2. Metabolomic Study of Polygonatum odoratum Polysaccharide on Hyperthyroidism Yin-deficiency Rats based on GC-MS Metabonomics

OBJECTIVE: To evaluate the effect of Polygonatum odoratum polysaccharide(POP) on hyperthyroidism Yin-deficiency rats based on GC-MS metabolomics and explore its mechanism. METHODS: SD rats were randomly divided into control, model and POP groups. The hyperthyroidism Yin-deficiency rats were treated by intraperitoneal injection of T3 for a week. The content of T3, T4 in serum and cAMP, cGMP in plasma was determined. A GC-MS based metabolic profiling method was employed to analyze the distinctive metabolic patterns of serum in control, model and POP. Multivariate statistical analysis was applied for multivariate analysis to find potential biomarkers of Yin-deficiency by SIMCA-P 12.0 software. RESULTS: Compared with the model group, polygonatum polysaccharide can significantly reduce the content of T3, T4, cAMP, the ratio of cAMP/cGMP and increase the content of cGMP, indicating it has certain Yin-nourishing action. The PCA score plot showed metabolic profiling of control, model and POP had significant differences. Nine potential biomarkers were identified by PLS-DA and variable importance for project values(VIP) based on the metabolomics. CONCLUSION: The endogenous metabolites were changed by POP intervention significantly in Yin-deficiency rats, and the preventive role may be related to the synergism of glucose and lipid metabolism, amino acid metabolism, energy metabolism with the multi-targets, which provides a basis for further study of the mechanism of POP treatment of Yin-deficiency.
3. Polar Chemical Constituents from Prunella vulgaris L. and Their Anti-human Breast Tumor Activities

OBJECTIVE: To study the polar chemical constituents from Prunella vulgaris L. METHODS: Silica gel, reverse-phase octadecylsilyl(ODS), Sephadex LH-20 chromatographic methods, MCI and HPLC were applied to isolate and purify compounds. MS and NMR methods were used to determine the structures of the compounds. Furthermore, the cytotoxicity of these chemical components for MCF-7, MDA-MB-231 and MCF-10A cell lines was measured by MTT method. RESULTS: A total of 12 compounds were isolated from the fruits of P. vulgaris and their structures were identified as methyl 3,4,α-trihydroxypropionate(1), danshensu(2), methyl rosmarinate(3),3,4-dihydroxybenzoic acid(4), quercetin-3-O-glycopyranoside(5), hyperoside(6), 2α,3α,19α,24-tetrahydroxylurs-12-en-28-oic acid(7), 2α, 3α, 24-trihydroxyolean-12-en-28-oic acid(8), cytidine(9), daucosterol(10), (3S,5R,10S)-7-oxo-12-methoxyabieta-8,11,13-triene-3,11,14-triol(11), and 2α,3α,24-trihydroxyolean-12,20(30)-dien-28-oic acid(12). The results of antitumor assay indicated that compound 2,3,5 and 6 significantly inhibited the activity of MCF-7, compound 3 could inhibit the activity of MDA-MB-231, but all of them also significantly inhibited the activity of normal cell lines MCF-10A. CONCLUSION: Compounds 9 and 11 are isolated from the genus of Prunella L. for the first time. Some chemical constituents form Prunella L. show certain anti-breast cancer activity.
5.Application of gyrA and pare gene detection in genotyping of Ureaplasma spp.

Objective To evaluate the contribution of gyrA and parE detection in Ureaplasma genotyping.Methods Sixty Ureaplasma isolates were selected with the Mycoplasma IST kit.The gyrA and parE were amplified by PCR.The DNA was sequenced and compared with the corresponding sequences in GenBank.Results The nucleotide sequence of gyrA had 100% identity in serovar 1,3,6,14 and 100%identity in serovar 2,4,5,7~13,too.But the sequence had 91%identity between the two groups.The nucleotide sequence of parE had 98%~99% identity in serovar 1,3,6,14.And it had 100% identity in erovar 2,5,7,8,11 and 100% identity in serovar4,12,13.But it had only 90% identity between the two groups.Ureaplasma parvum(Up),Ureaplasma urealyticum(Uu)and Up+Uu infection were found 68.3%(41/60),21.7%(13/60)and 10%(6/60) of clinical specimens,respectively.In Up isolates,serovar 3 was 48.8%(20/41).Conclusion Ureaplasma can be divided into two genotypes(Up and Uu)by gyrA analysis.And Up can be divided into four subtypes which correspond to serovar 1,3,6,14,respectively.Serovar 3 is the main isolate in our research.
7.In vitro establishment of a three-dimensional hepatocyte culture system using collagen hydrogel as scaffolds

