1.In situ rats single pass perfusion intestinal absorption of the effectivein components in Radix Angelicae Pubescentis.

In order to study the intestinal absorption behaviors of three active constituents, columbianetin acetate, osthole and columbianadin in Radix Angelicae Pubescentis extracts, in situ rat single pass intestine perfusion (SPIP) was carried out by the perfusion solution of the extract I containing less than 10% total coumarins. The absorption of extract II containing more than 60% total coumarins was compared with that of extract I by rat colon SPIP to elucidate the influence on absorption of the different coumarin content extracts of the Chinese traditional medicine. The samples of the perfusion solution were collected in certain intervals. The concentrations of three active components in the perfusion samples were determined by HPLC method. The results demonstrated that the absorption rate constants (Ka) or apparent permeability coefficients (Papp) of columbianetin acetate, osthole and columbianadin from extract I had no significant difference among concentration ranges of 62-555 microg x mL(-1), 101-887 microg x mL(-1), 19-186 microg x mL(-1), respectively. The absorption quantity of three components was proportional to its concentration respectively and the saturate absorption phenomena were not observed. This suggested that the absorption of columbianetin acetate, osthole and columbianadin showed the passive diffusion process. Three components could be absorbed in whole intestinal sections. The Ka and Papp of three components all showed colon > duodenum > jejunum > ileum in four different regions of rat intestine. At colon, Ka and P app were significant different from the others. The Ka or Papp of three components from the extract I was significantly more than that of same components from extract II. The extract I redounded to increase the absorption of three active components.

2.Comparative analysis of seven marine biological source of mineral drugs.

The marine biological source of mineral drugs recorded in Chinese Pharmacopoeia (2010 version) mainly including pearl, nacre, clam shell, common oyster shell, ark shell, cuttle bone, and sea-ear shell are widely used in clinical. Calcium carbonate and a small amount of protein are the main components in this type of drugs. In this paper, a systematical and comparable study were carried out by determination of calcium carbonate by EDTA titration method, the crystal of calcium carbonate by X-Ray powder diffraction and the total amino acids (TAAs) of the hydrolyzed samples by ultraviolet spectrophotometry method. As a result, the crystal structure is calcite for common oyster shell, mixture of calcite and aragonite for nacre and sea-ear shell, aragonite for the other drugs. The content of calcium carbonate ranged from 86% to 96%. Cuttle bone has the highest amount of TAAs among the seven drugs which reached 1.7% while clam shell has the lowest content of 0.16% on average. In conclusion, an effective method was developed for the quality control of marine mineral drugs by comprehensive analysis of calcium carbonate and TAAs in the seven marine mineral drugs.

3.Expression, purification of Staphylococcus aureus Efb protein and preparation of its functional antibody

Objective:To clone and express the Staphylococcus aureus Efb(extracellular fibrinogen-binding protein) protein in Escherichia coli, to purify the expression product and prepare its functional antibody and to detect the functions of Efb protein for further studies on S.aureus infection.Methods: Efb gene was amplified by PCR using S.aureus NCTC-8325 genome DNA as template and cloned into the recombinant expression vectors pET28a. E.coli BL21(DE3) with the plasmid was induced with IPTG for protein production. The protein was purified by Ni~(2+) affinity chromatography. The function of Efb protein was determined by complement activity assay and inhibition ELISA.The polyclonal antibodies were prepared by immunizing the animals. Results: The purified recombinant Efb was obtained, which could inhibit the CH50 and AH50 effectively. The functional poly-antibodies of Efb were prepared.Conclusion:Efb could inhibit the classical pathway and alternative pathway of complement activation, and the antibodies against to Efb could block the inhibition of the classical pathway of complement activation induced by Efb.

