1.Anti-proliferative and anti-metastatic effects of curcumin on oral cancer cells.

OBJECTIVEThe purpose of this article is to examine the effect of curcumin on the proliferation and metastasis of human tongue squamous cell carcinoma and analyze its mechanism.
METHODSSCC-4 were treated with curcumin of 0, 5, 10, 20, 30, 60, 100 micromol x L(-1) in 24 h. MTT assay, Matrigel invasion assay, flow cytometry and fluorescence microscopy were used to examine the effect of curcumin on the growth and metastasis of SCC-4. cDNA microarray and RT-PCR were employed to analyze the expression of genes treated by curcumin.
RESULTSThe results showed that curcumin could concentration-dependently inhibit SCC-4 cell proliferation at the concentration range from 20 to 100 micromol x L(-1). Furthermore, Matrigel invasion assay indicated that curcumin can reduce SCC-4 cell invasion under the dosage of 20, 30, 60 micromol x L(-1). Flow cytometry also showed that curcumin can influence the distribution of cell cycle of SCC-4 cell with the dosage of 20, 30, 60 micromol x L(-1). And the dosage of 30 micromol x L(-1) curcumin could lead to the recruitment of alpha-tubulin. cDNA microarray showed that 87 genes were activated and 198 genes were inhibited with the effect of curcumin. These results were validated by the real time quantitative RT-PCR.
CONCLUSIONAccording to the results, it suggests that curcumin has the potential as the leading compound for anti-cancer proliferation and invasion in oral cancer treatment, and cdc27, EGFR substrate 15, PPAR-alpha and H2A histone may play an important role among this multiple anticancer-targeting ability.

2.Clinical Value of Pulmonary Artery Parameters and Pulmonary Valve Annulus Z Scores for Choosing the Type of Operation in Complete Repaire of Pediatric Tetralogy of Fallot

Objective: To investigate the clinical value of transthoracic echocardiography (TTE) measured pulmonary artery parameters and pulmonary valve annulus Z scores for choosing the type of operation in complete repair of pediatric tetralogy of Fallot (TOF) Methods: A total of 143 patients received complete repair of pediatric TOF in our hospital were retrospectively studied. The patient's median age was 8 (1-171) months. Based on the type of operation, patients were divided into 2 groups: Trans-annular patch group, n=100 and Valve sparing repair group, n=43. Conventional TTE parameters, right ventricular outflow parameters and pulmonary valve annulus Z scores were compared between 2 groups; pulmonary valve annulus diameters measured by TTE and surgery were studied by correlation analysis. Results: Compared with Valve sparing repair group, Trans-annular patch group had decreased left ventricular end diastolic diameter (23.14±3.82) mm vs (25.23±4.56) mm, pulmonary valve annulus diameter (8.52±2.18) mm vs (10.01±2.61) mm and main pulmonary artery diameter (67.31 vs 82.91) all P<0.05. With reduced value of pulmonary valve annulus Z scores, the constituent ratio in Trans-annular patch group was gradually increasing and in Valve sparing repair group was gradually decreasing, all P<0.05. Correlation analysis indicated that pulmonary valve diameters measured by TTE and surgery had moderate agreement (r=0.581, P<0.001). Conclusion: TTE measured pulmonary valve annulus diameter, main pulmonary artery diameter and pulmonary valve annulus Z scores were the objective parameters provided to surgeon at pre-operation, which had clear clinical value.

To determine heat-shock protein (Hsp)90 expression is connected with cellular apoptotic response to heat stress and its mechanism, chicken (Gallus gallus) primary myocardial cells were treated with the Hsp90 promoter, aspirin, and its inhibitor, geldanamycin (GA), before heat stress. Cellular viability, heat-stressed apoptosis and reactive oxygen species level under different treatments were measured, and the expression of key proteins of the signaling pathway related to Hsp90 and their colocalization with Hsp90 were detected. The results showed that aspirin treatment increased the expression of protein kinase B (Akt), the signal transducer and activator of transcription (STAT)-3 and p-IKKα/β and the colocalization of Akt and STAT-3 with Hsp90 during heat stress, which was accompanied by improved viability and low apoptosis. GA significantly inhibited Akt expression and p-IKKα/β level, but not STAT-3 quantity, while the colocalization of Akt and STAT-3 with Hsp90 was weakened, followed by lower cell viability and higher apoptosis. Aspirin after GA treatment partially improved the stress response and apoptosis rate of tested cells caused by the recovery of Akt expression and colocalization, rather than the level of STAT-3 (including its co-localization with Hsp90) and p-IKKα/β. Therefore, Hsp90 expression has a positive effect on cellular capacity to resist heat-stressed injury and apoptosis. Moreover, inhibition of Hsp90 before stress partially attenuated its positive effects.

