OBJECTIVETo study the effect of ultrafiltration-membrane extracts of Radix Rehmanniae Praeparata (UMERRP) on theproliferation and genetic stability of bone marrow-derived mesenchymal stem cells (BMSCs) induced by cadmium chloride (CdCl2).

METHODSProtective effects on the proliferation, micronuclear rates, chromosome aberration rates, and apoptosis rates were observed by micronuclei test, karyotype analysis, and flow cytometry.

RESULTSCompared with the CdCl2 group, UMERRP with different molecular weights at 0. 8 g/L could obviously promote the proliferation (P <0. 05). Compared with the control group, micronuclear rates, chromosome aberration rates, and apoptosis rates were obviously enhanced in the CdCl2 group (P <0. 05). Compared with the CdCl2 group, UMERRP with different molecular weights could obviously decreased CdCl2 induced micronuclear rates, chromosome aberration rates, and apoptosis rates (P <0. 05). Of them, BMSC micronuclear rates and chromosome aberration rates decreased most obvious in UMERRP groups with molecular weight below 10 000 (P <0. 05). The apoptosis rate decreased most obviously in UMERRP groups with molecular weight ranging 100 000 and 200 000 (P <0. 05).

CONCLUSIONUMERRP could reduce CdCl2 induced micronuclear rates, chromosome aberration rates, and apoptosis rates.

To construct knockout vectors containing ampicillin resistant gene and partial sequence of hyaluronidase gene(Hyl)so that Hyl can be knock out by transforming the plasmid into Streptococcus zoopidemics mutans.First,partial sequence of Hyl(Hyl-1)was cloned into the vector of pMD19-T by using DNA of Streptococcus zoopidemics as template,and then a knockout vector pMD19T-SA was constructed,in which Hyl-1 gene was disrupted by inserting ampicillin resistant gene(Amp)from reverse PCR.As expected,the vector was proved to be consisted of Hyl-1-Amp-Hyl-1-pMD19-T.Thereafter,DNA fragment of Hyl-1-Amp-Hyl-1 was subcloned into pBR322 vector,the resulting construct was then checked by PCR and restriction analysis for the proper configuration of the knockout vector pBR322-SA.Both of the knockout vectors were used to transform Streptococcus zoopidemics and one recombinant was obtained in result.From results of PCR and Hyl activity assay,it was indicated that in the recombinant the Hyl gene was disrupted completely.

Background Our previous study demonstrated that curcumin can induce the apoptosis of retinal pigment epithelial (RPE) cells and herein inhibit the proliferation of RPE cells,and it is proved that the intravitreous injection of 0.1mg curcumin has less adverse effect to ocular tissue, inferring a good applicative prospect in clinic. Objective The goal of this experiment was to evaluate the effectiveness of curcumin on the prevention and treatment of experimental proliferative vitreoretinopathy (PVR). Methods PVR models were induced by injection of 0.1ml RPE cells (containing 2×106 cells) into vitreous cavity in 40 eyes of 20 healthy and mature New Zealand albino rabbits.0. 1ml curcumin(0. 1 mg) was then injected into lateral eye of each model rabbit immediately following the injection of RPE cells,and the equal volume of normal saline solution containing 0. 5‰ DMSO was injected into the fellow eye of each model rabbit as controls. On 1,3,7,14,21 and 28 days after injection, the changes of cornea, aqueous humor, lens, vitreous and fundus were examined and recorded by slit lamp biomicroscope, indirect ophthalmoscope,fundus color camera and B-type ultrasonograph to evaluate the inflammatory response. The incidence rate of retinal detachment was calculated and compared between curcumin group and control group. Results The inflammatory reaction in anterior chamber and misty opacity in vitreous were found from 1 day through 3 days after injection, but no obvious proliferative strap and retinal detachment in all of the experimental eyes. On the 7th day after injection, inflammatory reaction was extinct in the anterior chamber of rabbit eyes, and proliferative strap occurred in 14 eyes(75% ) in the control group but only 2 eyes (10% ) in curcumin group,showing significant difference between these two groups (P<0. 01). No retinal detachment was seen in both the two groups. On 14,21 and 28 days after injection, the incidence rate of retinal detachment was 55% ,80% ,95% respectively in control group and that of curcumin group was 10% ,15% ,15% respectively,presenting considerably differences between two groups (P<0. 01, P<0. 01 ,P<0. 01 ). Conclusion Injection of curcumin into vitreous cavity can effectively inhibit the occurrence and development of PVR in rabbit.