5.Path analysis of influencing factors correlated with test anxiety in middle school students

Objective To find some risk factors correlated with test anxiety of middle school students,and to find out influencing pathway for test anxiety. Methods 647 middle school students were investigated with Sarason' Test Anxiety Scale (TAS), Achievement Motivation Scale (AMS), Coping Style Scale for School Students ( CSS), Eysenck Personality Questionnaire( EPQ), Egma Minnen av Bardndosnauppforrestran(EMBU) and Family Environment Scale-Chinese Version(FES-CV). Statistics were done with version of SPSS14.0,and data were analyzed by Pearson correlation, multiple linenear stepwise regression and path analysis. Results The rates of test anxiety respectively was mild 25.97% ,moderate 45.65% ,severe 28.38%; there were no significant different between the male and female students anxiety ( 16.71 ± 6.44,17.01 ± 7.02, t = 1. 469, P = 0.334). Test anxiety positively correlated with Achievement motivation, reach motivation of competition, endurance, escape, expos, deny the fantasy,family conflicts,parental punished severely,excessive interference,objective deny,overprotective of father.( r 1-16 :0. 214,0. 135,0. 254,0. 216,0. 308,0.472,0. 492,0. 168,0. 249,0. 537,0. 282,0. 102,0. 238,0. 185,0. 233,0.301,0.273; P 1-16 = 0. 000 ~ 0. 030) , and negatively correlated with Problem-solving, rationalizing interpretation, family cohesion, informative, entertaining, emotional expression, organization, parental warmth and understanding ( r1-9: -0. 121, -0. 134, -0. 178, -0. 215, -0. 221, -0. 101, -0. 298, -0. 136, -0. 168; P 1-9 =0.000 ~0.007). Conclusion Test anxiety is commonly find in middle school students. Test anxiety is affected by some paths that are personality,achievement motivation,emotional coping style,family environment and parental education methods.

Objective To discuss the personality characteristic and personal social attributes of junior middle school students of online violent games addictive disorder (VOGAD).Methods From August to December at 2008,6000 students were randomly drown from 8 junior middle schools in Dalian ,then they were investigated with the self-made internet game survey form and the Chen internet addiction scale (CLAS).149( 2.5% ) internet game addicts were screen out from 6000 students, and were divided into the violence and entertainment game addicted group ( 66 students in per group,and the male is 55, female is 11 ).Average age was ( 14.36 ± 0.81 ) years old.No-play game group 66 students were mach according to the sex and the age.All objects were evaluated with Eysenck personality questionnaire (EPQ) and Minnesota Multiphasic Personality Inventory Abbreviation Form (MMPI-AF).Results There were significant different during the 3 groups in psychoticism (50.60 ± 5.10;50.40 ± 6.30; 46.81 ± 4.62 ), neuroticism ( 51.74 ± 5.70; 49.03 ± 6.85; 49.25 ± 6.09 ) , manifest anxiety (55.29 ±9.58;45.11 ±8.31; 43.24 ±8.32) ,dependence (58.90 ±9.09; 55.97 ±8.63; 54.05 ± 11.39) ,and social responsibility ( 37.47 ± 12.41; 43.25 ± 11.24 ;46.34 ± 14.37 ) (F1-5 = 10.32,3.83,36.17,4.11,8.23; P 1-5 < 0.05 ).Except of the social responsibility (P> 0.05 ) ,above results all suggested higher scores in the VOGAD group than other two groups (P1 < 0.05 ).Conclusion The results suggest higher scores in the psychoticism, neuroticism, dependency, manifest anxiety in VOGAD group, and lower Re in online violence game addict students.
7.PCR, clone and sequence analysis of rDNA-ITS of Nelumbo nucifera from different geographical origins in China.

OBJECTIVETo provide DNA molecular marker for identification of Nelumbo nucifera by exploring the differences of nrDNA-ITS sequence of N. nucifera originated from different habitats.
METHODTo compare nrDNA-ITS base sequence using specific PCR-ITS.
RESULTThe completed sequence of ITS and 5.8 S rDNA, and the partial sequences of 18S rDNA and 26S rDNA, totally 750 bp, from N. nucifera were obtained. The differences among N. nucifera from different habitats and from different cultivars were found.
CONCLUSIONThe method can be used to identify N. nucifera among different species and to distinguish their fakes. It provided the basis for identifying N. nucifera from different geographical regions by comparison of their ITS sequences.

