1.Clinical Observations on the Treatment of Ramsay Hunt Syndrome by Blood-letting Puncture and Cupping as Main Therapy

Purpose: To investigate the clinical efficacy of blood-letting puncture and cupping in cooperation with acupuncture and moxibustion for treating Ramsay Hunt syndrome. Atreatment group of 32 cases was treated by blood-letting puncture with plum-blossom needle and cupping in cooperation with acupuncture and moxibustion and a control group of 32 cases,with acupuncture and moxibustion, and andaciclovir. Results and conclusion: The cure and marked effectiveness rate was 81.2% in the treatment group and 56.2% in the control group after 4 courses of treatment. It was higher in the treatment group than in the control group (P<0.05). The short-term otalgia-relieving rate was also significantly higher in the treatment group than in the control group (P<0.01).
2.Multi-directional differentiation potential of subpopulations of mesenchymal stem cells isolated from human skeletal muscle expressing different myogenic and endothelial markers

BACKGROUND:Mesenchymal stem cells from human skeletal muscle exhibit multi-directional differentiation potential under the influence of osteogenic proteins such as bone morphogenetic protein 4 (BMP4). But the differentiation of a specific cell subpopulation is not yet clear.OBJECTIVE:To characterize the multi-directional differentiation potential of mesenchymal stem cells from human skeletal muscle based on the expression of different surface markers.METHODS:Four different subpopulations were isolated from the human skeletal muscle by fluorescence-activated cell sorting based on their expression of the myogenic-specific marker CD56 and the endothelial-specific markers CD34 and CD144, including CD56+, CD56+CD34+CD144+, CD34+CD144+, and unsorted groups. Osteogenic differentiation of the four groups of the cells was displayed by Von Kossa staining after the treatment with BMP4 alone or BMP4 plus transforming growth factor β3. Chondrogenic differentiation of these cells was displayed by Alcian blue staining. Bone metabolism was assessed by alkaline phosphatase staining.RESULTS AND CONCLUSION:No significant difference in the bone metabolism was found among four groups after the treatment with BMP4 (P > 0.05). Osteogenic and chondrogenic potentials of the four cell subpopulations were significantly different. Under the same osteogenic induction, the CD56+ cells exhibited strongest potential for osteogenic differentiation; and under the same chondrogenic induction, the CD56+CD34+CD144+ cells exhibited better potential for chondrogenic differentiation than the CD56+ cells. These findings indicate that the osteogenic and chondrogenic potentials are intimately associated with the type of mesenchymal stem cells from human skeletal muscle:the CD56+ cells are closely related to the osteogenic potential, while the CD56+CD34+CD144+ cells have stronger chondrogenic potential.
3.Alterations of plasma glucocerabrosidase,protein phosphastase 2A and ceramide levels in patients with ischemic stroke

Objective Toanalyzethealterationsofplasmaglucocerebrosidase(GBA),protein phosphatase 2A (PP2A)and its degradation product ceramide in patients with ischemic stroke. Methods Atotalof45inpatientswithischemicstrokeattheDepartmentofNeurology,theAffiliated Hospital of Logistics College of the CAPF were enrolled from May to September 2013,and 45 age-and sex-matched healthy subjects at the Physical Examination Center collected at the same time period were used as a control group. Blood samples of the patients and healthy subjects were obtained,anticoagulated, and the plasma was separated. H50 protein chip and laser matrix-assisted laser desorption/ionization top of flymassspectrometrywereusedtotestthelevelsofplasmaceramide.Results TheplasmaGBAand PP2A activities in patients with ischemic stroke were significantly lower than those of the control group;the GBA activities of the ischemic stroke group and the control group were 2 . 4 ± 0 . 8 and 3 . 1 ± 1 . 4 U/L respectively. There was significant difference (P<0. 05);the PP2A activities of the two groups were 6. 5 ± 2. 8 and 14. 5 ± 4. 7 U/L respectively (P<0. 01). The relative level of the plasma ceramide in patients with ischemic stroke was 1. 9 ± 0. 7,and it was significantly lower than 12. 2 ± 5. 0 of the control group (P<0.01).Conclusion ThedecreasedlevelsofplasmaGBAandPP2Aactivitiesaswellasthe ceramide in patients with ischemic stroke suggested that the abnormal phosphorylation of synuclein in the blood of patients with stroke.
4.DETECTION OF SPERMATOZOAL TOTAL RNAS BY LABORATORY ON CHIP GEL ELECTROPHORESIS

Objective Detecting of spermatozoal total RNAs by laboratory on chip gel electrophoresis so that it could provide better total RNAs for the sequent experiments, and spur the development of spermatozoal molecular biology. Methods Sperms of healthy adults were collected and then total RNAs were extracted by RNeasy mini kit(QIAGEN), detection and quality control were performed by loboratory on chip gel electrophoresis system. Meantime, the control RNAs were extracted from lymphocytes. Results It was found that there were a plenty of genes expressed in healthy sperms. Electrophoretic graphs showed that the total RNAs of spermatozoal had 2 bands which went ahead a little comparing to the normal somatic cells. The former peak appeared keenness, and the latter was broad and showed like a reversed U. The ratio of them was largely more than 2, no extra peaks were found in electrophoretic graph. Conclusion A simple,intuitionistic method to detect and control the quality of the healthy adults' spermatozoal total RNAs had been successfully constructed by using laboratory on chip gel electrophorosis.