Objective To evaluate the value of endobronchial ultrasound elastography in diagnosis of mediastinal lymph nodes by measuring the parameters of endobronchial ultrasound elastography. Methods Between 2016 and 2017, 46 patients with 63 lesions underwent EBUS-TBNA examination were included. Conventional ultrasound and ultrasound elastography were performed respectively to examine 63 lymph nodes before aspiration. Then various features of conventional ultrasound and parameters of ultrasound elastography were analyzed and recorded. Then evaluate the value of ultrasound elastography by pathological results and three-month follow-up data. Result The diagnostic value of ultrasound elastography image type, elasticity score, strain ratio and blue area ratio for malignant lymph nodes were higher than that of conventional ultrasound features in accuracy, sensitivity and specificity (P < 0.05); The accuracy of the boundary was the highest in the diagnosis of malignant lymph nodes. In the ultrasound elastography parameters, the accuracy of the elasticity score was the highest, and the blue area ratio had the largest area under the curve. When the boundary was used as the joint index to judge malignant lymph nodes with elasticity score and area ratio respectively, significantly higher than the diagnostic accuracy of conventional ultrasound and ultrasound elastography alone (P < 0.05). Conclusion The value of ultrasound elastography parameter in mediastinal lymph nodes diagnosis is higher than the conventional ultrasound features and a combination of both applications can improve discrimination accuracy of benign and malignant lymph nodes.

OBJECTIVE: To investigate the prevalence of social anxiety among the fourth-, fifth- and sixth-grade primary school students with myopia in Urumqi, China and the risk factors for social anxiety. METHODS: Stratified cluster random sampling was used to select 552 fourth-, fifth- and sixth-grade primary school students with myopia from four primary schools in Urumqi. A self-designed questionnaire, a social anxiety scale for children and a self-esteem scale were used to investigate the general demographic data and the current status of social anxiety and self-esteem. A multivariate logistic regression analysis was used to investigate the risk factors for social anxiety in primary school students with myopia. RESULTS: Of the 552 children, 173 (31.3%) were found to have social anxiety. The multivariate logistic regression analysis showed that a higher grade, female sex, autocratic family, high myopia, low self-esteem and wearing glasses for more than 2 years were risk factors for social anxiety (P<0.05). CONCLUSIONS There is a serious problem of social anxiety among the fourth-, fifth- and sixth-grade primary school students with myopia in Urumqi. The development of social anxiety is associated with age, sex, degree of myopia, time of wearing glasses, parental education style and self-esteem level.

OBJECTIVETo investigate the current status of social anxiety and depression among the fourth-, fifth-, and sixth-grade students in Urumqi, China, and to provide data support for mental health education for primary school students in the future.

METHODSStratified cluster random sampling was performed to select 919 fourth-, fifth-, and sixth-grade students from four primary schools for the Han and minority ethnic groups in Urumqi as research subjects. Social Anxiety Scale for Children and Children's Depression Inventory were used to evaluate the current status of social anxiety and depression.

RESULTSAmong the 919 fourth-, fifth-, and sixth-grade students, the detection rate of social anxiety was 28.5% (262 students). The fourth-grade students had significantly lower scores on each subscale of social anxiety and total score of social anxiety than the fifth- and sixth-grade students (P<0.05). Girls had significantly higher score on the subscale of fear of negative evaluation and total score of social anxiety than boys (P<0.05). The Uyghur students had significantly higher scores on each subscale of social anxiety and total score of social anxiety than the Han students and the students of other minority ethnic groups (P<0.05). Among these fourth-, fifth-, and sixth-grade students, the detection rate of depression was 11.2% (103 students). The fourth-grade students had significantly lower scores on the subscales of negative self-esteem, ineffectiveness, and interpersonal problems and total score of depression than the fifth- and sixth-grade students (P<0.05). The Uyghur students had significantly higher scores on the subscales of anhedonia and negative mood and total score of depression than the Han students and the students of other minority ethnic groups (P<0.05).

CONCLUSIONSThere are significant differences in the status of social anxiety and depression among the primary school students with different grades, sexes or ethnic groups in Urumqi. The fifth- and sixth-grade students, girls, and Uyghur students tend to have more serious psychological problems, which should be taken seriously by parents, schools, and the society.

OBJECTIVE: To investigate the association between parental feeding behaviors and body mass index (BMI) of children, and to provide a scientific basis for the prevention and intervention of overweight and obesity in children. METHODS: Stratified random cluster sampling was used to select 976 children from 7 kindergartens in Xinshi District of Urumqi, and a questionnaire survey and physical measurement were performed to obtain related data. RESULTS: A total of 976 questionnaires were distributed and 924 (94.7%) usable questionnaires were collected. The overall detection rates of underweight, overweight, and obesity in children were 3.1% (29 children), 9.2% (85 children), and 6.7% (62 children), respectively. The highest level of parental feeding behavior was monitoring, followed by restriction and pressure to eat. The parents of the Han children had significantly higher levels of restriction and pressure to eat than those of the Uygur children (P<0.01). As for parental feeding behaviors, restriction to eat was positively correlated with BMI of the Han and Uygur boys (P<0.01); pressure to eat was negatively correlated with BMI of the Han boys and girls (P<0.01) and was positively correlated with BMI of the Uygur boys and girls (P<0.01); monitoring was negatively correlated with BMI of the Han and Uygur boys and girls (P<0.05). Among the parents of the Han and Uygur boys, the parents of the overweight/obese children had significantly higher scores of restriction to eat than those of the children with normal weight (P<0.05); among the parents of the Uygur boys and girls, the parents of the overweight/obese children had significantly higher scores of pressure to eat than those of the children with normal weight (P<0.01); among the parents of the Uygur and Han boys and girls, the parents of the overweight/obese children had significantly lower scores of monitoring than those of the children with normal weight (P<0.01). CONCLUSIONS The parental feeding behavior is good in Urumqi, and the parental feeding behavior of the Uygur children is a little better than that of the Han children. Parental feeding behavior is closely correlated with BMI of children, and such correlation differs across ethnic groups and sexes. A high level of monitoring and low levels of restriction and pressure to eat may help to prevent and control the development of overweight and obesity in children.

