The thermostability of the inulinase was studied in this resea rc h. Some alcoholic materials and thickening agent could enhance the thermostabli lity of the inulinase. Using glycerol、xanthic pastern and though orthogonal ex periments of three elements and three levels, a satisfying protective agent, whi ch included glycerin(6%), xanthan gum(0.6%) and CaCl_2 (100mmol/mL) and ha d a significant effect on the enhancement of the inulinase thermostability, was acquired.
2.Creativity and personality analysis in 302 medical students using personality questionnaires.

OBJECTIVETo evaluate the creativity of medical students and study their personality characteristics using personality questionnaires.
METHODSThis investigation was conducted among 310 medical students using 16 Personality Factor Questionnaire (16PF) and Eysenck Personality Questionnaire (EPQ).
RESULTSA total of 302 students completed the questionnaires. The male and female students showed significant differences in two sub-factors with similar creativity. Four subfactors were identified to positively or inversely correlate to the creativity. The number of male students without either high or low scores was greater than that of female students. The incidences of abnormal scores in 16PF showed significant difference between students with typical EPQ scores and those with atypical scores for introversion-extroversion.
CONCLUSIONSThese medical students do not show high creativity in this study. Compared with the male medical students, the female students are more likely to have extroverted personality, and their scores for 16 basic personality factors easily exceed the normal ranges, suggesting the necessity of mental health education. The students with at least 5 abnormal scores in 16PF may show a typical introversion-extroversion personality, and individual psychological counseling can be considered when necessary.

3.hTERT single nucleotide polymorphism is associated with increased risks of hepatocellular carcinoma and tumor metastasis.

OBJECTIVETo investigate the effect of hTERT single nucleotide polymorphisms on the development and metastasis of hepatocellular carcinoma.
METHODSA total of 290 male patients were divided into hepatitis-induced primary hepatocellular carcinoma (HCC) group (n%162), metastatic HCC group (n%22), and control group (n%106). hTERT gene was amplified and hTERT single nucleotide polymorphisms (SNPs) were tested in these subjects.
RESULTSSignificant differences were found in rs2853690 and rs10069690 distribution, but the difference in rs6554743 remained uncertain. The C and T alleles of rs10069690 and rs6554743 showed significant differences between the 3 groups; the carriers of non-T allele of rs10069690 had higher frequencies in both primary and metastatic HCC groups.
CONCLUSIONSome of the polymorphisms of hTERT may increase the risks of development and metastasis of hepatocellular carcinoma.

4.Effects of PTK787 on cell proliferation and expression of fak mRNA in K562.

The aim of this study was to investigate the effects of tyrosine kinase inhibitor PTK787 on cell proliferation, cell cycle and the expression of fak mRNA of human chronic myeloid leukemia (CML) cell line K562, and to explore the mechanism of PTK787 against acute myeloid leukemia. The MTT method was used to detect the effects of PTK787 in various concentrations and at different time points on proliferation of K562 cells; the flow cytometry was used to determine the effects of PTK787 in different concentrations on cell cycle of K562 cells; the RT-PCR was used to assay the expression of fak mRNA in K562 cells treated with PTK787 for 48 hours. The results showed that along with increasing of the concentration and prolonging of time, the inhibitory rate of PTK787 on K562 proliferation was gradually enhanced. The comparison between various concentration groups at same time or comparison between various time groups in same concentration showed significant differences (p < 0.05), in which the effect of 320 micromol/L PTK787 on cells was strongest, while the continuous increase of PTK787 concentration or prolong of action time did not enhance the inhibitory rate on K562 proliferation. With increasing of drug concentration, the cell proportion in G(1) phase gradually increased, the cell proportion in S phase gradually decreased, the comparison between various groups revealed significant differences (p < 0.05), however the continuous increase of drug concentration from 160 micromol/L did not obviously change the cell proportion in phases of cell cycle. With increasing of drug concentration, the expression of fak mRNA in K562 cells gradually reduced with significant differences between various groups (p < 0.05), but with continuous increase of drug concentration from 160 micromol/L, the effect of PTK787 on the expression of fak mRNA in K562 cells also did not obviously change. It is concluded that the PTK787 shows effect of anti-leukemia cells through inhibiting transformation of the K562 cells from G(1) phase into S phase and decreasing the expression of fak mRNA in cells.

5.Establishment of a quality evaluation method for Angelica different processed products from genuine producing areas based on data mining.

The paper reports the development of a quality evaluation method for Angelica different processed products. The data of high-performance liquid chromatography, water, total ash and extract were analyzed with SPSS Clementine 11.0 software. Discriminant analysis (DA) established the classification model and parameter for Angelica different processed products. Fish's discriminant functions of Angelica different processed products were generated using 8 predictor variables selected from 59 indexes. The correct rate of discriminating back substitution is 96.7%. Angelica different processed products can be accurately and reliably recognized and validated with DA of SPSS Clementine 11.0 software.

6.Novel cationic liposome loading siRNA inhibits the expression of hepatitis B virus HBx gene.

