1.Study on Breeding of High-yield Avilamycin-producing Strains by Nitrogen Ion Implantation

To obtain high-yield avilamycin-producing strains,low energy N~+ ion implantation technology and screening of streptomycin-re- sistant mutants are used in the study on breeding mutation.The results show that,“saddle”region,which range is from 3?10~(15) to 5?10~(15) ions/cm~2,has got better induced mutation action.It also means that the strain's resistant mutation and yield mutation closely correlate to each other,and the method of streptomycin resistant screening is feasible.We have isolated a high-yield strain SVT-45 which the productivi- ty is 195% higher than the original strain's in the rotation-flask experiments.These results showed that the ion implantation was an effective method for microbe mutagensis.
2.An investigation on the life quality and mental health status of the disabled people in Zhejiang Province

Objective To learn the quality of life (QOL) and mental health status of the physically disabled people in Zhejiang Province,in order to provide evidence for improving the quality of life and mental health of the physically disabled people.Methods Using questionnaires and interviews to make household surveys of 950 physically disabled people.The questionnaire included the brief quality of life scale (SF-36) and the general health questionnaire (GHQ-28) in the anxiety and depression scale,physical and mental health status.The basic information and other living conditions of physically disabled people were collected through interviews.Results A total of 907 valid questionnaires were recovered,and the effective response rate was 95.47%.Scores of QOL of 907 physically disabled people was 60.89,and anxiety and depression scores were more than 4 points and there was a high score on anxious and depression.The scores of quality of life,anxiety and depression were not statistically significant between males and females(P >0.05).There were significant differences in all dimensions of SF-36 and anxiety and depression in different levels of physically disabled people(P < 0.01).There was a significant negative correlation between the quality of life and anxiety and depression in physically disabled people(rs =-0.626、-0.639,P < 0.01).The multiple linear regression showed that physiological function,role physical,physical pain and general health were the influencing factors of anxiety of the physically disabled people,and role physical,physical pain and general health were the influencing factors of depression of the physically disabled people.Condusion The quality of life of physically disabled people were comparably poor,and presented anxiety,depression and other psychological problems.Physical function status directly affects the quality of life and mental health status.
3.Effect of Action Observation Therapy on Upper Limb Function in Children with Spastic Hemiplegic Cerebral Palsy

Objective To explore the effect of action observation therapy (AOT) on upper limb function in children with spastic hemiplegic cerebral palsy,and the better program. Methods From March to November,2017,45 children with spastic hemiplegic cerebral palsy were randomly assigned to control group and AOT group.The control group was further divided into group A(n=11)and group B(n=11), and received conventional rehabilitation treatment and scenic-observation therapy, 20 minutes every time for group A,and 30 minutes every time for group B.AOT group was further divided into group C(n=10)and group D (n=11), and received AOT in addition, 20 minutes every time for group C, and 30 minutes every time for group D,five times each week for twelve weeks for all the groups.Before treatment,eight and twelve weeks af-ter treatment, they were assessed with hand grip strength, Upper Extremities Functions Test (UEFT), and Wee Functional Independence Measure(WeeFIM). Results After treatment,the hand grip strength and the score of UEFT were better in group D than in groups B and C(P<0.05),and were better at twelve weeks than at eight weeks(P<0.05).No significant difference was found in the score of WeeFIM among groups after treatment(P>0.05). Conclusion AOT could improve upper limp function in children with spastic hemiplegic cerebral palsy,and it's more ef-fective after more training.
4.Coal tar pitch smoke extract-induced pyroptosis in human bronchial epithelial cells.