BACKGROUNDTo evaluate the safety of intracytoplasmic sperm injection (ICSI) with epididymal or testicular sperm, this study compared children born after ICSI treatment with epididymal or testicular sperm with children conceived after ICSI with ejaculated sperm.

METHODSThis retrospective study included 317 children born after ICSI with percutaneous epididymal sperm aspiration (PESA), 103 children born after ICSI with testicular sperm aspiration (TESA), and a control group of 1008 children born after ICSI with ejaculated sperm. All of the patients received their assisted reproductive treatment in the Reproductive Medicine Center of the First Affiliated Hospital of Zhengzhou University from January 2004 to December 2011. Data, such as the rate of stillbirths, perinatal mortality, gestational age, birth weight, and the rate of congenital malformations of the three groups, were compared.

RESULTSPESA and TESA children were not different from ICSI children in the rate of stillbirths, perinatal mortality, infant mortality rate, gestational age, the rate of prematurity, and the rate of malformations (P > 0.05). A slight increase in birth defects was reported in the TESA group compared with those in the control group, but there was no significant difference between the groups (P > 0.05).

CONCLUSIONICSI with epididymal or testicular sperm does not lead to more stillbirths or congenital malformations compared with ICSI using ejaculated sperm.

Objective To test the reliability and validity of Chinese version of Illness Cognition Questionnaire(ICQ)in women with breast cancer. Methods From October,2016 to May,2017,297 women with breast cancer were investigated with the Chinese version of the ICQ and Brief Illness Perception Questionnaire(B-IPQ).The reliability and validity of the Chinese version of ICQ were tested. Results The Cronbach's α of three subscales of ICQ(helplessness,acceptance and perceive benefits)were 0.855,0.857, 0.878,and the test-retest reliability of three subscales were 0.946,0.963,0.901(P<0.001).A confirmatory factor analysis was tested,χ2/df=2.500,SRMR=0.063,CFI=0.920.The factor load capacity of each subscales was 0.37 to 0.84 and the correlation of helplessness,acceptance and perceive benefits was-0.49 to 0.81(P<0.001).The correlation coefficient of criterion-related validity of the helplessness subscale of ICP with B-IPQ was 0.727(P<0.001). Conclusion The Chinese version of ICQ has good reliability and validity,which is a valuable research tool in clinical studies of psychological cognition of patients with breast cancer.

BACKGROUNDDifferential diagnosis of isolated calf muscle vein thrombosis (ICMVT) and gastrocnemius hematoma is essential for early identification of deep vein thrombosis (DVT). This study aimed to investigate the diagnostic value of high-frequency color Doppler ultrasound for differential diagnosis of ICMVT and gastrocnemius hematoma.

METHODSA retrospective case series of 35 ICMVT (M:F, 21:14; mean age (64.5 ± 10.6) years) and 23 gastrocnemius hematoma (M:F, 16:7; mean age (75.4 ± 11.8) years) patients with bilateral/unilateral lower limb pain was conducted between January 2006 and September 2012. Characteristics and the morphology of high-frequency color Doppler ultrasonography of the lower limb deep vein, great saphenous vein, calf muscles, skin, and soft tissue were examined.

RESULTSICMVT hypoechoic signals were characterized by long, tube-like masses on longitudinal sections and oval masses on transverse sections, with apparent muscle thrombosis boundaries, distal and proximal venous connections, and, often, lower limb DVT. Gastrocnemius hematoma hypoechoic signals were characterized by large volumes, enhanced posterior hematoma echo, hyperechoic muscle boundaries, no hematoma blood flow, and no DVT, and clear differences in trauma/exercise- and oral anticoagulant-induced hematomas were readily apparent. According to the measurement, the ratio of long diameter/transverse diameter (D/T) in ICMVT patients was about less than 2.0, whereas in gastrocnemius hematoma patients the ratio was more than 2.0. Early stage isoechoic and hypoechoic signals were detected with gradually increasing ovular anechoic areas. Partial muscle fibers in the hematoma due to muscle fractures were apparent.

CONCLUSIONHigh-frequency color Doppler ultrasound was found to be a sensitive and reliable method for differential diagnosis of ICMVT and gastrocnemius hematoma due to trauma and exercise or prolonged oral anticoagulant use.