8.Effects of parental locus of control and mental health on test anxiety of middle school students

Objective To find out parental locus of control and mental health affecting students test anxiety. Methods The samples were from 5 middle schools including 1000 students,and their parents. They were investigated with the general life scales, Sarason's test anxiety scale, Symptom checklist 90 (SCL-90)and Parenting Locus of Control Scale(PLOC). Results The ratio of test anxiety: the mild was 28.8%, moderate was 45.0%, severe was 26.2% ,and felt anxiety was 55.8%. Ratio of test anxiety was higher in the boy students (X2=9.284, P =0.010) ,and heavier(F:16.42±6.70; M:15.18 ±6.51, t=2.716, P=0.007). Student test anxiety was significantly positively correlated with their parental mental health (r fatherl~11=0.166~0.272, Pfather l~11= 0.000;r mother 1~11=0.182~0.242, P mother 1~11= 0.000); Student test anxiety positively correlated with the effectiveness cof parents education, father's belief on the fate, eontrol of fathers for their children, locus of control from fathers (r1~5=0.075~0.143; P1-5=0.000~0.030);felt anxiety positively correlated with self-expect ,pressure from their mothers,over take care attitude from their parents,self-pressure from their parents( r1~6=0.068~0.230; P1-6=0.000~0.050) ,and negatively correlated with respect attitude of parents for their child, Marital relations,Communication time between the students and their parents,attitude of mother for learning( r1-7=-0.074~-0.140;P1~7=0.000~0.034). Felting test anxiety was served as a dependent factor, some factors enter the regression equation,and they were somatization of father, psychotic mother, the pressure from parents, mother over expectations, self-expectations ,the child control from student's father by Logistic regression analysis ( OR1~16=0.675~3.029;P1-6=0.000~0.007). Conclusion Students test anxiety is a common problem in male and female students. Mental health and locus of control from students' parents show test anxiety has somatization of father, psychotic mother, the pressure from parents, mother' expectations,self-expectations, the control of father for the child 6 risk factors.
9.Effects of 17β-estradiol on proliferation and migration of MCF-7 cell by regulating expression of claudin-6.

OBJECTIVETo explore the role of estrogen in the regulation of the expression of claudin-6 and biological behavior in MCF-7 cells.
METHODSRT-PCR and immunocytochemistry were conducted to analyze the expression and localization of claudin-6 in MCF-7 cells treated with 17β-estradiol. CCK-8 kit assay and Scratch Test were conducted to analyze the capability of proliferation and migration of 17β-estradiol treated MCF-7 cells.
RESULTSRT-PCR analysis and immunocytochemistry showed that 17β-estradiol induced a concentration-and time-dependent effect on claudin-6. At 5 nmol/L and at 24 h, 17β-estradiol treatment led to an increased level of claudin-6, which was located in the membrane of MCF-7 cells. CCK-8 analysis showed a significant decrease in the capability of proliferation of MCF-7 cells compared with the control group (P < 0.05). Cells Scratch Test showed decreased migration capability of MCF-7 cells compared with the control group (P > 0.05).
CONCLUSIONS17β-E2 might regulate the expression of claudin-6 and inhibit the proliferation and migration of MCF-7 cells. The inhibitory effects of 17β-E2 on growth and migration of MCF-7 cells may be mediated by claudin-6 expression regulation.

10.Pharmacokinetics of two recombinant humanized monoclonal antibodies against ricin in rhesus monkeys

Recombinant humanized anti-ricin monoclonal antibody (MIL50) is a recombinant humanized monoclonal antibody targeting ricin. In this study, an ELISA method was used to establish a method for the determination of MIL50 in macaque serum, and a cross design method was used. Twelve rhesus monkeys were intravenously injected 1 mg·kg-1 test preparation (MIL50 freeze-died powder injection) and reference preparation (MIL50 liquid preparation) to determine the plasma concentration of MIL50 at different time points, and the pharmacokinetic parameters were analyzed to compare the pharmacokinetic characteristics of MIL50 liquid preparation and freeze-died powder injection in rhesus monkeys. Animal welfare and experimental procedures follow the regulations of the Animal Ethics Committee of the Chinese Academy of Medical Sciences and Use of Laboratory Animals and the regulations derived by the Animal Care and Welfare Committee of the Institute of Radiation Medicine, Academy of Military Medical Sciences (IACUC-DWZX-2020-503).
