1.Protective effects of Bauhinia championii Benth flavones on oxidative stress of neonatal rat cadiocytes induced by H2O2

Objective To investigate the protective effects of Bauhinia championii Benth flavones(BCF) on oxidative stress of neonatal rat cadiocytes induced by H2O2.Methods Cadiocytes of neonate rat was cultivated for 72 hours and divided into six groups: a normal control group, a H2O2 group, BCF(60, 120 and 240μg/ml)+H2O2 groups and aShuxueninginjection (100μg/ml)+H2O2 group (n=8). 6 hours after the drugs were given, the morphology changes was observed and the survival rate was detected; the content of AST, CPK, LDH in culture medium were detected; the activity of SOD, CAT, GSH-Px and the content of MDA in cardiomyocytes were also determinted; the apoptosis rate were detected, and the expression of caspase-3 in cardiomyocytes was detected by Western blot.Results Compared with the H2O2 group, the activity of AST(28.8 ± 6.1 U/ml, 24.5 ± 5.3 U/mlvs. 36.2 ± 6.7 U/ml), CPK(1.8 ± 0.4 U/ml, 1.5 ± 0.3 U/mlvs. 2.5 ±0.4 U/ml), LDH(805.2 ± 160.9 U/L, 671.5 ± 128.7 U/Lvs. 916.5 ± 168.4 U/L) in culture medium were significantly decreased (P<0.05,P<0.01), the activity of SOD(84.8 ± 17.4 U/mg, 95.3 ± 18.2 U/mgvs. 55.7 ± 13.1 U/mg), CAT(23.4 ± 3.1 U/mg, 26.3 ± 3.5U/mgvs. 15.2 ± 3.0 U/mg) in cardiomyocytes were significantly increased and the content of MDA(8.1 ± 1.7 nmol/mg, 6.8 ± 1.5 nmol/mgvs. 11.1 ± 2.3 nmol/mg) were significantly decreased (P<0.05,P<0.01), the expression of c caspase-3(1.64 ± 0.16, 1.30 ± 0.12vs. 2.06 ± 0.25) and the apoptosis rate (24.2% ± 5.5%, 13.4% ± 3.9%vs. 51.2% ± 9.1%) were significantly decreased (P<0.05,P<0.01); the activity of GSH-Px (3.6 ± 0.9 U/mg vs. 2.4 ± 0.7 U/mg) in cardiomyocytes of BCF 240μg/ml treatment group was increased (P<0.05).Conclusions BCF could effectively improve the morphology of neonatal rat cadiocytes induced by H2O2, increase the survival rate, improve the activity of antioxidase, down-regulate the expression of caspase-3 and decrease the apoptosis rate, suggesting that BCF had dose-dependent protective effects on oxidative stress of neonatal rat cadiocytes induced by H2O2.
2.Thermal Desorption-Gas Chromatography or Gas Chromatograph-Mass Spectrometry for Analysis of Semi-Volatile Compounds on Atmospheric Particulate Matters

A thermal desorption ( TD) device was developed and coupled to gas chromatography ( GC) or gas chromatography-mass spectrometry ( GC-MS ) for the qualitative and quantitative analysis of semi-volatile organic compounds on atmospheric particulate matters ( PM ) . The TD was operated by direct heating and placed on the GC injector, leading to high heating rate and easy transfer of analytes to GC without focusing of analytes by cold trap. For establishing the TD-GC method, the materials used for supporting PM samples, temperature and time of thermal desorption, and types of sample injection were investigated for detection of sixteen polycyclic aromatic hydrocarbons ( PAHs) and nine n-alkanes. The limits of detection of the proposed TD-GC method were in the range of 0. 014-0. 093 ng for PAHs, and 0. 016-0. 026 ng for n-alkanes, respectively, with the correlation coefficients of correlation above 0. 9975. The TD-GC method was applied to the determination of trace PAHs and n-alkanes on PM10 samples from three cities. The recoveries were in the range of 95%-135% ( PAHs) and 95%-115% ( n-alkanes) , respectively. Finally, the TD was coupled to GC-MS for comparison of the contents of PAHs and n-alkanes on PMx with different particulate size ( x=10 , 5, 2, 1, 0. 5, 0. 25, 0. 1).
3.Relationship between slow coronary flow and vascular endothelial function

Objective:To study and analyze the relationship between slow coronary flow (SCF) and vascular endothelial func‐tion .Methods:A total of 88 patients ,who received coronary angiography in our hospital from Jan 2014 to Dec 2014 ,were selected .TIMI blood flow classification was used to assess coronary flow velocity of all patients .The CTFC (corrected TI‐MI frame count ) >27 frames was regarded as slow flow .The patients with slow flow were regarded as SCF group (n=43) , and those with normal blood flow were regarded as normal control group (n=45) .Levels of blood pressure ,blood glucose and blood lipids ,and vascular endothelial function were measured and compared between two groups .Logistic regression a‐nalysis was used to analyze the relationship between SCF and vascular endothelial function .Results:There were no signifi‐cant difference in levels of blood pressure ,blood glucose and blood lipids between two groups , P>0. 05 all .Compared with normal control group , there were significant reductions in fore brachial artery flow‐mediated vascular diastolic function [FMD ,(8. 33 ± 2. 04 )% vs . (7. 06 ± 1. 78 )% ] and nitroglycerin mediated vasodilation [NMD , (20. 39 ± 4. 13 )% vs . (16.10 ± 5.22)% ] in SCF group ,P<0.01 both .Logistic regression analysis indicated that reduced FMD (OR=1.069 ,P=0.011) and NMD (OR=1.183 ,P=0.014) were risk factors for SCF .Conclusion:The vascular endothelial dysfunction is a risk factor of slow coronary flow .
4.Effect of Acupuncture at Sanyinjiao (SP6) on the Infrared Temperature of Guanyuan (CV4) and Sanyinjiao in Dysmenorrhea Patients

