1.Clinical significance of fetal weight estimation by modiefied uterine fundal height

Objective To investigate the accuracy of fetal weight estimation by modiefied uterine fundal height.Methods Totally 547 pregnant women admitted to maternity ward of the Second Affiliated Hospital of Chongqing Medical University from December 1,2013 to April 30,2014 with childbirth were chosen as the subject.The modified uterine fundal height were measured by the uterus inclination and modiefied fundal height admeasuring apparatus,normal uterine fundal height and and abdomen circumfer-ence were measured by tape.The fetal weight was estimated by utilizing modified uterine fundal height.Results The modified fun-dal height was(31.7±2.30)cm,which was significantly higher than that normal fundal height(34.7 ± 2.46 )cm(t = 54.25,P <0.01).The accuracy rate of modified uterine fundal height for fetal weight estimation with empirical formulas,Yuan′s,Zhuo′s,Lin′s,Zen′s,Gu′s were 68.4%,69.7%,59.4%,67.8%,69.3%,78.4%,respectively.The accuracy rate of normal uterine fundal height for fetal weight estimation with the same formulas were 53.4%,34.4%,54.3%,46.1%,38.8%,45.9%,respectively.The accuracy rate of modiefied uterine fundal height for fetal weight estimation with empirical formulas,Yuan′s,Lin′s,Zen′s,Gu′s were significantly higher than that of utilizing normal uterine fundal height estimation with the same formulas(P <0.01).Conclusion Modified uterine fundal height has better accuracy in estimating fetal weight,it has a important clinical significance.
2.The establishment of the three-dimensional culture model of the human skin squamous-celled carcinoma and their biological evaluation of the high-invasive squamous carcinoma cells

Objective To establish the three-dimensional culture model of the human skin squamous-celled carcinoma ,and isolate the highly invasive cell subsets ,and compare their biological characteristics .Methods By using the composite chitosan tissue engi-neering skin ,the three-dimensional culture model of the human skin squamous-celled carcinoma was successfully established , through digestion ,we divided and got the high-invasive squamous carcinoma cells subsets .After that ,the different invasive proper-ties of subpopulations of cells in nude mice tumor effects were compared .Results Different concentrations of cinnamaldehyde and interferon on skin squamous-celled carcinoma cell proliferation inhibition had statistical significance (P<0 .01) ,the different invasive characteristics of skin squamous cells carcinoma between subpopulations of cells proliferation inhibition also was statistically signifi-cant(P<0 .01) ,and the two different invasive properties of subpopulations of cells in the tumor characteristics also had statistical significance(P<0 .01) .Conclusion We established the three-dimensional culture model of the human skin squamous-celled carcino-ma successfully ,by using this model ,we could screen the higher invasive cells subsets .
3.Experimental study on the effect of VIP on MVD in the process of fracture healing in rats with high selectiveneurotomy

Objective The mechanism and signaling mechanisms of fracture healing by vasoactive intestinal peptide (VIP) regulating are not clear.We established an selective cut off sensory/motor nerve combined with tibial fracture rat model to study the effects of VIP and microvessel density(MVD) during the fracture healing process after nerve injury.Methods 60 SD rats were randomly divided into 3 groups: tibial fracture + motor nerve (anterior root) cutting group (group ART);tibial fracture + posterior root (sensory nerve) cutting group (group PRT);tibial fracture group (group SO),and each group were 20.Three groups of rats in the model after the establishment of 1W, 2W, 3W, 4W these time points, were killed.The skeleton specimens were obtained in the vicinity of the fracture 5mm, to observe the expression of VIP and microvessel density of the law of change.Results Histology observation showed: new bone trabecular bone callus maturity of PRT group is lower than SO group and ART group at each time points;the immunohistochemical staining showed that compared with callus VIP average optical density of PRT group at 2w and 3w(0.156±0.015、0.163±0.012), SO group(0.216±0.012、0.223±0.010) and ART group(0.198±0.014、0.212±0.016) increased obviously(P<0.05).The detection of callus MVD indicated that compared with callus MVD average density of PRT group at 2w and 3w(26.4±2.2、32.3±2.0), SO group(38.2±2.3、40.6±2.6) and ART group(36.6±2.2、38.5±2.1) significant increased (P<0.05).At the 2nd, 3rd and 4th week, the wet weight of tibia was significantly increased in group ART and SO compared with group PRT(P<0.05),which increased more significantly at 2 weeks.Conclusion The effect of loss of sensory nerve innervation on the formation of microvessel density, callus maturity and fracture healing rate were more pronounced than loss of motor nerve innervation.
4.Effects of curcumin on proliferation and COL I expression of human circulating fibrocytes

