1. Effect of gene silencing of Golgi a-mannosidase . on the invasion and migration of human gastric carcinoma BGC-823 cells

Objective: To investigate the effect of gene silencing of Golgi a-mannosidase II(GM II) by RNA interference on the invasion and migration of human gastric carcinoma BGC-823 cells. Methods: Four targeted locations were designed and four plasmid vectors expressing short hairpin RNA (shRNA) for gene silencing of GM II were constructed, and the negative control plasmid vector was also constructed. The plasmids were transfected into human gastric carcinoma BGC-823 cells by Lipofectamine 2000. The expression levels of GMIImRNA and protein were detected by RT-PCR and Western blotting, respectively. The best plasmid for gene silencing of GMIIwas selected. The invasive and migration capabilities of BGC-823 cells induced by gene silencing of GMIIwere detected by Transwell invasion assay and scratch-wound assay, respectively. Results: Plasmid vector pGPU6/GFP/Neo-1303 was the best one for the gene silencing of GM .. The number of cells invading through the matrigel in the plasmid vector pGPU6/GFP/Neo-1303-transfected group was much lower than those in the blank control group and the pGPU6/GFP/Neo-shNC-transfected group (P < 0.05). After the cells were cultured without serum for 24 h, the invasive and migration capabilities of the cells transfected with pGPU6/GFP/Neo-1303 were significantly weakened than those of the blank control group and the pGPU6/GFP/Neo-shNC- transfected group (P < 0.05). Conclusion: The expressions of GMIImRNA and protein in human gastric carcinoma BGC-823 cells can be silenced by RNA interference, which results in the inhibition of invasive and migration capabilities of BGC-823 cells. These results suggest that GMIImight be a novel target for the treatment of human gastric carcinoma. © 2012 by Tumor.
2.Gastrointestinal stromal tumors:imaging features with intraoperative and clinicopathological correlation

Objective To analyze imaging of gastrointestinal stromal tumors(GIST),and to compare their imaging features with operational and pathological findings.Methods Clinical,imaging,and pathological data of 20 patients with GIST were collected.Results Imaging findings were endophytic or exophytic tumors with heterogeneous density or signal intensity,corresponding to hemorrhage,necrosis,and cystic changes.Imaging was correct for the location of the lesion in 11 of 16 primary GIST and 4 cases of relapsed tumors.Preoperational CT did not detect mesenteric,peritoneal,and omental metastasis in 5 cases. Hepatic metastases detected at CT (3 cases )were identified by operational findings.Conclusions GIST has some imaging features.CT is a useful tool in detecting and characterizating of lesions rather than detecting mesenteric,peritoneal,and omental metastasis.
3.No relation between ACE-I/D polymorphism and high altitude pulmonary edema in the Han Chinese.

OBJECTIVESTo explore whether the angiotensin I -converting enzyme (ACE) I/D (insertion/ deletion) polymorphism is associated with the susceptibility to high altitude pulmonary edema (HAPE) in the Han Chinese.
METHODSOne hundred and forty-seven HAPE-p (HAPE patients) and 193 HAPE-r (HAPE resistants) were enrolled from the Yushu earthquake reconstruction workers in Qinghai province where the altitude is over 3 500 m above sea level. Blood samples were collected from each of the HAPE-p and HAPE-r groups. Information about physiological phenotypes was obtained via fieldwork investigation. The ACE-I/D polymorphism in HAPE-p and HAPE-r was detected by polymerase chain reaction (PCR).
RESULTSThe SaO2 was significantly lower while HR was significantly higher in HAPE-p group than those in HAPE-r group. The genotype frequencies of ACE-I/D for II, ID, DD in HAPE-r and HAPE-p groups were 0.430, 0.446, 0.124 and 0.435, 0.469, 0.095, respectively, the allelic frequencies of I and D were 0.650, 0.350 and 0.670, 0.330, respectively. The OR of ID, DD and D alleles relative to II for HAPE was 0.961 (0.610-1.514), 1.322 (0.634-2.758) and 1.080 (0.783-1.489). There was no significant difference of the genotypic and the allelic frequencies in ACE-I/D polymorphism between HAPE-p and HAPE-r groups.
CONCLUSIONSThere is no relation between ACE-I/D polymorphism and HAPE in the Han Chinese.

4.Evaluation of right ventricular volume and systolic function by realtime three-dimensional echocardiography.

The optimal plane for measurement of the right ventricular (RV) volumes by real-time three-dimensional echocardiography (RT3DE) was determined and the feasibility and accuracy of RT3DE in studying RV systolic function was assessed. RV "Full volume" images were acquired by RT3DE in 22 healthy subjects. RV end-diastolic volumes (RVEDV) and end-systolic volumes (RVESV) were outlined using apical biplane, 4-plane, 8-plane, 16-plane offline separately. RVSV and RVEF were calculated. Meanwhile tricuspid annual systolic excursion (TASE) was measured by M-mode echo. LVSV was outlined by 2-D echo according to the biplane Simpson's rule. The results showed: (1) There was a good correlation between RVSV measured from series planes and LVSV from 2-D echo (r = 0.73; r = 0.69; r = 0.63; r = 0.66, P < O. 25-0. 0025); (2) There were significant differences between RVEDV in biplane and those in 4-, 8-, 16-plane (P < 0.001). There was also difference between RV volume in 4-plane and that in 8-plane (P < 0.05), but there was no significant difference between RV volume in 8-plane and that in 16-plane (P > 0.05); (3) Inter-observers and intro-observers variability analysis showed that there were close agreements and relations for RV volumes (r = 0.986, P < 0.001; r = 0.93, P < 0.001); (4) There was a significantly positive correlation of TASE to RVSV and RVEF from RT3DE (r = 0.83; r = 0.90). So RV volume measures with RT3DE are rapid, accurate and reproducible. In view of RV's complex shape, apical 8-plane method is better in clinical use. It may allow early detection of RV systolic function.