BACKGROUND:Rapid loss of liver-specific functions of the cultured hepatocytes limits the development of hepatocyte-based therapies. OBJECTIVE:To establish a three-dimensional culture system based on col agen hydrogel that enables to enhance liver-specific functions for a long period during culture of hepatocytes. METHODS:Hepatocytes were isolated from Sprague-Dawley rats and then encapsulated into liquid type Ⅰcol agen solution that was premixed with hepatocyte growth factor and Dulbecco's modified Eagle’s medium to create hepatocyte/col agen hydrogel compounds. The compounds were inoculated into a 96-wel plate. After gelation, culture medium was added. Light microscope, hematoxylin-eosin staining and transmission electron microscopy were used to examine the morphological characteristics and ultrastructure of the cultured hepatocytes. The cellsupernatant was col ected and tested for albumin secretion and urea synthesis. Periodic acid-schiff staining, immunofluorescence staining and quantitative real-time PCR were also used to further clarify liver-specific phenotype or function of the hepatocytes.RESULTS AND CONCLUSION:(1) Light microscope revealed that hepatocytes were round shape and distributed uniformly in col agen hydrogel. The three-dimensional hepatocyte culture system exhibited similarities to liver-like structure and tight junction were formed between hepatocytes after 14 days of culture. (2) Within the three-dimensional culture system, hepatocytes remained positive for periodic acid-schiff staining, albumin and hepatocyte nuclear factor-4αpositive after 14-day culture, which provided the convincing evidence of highly differentiated primary hepatocytes with functions of glycogen and albumin synthesis. (3) The albumin and urea productions in the three-dimensional culture system had a significantly higher level than in the two-dimensional culture, and could remain at a high level at least for 15 days. (4) The expression levels of hepatocyte-specific genes including Albumin, HNF-4α, Claudin-3, CYP1A1, CYP3A1 and G6P were significantly improved in the three-dimensional culture as compared with the two-dimensional culture. The col agen hydrogel based three-dimensional culture system provides a valuable model to enhance the hepatocyte functional maintenance and lay the foundation for the development of hepatocyte-based therapy for liver disease.
8.Design of a noninvasive ventilator's turbine.

The design principles of a noninvasive ventilator's turbine are studied and discussed in this paper. The design is completed from its several aspects and in combination of related theories, using SolidWorks tools. Abundant experimental results prove that this design's technical specifications meet all the requirements.

10.Endoscopic and histopathological characteristics of chronic gastritis,duodenitis and peptic ulcer in children

Objective To investigate the relationship between the endoscopic characteristics of chronic gastritis,duodenitis,peptic ulcer and their histopathologic findings in children,and explore the relationship between Helicobacter pylori(Hp) infection and the severity of histopathologic changes of gastroduodenal mucosa in children. Methods Three hundreds and sixty-six children with chronic upper gastrointestinal symptoms who were examined by gastroscopy were enrolled.The gastric and duodenal mucosal biopsy specimens of all the patients were studied histopathologically. ResultsTypes of chronic gastroduodenal diseases in all these patients were: chronic gastritis(n=206,56.3%),chronic gastritis combined with duodenitis(n=112,30.6%),chronic gastritis combined with peptic ulcer(n=48,13.1%).There was chronic inflammation in gastric mucosa and duodenal bulb mucosa in all the cases examined by histopathologic examination.Hp infection was found in 106 cases(28.96%).The gastric mucosal inflammation was more severe in those with Hp infection than those without(P0.05).The were significant differences in the incidences of inflammation activity,atrophia and nodulus lymphaticus of gastric mucosa between those with Hp infection and those without(P