The pediatric reference intervals in clinical laboratory play an important role in diagnosis of illness,therapeutic monitoring,prediction of prognosis and health evaluation.Compared with establishing reference interval for adults,there are more challenges to establish pediatric reference intervals.Therefore,the procedure and key technologies of direct method and indirect method are stated based on the characteristics of children population and pediatric,by which to define,transfer and validate pediatric reference intervals.This study will provide systematically methodological ideas for clinical laboratories to establish pediatric reference intervals.
6.Establishment and validation of reference intervals for reticulocyte parameters in children from Beijing

Objective:To establish the reference intervals of reticulocyte parameters for children in the Beijing area.Methods:1 766(856 males and 910 females) healthy children aged from 1 to 18 years old in the Beijing area were studied, the infant group (≥1-3 years old) included 146 participants; the preschool group (>3-6 years old) had 449 participants; the school age group (>6-13 years old) had 646 participants and the adolescent group (>13-18 years old) had 525 participants. Seven parameters were measured in venous blood by SySmex XN-20 (A1) automatic blood cell analyzer, which included reticulocyte percentage (RET%), absolute reticulocyte count (RET#), low fluorescence reticulocyte (LFR), medium fluorescence reticulocyte (MFR), high fluorescence reticulocyte (HFR), immature reticulocyte fraction (IRF) and reticulocyte hemoglobin equivalent (RET-He). After the test results were collected, the reference intervals were established according to the percentile ( P2.5, P97.5). As the reference intervals were established, venous blood samples from 109 healthy children in Beijing were collected to verify the reference intervals according to WS/T 402-2012 "Define and determine the reference intervals in clinical laboratory". Results:The reference interval of 7 Reticulocyte parameters for children aged 1-18 years without age and sex grouping,reference interval (RET%): 0.74%-2.06%; absolute reticulocyte count (RET#): (34.5-101.5)×10 9/L; low fluorescence reticulocyte (LFR): 86.6%-96.5%; medium fluorescence reticulocyte (MFR): 3.2%-11.5%; high fluorescence reticulocyte (HFR): 0%-2.1%; immature reticulocyte fraction (IRF): 3.5%-13.4%; reticulocyte hemoglobin equivalent (RET-He): 28.5-34.2 pg. Over 90% test results of samples from the 109 children ranged within the reference ranges. Conclusion:This study established convincible reference intervals of seven reticulocyte parameters for healthy children aged from 1 to 18 in the Beijing area was established, and reference interval verification passed.

In 2022,breast cancer ranks the first in incidence among all malignant tumors worldwide,and it's high recurrence and fatality rate has posed serious threat to human life and health.Triple negative breast cancer(TNBC)is known for its poor prognosis among all types of breast cancer,and is often associated with elevated expression of epidermal growth factor receptor(EGFR).EGFR is widely expressed on the surface of various cells.The signaling pathway mediated by EGFR is mainly involved in cell growth,proliferation and differentiation.EGFR overexpression is often closely related to tumor invasion,metastasis and prognosis.Therefore,elucidating the roles of EGFR in TNBC and developing prevention and treatment strategies correspondingly are helpful to prevent and control the occurrence of breast cancer,and improve patients'quality of life.This review summarizes the research progress in the roles of EGFR,the treatment strategies and drug resistance in TNBC,in order to provide more refence for treating and improving the prognosis of breast cancer.

9.Value of CT Features on Differential Diagnosis of Pulmonary Subsolid Nodules and Degree of invasion Prediction in Pulmonary Adenocarcinoma.

BACKGROUND:
Subsolid pulmonary nodules are common computed tomography (CT) findings of primary lung adenocarcinoma. It is of clinical value to determine the clinical treatment strategies based on CT features. The aim of this study is to find the valuable CT characteristics on differential diagnosis and the degree of invasion prediction by a retrospectively analysis of three groups subsolid nodules, including benign, and invasive adenocarcinoma.
METHODS:
The CT findings of 106 cases of resected sub-solid nodules were retrospectively analyzed. The nodules were firstly divided into benign and malignant groups and the malignant group was further divided into non/micro-invasive group (atypical adenomatous hyperplasia/adenocarcinoma in situ/minimally invasive adenocarcinoma) and invasive adenocarcinoma group. The nodule size, proportion of solid components, tumor-lung interface, shape, margin, pleural traction, air bronchus sign, vascular abnormalities inside the nodule were evaluated. The univariate analysis (χ2 test, non-parametric test Mann-Whitney U test) was performed to screen statistically significant variables and then enrolled in further multivariate Logistic regression analysis.
RESULTS:
Multivariate logistic regression analysis showed that a clear tumor-lung interface, air bronchus sign, and pulmonary vascular abnormalities were important indicators of malignant nodules with hazard ratios of 38.1 (95%CI: 5.0-287.7; P<0.01), 7.9 (95%CI: 1.3-49.3; P=0.03), 7.2 (95%CI: 1.4-37.0; P=0.02), respectively. The proportion of solid components was the only significant indicator for identifying invasive adenocarcinoma from AAH/AIS/MIA , with a risk ratio of 1.04 (95%CI: 1.01-1.06, P=0.01).
CONCLUSIONS
SSNs with clear tumor-lung interface, air bronchus sign, and pulmonary vascular abnormality inside nodule are more likely to be malignant. A higher percentage of solid components indicates a higher likelihood to be an invasive lesion in malignant SPNs.
