1.Protective Effect of Gingko Biloba Extract on Acute Lung Hemorrhage Induced by Lipopolysaccharide in Newborn Rats

Objective To investigate the protective effect of Gingko Biloba extract (GBE) on acute lung hemorrhage induced by Lipopolysaccharide(LPS) in newborn rats. Methods 1. Acute lung hemorrhage models were reproduced by intraperitoneal injection with LPS (5 mg/kg). 2. Thirty two rats were randomly divided into 4 groups,GBE groups (4 mg/kg,8 mg/kg, 16 mg/kg) and LPS group 5 mg/kg. Results In group LPS, extensive lung hemorrhage was observed after 4 hours of injection . TNF - ? iung content was obvious in LPS group. The expression of lung nuclear factor(NF-kB )immunohistochemistry wasobvious. While the parameters were obviously attenuated by GBE before LPS. Conclusion GBE may be useful in the treatment of acute pulmonary inflammatory disease.
2.Quantitative changes of peripheral blood dendritic cells in patients with different degrees of cerebrovascular stenosis

Objective To study the changes of proportion of peripheral blood dendritic cells (DCs) in patients with different degrees of cerebrovascular stenosis and after stenting,and explore the relation between DCs level and cercbrovascular stenosis. Methods Sixty-three patients,admitted to our hospital from November 2009 to February 2010 and from August 2010 to December 2010,were divided into control group (CG,n=9),mild/moderate stenosis group (MsG,n=17),severe stenosis group (SsG,n=30) and stent implanted group (SiG,n=7) according the results of digital subtraction angiography (DSA) and their clinical situation.Flow cytometry 4-color analysis was employed to detect the proportion of DCs subtypes in the peripheral blood. Results The percentage of peripheral blood myeloid dendritic cells (mDCs) was significantly different in the above groups (P=0.000):that between CG and SsG,and that between MsG and SsG were significantly different (P<0.05).Whereas, no obvious difference was founded in plasmacytoid dendritic cells (pDCs) between each 2 groups (P=0.065).Conclusion The percentage of mDCs suggests the alterations of different degrees of cerebrovascular stenosis:the severer the stenosis,the lower the percentage of mDCs; stent implantation will not affect the distribution of DCs subtypes
3.Determination of eleven major components and fingerprint chromatography for Reduning injection by UPLC.

A reliable method for simultaneous determinition of eleven representative components (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C, shanzhiside, geniposidic acid, genipin-1-β-D-gentiobioside, geniposide and secoxyloganin) in combination of chromatographic fingerpint analysis for Reduning injection was developed by ultra high-performance liquid chromatography (UPLC). The method was performed on an Agilent ZORBAX SB-C18 anlytical column (3. 0 mm x 100 mm, 1. 8 µm) with a guard column of Agilent UPLC Guard ZORBAX SB-C18 (3.0 mm x 5 mm) at the column temperature of 30 °C. The gradient mobile phase consisted of acetonitrile (A)-0. 1% phosphoric acid (B) with a flow rate of 0. 4 mL . min-1. The injection volumn was 2 µL. The detection wavelengths were set at 324 nm and 238 nm for quantit tive analysis and 225 nm for fingerpint chromatography. Neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C, shanzhiside, geniposidic acid, genipin-1-β-D-gentiobioside, geniposide and secoxyloganin were baseline seperated with good linearity relationships (r >0. 999) between concentration and peak areas over the linear ranges. The average recoverys of the investigated compounds were 103.5%, 100. 2%, 103. 3%, 102. 8%, 101. 3%, 102. 8%, 97. 36%, 99. 62%, 98. 16%, 102. 8%, 99. 27%, respectively. Reduning injection of forty-five batches was analyzed by UPLC finge print chromatography. Thirty batches were selected to generate the reference fringerprint chromatography with fourteen common peaks. The similarity values between the reference fringerprint chromatography and the remaining fifteen batches were higher than 0. 99. The developed method was fast, accurate and sensitive. It could be used as a reference for the quality control of multiple components determination and fingerprint chromatography for Reduning injection in future.