OBJECTIVETo explore whether coal tar pitch smoke extract (CTP) induced pyroptosis in human bronchial epithelial cells (BEAS-2B).
METHODSBEAS-2B cells were treated with different concentrations of CTP (1, 3 µg/ml) for 8h and 24 h, respectively. Lactic dehydrogenase (LDH) activity and interleukin-1 beta (IL-1β) levels in the supernatants of cell culture media were measured with LDH activity or human IL-1β ELISA kit, respectively. The activity of Caspase-1 was measured with Caspase-1 colorimetric assay kit.
RESULTSThe activity of caspase-1 in 1 and 3 µg/ml CTP groups were (9.29 ± 0.30) and (8.67 ± 0.59) µmol/ml respectively which were both significantly increased compared to that (7.42 ± 0.59) µmol/ml in the control group (P < 0.05) after 8 h exposure, but there was no significant difference in the activity of LDH and levels of IL-1β in the cell culture media among the CTP and control groups. 24 h after exposure, the activity of LDH in the CTP (1, 3 µg/ml) groups were (1323.03 ± 28.53) and (1148.45 ± 16.42) U/dl respectively which were significantly higher than that (1091.93 ± 26.64) U/dl in the control group (P < 0.05), and the levels of IL-1β in the CTP (1 and 3 µg/ml) groups were (125.37 ± 25.00) pg/ml and (92.04 ± 19.09) pg/ml respectively which were significantly higher than that (46.20 ± 14.43) pg/ml in the control group (P < 0.05), but there was no significant difference in the activity of Caspase-1 among CTP and control groups (P < 0.05).
CONCLUSIONCTP treatment induced early increase in caspase-1 activity followed by the increase in LDH activity and IL-1 levels, indicative of pyroptosis in human bronchial epithelial cells.

5.Studies on the flavonoids in herb from Scutellaria barbata.

OBJECTIVETo study flavonoids in Scutellaria barbata and elucidate their structures.
METHODCompounds were isolated and purified by normal column chromatography, paper thin layer chromatography and Sephadex chromatography. The chemical structures were elucidated by ESI mass and NMR spectra.
RESULTFive flavonoids were isolated from S. barbata and identified as apigenin (1), luteolin (2), ethyl-7-O-apigenin-glucuronate (3), apigenin-7-O-beta-glucoside (4), and apigenin-7-O-neohesperidoside (5), in addition to benzyaldehyde (6).
CONCLUSIONFlavonoidglycoside (3), (4) and (5) were isolated from S. barbata for the first time.

6.Expression of microRNA-15b in endometrial carcinoma and its effect on proliferation of endometrial carcinoma cells

Objective To investigate the expression of microRNA-15b (miR-15b) in endometrial carcinoma and its effect on proliferation of endometrial carcinoma cells.Methods Eighty-seven cases of endometrial cancer tissues were selected as the research objects from patients with endometrial cancer undergoing endometrial cancer staging surgery in Xinxiang Central Hospital from February 2013 to February 2017.Another 45 cases of normal endometrial tissues from patients with uterine fibroids undergoing line hysterectomy were collected as the control.The expressions of miR-15b in endometrial carcinoma tissues and normal endometrial tissues were detected by real-time quantitative polymerase chain reaction(PCR) and relationship between the miR-15b expression and clinicopathological feature was analyzed.The HEC-1A cells were cultured and were randomly divided into miR-15b mimics group,control sequence group and blank control group.The expression of miR-15b in HEC-1A cells was detected by real-time quantitative PCR.Tetramethylazolazole blue (MTT) assay was used to detect the cell proliferation.Flow cytometry was used to detect the apoptotic rate and cell cycle.Results The relative expression levels of miR-15b in endometrial carcinoma tissues and normal endometrial tissues were 1.32 ±0.13 and 2.49 ±0.16,respectively.The relative expression level of miR-15b in endometrial carcinoma tissues was lower than that in the normal endometrial tissues (t =46.184,P <0.05).The relative expression level of miR-15b in endometrial carcinoma was correlated with International federation of gynecology and obstetrics staging,myometrial invasion and lymph node metastasis (P < 0.05),but it was not correlated with age,histological type,differentiated degree,in filtrating lymph vessel and blood vessel or not.Compared with the blank control group and the control sequence group,the relative expression level of miR-15b in miR-15b mimics group was significantly increased (P < 0.05),the cell proliferation abilities at cultured 24,48,72 and 96 h were decreased (P < 0.05),while the apoptosis rate increased (P <0.05),the proportion of G0 + G1 phase was significantly increased,while the proportion of G2 + M phase was decreased (P <0.05).There was no significant difference in the relative expression level of miR-156,the cell proliferation ability at cultured 12,24,48,72,96 h,the apoptosis rate,the proportion of G0 + G1,S and G2 + M phase between the blank control group and the control sequence group(P <0.05).Conelusion miR-15b is low expression in endometrial carcinoma tissue.Up-regulation of miR-15b expression in endometrial carcinoma cells can inhibit cell proliferation,it may be related to block cell cycle to promote cell apoptosis.
7.Regulatory Actions of Intestines-unblocking and Turbid-purging Recipe on Colonic 5-Hydroxytryptamine and Its Receptor in Rats with Constipation-dominant Irritable Bowel Syndrome