In order to solve the problem of selection and in vivo delivery problem in siRNA treatment, hepatitis B virus (HBV) HBx gene which could be targeted by siRNA was studied. The siRNA expression plasmid which specific inhibits HBx expression was obtained by in vitro selection via a dual-luciferase plasmid including HBx-Fluc fusion protein expression domain. The selected siRNA expression plasmid was then encapsulated in PEG-modified cationic liposome, which was devoted into pharmacodynamic studies at both cellular and animal level. The results illustrated that the cationic liposome which encapsulated siRNA expression plasmid could effectively inhibit HBx gene expression both in vitro and in vivo.

7.Anti-mutagenesis effects of total flavonoids of Astragalus.

OBJECTIVETo investigate the anti-mutagenic action of the total flavonoids of Astragalus (TFA).
METHODThree groups of concentrations of TFA and one inducer group were used. The anti-mutagenic action of TFA was studied by experiments in vitro and in vivo including miconucleolus assay, semen teraogenesis test and gene mutagenesis of V79/HGPRT.
RESULTTFA did not affect the weight of the mice during the experiment period. The high concentration of TFA could significantly reduce cyclohophamide-induced miconucleolus number and gene mutagenesis of V79/HGPRT compared with inducer group. However, TFA did not make statistic change in the semen teraogenesis induced by mitomycin C.
CONCLUSIONTFA has antimutagenesis effect.

8.Effects of Notch-1 down-regulation on malignant behaviors of breast cancer stem cells.

This study examined the effect of Notch-1 signaling on malignant behaviors of breast cancer cells by regulating breast cancer stem cells (BCSCs). BCSCs were enriched by using serum-free medium and knocked out of Notch-1 by using a lentiviral vector. Real-time polymerase chain reaction (RT-PCR) and Western blotting were used to detect the Notch-1 expression levels in breast cancer cell lines and BCSCs, and flow cytometry to detect the proportion of BCSCs in BCSC spheres. The BCSC self-renewal, migration, invasion, and tumorigenicity were examined by the tumor microsphere-forming assay and transwell assay and after xenotransplantation. The results showed that the Notch-1 silencing reduced the number of BCSC spheres, the proportion of BCSCs, and the number of cells penetrating through the transwell membrane. It also decreased the size of tumors that were implanted in the nude mice. These results suggest that Notch-1 signaling is intimately linked to the behaviors of BCSCs. Blocking Notch-1 signaling can inhibit the malignant behaviors of BCSCs, which may provide a promising therapeutical approach for breast cancer.
9.The study on metabolic difference of human body affected by active stress and passive stress under special events.

OBJECTIVETo study the metabolic difference of body influenced by active stress and passive stress under special events.
METHODSTo detect serum multiple biochemistry index of 57 earthquake rescue medical team and 13 victims of a natural calamity in Wenchuan earthquake by using Hitachi 7600 automatic analyzer.
RESULTSStress affected biochemistry index deeply. To compared with rescue medical team, the serum ADA, ALP and TG of victims increased obviously and TP, ALB, MAO, Cr, UA, K, Na, Cl, Ca, ApoA1 and HDL decreased obviously.
CONCLUSIONMany biochemistry index have been changed under stress and it relate with stress extent. The human body function status was better in active stress than in passive stress.

10.Genetic study on somatotype of child and adolescent twins in Han nationality.

OBJECTIVETo assess the genetic and environmental influences on the somatotype of children and adolescents, and the effects of sex and age.
METHODSThe components of somatotype were calculated by using Heather-Cater method in a total of 376 twin pairs of Han nationality, including 245 monozygotic (MZ) and 131 like-sex dizygotic (DZ) twin pairs aged 6 to 18 years. Model-fitting method by Mx package was performed to evaluate the proportion of variance components and to analyze the effects of sex and age on each component of somatotype using the adjusted data for other two somatotype components. The heritability of each component in different development periods divided by growth spurt was also evaluated.
RESULTSThe estimated heritabilities of endomorphic, mesomorphic and ectomorphic components were 0.45, 0.80, 0.44 in boys, 0.82, 0.79 and 0.81 in girls respectively after adjusting age. In boys, the heritability of endomorphic component during late puberty was significantly higher than that during pre-puberty (t = 4.99, P < 0.01) and puberty (t = 6.16, P < 0.01), while the heritability of ectomorphic component during late puberty was significantly lower than that during pre-puberty (t = 3.35, P < 0.01) and puberty (t = 4.12, P < 0.01). In girls, the heritability of endomorphic (t = 2.77, P < 0.01) or mesomorphic (t = 2.08, P < 0.05) component during pre-puberty was significantly higher than that in early puberty.
CONCLUSIONThe genetic influence on somatotype of girls should be much more than that of boys, especially on the endomorphic and ectomorphic components. For boys, the mesomorphic component is mainly determined by genetic factors, but the other components are mainly affected by environmental ones. The effects of the development periods on the heritability of somatotype should be paid much attention to.
