco-py.And the expression of Cx43, P-AMPK, AMPK and GAPDH was determined by Western blot .Results The viability of H9c2 cells was increased and the level of intracellular ROS was decreased in Met group , while there existed no significant difference in LDH release among groups.Met significantly increased the phos-phorylation of AMPK and the expression of Cx 43, and improved the distribution of Cx 43 also.Conclusion The beneficial effect of Met on cardiovascular system in diabetic patients may via up-regulating the expression of Cx43 and down-regulating of intracellular ROS through activation of AMPK .
7.Effects of Sodium Tanshinone ⅡA Sulfonate on Cardiac Hypertrophy and MAPK Signaling Pathway

Objective To determinate the effects of sodium tanshinone ⅡA sulfonate(STS)on cardiomyocyte hypertrophy and explore the relative effects of STS on mitogen-activated protein kinase signal transduction system in rats with cardiomyocyte hypertrophy through constricting the thoracic aorta.Methods The models of cardiomyocyte hypertrophy were established in vivo,and the thoracic aorta was partially tied between the right innominate and the left common carotid arteries.The rats were randomly divided into 6 groups(n=8/group)as follows:①sham,②transverse aortic constriction(TAC),③TAC+low-dose Tan(TAC+LT)(5 mg/kg),④TAC+middle-dose Tan(TAC+MT)(10 mg/kg),⑤TAC+high-dose Tan(TAC+HT)(20 mg/kg),and ⑥ TAC+Val(10 mg/kg).After treatment for 8 weeks,echocardiography was performed to observe the changes in hypertrophy and heart function,and heart samples were cut into transverse sections and stained with hematoxylin and eosin(H&E).The MAPKs protein expression in the cardiomyocytes was detected by Western blot.Results The heart weight index(HWI),left ventricular mass index(LVMI)and cross-sectional diameter of cardiomyocytes(CD),left ventricular posterior wall thickness(LVWT),and interventricular septal thickness(IVS)were significantly increased in TAC group as compared with sham group.The relative parameters in STS groups and Val group were reduced as compared with those in TAC group.Western blot analysis revealed the p-ERK and p-p38 expression was significantly decreased in TAC group as compared with sham group(P<0.01).The p-ERK expression was significantly decreased in STS groups and Val group as compared with TAC group(P<0.05).The TAC+HT group,TAC+MT group and Val group had significantly higher p-p38 expression than TAC group(P<0.05).Conclusion Tanshinone ⅡA could regulate the expression of protein in MAPK pathway to exert its inhibitory effects on hypertrophy of cardiomyocytes.
8.Study on complications and compliance of different lying time after intrathecal injection in children with leukemia

Objective:To investigate the complication and compliance of different supine time and the degree of obedience in children with leukemia after intrathecal chemotherapy.Methods:A total of 553 children with leukemia after intrathecal chemotherapy from April 1, 2017 to March 28, 2019 in Beijing Children′s Hospital Affiliated to Capital Medical University were selected. Children who received intrathecal injection from April 1, 2017 to December 31, 2017 were selected as control group(274 cases), from January 1, 2018 to March 28, 2019 were selected as research group(279 cases). The children in research group were supine 2 hours but control group were supine 4 hours after intrathecal. The complications that occurred after injection and within one week after injection and compliance were observed.Results:Finally, 457 children included in this study, there were 235 cases in research group and 222 cases in control group. The number of cases of limb numbness and local pressure reddening in the research group and the control group were 18, 20 and 31, 34, respectively, with statistically significant differences between the two groups( χ2 values were 4.74, 5.07, P<0.05). After intrathecal injection, the patients' compliance with the time of supine removal was completely acceptable to the research group, relatively acceptable, acceptable, and required efforts to adhere to the number of cases were 65, 83, 42, 45, respectively, while the control group were 34, 50, 76, 62, respectively, the differences were significant ( χ2 value was 30.04, P<0.05). Conclusions:Supine for 2 hours after intrathecal injection can reduce the incidence of complications and improve compliance, which is safe and feasible. So, supine for 2 hours after intrathecal injection is recommended.
9.Study on the clinical features and prognostic factors of primary anorectal malignant melanoma