5.Influence of combination of external prostaglandin and narrow band ultraviolet B on skin melanin and tyrosinase level in Guinea pigs with vitiligo

Objective To investigate the influence of combination of external prostaglandin and narrow band ultraviolet B (NB-UVB) on skin melanin and tyrosinase level in models with vitiligo.Methods Guinea pigs models with vitiligo were produced by 3% hydroquinone cream.And then all the animal models were randomly divided into six groups according to the weight:normal group,model group,control group,medication alone group,light alone group and combination group.Normal group and model group were not treated.Control group was used the same amount of vaseline,2 times per day;the 0.25 mg · mL-1 dipropionazone tincture skin smear in medication alone group,2 times per day,stay on 2 months;the NB-UVB irradiation treatment instrument was exposed to 1 J · cm-1 at about 20 cm from the skin of the white spot,and the wavelength was 310 ~315 nm in light alone group,and once every other day,stay on 2 months;combination group,topical prostaglandina combination NB-UVB irradiation,frequency,method and time same to medication alone group and light alone group.After administration,the observation of vitiligo guinea pig skin tissue morphology and melanin distribution in the light microscope.Results The percentage of melanin hair follicles in model group,control group,medication alone group,light alone group and combination group were (37.50 ± 20.43) %,(53.33 ± 14.38) %,(55.00 ± 16.12) %,(46.67 ± 19.66)%,(71.67 ± 7.53)%,respectively.The combined group is compared to the model group,the difference is significant in statistics (P < 0.05).But comparison between control group,medication alone group,light alone group and model group,the difference were not significant in statistics (all P > 0.05).Conclusion Combination of external prostaglandins and NB-UVB have some effect on vitiligo treatment.
6.Effects of combined tacrolimus ointment and narrow band ultraviolet B on the changes of the level of interleukin-10 and tyrosinase in Guinea pig with vitiligo

Objective To investigate the effect of combined narrow band ultraviolet B ( NB-UVB) and tacrolimus on the changes in the level of tyrosinase and interleukin -10 ( IL -10 ) in Guinea pig with vitiligo. Methods Animal models with vitiligo were built first .And then all the animal models were randomly divided into six groups of eight according to the principle of weight balance: normal group , model group , control group(2 times per a day), medication alone group (0.03% tacrolimus ointment skin smear , 2 times per a day ) , light alone group ( white spot area use NB -UVB irradiation , once every other a day ) , combination group(topical tacrolimus combination NB -UVB irradiation, frequency , method and time same to medication alone group and light alone group ). Colorimetric method and ELISA method were used to detect the changes in the level of tyrosinase in serum and IL -10 in skin tissue homogenates and serum respectively , before and after the administration and irradi-ate.Results No significant changes in the level of tyrosinase in serum were observed before and after combined administration of NB -UVB and tacrolimus in animal models with vitiligo .While IL-10 contents in skin tissue homogenates and serum were significantly higher than that before administration , and the difference was statistically significant (P<0.01).Conclusion Combined treatment of NB-UVB and tacrolimus can significantly improve the level of IL -10 in skin tissue homogenates and serum for animal models with vitiligo .

Objective To investigate the changes in the number of skin melanin after combined treatment of narrow band ultraviolet B ( NB-UVB) and tacrolimus ointment in animal models with vitiligo. Methods Animal models with vitiligo were built first.And then all the animal models were randomly divided into six groups of eight according to the principle of weight balance:normal group,model group,control group ( where vaseline was used in the shed white spot areas in the back of animal model, these two methods were implemented 2 times per day ), medication alone group(0.03%tacrolimus ointment skin smear,these two methods were implemented 2 times per day), light alone group(white spot area use NB-UVB irradiation, these two methods were irradiation once every other day ) , combination group ( topical tacrolimus combination NB-UVB irradiation,frequency,method and time same to medication a-lone group and light alone group).The differences of the number of mela-nin in the skin tissue before and after medication were checked and com-pared and irradiation by histopathology.Results The effect of the com-bined group on skin melanin express was significantly different compared with model group ( P<0.01 ) .Conclusion Combination of NB -UVB and tacrolimus ointment has a significant effect on vitiligo treatment.
8.Study on Quality Control Method of Yinqiao Chaihuang Granules

