The ubiquitin-proteasome pathway is one of important pathways during selective protein degradation,which participates in many intracellular physiological and biochemical processes.Bortezomib,a kind of proteasome inhibitor,can inhibit cell growth and proliferation,induce cell apoptosis and overcome drug resistance in chemotherapy.The mechanisms of bortezomib on the therapy of multiple myeloma are reviewed in this paper.
2.Effect of Glutamine on Expressions of Nuclear Factor-?B and Heat Shock Protein 70 in Brain of Young Rats Induced by Endotoxin

Objective To explore the effect of glutamine(Gln) on expression of nuclear factor kappa B(NF-?B) and heat shock protein 70(HSP70) in brain of young rats induced by lipopolysaccharide(LPS).Methods Ten days old Wistar rats were randomly divided into 3 groups by injection intraperitoneally different agonts,LPS group,normal saline control group(NS group) and Gln group(Gln 1.346 g/kg,1 hour before LPS).NF-?B and HSP70 distribution and expression in brain were deteted by immunohistochemistry.The levels of HSP70 in rats brain induced by LPS were detected by Western blot.SPSS 12.0 software was used.Results The nuclei of neuron in cerebral cortex in LPS group obviously cleared at 6 hours.The positive stain of nuclei in Gln group at 2 hours could not be seen.The stain of nuclei in cerebral cortex was weakened in LPS group at 6 hours by immunohistochemistry.HSP70 protein expression decreased with the measurement of Western blot,especially at 24 hours.HSP70 expression in LPS group was similar as that in NS group.The stain of nuclei in neuron in Gln group at 2 hours increased.It also showed the amount of protein expression increased in Western blot in group Gln at 2,6,12,24 hours(Pa

Objective To establish the clinical reference of serum homocysteine in Xi’an region.Methods 310 cases of serum of healthy persons were collected to test the homocysteine concentrations using Enzyme circulation method.Results Health-y adult male homocysteine value was significantly higher than female and its reference range was:men 0~1 6.3 5μmol/L and women 0~12.89μmol/L.Conclusion Have established the healthy crowd in Xi’an region serum HCY reference for the re-gion’s heart cerebrovascular disease treatment and prognosis.
7.Management of medical equipment in hospital ship

Objective To explore the medical equipment management to facilitate the medical support in the hospital ship.Methods The influences and the ship-mounted equipment on medical equipment were analyzed,the requirements of sea conditions for medical equipment management were explored,the hidden risks and present situation of medical equipment were introduced,and some countermeasures were put forward accordingly.Results The problems of equipment management were analyzed in hospital ship,and some countermeasures were brought out including bringing into better balance the relations between various sectors,enhancing personnel,introducing professional talents,trying for the support from the director and etc.Conclusion The safety of hospital medical equipment is of great significance to the hospital ship itself and the patient.Only to strengthen medical equipment management while focusing on the qualified personnel contributes tobetter fulfilling its mission.
8.Effect of total flavonoids of astragalus on endoplasmic reticulum chaperone, calumenin and connecxin 43 in suckling mouse myocardium with myocarditis caused by coxsackievirus B3.

OBJECTIVETo investigate the effect of total flavonoids of astragalus on the expression of endoplasmic reticulum chaperone, calumenin and connecxin 43 (CX43) in suckling mouse myocardium with myocarditis caused by coxsackievirus B3 (CVB3).
METHODSThe primary culture of suckling mouse myocardium cells were randomly divided into control group, CVB3 infected group and total flavonoids of astragalus group. Firstly, to confirm the identity of the suckling mouse myocardium, α-SMA was monitored by immunohistochemistry method. Then the protein expression changes of endoplasmic reticulum chaperone-glucose regulatory protein 78 ( GRP78), calumenin and CX43 were detected by Western blot.
RESULTS(1) Compared with that of the control group, the GRP78 expression level in CVB3 infected group was improved, the expression levels of calumenin and CX43 were all reduced. (2) Compared with that of CVB3 infected group, GRP78 expression level was decreased, and the expression levels of calumenin and CX43 were increased in total flavonoids of astragalus group.
CONCLUSIONCVB3 infection may cause endoplasmic reticulum stress of rat myocardium cells by increasing the expression of GRP78 and decreasing the expression of calumenin and CX43. On the other hand, total flavonoids of astragalus can reduce the expression of GRP78 and increase the expression of calumenin and CX43.The results of this experiment may be closely related to the effects of anti-arrhythmia with viral myocarditis caused by CVB3.

9.Acupuncture plus bloodletting therapy for insomnia in blood stasis constitution: a clinical study

Objective: To observe the clinical efficacy of acupuncture plus bloodletting therapy in treating insomnia in people with blood stasis constitution. Methods: Seventy-two patients were randomized into a treatment group and a control group by using the random number table, 36 cases each. The treatment group received acupuncture plus bloodletting therapy, while the control group was intervened by oral administration of estazolam. One-week treatment was taken as a course, for 3 successive courses in total. A follow-up study was conducted 3 months later. The Pittsburgh sleep quality index (PSQI) was evaluated before and after the intervention, as well as in the follow-up. The clinical efficacies were also compared. Results: The total effective rate was 88.9% in the treatment group versus 83.3% in the control group, and the between-group difference was statistically significant (P<0.05). After the intervention, the PSQI scores dropped significantly in both groups (both P<0.05); the between-group difference in PSQI score was statistically significant (P<0.05). The follow-up study showed that the between-group difference in the global score of PSQI was statistically significant (P<0.05). Conclusion: Acupuncture plus bloodletting therapy can produce a more significant efficacy than oral administration of estazolam in treating insomnia in people with blood stasis constitution.
10.The potential effects of endoplasmic reticulum stress on the apoptosis of myocardial cells from mice with heart failure induced by acute viral myocarditis caused by B 3 Coxsackie virus.

OBJECTIVETo explore the apoptotic pathway mediated by endoplasmic reticulum stress in the mouse myocardium with heart failure induced by acute viral myocarditis caused by B-3 Coxsackie virus.
METHODSForty BALB/c male mice were randomly divided into 2 groups (n = 20): the control group and the virus infection group. The BALB/c mouse myocarditis was induced by B-3 Coxsackie virus and the mouse behavior was observed conventionally. All the mice were sacrificed on day 7 and the changes of left ventricular pressure (LVP) and the rate of change of left ventricular pressure (LV dp/dt) were measured. The cardiomyocytic apoptosis was analyzed by TUNEL method and the mRNA expression level of endoplasmic reticulum haperones glucose-regulated protein (GRP)78 and GRP94 was detected by RT-PCR.
RESULTS(1) Compared with those of control group, the parameters of cardiac hemodynamics in the virus infection group were significantly decreased (P < 0.01); (2) Compared with that of control group, myocardial apoptosis was significantly increased in the myocardial cells from mice with heart failure induced by acute viral myocarditis (P < 0.01); (3) The mRNA expression level of GRP78 and GRP94 were increased significantly in the virus infection group compared with the control group.
CONCLUSIONThese findings suggest the endoplasmic reticulum stress may mediate the apoptosis of myocardial cells in the mice myocardium of heart failure induced by acute viral myocarditis caused by B-3 Coxsackie virus.
