1.The effect of medical assessment on the rehabilitation training of autism spectrum disorder children

Objective To explore the effect of medical assessment on the rehabilitation training of children with autism spectrum disorders (ASD). Methods There were 67 children with ASD selected from special education schools .Their developmental level , ability of social life and social communication were evaluated by child health care physicians .There were 48 children classified as intervention group:individualized rehabilitation training targets were designed based on the assessment results and individualized rehabilitation training program was carried out .The other 19 children were served as controls and routine rehabilitation training was carried out .All children were reevaluated by physicians with the same methods after training for one year . Results The adaptability , language level and communication ability of children in the intervention group improved better than the control group , with statistically significant difference. Conclusion It is beneficial to develop training programs for children with ASD on the basis of medical assessment which can promote the development level of ASD children , social life ability and communication ability .
2.Effect of high glucose on the expression of transcription factor Scleraxis in periodontal ligament cells in vitro.

OBJECTIVETo approach the mechanisms of effects of high glucose on the differentiation of periodontal ligament cells(PDLC) by investigating the changes of Scleraxis mRNA expression in high glucose condition in vitro.
METHODSHuman PDLC were cultured in high glucose medium (4500 mg/L glucose) and normal glucose medium (1000 mg/L glucose), respectively. High glucose was used to inhibit the osteogenic differentiation of PDLC. PDLC cultured in normal glucose medium served as control. Alkaline phosphatase (ALP) activity, the early parameter of osteogenetic differentiation of cells and the expression of Scleraxis mRNA were detected in each group. ALP activity was measured colorimetrically by using nitrophenyl phosphate as a substrate and the expression of Scleraxis mRNA was analyzed by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR).
RESULTSALP activity of PDLC was lower in high glucose medium than in normal glucose medium, and the values were 0.113 +/- 0.068 and 0.218 +/- 0.012, respectively. However, the level of Scleraxis mRNA was quite higher in high glucose medium compared with in normal glucose medium, and the values were 0.973 +/- 0.055 and 0.611 +/- 0.205, respectively. The values of ALP activity and the expression of Scleraxis mRNA were significantly different between the two groups.
CONCLUSIONSHigh glucose inhibited osteogenetic differentiation of PDLC and up-regulated Scleraxis expression. The adverse changes of Scleraxis expression and osteogenic differentiation of PDLC suggest that Scleraxis may regulate the osteogenic differentiation of PDLC negatively and the inhibition of high glucose on osteogenetic differentiation of PDLC may be regulated by Scleraxis in transcription level.

OBJECTIVETo provide reference for the safe needling depth of Dazhui (GV 14), Jianzhongshu (SI 15), Xuanshu (GV 5) and Mingmen (GV 4) in clinical acupuncture and moxibustion treatment.
METHODSThirty-two adult volunteers were divided into 3 groups, thin person group, moderate person group and fat person group according to Luo's indexes, and computer-aided tomography was used to measure the needling depth of Dazhui (GV 14), Jianzhongshu (SI 15), Xuanshu (GV 5) and Mingmen (GV 4).
RESULTSThe safe depths of perpendicular needling were different for persons of different somatotypes, for example the needling depth for Dazhui (GV 14) was (32.86 +/- 3.96) mm for the thin person group, (37.76 +/- 4.91) mm for the moderate person group, and (47.93 +/- 5.30) mm for the fat person group.

4.Study on needling depth of Fengfu (GV 16) with CT.

OBJECTIVETo study on needling safe depth of Fengfu (GV 16) with CT, so as to provide reference for safe needling depth of Fengfu (GV 16) in clinical acupuncture treatment.
METHODSForty-one adult volunteers were divided into 3 groups, a thin person group, a moderate person group and a fat person group according to Luo's indexes, and computer-aided tomography (CT) was used to measure the needling depth of Fengfu (GV 16).
RESULTSThe safe depths of perpendicular needling of Fengfu (GV 16) were different for persons of different somatotypes. The safe needling depth was (27.73 +/- 3.45) mm for the thin person group, (30.78 +/- 2.90) mm for the moderate person group, and (33.39 +/- 4.27) mm for the fat person group.
CONCLUSIONThe safe needling depth < or = the dangerous depth x 75% can be used for reference for the safe needling depth of Fengfu (GV 16) for different somatotypes persons.