Objective To investigate the regulatory actions of Intestines-unblocking, Turbid-purging Recipe (ITR) on colonic 5-hydroxytryptamine (5-HT) and its receptor 5-hydroxytryptamine 3 (5-HT3) in rats with constipation-dominant irritable bowel syndrome (IBS-C), and to explore the therapeutic mechanism of ITR in treating IBS-C. Methods Forty-two male SD rats were randomly divided into 6 groups, namely normal group, model group, western medicine group, high-, middle- and low-dose Chinese medicine groups, 7 rats in each group. IBS-C rat model was established by intragastric administration of ice water. After establishment of the model, western medicine group was given intragastric administration of Cisapride Tablets (at the dosage of 3.6 mg·kg-1·d-1), Chinese medicine groups were given intragastric administration of various dosages of ITR granules (18.5, 9.25, 4.625 g·kg-1·d-1 respectively) , and the model group was given intragastric administration of normal saline, the treatment lasting 14 d. The rats in various groups were given normal feeding and drinking. After treatment, HE staining method was used to observe pathological changes in the intestinal tissue, immunohistochemistry method was used to observe the expression levels of intestinal 5-HT and 5-HT3 receptor. Results Compared with the normal group, the expression level of rat intestinal 5-HT was increased (P < 0.05) and that of 5-HT3 receptor was decreased (P < 0.05) in the model group and the medication groups. Compared with the model group, 5-HT expression level was decreased significantly (P<0.05) and 5-HT3 receptor expression level was increased (P < 0.05) in the medication groups, and the improvement of the middle-dose Chinese medicine group was more obvious (P < 0.05). Conclusion ITR has therapeutic efficacy for IBS-C rats through lowering 5-HT expression and increasing 5-HT3 receptor expression, which results into the improvement of intestinal sensitivity and abnormal dynamic of the rats.
8.Clinical characteristics and treatments of patients with multiloculated pyogenic brain abscess

Objective To investigate the clinical characteristics and treatment choice of patients with multiloculated pyogenic brain abscess (MLPBA).Methods The clinical data of 89 patients with pyogenic brain abscess (including 20 with MLPBA and 69 with uniloculated pyogenic brain abscess [ULPBA]) treated during the recent 21 years (1991-2011) were collected and analyzed retrospectively.Results MLPBA patients counted for 22.5% of our patients with pyogenic brain abscess.The male-female ratio,age distribution,history duration,location of abscess,kinds of isolated microorganisms,predisposing factors and clinical manifestations of patients with MCPAB were similar to those of patients with ULPBA.The ratio of positive isolated microorganism and ratio breaking into ventricles in patients with MLPBA were significantly higher than those in patients with ULPBA (P<0.05); and the average volume of abscess was 8.8 mL in patients with MLPBA and 13.3 mL in patients with ULPBA.A higher rate of abscess recurrence after stereotactic surgery in patients with MLPBA (26.7%) was found as compared with that in patients with ULPBA (3.2%).The mortality in patients with MLPBA was 0% and that in patients with ULPBA was 4.3%.Conclusion MLPBA is not rare; the volume of abscess in patients with MLPBA is significantly smaller than that in patients with ULPBA,but the abscess in patients with MLPBA is significantly easier broken into the ventricles than that of ULPBA.Stereotactic operation is the first treatment choice; the prognosis for patients with MLPBA can be as good as that for patients with ULPBA,although the possibility of recurrent abscess formation after surgery is higher.
9.Value of MRI in the diagnosis of cerebral abscess caused by Candida albicans in premature infants.

