Objective To explore the effect of glutamine(Gln) on expression of nuclear factor kappa B(NF-?B) and heat shock protein 70(HSP70) in brain of young rats induced by lipopolysaccharide(LPS).Methods Ten days old Wistar rats were randomly divided into 3 groups by injection intraperitoneally different agonts,LPS group,normal saline control group(NS group) and Gln group(Gln 1.346 g/kg,1 hour before LPS).NF-?B and HSP70 distribution and expression in brain were deteted by immunohistochemistry.The levels of HSP70 in rats brain induced by LPS were detected by Western blot.SPSS 12.0 software was used.Results The nuclei of neuron in cerebral cortex in LPS group obviously cleared at 6 hours.The positive stain of nuclei in Gln group at 2 hours could not be seen.The stain of nuclei in cerebral cortex was weakened in LPS group at 6 hours by immunohistochemistry.HSP70 protein expression decreased with the measurement of Western blot,especially at 24 hours.HSP70 expression in LPS group was similar as that in NS group.The stain of nuclei in neuron in Gln group at 2 hours increased.It also showed the amount of protein expression increased in Western blot in group Gln at 2,6,12,24 hours(Pa

Abstract? AIM: To investigate changes of anterior chamber parameters after posterior chamber phakic implantable collamer lens ( ICL ) surgery and its correlation with intraocular pressure ( IOP) .?METHODS: This was a retrospective case series study. Seventy four eyes in 43 myopia patients were examined by Allegro Oculyzer anterior segment tomography to obtain the changes of anterior chamber volume ( ACV ) , anterior chamber angle ( ACA) , central anterior chamber depth ( ACD) and vault, meanwhile, to measure the IOP to analyze the correlation with anterior chamber parameters.?RESULTS: Compared with preoperative, ACV, ACA, ACD all decreased apparently ( P <0.01 ) in the postoperative period of 1, 3, and 6mo.ACV, ACA and ACD showed their stability in each period after the surgery.There were changes of vault in 1 and 6mo ( t=27.66, P=0.01).IOP had not altered when compared with the preoperative.This research revealed that IOP had no relation with ACV, ACA, ACD and vault in every phase( all P>0.05).?CONCLUSION:For patients underwent ICL, the anterior chamber parameters all decreased which included ACV, ACA, ACD, and had stabilized since early postoperative period. Correspondingly, IOP was stable and had not correlate with ACV, ACA, ACD and vault, however the long-term observation is still necessary.
4.Assessment of Oxygen Uptake Efficiency Slope on Cardiopulmonary Functional Reserve for Patients with Chronic Obstructive Pulmonary Disease

Objective To investigate the efficacy of oxygen uptake efficiency slope (OUES) on evaluation the cardiopulmonary function of patients with chronic obstructive pulmonary disease (COPD). Methods The cardiopulmonary function of 54 stable COPD patients with the cardiopulmonary function of Ⅱ~Ⅳ were evaluated, following a symptom-limited Steep protocol with simultaneous respiratory gas measurement,they were performed exercise tests on a treadmill, simultaneously the oxygen uptake (VO2), carbon dioxide production (VCO2),peak oxygen uptake (VO2peak), minute ventilation (VE), and respiratory gas exchange rate (RER) were measured. OUES was derived from the relation between VO2 and VE during incremental exercise and was determined by VO2=algVE+b, where a=OUES, to measure anaerobic threshold (VAT) meanwhile. Results OUES correlated with the VO2peak (P<0.001). 75% OUES, 90% OUES and 100% OUES were not significantly different (F=0.239, P=0.830). Conclusion OUES can respond the cardiopulmonary function in patients with COPD, 75% OUES from sub-maximal exercise can be an index for cardiopulmonary function.

Anoikis is a form of apoptosis induced upon cell detachment from extracellular matrix.It has been determined that acquisition of resistance to anoikis is a critical step for tumor cell metastasis.MiR-21,the most prominent oncomiR,plays an important role in tumor progression.In this study,we revealed that up-regulation of miR-21 in human esophageal adenocarcinoma (EA) is associated with lymph node metastasis and poor survival rate.Because of the established anti-apoptosis effect of miR-21,it is tempting to speculate that miR-21 might contribute to tumor metastasis by regulating anoikis,qRT-PCR analysis demonstrated that miR-21 expression in OE33/AR cells (subpopulation of human EA OE33 cells that acquired resistance to anoikis) was significantly increased.Also,transfection of miR-21 mimics provided OE33 cells resisting to anoikis.By luciferase assays,we verified that PDCD4 and PTEN were the functional targets of miR-21.In mouse model,via tail vein injection experiment,we showed that the metastasis formation of OE33 cells in vivo could be mediated by changing the miR-21 expression pattern.Taken together,our findings suggested that miR-21 was involved in the regulation of anoikis in human EA cells.Targeting miR-21 may provide a novel strategy to prevent metastasis.