4.Analysis on the current situation of neglected rural children aged 0-6 vears and its impact factors in the western areas of China

Objective To learn the current neglected situation and its impact factors on rural children in two provinces in Western China.Methods The investigation was conducted by using multistage stratified cluster sampling method in some parts of the Shaanxi province and the city of Chongqing.Results 1488 subjects,aged from 0 to 6,were recruited in the present study.Results showed that the total prevalence rates of neglected rural children in the two research sites were 31.59% and 48.32 respectively.No significant difference was found on the prevalence of neglected for boys and girls ( x2 =0.86,U=1.51,P > 0.05 ).The degrees of negligence in the older children showed a significant increase than in the younger children (x2 =13.36,F=33.45,P< 0.05 ).The prevalence and degree of negligence in families with three generations were lower than that of a single-parent families and remarried families (H=10.03,F=2.83,P<0.05).The one-child-families were significantly lower than multiple children families,both in terms of prevalence and degree of negligence.Our data demonstrated the degrees of negligence in children whose parents were away from home,were higher than the children whose parents stayed with them ( x2=30.30,U=6.76,P<0.05).The degree of negligence in Shaanxi was lower than that in Chongqing (x2=9.42,U=8.40,P<0.05).Based on multivariate non-conditional logistic regression analysis,the determinants of stunting among the stranded children were as follows:parents were away from home (OR=1.54,95% CI:1.20-1.97) ;structure of the family (OR=1.38,95% CI:1.16-1.65) ; father' s occupation (OR=0.87,95% CI:0.78-0.97) ; schooling of the children' s mother (OR=1.27,95%CI:1.07-1.52) ; relationship between children and their fathers (OR =1.43,95% CI:1.07-1.91 ) etc.Conclusion The situation of negligence in children living in the rural areas,were serious in Shaanxi and Chongqing provinces,which called for the strengthening on the publicity and education of the issue.School and the society should also pay more attention to this problem.
5.Results of accelerated trans - epithelial corneal collagen cross-linking protocol for the treatment of progressive keratoconus

AIM: To report the long- term clinical outcomes of accelerated trans-epithelial corneal cross-linking ( CXL ) protocols using KXL System ( Avedro, USA ) in the treatment of progressive keratoconus. · METHODS: Totally 52 patients ( 102 eyes ) with progressive keratoconus between December 2014 and February 2017 [ maximum keratometry values ( Kmax) ≤60.0D, minimum corneal thickness(Thk) ≥400m] were treated with an accelerate trans-epithelial CXL protocol (UV-A irradiation intensity 45mW/cm2 with a total fluence of 7. 2J/cm2 ) using KXL system ( Avedro, USA ) in Southwest Hospital. The average follow-up time was 11. 65mo (range: 9-26mo). Uncorrected distance visual acuity ( UDVA) , corrected distance visual acuity ( CDVA) , intra- ocular pressure ( IOP ) , slit-lamp microscope examination, Kmax and average keratometry values ( AveK ) , corneal stromal demarcation line depth and endothelial cell density ( ECD) were evaluated. ·RESULTS:The 52 patients (102 eyes) were included in this research, male 36 (70 eyes) and female 16 (32 eyes), average age was 19. 5±4. 6 years. Preoperative CDVA was 0. 84±0. 89 (LogMAR), postoperative CDVA was 0. 69±0. 72 ( P = 0. 398 ). Preoperative UDVA was 1. 02 ± 0. 62 (LogMAR), postoperative UDVA was 0. 85 ± 0. 59 ( P =0. 154 ). Preoperative IOP was 12. 95 ± 4. 40mmHg, postoperative IOP was 11.92±3. 66mmHg (P=0. 272). No statistical difference (P=0. 552) has been found between preoperative and postoperative ECD. Nevertheless, on the Sirius anterior system ( Sirius, CSO, Itlay) , significant statistical difference (P=0. 017) was confirmed between preoperative Kmax ( 50. 83 ± 3. 48D ) and postoperative Kmax (52. 05±3. 63D). Meanwhile, the postoperative Avek (47.74±2. 51D) was significantly lower (P=0. 041) than the preoperative Avek ( 48. 73 ± 4. 33D ). The average corneal stromal demarcation line depth ( 192 ± 23. 6μm ) was detected by the anterior segment OCT. No statistical difference ( P = 0. 816 ) has been found between preoperative and postoperative Thk. No severe complication was observed in all cases. ·CONCLUSION: Accelerated trans-epithelial CXL was effective in decreasing keratometry values for progressive keratoconus in this research, and the outcomes remained stable during the follow-up time. No endothelium damage or other severe complications were observed in this clinical research. The accelerated trans-epithelial CXL is as effective as the standard CXL.
6.Expressions of myogenic markers in skeletal muscle differentiation of human bone marrow mesenchymal stem cells.