OBJECTIVETo explore the value of serial magnetic resonance imaging (MRI) in the diagnosis of cerebral abscess caused by Candida albicans in premature infants.
METHODSThe clinical data of 8 premature infants with central nervous system invasive fungal infection (IFI) were retrospectively studied. The infants underwent serial cerebral MRI scans (T1WI, T2WI and DWI).
RESULTSCandida albicans was found as pathogen in all of the 8 infants. Seven infants presented with cerebral abscess and 4 infants had concurrent meningitis. Widespread involvements were found on MRI, particular in white matter area of subcortex, centrum semiovale and periventricle. The MR imaging findings in 4 infants within 11 days after IFI showed diffusive and multiple miliary nodes and hyperintense signals on DWI, but obvious changes were not found on T1WI and T2WI. The most striking hyperintense signals on T1WI and hypointense signals on T2WI appeared between 2 and 4 weeks after IFI, and some nodes of rim-like hyperintensity and marked contrast enhancement were also noted on T1WI. Smaller and smaller changes of the miliary foci were seen on T1WI and T2WI 4 weeks later. Delayed myelination and thinner corpus callous were shown in 2 patients at three months.
CONCLUSIONSMRI-DWI and serial MRIs are helpful in the early diagnosis of candida cerebral abscess and the evaluation of treatment outcome in premature infants.

10.Reconstruction of mandibular defects with tissue-engineered bone using 3D bionic printing technology

BACKGROUND: The 3D printed polylactic-co-glycolicacid/nano-hydroxyapatite (PLGA/nHA) scaffold carrying human recombinant bone morphogenetic protein 2 (rhBMP-2)/chitosan (CS) sustained release tissue-engineering bone has good biological activity, mechanical properties, and biological activity of its controlled release rhBMP-2. OBJECTIVE: To investigate the repair of mandibular defects with PLGA/nHA scaffold/rhBMP-2/CS sustained release tissue-engineering bone manufactured using 3D bionic printing technology. METHODS: Animal models of bilateral critical mandibular bone defects were established in 27 New Zealand white rabbits, followed by implantation of a 3D-printed PLGA/nHA scaffold/rhBMP-2/CS sustained release tissue-engineering bone on one side (experimental side), and a 3D-printed PLGA/nHA scaffold on the other side (control side). Mandibular specimens were harvested at postoperative days 30, 60 and 90 to carry out cone-beam CT, Micro CT, histological and immunohistochemical examinations. RESULTS AND CONCLUSION: The results from micro-CT analysis revealed that the volume of newly formed bone volume and the amount of bone trabeculae on the experimental side were significantly higher than those on the control side at different postoperative time points (P < 0.05). The results from cone-beam CT examination showed that at 90 postoperative days, bone density of the bone defect on the experimental side was close to that of the surrounding bone, new bone tissues were full of the original bone defect area, and the trabecular bone arranged regularly, while on the control side, worm-eaten discontinuous low-density osteoid tissues were visible in the bone defect area. Osteogenesis on the experimental side was better than that on the control side. Histological findings demonstrated that on the experimental side, a large amount of mature lamellae were detected in the bone defect area, with well-arranged trabecular bones and abundant capillaries, and moreover, the scaffold material had been completely absorbed. However, low-density, loose-meshed, irregular braided bone tissues with rare capillaries were observed on the control side, and the scaffold material had been mostly absorbed. Immunohistochemical findings indicated that the osteocalcin-dyed area on the experimental side was significantly larger than that of the control side at postoperative 90 days. To conclude, 3D-printed PLGA/nHA scaffold/rhBMP-2/CS sustained release tissue-engineering bone is favorable for the repair and reconstruction of experimental mandibular defects in rabbits.
