1. The value of adenosine deaminase, interferon-gamma, and interferon-gamma induced protein of 10KD in the diagnosis of tuberculous pleuritis

Objective To explore the value of adenosine deaminase (ADA) activity, interferon-gamma (IFN-γ) and IFN-γ induced protein of 10kD (IP-10) levels in pleural effusion for the diagnosis of tuberculous pleuritis. Methods ADA activity, IFN-γ and IP-10 levels in pleural effusion were determined in sixty-three patients with tuberculous pleuritis and 50 patients with malignant pleural effusion. Results The mean levels of ADA, IFN-γ and IP-10 in the tuberculous pleural effusion were significantly higher than those in malignant pleural effusion (P<0.01). When 45U/L was regarded as cut off value for ADA, the sensitivity, specificity and diagnostic odds ratio in the diagnosis of tuberculous pleurisy were 71.4%, 94.0% and 39.17 respectively. When 138.5pg/ml was regarded as cut off value for IFN-γ in tuberculous pleural effusion, the sensitivity, specificity and diagnostic odds ratio were 93.7%, 82.0% and 67.19 respectively. When 9.21μg/ml was regarded as cut off value for IP-10 in tuberculous pleural effusion, the sensitivity, specificity and diagnostic odds ratio were 85.7%, 90.0% and 54.00 respectively. The combined determination of the three markers for the diagnosis of tuberculous pleurisy had a sensitivity of 95.2%, specificity of 96.0% and diagnostic odds ratio of 72.16. Conclusion The accuracy of diagnosis for tuberculous pleurisy can be improved by combined determination of ADA, IFN-γ and IP-10.
2.Identication of pearl powder and conch powder from different origins by differential scanning calorimetry.

The paper is aimed to establish a methods for identication of pearl powder and conch powder from different origins. Hermetic aluminum pan was used to encapsulate samples. The optimal testing conditions were: heating rate 10 degrees C x min(-1), sample weight 3 mg and nitrogen gas flow rate 40 mL x min(-1). The enthalpy values of pearl powder and conch powder was obvious different. Identication of pearl powder and conch powder by DSC is a practical method for its accuracy, convenience and practificality.

3.Pharmacokinetic study of recombinant human acidic fibroblast growth factor in rabbits by skin external use.

AIMTo investigate the pharmacokinetic characteristics of recombinant human acidic fibroblast growth factor (rhaFGF) after external use in rabbits.
METHODS125I-rhaFGF 180 U.cm-2 was daubed to normal skin and scathed skin in rabbits. The radioactivity and paper chromatography were used to determine the 125I-concentrations and distribution in plasma and organs at different times.
RESULTSThe plasma concentration of 125I-rhaFGF increased rapidily, and reach peak plasma level (73.03 pg.mL-1) thirty minutes after administration. Then the concentration of 125I-rhaFGF decreased quickly after thirty minutes, and approached to zero after three hours. Highest radioactivity accumulated in the skin, followed by kidney, lowest in the brain 96 h after administration.
CONCLUSIONrhaFGF can not be absorbed from the normal skin, whereas a small amount of rhaFGF can be absorbed through scathed skin. The t1/2 of rhaFGF in plasma was very short. Cumulative effect of rhaFGF was not observed. Absorbed rhaFGF showed high affinity to skin, and can be distributed to skin far from the site of administration.

4.Determination of the fluorine release of fluoride varnishes by neutron activation method.

OBJECTIVETo determine the fluorine content in enamel before and after besmearing fluoride varnishes by neutron activation method.
METHODSA pair of mandibular deciduous central incisor teeth were chosen. One was removed, and the other was besmeared with fluoride varnishes and removed 24 hours later. The contents of fluorine were determined and analyzed statistically.
RESULTSThe fluorine content in the enamel of the tooth besmeared with fluoride varnishes was higher than that in the control tooth (P < 0.05).
CONCLUSIONSThe application of the fluoride varnishes can effectively increase the content of fluorine in the sclerous tissues.

5.Identification of Max binding protein as a novel binding protein of Nck1 and characterization of its role in inhibiting human liver cancer SK-HEP-1 cells.

BACKGROUNDThe tendency of tumor cells to disperse throughout the liver is a distinct feature of hepatocellular carcinoma (HCC). Nck family adaptor proteins function to regulate actin cytoskeletal reorganization that leads to cell motility. We previously found that Max binding protein (MNT) was differentially expressed in HCC, and interacted with Nck1 by 2-DE. MNT is a protein member of the Myc/Max/Mad network which plays roles in cell proliferation, differentiation, and death. We investigated the effects of MNT on migration of human liver cancer SK-HEP-1 cells to study the migration regulatory role of MNT in HCC cells.
METHODSInteraction between MNT and Nck1 was further validated in hepatoma cells by GST-pull down assay and immunoprecipitation. siRNAs specific to MNT (MNT siRNA) were used to knockdown MNT expression. Western blotting, transwell assay were used to determine the migration potential of cells.
RESULTSInteraction between MNT and Nck1 was validated in hepatoma cells. MNT knockdown promoted the migration of human liver cancer SK-HEP-1 cells (P < 0.01).
CONCLUSIONThe results suggest that MNT, via interaction with Nck1, inhibits hepatoma cell migration.