Objective To establish the quality control method of Yinqiao Chaihuang Granules.Methods Lonicerae Japonicae Flos, Bupleuri Radix, Scutellariae Radix and Glycyrrhizae Radix et Rhizoma in Yinqiao Chaihuang Granules were identified by TLC, and the content of forsythoside A was detected by HPLC. Results Under the selected chromatographic conditions, TLC spots were clear, the separating was good, and non-interference was found in negative control. Forsythoside A in calibration curve showed good linearity in the range of 0.029 6–0.474 μg, with average recovery of 99.73%, RSD=1.18%. Conclusion The method is accurate and simple, which can be used for the quality control of Yinqiao Chaihuang Granules.

OBJECTIVETo evaluate the efficacy and safety of trastuzumab in combination with chemotherapy versus chemotherapy alone in the first-line treatment of HER-2-positive advanced gastric or gastro-oesophageal junction cancer.
METHODSFifteen Chinese research centers are involved in the BO18255 (ToGA) study. Patients with gastric or gastro-oesophageal junction cancer were eligible for inclusion if their tumor showed overexpression of HER-2 protein by immunohistochemistry +++ or FISH-positive. Patients were randomly assigned in a 1:1 ratio to receive a chemotherapy regimen consisting of capecitabine or 5-FU plus cisplatin or chemotherapy in combination with intravenous trastuzumab. The primary endpoint was overall survival.
RESULTSEighty-five Chinese patients were enrolled in this study, of whom 84 were included in the primary analysis: trastuzumab plus chemotherapy (FP/H) (n = 36) and chemotherapy alone (FP)(n = 48). The median follow-up was 15.2 months in the FP/H group and 14.2 months in the FP group. The median survival time was 12.6 months in the FP/H group compared with 9.7 months in the FP group [hazard ratio 0.72, 95%CI (0.40; 1.29)]. Grade 3/4 adverse events were higher in the FP/H(63.9%)than FP (47.9%) groups, including neutropenia, vomiting and nausea. Two mild cardiac adverse events occurred in the FP/H group. Severe adverse events occurred in 3 cases of both two groups, respectively.
CONCLUSIONSAddition of trastuzumab to chemotherapy is well tolerated and shows improved survival in Chinese patients with advanced gastric or gastro-oesophageal junction cancer. These results are consistent with the results of ToGA whole population trial. Trastuzumab in combination with chemotherapy can be considered as a new option for patients with HER-2-positive advanced gastric or gastro-oesophageal junction cancer.

10.Venetoclax Combined with CACAG Regimen in the Treatment of Patients with Refractory/Relapse Acute Myeloid Leukemia:A Prospective Clinical Study

Objective:To investigate the efficacy and safety of Venetoclax combined with CACAG regimen in treatment of patients with refractory/relapse acute myeloid leukemia(R/R AML).Methods:The study was a singlecenter prospective clinical trial.The enrolled patients met the criteria for R/R AML.Treatment included Azacidine(75mg/m2,d1-7),Ara-C(75-100 mg/m2,q12h,d1-5),Aclacinomycin(20 mg d1,d3,d5),Chidamide(30 mg d1,d4),Venetoclax(100 mg d1,200 mg d2,400 mg d3-d14,in combination with Triazole Drug,reduced to 100 mg/d),and granulocyte colony-stimulating factor(300 μg/d until neutrophil recovery).The primary endpoint of observation was overall response rate after 1 course of treatment.Results:A total of 19 patients were enrolled from January 2022 to April 2023.After 1 course of treatmen,the overall response rate was 81.3%(13/16),the CR rate was 68.8%(11/16),and the PR was 12.5%(2/16).Among the 11 patients who got CR/CRi,8 cases achieved CRm(minimal residual disease negative CR)and 3 cases did not.As of March 27,2023,the median follow-up time was 111(19-406)days.The six-month overall survival and progression-free survival rates were both 55.7%,the 1-year overall survival and progression-free survival rates were 46.4%and 47.7%,respectively.In addition,compared with the non-CRm group,CRm patients had a better PFS(377 days vsi11 days,P=0.046).Treatment-related adverse events were mainly 3-4 degrees of bone marrow suppression,complicated by various degrees of infection(n=12),hypokalemia(n=12)and hypocalcemia(n=10)and elevated liver enzymes(n=8),of which 3/4 degrees accounted for 47.4%(9/19).Conclusion:The Venetoclax combined with CACAG regimen is an effective salvage therapy for patients with R/R AML,with high remission rate and safety profile.