9.Expressions of Telomerase Reverse Transcriptase and Heparanase in Prognosis of Patients with Colorectal Cancer

Objective:The aim of this study was to investigate the relationship among the telomerase reverse transcriptase (TRT), the heparanase gene expressions, and the metastasis and prognosis of colorectal cancer. Method: The expression of TRT in 33 cases of formalin fixed, paraffin embedded colorectal adenocarcinoma tissues was examined using antibody of human TRT and Streptavidin HRP(SP) immunohistochemical method, meanwhile metastasis and 5 year survival rate of the patients were analyzed. The expression of heparanase gene in tweenty fresh colorectal carcinoma tissues were also analyzed by reverse transcriptase polymerase chain reaction (RT PCR). Results: The expression of TRT was found positive in all 33 patients with colorectal cancer, of which 14 cases were strongly positive and 19 weakly positive. TRT was expressed diffusely in the cytoplasm and partly in the nuclei of the cancer cells. Whereas only minimally or weakly positive signals for TRT located mainly in the nuclei with few cytoplasm around, were found in normal or in proliferative colorectal mucosa. The metastatic rate and the 5 year survival rate in tumors with strong expressing TRT were 50.0% (7/14) and 21.4% (3/14), respectively, whereas in those with weak expressing TRT were 15.8% (3/19) and 84.2% (16/19), respectively. There were significant differences between two groups in the metastatic rate (P< 0.05) and the 5 year survival rate (P < 0.001).The expression of heparanase gene was found positive in 20/20 cases of colorectal carcinoma when RT PCR using 0.5 μ g total RNA but positive was only in 11/20 cases of the cancer when using 0.1 μ g total RNA. The positive rate for metastatic tumors was 90.0% (9/10), whereas for non metastatic tumors it was only 20.0% (2/10) when using 0.1 μ g templet of total RNA. Five cases of normal colorectal mucosa were negative for heparanase. Conclusions: The expression of TRT may be corresponding to metastatic rate and 5 year survival rate. The expression of heparanase gene may be related to the malignant degree and metastasis of colorectal cancer. Both TRT and heparanase may be important new markers for predicting the prognosis of the patients with colorectal cancer.
10.Reversion of multidrug resistance (MDR) in human glioma cells by RNA interference (RNAi).

OBJECTIVETo explore whether the constructed vector of short haprin in vivo can induce human glioma cell line BT325 to produce RNAi duplexes and reverse the expression of MDR1 gene.
METHODSThree 62nt oligonucleotide fragments (shRNA) were constructed according to GenBank MDR1 sequence and were cloned to the retrovirus-delivered vectors. After transfected these vectors directly into the human malignant glioma BT325 cells by lipofectamine 2000 with enhanced green fluorescence protein (EGFP) co-transfecting, the MDR1 gene silence effects were detected by the changing level of mRNA and P-glycoprotein including real time PCR (RT-PCR), Northern blot and Western blot analysis. To assess the multidrug resistance against adriamycin (ADR) and VCR, cell proliferation assays were performed by cell counting kit-8.
RESULTSThe RNAi plasmid vectors were constructed successfully. RT-PCR showed MDR1 mRNA was significantly reduced (P < 0.05). Northern blot analysis showed that the gene silence became most intense at 48 hours after transfection. Western blot analysis demonstrated that P-gp expression was reduced at different time to 12.9%, 30.3% and 4.8%, respectively. The chemosensitivity assays indicated that the transfected cells showed an enhanced sensitivity to ADR and VCR. Based on the value of IC(50), BT325 cells had significantly increased sensitivity to the drugs.
CONCLUSIONThe sequence specific RNAi can inhibit MDR1 mRNA and P-gp expression in the glioma cell line. It may reverse multidrug resistance phenotype, therefore, may provide promising therapeutic modalities in the treatment of human glioma.