5.Relationship between Childhood Abuse Experience,Parent Attachment and Depression in Adolescents

Objective To explore the relationship between childhood abuse experience,parent and peer attachment and depression in adolescents.Methods A total of 736 junior students from the 7th,8th and 9th grades of 2 public schools in Xinxiang City and Jiyuan City Henan province completed the questionnaires.Childhood Trauma Questionnaire(CTQ),Inventory of Parent and Peer Attachment(IPPA) and Depression Self-Rating Scale for Children(DSRSC) were employed the childhood abuse experience,parent attachment quality and depressive mood in adolescents,respectively.The hierarchical regression analysis was used to examine the relationship between parent and peer attachment,child abused experience and depression.Results Male participants reported high score of childhood abused than female participants(33.29±7.89 and 31.05±6.01,t=4.343,P=0.000).There were no significantly gender difference in mother attachment(male:59.89±15.86,female:61.45±16.48,t=-1.303,P=0.193),father attachment(male:59.32±17.17,female:60.28±16.95,t=-0.764,P=0.445) and depression(male:7.68±5.27,female:7.31±4.21,t=1.057,P=0.291).Childhood abuse had a positive correlation with adolescent depression(r=0.408,P=0.000),which negatively correlated with mother and father attachment(r=-0.544,-0.417,respectively,Pa=0.000).There was a positive correlation between mother attachment and father attachment(r=0.604,P=0.000),and both of them had negative correlation with depressive disorder(both r=-0.426,Pa=0.000).Mother and father attachment were the partial mediation of the relationship between childhood abuse and adolescent depression.Conclusions Male adolescents suffered more maltreatment than female adolescents.There are no gender difference in parent attachment and depression.Parent attachment plays the partial mediator role in childhood abuse and adolescent depression.Results reveal the importance of assessment and consideration of parent attachment in the intervention of depression in abused adolescents.
6.Expression of matrix metalloproteinase 1 and tissue inhibitor of metalloproteinase 1 in biopsy liver from patients with fibrosing cholestatic hepatitis

Objective To explore the fibrosis mechanism of patients with fibrosing cholestatic hepatitis(FCH) in the way of degradation of collagen.Methods The expressions of matrix metalloproteinase 1(MMP1) and tissue inhibitor of metalloproteinase 1(TIMP1),and contents of type I,III collagen proteins were detected by immunohistochemical staining in the liver tissues of 9 cases with FCH associated with HBV developed following renal transplantation and 5 cases without liver disease as controls.Results The expressions of MMP1 and TIMP1,and type I,III collagen proteins in the patients with FCH were significantly higher than those in the control group.There was a positive correlation between the expressions of type I,III collagen proteins and TIMP1/ MMP1 ratio.Conclusion Hepatic fibrosis in the patients with FCH associated with HBV developed following renal transplantation may be relative to the increase of TIMP1 expression which inhibit the degradation of collagen.
7.Expression of collagen type Ⅰ, Ⅲ, MMP1, and TIMP1 in liver tissues of patients with chronic hepatitis B

Objective To observe the expressions of matrix metalloproteinase 1(MMP1) and tissue inhibitor of matrix metalloproteinase1(TIMP1) and the collagen type Ⅰ,Ⅲ deposition in the liver tissues,and evaluate the possible fibrosis mechanism of patients with chronic hepatitis B(CHB) in the way of degradation of collagen. Methods The specimens of the biopsy liver in 50 cases with CHB were detected for the expressions of type Ⅰ,Ⅲ collagen proteins,MMP1 and TIMP1 by immunohistochemical staining. Results The expressions of type Ⅰ,Ⅲ collagen proteins and TIMP1,were significantly increased along with the advancing of hepatic fibrosis.There was a positive correlation between the expressions of type Ⅰ,Ⅲ collagen proteins and TIMP1. Conclusions Hepatic fibrosis in the patients with CHB may be related to increase of TIMP1 expression that inhibit the degradation of collagen.

9.Relationship between MspⅠ polymorphism of CYP1A1 gene and susceptibility to breast cancer in Yi nationality in Yunnan province

Objective To explore the relationship between MspⅠpolymorphism of CYP1A1 gene and susceptibility to breast cancer in Yi nationality in Yunnan province. Methods The gene polymorphism of restriction enzyme of 3′-terminal of CYP1A1 was detected by PCR-RFLP in 60 healthy Yi women , 51 Yi women with breast cancer, 235 healthy Han women, and 250 Han women with breast cancer. Results The distribution frequency of CYP1A1-MspⅠgenotypes was significantly higher in Yi women with breast cancer (51.0%) than in the healthy Yi women (33.3%) (P < 0.05), an the allele C had a higher frequency in women with breast cancer than in healthy controls (P < 0.05). Significant differences of frequencies were found in genotypes TT, TC and CC between women with breast cancer and healthy controls (P < 0.05). The risks of TC and CC for breast cancer were 1.19 and 1.95 folds respectively to TT genotype. But as compared with Yi women , the distribution frequency of CYP1A1-MspⅠ genotypes did not differ between Han women with breast cancer and in the healthy control (P > 0.05), and there were no differences in three genotype frequencies (P > 0.05). Conclusions Gene polymorphism of CYP1A1 genotypes may be associated with the risk of breast cancer in Yi nationality but not in Han nationality in Yunnan. The mutation of CYP1A1 gene may increase the incidence of breast cancer in Yi nationality.

Lots of literature showed that biotechnics played important roles in the quality identification of Chinese medicinal herbs and the researches on new drugs of TCM.