Objective By using infrared thermal imager (Flir-SC620), to observe the effect of needling Sanyinjiao (SP6) on the skin temperature at Guanyuan (CV4) and Sanyinjiao in patients with primary dysmenorrhea (PD) of cold and dampness stagnation pattern, and to explore the probability of using infrared thermal imaging for diagnosis and as an objective index for evaluating the action and needling qi of acupuncture. Method Thirty-six subjects were recruited and divided into four group, a health control group (group A), a control group of PD of cold and dampness stagnation pattern (group B), a needling-qi-expected group (group C) and a needling-qi-unexpected group (group D). Group A and B were not given acupuncture treatment, while group C and D were treated with acupuncture at bilateral Sanyinjiao with needles retained for 30 min, and the needling sensations were recorded. The infrared thermal imager was used to detect the skin temperature at Guanyuan and bilateral Sanyinjiao for 40 min for each group, and the temperature was recorded every 10 min. The temperature during different periods of time, 0-10 min, 20-20 min, 20-30 min, 30-40 min, 0-30 min, and 0-40 min were then calculated. In group C, those obtained the needling qi sensation were further grouped into C-1 and those didn’t obtain the sensation were into C-2; in group D, those obtained needling qi sensation were further grouped into D-1 and those didn’t obtain the sensation were into D-2. SPSS 17.0 was adopted for data processing, and the data were analyzed by using MANOVA of repeated measuring. Result Compared to group A (6 cases), the temperature at Guanyuan in group B (6 cases) was significantly decreased during 0-30 min and 0-40 min (P<0.05), the temperature at the left Sanyinjiao during 0-40 min in group B was significantly decreased (P<0.05), and the temperatures at bilateral Sanyinjiao during 30-40 min in group B significantly dropped (P<0.05). Compared to group B, the temperatures at Guanyuan during 0-30 min and 0-40 min in group C1 (12 cases) and group D1 (11 cases) were significantly increased (P<0.05), the temperature at left Sanyinjiao during 0-40 min in group D1 was significantly increased (P<0.05), and the temperatures at bilateral Sanyinjiao during 30-40 min in group D1 were significantly increased (P<0.05). There was no case in group C2 and only 1 case in group D2, hence, the data were not enough for analysis. Conclusion Decrease of the infrared temperature at Guanyuan and Sanyinjiao can be taken as one of the diagnostic criteria for dysmenorrhea of cold stagnation pattern. Increase of the infrared temperature at Guanyuan can be regarded as one of the objective evidences for the along-meridian transmission characteristic in needling Sanyinjiao.
5.Quantification of Deqi (arrival of qi) by Short-latency Somatosensory Evoked Potentials:A Randomized Crossover Controlled Trial Plan

Objective To explore the feasibility of using short-latency somatosensory evoked potentials (SLSEP) to quantitate Deqi.Methods A randomized crossover controlled trial was carried out. Healthy subjects were enrolled and allocated to treatment (thick needle, deep insertion and manipulation for Deqi) and control (thin needle, shallow insertion and no manipulation without Deqi) groups. Somatosensory evoked potentials were recorded before, during and after acupuncture. Deqi was assessed using the score scale in the subjets. The effects of Deqi and no Deqi at point Sanyinjiao (SP 6) on the potentials were observed.Results The preliminary exploration of the feasibility by the trial test showed that the effect of Deqi on short-latency somatosensory evoked potentials had certain regularity. It was worthy to be observed.Conclusion The plan is feasible. The formal test can be conducted.
6.Optimization of Menin inhibitors based on artificial intelligence-driven molecular factory technology

Abstract: The new generation of artificial intelligence technology, represented by deep learning, has emerged as a crucial driving force in the advancement of new drug research and development. This article creatively proposes a workflow named “Molecular Factory” for the design and optimization of drug molecules based on artificial intelligence technology. This workflow integrates intelligent molecular generation models, high-performance molecular docking algorithms, and accurate protein-ligand binding affinity prediction methods. It has been integrated as a core module into DrugFlow, a one-stop drug design software platform, providing a comprehensive set of mature solutions for the discovery and optimization of lead compounds. Utilizing the “Molecular Factory” module, we conducted the research of second-generation inhibitors against Menin that can combat drug resistance. Through the integration of computational and experimental approaches, we rapidly identified multiple promising compounds. Among them, compound RG-10 exhibited the IC50 values of 9.681 nmol/L, 233.2 nmol/L, and 40.09 nmol/L against the wild-type Menin, M327I mutant, and T349M mutant, respectively. Compared to the positive reference molecule SNDX-5613, which has entered Phase II clinical trials, RG-10 demonstrated significantly enhanced inhibitory activity against the M327I and T349M mutants. These findings fully demonstrate the unique advantages of the "Molecular Factory" technology in practical drug design and development scenarios. It can rapidly and efficiently generate high-quality active molecules targeting specific protein structures, holding significant value and profound implications for advancing new drug discovery.
