1.Development,Reliability and Validity of Child-Neglect Scale

Objective To develop a child neglect scale with Chinese culture background to assess the status of the neglected children in China,and examine the reliability and validity of the child-neglect scale(CNS).Methods Considering the cultural background of China,an item pool was established by revising items in correlative literatures and scales.Then,the first draft of the CNS was improved by reserving the effective items well graded by professional experts.A total of 871 students from 2 junior high schools and a vocational and technical college were involved in the study.Those students were surveyed with Parental Rearing Patterns scale and child neglect scale.Exploratory and confirmatory factor analysis were applied to the development and evaluation of the structure of the scale.Results The findings were as follows:the general Cronbach′s Alpha reliability was 0.85,the split-half reliability was 0.81,the test-retest reliability was 0.90. The CNS was made of the 4 sub-scales which was safe neglect scale,physical neglect scale,communion neglect scale and affection neglect scale.the general Cronbach′s Alpha reliability of the child neglect scale was 0.79-0.85,the split-half reliability was 0.64-0.81,and the test-retest reliability was 0.82-0.90.The item loadings of the neglect scale were over 0.30.The confirmatory factor analysis indicated that the ratio of Chi-square to degrees of freedom were 1.766,the goodness of fit index was 0.917,the Tucker-Lewis index was 0.916,and the root mean square error of approximation was 0.047.Criterion-related validity studies indicated that the scores of the CNS were significantly correlated with the rearing patterns as well(r=0.049,-0.465 P

3.Improvement and optimization of performance verification on enzyme linked immunosorbent assay for determination of hepatitis B markers

Objective To evaluate the performance of enzyme linked immunosorbent assay (ELISA) kit in detection of Hepatitis B virus(HBV) markers by using improved and optimized method ,so as to provide a practical and feasible method and reagents for clinical laboratory .Methods ELISA test was used for the detection of HBV markers .The gradient dilution method was used to e‐valuate the lower limit .The verifiation of cut off value was carried out based on clinical and laboratory standards institute (CLSI) EP12‐A2 document .Samples with cut off values were collected to evaluate the precision ,including repeatability and intermediate precision .The coincidence rates were counted through comparing the results of ELISA with those of external quality assessment and those detected by using Abbott i4000SR chemiluminescence instrument .Results The lower detection limit of HBsAg ,HBsAb , HBeAg ,HBeAb and HBcAb were 0 .2 IU/mL ,20 mIU/mL ,1 NCU/mL ,0 .75 NCU/mL and 0 .05 NCU/mL respectively .The cut‐off value(C50 )± 20% concentration included the concentration range between C5 and C95 .The within‐run coefficient of variation (CV)≤15% ,in sandwich method the between‐run CV≤25% ,in competition method the between‐run CV≤35% .The positive and negative coincidence rates in accuracy and comparing with i 4000SR all were more than 95% and all κ>0 .75 .Conclusion ELISA tests for HBV markers in our laboratory could meet the requirements of the detection performance and clinical needs .

5.Analysis and Discussion on Calculating Likelihood Ratio of DNA Mixture.

OBJECTIVE:
To analyze and discuss four methods of calculating likelihood ratio of DNA mixture.
METHODS:
In the case with CNAS-T0757 proficiency testing in 2013, the likelihood ratios were calculated and compared among four methods, including unrestricted combinatorial method, Clayton's method, p2 principle method, and recommendations from ISFG.
RESULTS:
The likelihood ratios were maximum by Clayton's method and recommendations from ISFO, followed by result of the unrestricted combinational method. The minimum likelihood ratio was obtained by p2 principle.
CONCLUSION
The unrestricted combinational method could give fUrthest consideration to both information preservation and appraiser protection.

6.Correlation analysis of G870A CCND1 gene polymorphism with digestive system tumors.

OBJECTIVETo study the correlation of G870A CCND1 gene polymorphism and digestive system tumors.
METHODSFrom August 2010 to August 2014, 164 digestive system cancer patients (including 82 patients with gastric cancer and 82 with colorectal cancer) and 82 healthy subjects (control group) were examined with PCR-restriction fragment length polymorphism (PCR-RFLP). The distribution of CCND1 gene G870A frequency in the 3 groups and its association with tumor staging and grading were analyzed.
RESULTSThe frequencies of the GG, GA and AA genotypes in G870A CCND1 gene loci in patients with gastric cancer and colorectal cancer differed significantly from those in the control group (P<0.05). G870A CCND1 gene polymorphism was closely associated with an increased risk of digestive system tumors (P<0.05). The GA and AA genotypes were associated with a significantly higher risk of digestive system cancer risk than the GG genotype (P<0.05), and their frequencies were significantly higher in patients with tumors of higher pathological grade and in those in advanced tumor stages (P<0.05).
CONCLUSIONG870A CCND1 gene polymorphism is associated with the risk of digestive system tumors. The allele A is associated with an increased risk of digestive system tumors and correlated with the tumor differentiation and staging of the tumor.
7.Relationship between Childhood Abuse Experience,Parent Attachment and Depression in Adolescents