5.Efficacy and safety of paroxetine in treatment of post-stroke depression and anxiety

Objective To investigate the efficacy and safety of paroxetine in the treatment of post-stroke depression and anxiety.Methods Patients with post-stroke depression and anxiety (312 cases) were randomly divided into observation and control groups.All patients were given routine,symptomatic,supportive treatment and rehabilitation training,psychological intervention,in addition,the observation group was treated with paroxetine hydrochloride.The Hamilton Depression Scale (HAMD) score,anxiety scale (HAMA) score,plasma substance P (SP),neuropeptide Y (NPY),cortisol (Cor) content and side effects scale (TESS) score of both groups before and after treatment were analyzed and compared.Results After treatment,the HAMD and HAMA scores of two groups were significantly decreased,and those of the observation group was significantly lower than the control group (P < 0.05).The clinical curative effect of the observation group was better than that of the control group (P < 0.05).In both groups,the levels of SP and Cor were significantly decreased,NPY was significantly increased,and the improvement of the observation group was higher than that of the control group (P < 0.05).In the observation group,some patients had anorexia,nausea,dizziness,insomnia and other reactions,but they were tolerant,and there was no significant difference in the TESS score between the observation group and control group.Conclusion Paroxetine treatment of post-stroke depression/anxiety can reduce depression and anxiety symptoms,improve neuropeptide and cortisol levels,and have good curative effect and good safety.It is worthy of clinical application and further study.
6.MACF1 knockdown in glioblastoma multiforme cells increases temozolomide-induced cytotoxicity.

OBJECTIVETo investigate the role of microtubule-actin crosslinking factor 1 (MACF1) in the response of glioma cells to temozolomide (TMZ).
METHODSTMZ was applied to a human gliomablastoma cell line (U87) and changes in the protein expression and cellular localization were determined with Western blot, RT-PCR, and immunofluorescence. The responses of the cells with MACF1 expression knockdown by RNA interference to TMZ were assessed. TMZ-induced effects on MACF1 expression were also assessed by immunohistochemistry in a nude mouse model bearing human glioblastoma xenografts.
RESULTSTMZ resulted in significantly increased MACF1 expression (by about 2 folds) and changes in its localization in the gliomablastoma cells both in vitro and in vivo (P<0.01). Knockdown of MACF1 reduced the proliferation (by 45%) of human glioma cell lines treated with TMZ (P<0.01). TMZ-induced changes in MACF1 expression was accompanied by cytoskeletal rearrangement.
CONCLUSIONMACF1 may be a potential therapeutic target for glioblastoma.
7.A five-year follow-up study of hypothyroidism in areas with different iodine intakes

Objective To investigate epidemiologieal characteristics of hypothyroidism and factors influencing its outcome in the areas with different iodine intakes.Methods An epidemiologic follow-up study of thyroid diseases was performed in Panshan (iodine deficient),Zhangwu [mild iodine deficiency before universal salt iodination (USI) and more than adequate iodine intakes after USI ] and Huanghua ( iodine excessive) in China.A total of 3 761 subjects were investigated in 1999,and 3 018 (80.2% ) of them were followed up in 2004.FT_4,FT_3,TSH,thyroid autoantibodies and urinary iodine concentration were determined and thyroid B ultrasound was performed in all participants.Results Five-year cumulative incidence of overt hypothyroidism was 0.23%,0.47% and 0.35% in Panshan,Zhangwu and Huanghua,respectively.Twenty patients with autoimmune- induced overt hypothyroidism did not comply with the physician's orders to take thyroxine.Thyroid function spontaneously turned to normal in 55% of them,turned to subclinical hypothyroidism in 20% and maintained overt hypothyroidism in 25%.Five-year cumulative incidence of subclinical hypothyroidism in Zhangwu (2.60%) and Huanghua (2.89%) were significantly higher than that in Panshan(0.23% ) (both P<0.01 ).One hundred patients with subclinical hypothyroidism were followed up,and 5% of them developed overt hypothyroidism,66% turned to euthyroidism and 29% maintained subclinical hypothyroidism.Raised serum TSH (>6 mU/L) in the original survey ( OR = 3.4),positive thyroid autoantibodies in the follow-up study ( OR = 5.3 ) and more than adequate iodine supplementation in the mild iodine-deficient population ( OR = 8.0) were risk factors influencing outcome of subclinical hypothyroidism.Conclusion More than adequate iodine supplementation among a population in the mildly iodine-deficient area may increase the incidence of overt and subolinical hypothyroidism, and also influence outcome of subclinical hypothyroidism.