OBJECTIVETo investigate the expressions of myogenic markers MyoD, myogenin,and desmin in skeletal muscle differentiation of human bone marrow mesenchymal stem cells (hBM-MSCs).
METHODSMyogenic markers MyoD, myogenin,and desmin of hBM-MSCs cultured in vitro were detected by immunofluorescence and RT-PCR. A total of 21 8-to-10 week-old immunosuppressed mdx mice were transplanted with 1x107 passage 5 of hBM-MSCs. The mice were euthanized 2-24 weeks after transplantation,and gastrocnemius muscle were analyzed for human MyoD, myogenin,desmin,and dystrophin (Dys) expressions by immunohistochemistry and RT-PCR.
RESULTSThe numbers of MyoD-,myogenin-,and desmin-positive cells per 100 hBM-MSCs were 23.5∓5.3, 30.7∓6.2, and 28.4∓5.7, respectively. MyoD, myogenin, and desmin mRNA was observed in passage 5 of hBM-MSCs. After two weeks of hBM-MSCs transplantation,a small number of MyoD-and myogenin-positive cells were observed in skeletal muscle of mdx mice,and desmin-positive cells were observed 4 weeks after transplantation. Expressions of MyoD and myogenin were detected in the muscle of mdx mice 2-4 weeks after hBM-MSCs transplantation, which reached a peak 12-16 weeks later. Desmin was expressed in the muscle of mdx mice 4-8 weeks after transplantation,with much more expression after 16 weeks of transplantation. A small number of Dys-positive cell and Dys mRNA expression were presented in the muscle of mdx mice 4 and 8 weeks after hBM-MSCs transplantation,respectively. The expression of Dys in the muscle of mdx mice increased gradually after transplantation.
CONCLUSIONhBM-MSCs have the potential of myogenic differentiation in vitro and contribute to myogenic conversion in xenogeneic animal,during which the up-regulation of MyoD and myogenin expressions may play an important role.

7.Relationship between multi-slice spiral CT pulmonary perfusion imaging and the expression of EphB4 and ephrinB2 in non-small cell lung cancer.