Objective To explore the relationship between childhood abuse experience,parent and peer attachment and depression in adolescents.Methods A total of 736 junior students from the 7th,8th and 9th grades of 2 public schools in Xinxiang City and Jiyuan City Henan province completed the questionnaires.Childhood Trauma Questionnaire(CTQ),Inventory of Parent and Peer Attachment(IPPA) and Depression Self-Rating Scale for Children(DSRSC) were employed the childhood abuse experience,parent attachment quality and depressive mood in adolescents,respectively.The hierarchical regression analysis was used to examine the relationship between parent and peer attachment,child abused experience and depression.Results Male participants reported high score of childhood abused than female participants(33.29±7.89 and 31.05±6.01,t=4.343,P=0.000).There were no significantly gender difference in mother attachment(male:59.89±15.86,female:61.45±16.48,t=-1.303,P=0.193),father attachment(male:59.32±17.17,female:60.28±16.95,t=-0.764,P=0.445) and depression(male:7.68±5.27,female:7.31±4.21,t=1.057,P=0.291).Childhood abuse had a positive correlation with adolescent depression(r=0.408,P=0.000),which negatively correlated with mother and father attachment(r=-0.544,-0.417,respectively,Pa=0.000).There was a positive correlation between mother attachment and father attachment(r=0.604,P=0.000),and both of them had negative correlation with depressive disorder(both r=-0.426,Pa=0.000).Mother and father attachment were the partial mediation of the relationship between childhood abuse and adolescent depression.Conclusions Male adolescents suffered more maltreatment than female adolescents.There are no gender difference in parent attachment and depression.Parent attachment plays the partial mediator role in childhood abuse and adolescent depression.Results reveal the importance of assessment and consideration of parent attachment in the intervention of depression in abused adolescents.
8.Preliminary Study on Personality Characteristics of Abused Children

Objective To explore the personality characteristics of abused children in order to reduce the incidence of child abuse.Methods Two hundred and ninty five middle school students were investigated with general questionnaire and Eysenck Personality Questionnaire of children. Eighty six students experiencing child abuse (CA) last year as study group and one hundred and ninety six non-abuse children as controls (NCA) were analyzed by means of Eysenck Personality Questionnaire of children.Results The score of neuroticism in CA group was significantly higher than that in the control group (55.62±10.60/52.65±10.98,t=-2.114 P=0.035). The score of lie in CA group was significantly lower than that in control group (42.21±9.87/46.04±9.20,t=3.184 P=0.002). On the impact of different sex, the psychoticism score of male was significantly higher than that in the control group(52.37±11.49/48.04±9.97,t=-2.227 P=0.028), and the lie score was significantly lower than that in control group(41.03±9.18/46.18±8.79,t=3.125 P=0.002).The scores of those in the female were not significant.Conclusions There is a close association between the unstable emotion and child abuse in children, so training emotional self-control and emotional expression of children might be a intervention strategy in the future. In addition, the frequency of lie in children is probably one of factors that determine whether children are abused or not.
9.Treatment of lumbar isthmic spondylolisthesis using titanium-coated interbody fusion system

Objective To study the clinical outcomes of titanium-coated interbody fusion system in treating lumbar isthmic spondylolisthesis.Methods A retrospective study was made in 22 cases with lumbar isthmic spondylolisthesis including 15 with degree I spondylolisthesis and seven with degree II spondylolisthesis treated by posterior lumbar interbody fusion using titanium-coated fusion cages from June 2002 to December 2003 in our hospital.There were six males and 16 females,with age range of 41-68 years(mean 52.5 years).In all cases,double cages were implanted with additional posterolateral bone graft,supplemented with pedicle screw systems.The average preoperative and postoperative disc space height,foraminal height,segmental and lumbar lordosis,spondylolisthesis and domain of sagittal rotation on flexion-extension radiographs were measured and fusion state observed.The Oswestry disability index (ODI)was used to evaluate the clinical outcome.Results The fusion rate was 95.4%.There was significant recovery of disc space height,foraminal height and domain of sagittal rotation.ODI decreased significantly.Conclusion The anatomic titanium-coated fusion system is effective and safe in treatment of lumbar isthmic spondylolisthesis because it has advantages in restoring sagittal alignment and keeping bony endplate intact.
10.Identification of atractylodis macrocephalae rhizoma and atractylodis rhizoma from their adulterants using DNA barcoding.

Atractylodis Macrocephalae Rhizoma and Atractylodis Rhizoma were widely used in strengthening spleen under different disease conditions, and were easily and often misused each other. Therefore, DNA barcode was used to distinguish Atractylodis Macrocephalae Rhizoma and Atractylodis Rhizoma from their adulterants to ensure the safe use. The sequence lengths of ITS2 of Atractylodes macrocephala, Atractylodis Rhizoma (A. lancea, A. japonica and A. coreana) were both 229 bp. Among the ITS2 sequences of A. macrocephala, only one G/C transversion was detected at site 98, and the average GC content was 69.42%. No variable site was detected in the ITS2 sequences of A. lancea. The maximum K2P intraspecific genetic distances of both A. japonica and A. coreana were 0.013. The maximum K2P intraspecific genetic distances of A. macrocephala, A. lancea, A. japonica and A. coreana were less than the minimum interspecific genetic distance of adulterants. The ITS2 sequences in each of these polytypic species were separated into pairs of divergent clusters in the NJ tree. DNA barcoding could be used as a fast and accurate identification method to distinguish Atractylodis Macrocephalae Rhizoma, Atractylodis Rhizoma, from their adulterants to ensure its safe use.