The histological hidradenoma framework consisting of solid and cystic components, and the presence of poroid and cuticular cells resembling a poroid neoplasm. 8.Clinical trial of Qishen Tongluo granules in the treatment of diabetic nephropathy in stage Ⅲ

Objective To observe the clinical efficacy and safety of Qishen Tongluo granules in the treatment of diabetic nephropathy (DN) in stage Ⅲ.Methods One hundred and four patients with DN in stage Ⅲ were randomly divided into control and treatment groups with 52 cases per group.Control group was given potassium choride tablets 100 mg per time,qn,orally.Treatment group was given Qishen Tongluo granule 20 g per time,bid,orally.Two groups were treated for 12 weeks.The clinical efficacy,fasting plasma glucose (FPG),hemoglobin A1 c (HbA1c),homostasis model assessment of insulin resistance index (HOMA-IR),urinary albumin excretion rate (UAER),serum creatinine (SCr) and adverse drug reactions (ADR) were compared between two groups.Results After treatment,the total effective rates of treatment and control groups were 86.54% (45 cases/52 cases) and 69.23% (36 cases/52 cases) with significant difference (P < 0.05).After treatment,the main indexes in treatment and control groups were compared:FPG were (6.54±0.68) and (7.16 ±0.74) mmol · L-1,HbA1c were (7.18 ±0.72)% and (7.62-± 0.79)%,HOMA-IR were (4.71 ±1.06) and (5.65±1.10),UAERwere (58.34 ±16.52) and (82.15±19.05) mg· 24 h-1,SCr were (85.60 ± 15.69) and (94.25 ± 16.77) μmol · L-1,the differences were statistically significant (all P <0.01).The adverse drug reactions in two groups were nausea,abdominal discomfort,lack of appetite and dizziness.The total incidences of ADR were 9.62% and 13.46% without significant difference (P > 0.05).Conclusion Qishen Tongluo granules have a definitive clinical efficacy and safety in the treatment of DN in stage Ⅲ,which can effectively improve the renal function and insulin resistanc,and reduce blood glucose.
9.Solitary sclerotic fibroma on the thigh

Sclerotic fibroma is a rare fibrous tumor of the skin associated with Cowden’s disease. In 1989, Rapini described sclerotic fibroma without Cowden’s disease as solitary sclerotic fibroma of the skin. It is a solid, well-circumscribed, slow-growing nodular tumor and it looks similar to a keloid scar. Consequently, it is extremely difficult to make a differential diagnosis of solitary sclerotic fibroma with keloid scar based on clinical findings only. The authors report a case of solitary sclerotic fibroma arising at the left lateral thigh of a 25-year-old man.
10.Establishment of an erythroleukemia model in CB6F1 mice by transplant with haploidentical mouse leukemic cell line FBL-3.

The purpose of study was to investigate the feasibility for establishing erythroleukemia model in CB6F1 mice by transplant with haploidentical mouse leukemic cell line FBL-3 and to explore the biological characteristics of FBL-3 cells in CB6F1 mice, CB6F1 and C57BL/6 mice were inoculated intravenously at doses of 1×10(3)-1×10(7) FBL-3 cells respectively. The survival time, the count of peripheral white blood cells, the percentage of erythroblasts and chromosome of these mice were observed. The liver, spleen, lung and kidney were obtained from the dying CB6F1 mice for pathological examination. The ultrastructure of erythroblasts in bone marrow and spleen was observed by transmission electron microscopy as soon as these mice died. Expression of MHC molecules and karyotype of spleen and bone marrow cell were measured. The results showed that 100% and 92.5% incidences of erythroleukemia were observed when 1×10(3)-1×10(7) FBL-3 cells had been administrated intravenously to CB6F1 and C57BL/6 mouse, respectively. There was a linear relationship between the survival time and the number of inoculated leukemic cells. The survival time of CB6F1 was longer than C57BL/6 mice inoculated the same number cell. The main targets for FBL-3 cell infiltration were liver, spleen, marrow, lung and kidney. The reaction of FBL-3 cells to glycogen staining was positive, while the to reaction peroxidase, alkaline phosphatase and butyric acid staining were negative, reaction to chloroacetic acid staining partially was positive. Virus-like particles were found in the spleen and bone marrow cells under electron microscope. Chromosomes of spleen and bone marrow cells in the majority were non-diploid, and the expression of H-2b increased, H-2d expression decreased. It is concluded that the erythroleukemia mouse model can be established in CB6F1 mice transplanted with leukemic FBL-3 cells, that provides a convenience experimental erythroleukemia model for study.