Aim To isolate and characterize the human circulating fibrocytes from human peripheral blood and explore the effects of curcumin on human circulating fi-brocytes.Methods The cells were isolated and puri-fied by density gradient centrifugation,and identified by flow cytometry and immunocytochemistry .Then , CCK-8 and flow cytometry were used to study the effect of curcumin on the proliferation as well as COL I ex-pression of human circulating fibrocytes,respectively. Results After being isolated the cells expressed CD34,CD45 and COLⅠ,among which 79.7% were both CD45 and collagen I positive,typical of human circulating fibrocytes.Curcumin could exert regulatory effects on proliferation of human circulating fibrocytes. Exposure of the cells to curcumin for as short as 24 hours promoted their growth,while prolonged treatment (72 h ) significantly inhibited cell propagation and downregulated the COLⅠ levels,best manifested at a concentration as high as 20 μmol · L-1 .Conclusion The proliferation of cells and COLⅠexpressions can be effectively inhibited by curcumin with the prolonged action period and high concentrations.

7. Clinical effect of combined artificial extracorporeal liver support therapy for toxic hepatic failure

Objective:
To investigate the clinical effect of plasma exchange (PE) versus double plasma molecular adsorption system combined with PE (DPMAS+PE) in the treatment of toxic hepatic failure.
Methods:
A total of 67 patients with toxic hepatic failure who were admitted during the same period of time were divided into PE group, DPMAS+ PE group, and control group. The 22 patients in the PE group were treated with PE alone, and the 24 patients in the DPMAS+PE group were given DPMAS combined with PE. The clinical out-come was compared between the three groups.
Results:
Both treatment groups had significantly higher clinical response rate and 24-week survival rate than the control group. After treatment, both treatment groups had significant reductions in the serum levels of total bilirubin (TBil) , direct bilirubin (DBil) , alanine aminotransfer-ase (ALT) , and aspartate aminotransferase (AST) , and the PE group had significant reductions in the albumin (Alb) level and activated partial thromboplastin time (APTT) (

With the rapid development of sequencing technologies towards higher throughput andlower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficientand easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive (GSA; http://bigd.big.ac.cn/gsa or http://gsa.big.ac.cn), a data repository for archiving rawsequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration (INSDC), GSA adopts four data objects (BioProject, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. In the era of bigdata, GSA is not only an important complement to existing INSDC members by alleviating theincreasing burdens of handling sequence data deluge, but also takes the significant responsibilityfor global big data archive and provides free unrestricted access to all publicly available data insupport of research activities throughout the world.
9.Preparation of hydroxyapatite coating in concentrated simulated body fluid by accelerated biomimetic synthesis.

In the present work, NiTi alloy substrates were activated by three different pretreatment processes. 5 X SBF1 and 5 X SBF2 concentrated simulated body fluids were prepared with citric acid buffer reagent, and then calcium phosphate coatings were formed quickly on NiTi alloy surface by accelerated biomimetic synthesis after pretreatment. The microstructure, composition and surface morphology of calcium phosphate coatings were studied. The results indicate that calcium phosphate coatings possess porous and net structure, which are composed of precipitated spherical particles with diameter less than 3 microm. The analysis of XRD shows that the main component of calcium phosphate coatings is hydroxyapatite, whereas the concentrated 5 x SBF simulated body fluid, which is in the absence of Mg2+ and HCO3- crystal growth inhibitors, apparently accelerates the growth rate of hydroxyapatite coatings.

10.Identification of common Sarcophagidae by short cytochrome oxidase I fragment.

OBJECTIVE:
To identify the common Sarcophagidae with a 278 bp fragment of cytochrome oxidase I in mitochondrial DNA and to obtain an unambiguous and rapid identification method for Sarcophagidae in forensic investigations.
METHODS:
Nineteen Sarcosaprophagous flies were collected from 16 locations in 12 Chinese provinces. All specimens were comprised of 4 species. The mtDNA of flies was extracted with SDSPK extraction method. Polymerase chain reaction was conducted in an Eppendorf 5331 thermal cycler. The PCR products were purified and sequenced and the obtained sequences were uploaded to GenBank. A neighbor-joining tree was constructed with MEGA4.0 package.
RESULTS:
The 19 Sarcosaprophagous flies were well clustered. The intraspecific variation within species varied from 0% to 3%, while the interspecific variations between species varied from 8% to 12%.
CONCLUSION
Congeneric species can be separated by the short fragment (278 bp region in the cytochrome oxidase subunit I gene), which will be instrumental for implementation of the Chinese Sarcophagidae database and lay a foundation for post mortem interval estimation in future forensic cases.
