2.The effect of wet hyperthermia combined with microwave ablation on the proportion and function of CD 4 +CD25 +regulatory T cell in animal model of mouse mammary carcinoma

Objective To investigate the effects of wet hyperthermia combined with microwave ablation on the pro-portion and function of CD 4 +CD25 +regulatory T cell in animal model of mouse mammary carcinoma and explore its significance .Methods An experimental model of mouse mammary carcinoma was established and then treated with microwave ablation on day 14.Local wet hyperthermia was also performed .The proportion of CD4 +CD25 +regula-tory T cells (Tregs) in splenocytes was analyzed by FACS.The relative expression of Foxp3, as well as the expres-sion of CTLA-4 and GITR, on Tregs was determined by real-time PCR assay and FACS respectively .Moreover, the secretion of TGF-βand IL-10 from Tregs also were detected by ELISA .In addition , the suppressive capacity of Tregs on CD4 +CD25 -T cell was also examined by MTT assay .Results The proportion of Tregs in wet hyperther-mia combined with microwave ablation treated group decreased significantly compared with that in wet hyperthermia or microwave ablation treated alone group ( P<0.05 ) .Moreover , the expression level of Foxp 3 on Tregs decreased significantly (P<0.05).Furthermore, the expression of CTLA4 and GITR, as well as the secretion of TGF-βand IL-10, on Tregs also obviously decreased (P<0.05).Finally, the suppressive capacity of Tregs on the proliferation of CD4+CD25-T cells were also significantly impaired ( P<0.05) .Conclusions Wet hyperthermia combined with microwave ablation os more effective than single wet hyperthermia or microwave ablation as a mono -therapy alone on the function of Tregs , which may be closely related to the enhanced suppression of tumor growth .
3.The effect of microwave ablation combined with dendritic cell tumor vaccine treatment on mouse mammary carcinoma and its immunological mechanism

Objective To investigate the therapeutic effects of microwave ablation combined with dendritic cell (DC) tumor vaccine on a murine breast cancer model and to explore its immunological mechanism.Methods Forty-eight Balb/c mice were used to establish models of mouse mammary carcinoma,and then randomly divided into a sham-operation group,a microwave ablation treated group,a DC tumor vaccinated group and a microwave ablation plus DC tumor vaccinated group.Microwave ablation was conducted with the microwave ablation group on day 14 after establishing the model.DC tumor vaccination was performed on day 7 in the vaccinated group.In the microwave ablation plus DC tumor vaccinated group,DC tumor vaccination and microwave ablation were consecutively performed on days 7 and 14,respectively.Tumor growth,lung metastasis and survival were observed.The proportion of CD3+ and CD4+ T cells in splenocytes were analyzed by fluorescence Activated Cell Sorter (FACS) on the 14th day after microwave ablation.Serum levels of interferon-γ (IFN-γ) and interleukin-4 (IL-4) were also detected using ELISA.Results The survival time (56.4 ± 4.3 days),tumor size (67.3 ± 6.1 mm2) and lung metastasis index (0.6 ± 0.4) in the microwave ablation plus DC tumor vaccinated group were all significantly lower than the corresponding averages in the microwave ablation and DC tumor vaccinated group.The proportions of splenic CD3 + T cells (27.8 ±10.3 %) and CD4+ T cells (16.7 ± 8.3 %) in the microwave ablation plus DC tumor vaccinated group were significantly larger than in the control group.Average serum levels of IFN-γ (41.8 ± 22.9 pg/ml) in the microwave ablation plus DC tumor vaccinated group were also significantly larger than in the control group,while the average serum level of IL-4 (15.4± 6.4 pg/ml) was significantly lower.Conclusions Microwave ablation and DC tumor vaccination together could have synergistic effects in treating experimental breast cancer,which might be closely related to the enhanced immune response.

5.Relationship between condition of hypoxia and prognosis in patients with renal clear cell carcinoma

Objective To explore the relationship between condition of hypoxia and prognosis in patient with renal clear cell carcinoma (RCCC).Methods The expression of hypoxia-inducible factor-lo( HIF-1 α) protein in cancer tissue of 89 RCCC cases was examined by streptavidin-biotin complex immunohistochemistry.Clinical and pathological data and prognosis of 89 cases were analyzed retrospectively.There were 66 males and 23 females,with an average age of 57 years.The patients were divided into two groups,the chronic pulmonary disease (CPD) group ( 19 cases) and non CPD (NCPD) group (70 cases),with 46cases in clinical stage I,15 cases in stage Ⅱ,26 cases in stage Ⅲ,and 2 cases in stage Ⅳ.The relationship between survival time and clinicopathological variables including the presence of CPD,the positive rate of HIF-1α protein,smoking history and hemoglobin level were evaluated by the Kaplan-Meier method.And the Cox proportional hazards regression model was build to analyze the correlation between each variable and survival time.Results The 89 cases were followed up for a median time of 19 months (6 to 84 months).Twenty cases died,and 69 cases survived.Between the CPD group and NCPD group,clinical stage,hemoglobin level and the degree of expression of HIF-1 α were significantly different (P < 0.05 ).The median survival time was 44 and 71 months in CPD group and NCPD group,respectively,and the difference was significant ( P < 0.05 ).The median survival time was 43 and 70 months in Hb≤ 110 g/L group and Hb > 110 g/L group,respectively,and the difference was significant ( P < 0.05).The stronger the degree of expression of HIF-lα was,the shorter the overall survival was.And the difference was significant ( P <0.05 ).Associated with CPD,hemoglobin level,the expression of H1F-1α were independent factors affecting overall survival of the patients (P <0.05 ).CPD and HIF-1 α expression were positively related to disease-free survival time,and hemoglobin level was negatively related to disease-free survival time.Conclusions Systemic hypoxia caused by CPD may aggravate the hypoxie state in the organization of the patients with RCCC.The condition of hypoxia and prognosis in patient with RCCC is negatively correlated.
6.Sophoridine inhibits NF-kappaB signaling pathway activation in kidney tissue of endotoxemia mice.

This study is to investigate the effects of sophoridine on NF-kappaB signaling pathway in kidney tissue of endotoxemia mice and the mechanism involved. BALB/c mice were challenged with lipopolysaccharide (LPS) caudal vein injection, then sophoridine was administered by intraperitoneal injection. Totally 50 mice were divided into 5 groups: control group, LPS model group, sophoridine treatment 12 mg x kg(-1) group, 6 mg x kg(-1) group and 3 mg x kg(-1) group. All animals were sacrificed at 6 hours after treatment. Kidney and blood samples were harvested. IKKbeta mRNA and TNF-alpha mRNA expression of renal tissue was measured by the reverse transcription polymerase chain reaction (RT-PCR), and phosphorylation IKKbeta protein (pIKKbeta) was detected by immunohistochemistry. NF-kappaB P65 protein expression and distribution of renal tissue were observed by Western blotting and immunofluorescence laser confocal microscopy. Serum TNF-alpha level was detected by radioimmunoassay. The results showed that the sophoridine significantly reduced the expression of IKKbeta mRNA and pIKKbeta protein, and inhibited the expression of NF-kappaB P65 protein and decreased the entry nuclear rate of NF-kappaB P65 in the renal tissue of endotoxemia mice. Thereby the renal TNF-alpha mRNA expression and serum TNF-alpha level were significantly reduced. These results suggest that sophoridine could inhibit inflammatory reaction induced by LPS through inhibiting activation of NF-kappaB signaling pathway.