Objective:To analyze the influence of clinicopathological features on the prognosis of anorectal malignant melanoma (AMM), and to establish a more accurate prognosis prediction model.Methods:From January 1, 2006 to December 31, 2018, at Fudan University Shanghai Cancer Center, the data of 89 patients diagnosed with AMM and underwent operation were retrospectively analyzed. The Cox proportional hazard regression model was used to analyze factors affecting the prognosis. Backward elimination was used to select variables, Nomogram prognosis prediction model was established and verified internally, and the consistency index was calculated.Results:Among 89 AMM patients, 65 (73.0%) were female, 78(87.6%) were <70 years old, and the most common tumor location was the rectum (48.3%, n=43), followed by the anal canal (31.5%, n=28) and the anorectal canal (20.2%, n=18). Thirty-eight (42.7%) patients directly received abdominal-perineal resection (APR), 37 patients (41.6%) received interferon-based immunotherapy, and 11 patients (12.4%) received both systemic chemotherapy and immunotherapy. The 3-year disease-specific survival (DSS) rate was 41.6%(37/89), and the 5-year DSS rate was 31.5%(28/89). The results of multivariate Cox analysis showed that age ≥70 years old and T2 stage were risk factors of the prognosis of AMM patients (hazard ratio ( HR)=11.29, 4.83; 95% confidence interval ( CI) 2.89 to 44.13, 1.66 to 14.11; both P<0.01), while neurovascular invasion, immunotherapy treatment, systemic chemotherapy combined with immunotherapy, APR after extensive local resection, directly APR and APR combined with inguinal lymphadenectomy were protective factors of the prognosis ( HR=0.09, 0.23, 0.10, 0.13, 0.26, 0.02; 95% CI 0.02 to 0.34, 0.10 to 0.57, 0.02 to 0.49, 0.03 to 0.52, 0.08 to 0.90, 0.00 to 0.27; all P<0.05). The Nomogram model was further established with age, gender, tumor location, T stage, distant metastasis, medication chemotherapy and surgical treatment. The results of the Nomogram model internal verification indicated that the accuracy of the model in predicting 1-year, 3-year and 5-year DSS was good, and the consistency index was 0.749, which was significantly higher than the consistency index of traditional TNM stage (0.607). Conclusions:Most AMM patients are <70 years old, and the majority of them are female. The common location of AMM is rectum, and many patients receive immunotherapy. Age ≥70 years old and T2 stage are risk factors affecting the prognosis of AMM patients. Neurovascalar invasion, immunotherapy based comprehensive treatment, APR after extensive local resection, directly APR and APR combined with inguinal lymphadenectomy are protective factors of the prognosis. Nomogram prognosis prediction model established based on the clinicopathological features and treatment of AMM patients has higher accuracy and clinical reference value than the traditional TNM stage system.
10.Simvastatin improves cardiac function in acute phase after myocardial infarction through Akt/eNOS pathway

Aim To investigate the effect of simvastatin ( Sim ) on endogenous antioxidant system after acute myocardial infarction ( AMI ) and its potential mecha-nisms. Methods The acute myocardial infarction ( AMI ) rat models were made by ligation left anterior descending of coronary artery. Then the successful models were randomly divided into myocardial infarc-tion group ( MI group) and simvastatin group ( Sim,20 mg·kg-1·d-1), another group without ligation left anterior descending of coronary artery served as sham group(Sham group). The Sim group was administered simvastatin by gavage for 7 days. MI group and Sham group received saline. Hemodynamic parameters, lipid levels, troponinI ( c-TnI ) and lactate dehydrogenase ( LDH) concentrations were examined after 7days, and the levels of superoxide dismutase ( SOD) and glutathi-one peroxidase ( GP) of myocardial antioxidant system were detected by ELISA. The expression of cardiac p-Akt and p-eNOS protein were detected by Western blot. Results Acute myocardial infarction significant-ly lowered cardiac hemodynamic parameters, increased serum c-TnI and LDH levels, lowered levels of SOD and GP, and lowered the expression of p-Akt and p-eNOS protein. However, Sim could effectively prevent the deterioration of cardiac function, reduce serum c-TnI and LDH levels, increase levels of SOD and GP, and increase p-Akt and p-eNOS protein expression. Conclusion Early using Sim can effectively improve heart function after acute myocardial infarction, acti-vate myocardial antioxidant system,and reduce myocar-dial necrosis, which may be related to increasing the expression of p-Akt and p-eNOS.