OBJECTIVETo investigate the role of the expression of ephrinB2 and EphB4 in non-small cell lung cancer (NSCLC), and their relationship with multi-slice spiral CT pulmonary perfusion imaging.
METHODSThirty-one nodules with pathologically proven NSCLC underwent CT perfusion scan. The perfusion parameters including blood flow (BF), blood volume (BV), peak enhancement image (PEI) were collected. The expression of ephrinB2 and EphB4 in tumor cells and interstitial vasculature were detected by immunohistochemistry. Correlation analysis and trend test were used to assess the relationship between ephrinB2/EphB4 expression and clinicopathological features, and between ephrinB2/EphB4 expression and perfusion parameters.
RESULTSPositive expression of ephrinB2 and EphB4 in the NSCLC group was 83.9% and 71.0%, respectively, significantly higher than that in the internal control group (P < 0.01). The expression of ephrinB2 and EphB4 was consistently in tumor parenchyma but differently in tumor vessels. The expressions of ephrinB2 and EphB4 were positively correlated with lymphatic metastasis (P < 0.05). The expression of EphB4 was negatively correlated with blood flow (BF) and blood volume (BV), respectively (P < 0.05). There was a significant positive correlation between ephrinB2 expression and BF (r = 0.516, P = 0.003), and a positive correlation between ephrinB2 expression and BV (r = 0.448, P = 0.013). The expressions of ephrinB2 and EphB4 were not correlated with PEI (P > 0.05). The values of BF and BV in the high and moderate EphB4 expression groups were significantly decreased compared with that in the negative group (P < 0.01). The value of BF in the high ephrinB2 expression group was significantly increased compared with that in the moderately positive group and negative group (P < 0.01). The value of BV in the high ephrinB2 expression group was significantly increased compared with that in the negative group (P < 0.01).
CONCLUSIONThe CT pulmonary perfusion imaging reflects the density difference of blood vessels with functional lumen, and such difference also depends on the quantity and quality of vasculature with functional lumen.

8.Curative efficacy analysis of allo-HSCT using conditioning regimen of FLAG combined with modified BUCY for refractory and relapsed patients with hematologic malignancies.

This study was aimed to investigate the curative efficacy of allogenetic hemopoietic stem cell transplantation (allo-HSCT) using FLAG and modified BUCY conditioning regimen for patients with refractory and relapse or high risk hematologic malignancies. The therapeutic effect of allo-HSCT with FLAG and modified BUCY conditioning regimen on 10 patients with hematologic malignancies was analyzed. 10 patients included 2 cases of relapse (1 early relapse, 1 progression) and 7 cases of refractory (2 CR(2), 1 CR(3) and 2 PR and 1 NR) and 1 CR(1) with high risk. Among 10 cases 8 cases was diagnosed as AML, 1 case as pro-T lymphoblast lymphoma/leukemia and 1 case as spleen marginal zone lymphoma. The results showed that implantation of all the patients was successful. The relapse-free median survival time of 8 cases was 164 days (57 - 442 days) and survived up to now, 2 cases died (1 case died of pulmonary infection, 1 case died of fungus pulmonitis). In conclusion, the curative efficacy of allo-HSCT using FLAG and modified BUCY conditioning regimen for patients with refractory and relapsed hematologic malignancies is satisfactory.

9.Effect of shRNAmir lentiviral vector-mediated COX-2 silencing on the radiosensitivity of nasopharyngeal carcinoma cells.

OBJECTIVETo evaluate the effect of COX-2 silencing on the radiosensitivity of a nasopharyngeal carcinoma (NPC) cell line C666-1.
METHODSAnti-COX-2 C666-1 cell line with COX-2 gene silencing mediated by shRNAmir lentiviral vector and the control cell line Anti-GL-2 C666-1 were exposed to various radiation doses. The clonogenic survival assay and curve fitting was used to calculate the radiobiological parameters and the sensitization enhancement ratio after the radiation. Cell cycle changes were assessed after the exposure by flow cytometric analysis. In a BALB/c nude mouse model, the growth curve of the xenografts was generated and the tumor growth inhibition rate was calculated.
RESULTSCompared with the control cells, Anti-COX-2 C666-1 cells showed obviously lowered values of SF2, D0 and Dq but significantly increased α/β with a sensitivity enhancement ratio of 1.4014. COX-2 gene silencing increased the inhibition rate of the tumor xenografts after the radiation, and caused also decreased percentage of G2/M arrest resulting from the exposure.
CONCLUSIONStable COX-2 silencing in NPC cells can improve the effect of radiotherapy both in vitro and in vivo. By changing the radiobiological parameters, genetically based COX-2 inhibitor may be a potentially promising radiosensitizer of NPC.

Beclomethasone is an effective glucocorticoid, and beclomethasone-aptamer is a short single-stranded DNA with affinity and specificity to beclomethasone. The interaction between them is still unclear. The study of the interaction between aptamers and beclomethasone has a certain significance for the application of aptamers.