Objective To evaluate the relationship between iodine intake level and the occurrence and development of thyroid diseases.Methods During the period of 1999-2004,a cross-sectional survey in 1999 and a 5-year follow-up survey were conducted in the 3 communities with different iodine intake levels in China:Panshan (mild deficiency),Zhangwu (more than adequate) and Huanghua (iodine excess).A total of 3 761 subjects were included in 1999 and 80.24% of them participated in the 5-year follow-up survey in 2004.The levels of serum thyroid hormones and thyroid autoantibodies were measured,and thyroid B ultrasound was performed.Results (1) The prevalence of overt hypothyroidism in 1999 in Zhangwu and Huanghua was 3.5 and 7.3 times,and that of subclinical hypothyroidism was 3.2 and 6.6 times as high as in Panshan,respectively.The 5-year cumulative incidence of subclinical hypothyroidism was 11.3 and 12.6 times as high as in Panshan,but no difference of incidence of overt hypothyroidism was found.(2) The prevalence of autoimmune thyroiditis (AIT) in 1999 in Zhangwu and Huanghua was 3.8 and 6.2 times,and the S-year cumulative incidence of AIT was 4.4 and 5.5 times as high as in Panshan.(3) During the 5-year follow-up period,the incidences of hypothyroidism in subjects with positive thyroid peroxidase antibody and euthyreidism in Zhangwu and Huanghua were 4.2 and 10.3 times as high as in Panshan.(4) No significant differences in both prevalence in 1999 and 5-year cumulative incidence of overt hyperthyroidism were found in the 3 communities.(5) The incidence of thyroid cancer was 19.37/100 000 per year in Huanghua,and all of the cases were papillary thyroid cancer,and no thyroid cancer was found in Zhangwu and Huanghua.Conclusion More than adequate or excessive dietary iodine intake may induce and promote the occurrence and development of hypothyroidism and AIT,therefore,median urinary iodine excretion (MUI)more than 200?g/L in the population appears not safe.The observation that a higher incidence of thyroid cancer occurred in the iodine excessive areas with MUI 600?g/L deserves further investigation.
9.Tissue culture of safflower and analysis of secondary metabolites in suspension cells.

Safflower(Carthamus tinctorius), a valuable traditional Chinese medicinal plant, has attracted much attention in recent years. This study established a stable tissue culture system of safflower and analyzed the chromatogram of its secondary metabolites, providing high-quality experimental materials for further research on natural products in safflower. The calluses were established from the safflower seeds germinated in a sterile environment, and then they were differentiated into the aseptic seedlings, or cultured to obtain suspension cells in liquid medium. The ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS), Progenesis QI, and principal component analysis(PCA) were used to detect and analyze the secondary metabolites in the suspension cells before and after induction with different elicitors(methyl jasmonate, silver nitrate, salicylic acid and yeast extract). A total of 23 secondary metabolites including flavonoids, phenylpropanoids, alkaloids, fatty acids and aromatic glycosides were detected in safflower suspension cells. In response to the four elicitors, 11 compounds showed increased or decreased relative content. The results indicate that different elicitors have various effects on the accumulation of secondary metabolites in safflower suspension cells, and yeast extract shows more obvious positive induction. Therefore, different elicitors may play a role in the expression of related genes in the biosynthetic pathway of specific secondary metabolites. The results facilitate the discovery of targeted elicitors and the large-scale production of valuable secondary metabolites in the future.

10.Discussion on research idea of quality marker of Salvia miltiorrhiza based on biosynthetic pathway of tanshinone compounds.

Based on the theory of Q-marker, the hairy root of Salvia miltiorrhiza and S. miltiorrhiza in many provinces were studied. The relative expressions of SmCPS, SmKSL and CYP76AH1 genes in hairy roots were detected by real-time fluorescence quantitative PCR and the contents of tanshinoneⅡ_A, cryptotanshinone, tanshinoneⅠ, 1,2-dihydrotanshinone, ferruginol and miltiradiene were detected by UPLC and GC-MS, respectively. Statistical analysis shows as fllows: in the hairy root of S. miltiorrhiza, the content of miltiradiene and ferruginol is positively correlated with the content of tanshinone compounds in the downstream, and the relative expression of important genes in the biosynthetic pathway of tanshinone can reflect the content of tanshinone compounds to a certain extent; in many provinces of S. miltiorrhiza, the content of ferruginol and tanshinone compounds can also be found that there is a positive correlation between the contents. Based on the biosynthetic pathway of tanshinone compounds, which is a special index component in S. miltiorrhiza, this study focused on the important relationship between the upstream gene, the middle intermediate compound and the downstream tanshinone compound content of the biosynthetic pathway, and explored the possible research ideas of improving the quality marker system of S. miltiorrhiza, and then provided the possible research ideas for understanding and studying the quality marker of traditional Chinese medicine from the biosynthetic pathway.
