2.Coordination mechanism and antioxidant activity of L-hydroxyproline zinc(II).

The coordination compound of L-hydroxyproline (Hyp)-Zn (II) was synthesized with Hyp and zinc sulfate as raw materials in water medium, coordination Synthesizing Mechanism and Antioxidant Activity of Hyp-Zn(II) coordination compound has been researched. Compared with Hyp, the infrared spectrogram of Hyp-Zn (II) coordination compound emerge a new absorption peak at 1100 cm(-1). Conclusion could be obtained that there exists a coordination effect between Hyp and ZnSO4; TG and DSC curve of Hyp and Hyp-Zn(II) coordination compound were analysed. Compared with Hyp, the peak of Hyp-Zn(II) disappear at 290 degrees C and 375 degrees C. This phenomenon confirmed the front conclusion; At the NMR graph of Hyp-Zn(II) coordination compound, the disappearance of the alpha-carboxyl-hydrogen and alpha-hydroxyl-hydrogen's peak at 3.5-3.9 ppm could indicate that combination's position of Hyp is alpha-carboxyl and alpha-hydroxyl; Structure of Hyp-Zn(II) coordination compound were exosyndrome by the Atomic Force microscopy. It is showed that centr-atom Zn(II) was surrounded by several Hyp at Hyp-Zn(II) coordination compound's phase diagram; The proportion of Hyp-Zn(H) coordination compound was determined by dialysis experiment, the proportion is 4:1; Above-mentioned determination and exosyndrome indicated that the molecular formula of Hyp-Zn (II) coordination compound is Zn(Hyp)4.H2O. The results indicated that the Hyp-Zn(II) coordination compound can inhibit hydroxyl free radicals of Zn(II), and the Percentage of Inhibition is 75.5%; the total antioxidant activities of Hyp-Zn(II) coordination compound is 80.167 u/mL, the anti-superoxide activities of Hyp-Zn (II) coordination compound is 53.19 u/mL.

3.Correlation between DNA load of human papillomavirus and recurrence of condyloma acuminata.

OBJECTIVETo determine the correlation between DNA load of human papillomavirus (HPV) and recurrence of condyloma acuminata (CA).
METHODSThe HPV6/11 and HPV16/18 DNA load of 31 cases of primary CA and 32 cases of recurrent CA were detected by real-time fluorogenic quantitative PCR.
RESULTSAmong the 63 CA patients, 62 cases were HPV6/11 DNA positive. The positive rate was 98.4%. The ranges of HPV6/11 DNA load in primary and recurrent CA were 1.4x10(3)-6.7x10(7) copies/ml and 1.2x10(4)-3.6x10(8) copies/ml respectively. Of 62 cases with HPV6/11 DNA positive, 7 cases were HPV16/18 DNA positive (11.3%). The ranges of HPV16/18 DNA load in primary and recurrent CA were 1.9x10(3)-1.6x10(4) copies/ml and 1.4x10(5)-1.7x10(7) copies/ml respectively. The HPV6/11 and HPV16/18 DNA load in recurrent CA were higher than in primary CA (P < 0.05). The DNA load of HPV6/11 was positively correlated with times of recurrence and course of disease (r=0.37 and 0.30 respectively).
CONCLUSIONCertain correlation exists between DNA load of HPV and recurrence of CA.

4.Relationship between Helicobacter pylori infection and Henoch-Schonlein purpura with gastrointestinal involvement in children.

OBJECTIVETo study the relationship of Helicobacter pylori (H. pylori) infection with the development and relapse of Henoch-Schonlein purpura (HSP) with gastrointestinal involvement in children.
METHODSThirty-six HSP children with gastrointestinal manifestations and 16 of 32 HSP children without gastrointestinal involvement underwent gastroscopy and rapid urease test for H. pylori detection. Thirty healthy children served as the control group. All of the patients received 14C urea breath test and serum H. pylori antibody detections. H. pylori infection was definited when two of detection approaches demonstrated positive.
RESULTSTwenty-one of 36 HSP patients with gastrointestinal manifestations were confirmed with H. pylori infection (58.3%). Of them, the relapsed patients had an H. pylori positive rate of 81.3% (13/16), which was significantly higher than that of the newly diagnosed patients (45.0%, 9/20) (chi(2)=4.49, P < 0.05). Nine of 32 HSP patients without gastrointestinal manifestations were confirmed with H. pylori infection (28.1%); 2 of 30 healthy children showed H. pylori positive (6.7%, 2/30). There was a significant difference in the H. pylori positive rate among the three groups (chi(2)=14.7, P < 0.01).
CONCLUSIONSH.pylori infection may be associated with the development and relapse of HSP with gastrointestinal involvement in children.

5.Two novel STK11 mutations in three Chinese families with Peutz-Jeghers syndrome.

BACKGROUNDPeutz-Jeghers syndrome (PJS) is an autosomal dominantly inherited disease. STK11/LKB1 gene germline mutations have been identified as responsible for PJS. In our study, we investigated the molecular basis of PJS and evaluated correlation between the STK11 mutations and the Chinese population.
METHODSWe collected three pedigrees of PJS and screened the 9 exons and their flanking intronic sequences of STK11/LKB1 gene in the probands and normal individuals in the families using polymerase chain reaction (PCR) and direct sequencing.
RESULTSSequencing of the STK11 gene in the probands of 3 families revealed two novel mutations (c180C-->G and c998-1002delGCAGC) in exon 1 and exon 8, respectively. The mutation of c180C-->G resulted in a premature termination codon. The other mutation, a deletion of five nucleotides (998-1002delGCAGC) in exon 8, predicted to generate a translational frameshift and a termination at codon 1070.
CONCLUSIONSThe growing number of mutations in PJS pedigrees suggests the molecular basis of PJS. STK11 gene mutation can be detected in most patients with PJS.

6.Comparison between immunofluorescence and PCR in detecting human papilloma virus in condyloma acuminata.

OBJECTIVETo compare the effectiveness of immunofluorescence and polymerase chain reaction (PCR) in detecting human papilloma virus (HPV) in condyloma acuminata (CA).
METHODSHPVs in CA tissues from 60 patients were detected by immunofluorescence and PCR, respectively. Different subtypes of HPVs were also identified with restriction fragment length polymorphism (RFLP).
RESULTSThe positive detective rates of immunofluorescence and PCR were 56.67% (34/60) and 96.67% (58/ 60), respectively (P < 0.01). RFLP results showed HPV6 and HPV11 were the main subtypes in the detected virus, which accounted for 98.28%.
CONCLUSIONThe sensibility of PCR is superior to that of immunofluorescence.

7.Expressions of CD1a and CD83 of Langerhans cells in the local lesions of epidermodysplasia verruciformis patients.

OBJECTIVETo investigate the expressions of CD1a and CD83 of Langerhans cells (LC) in the lesions of epidermodysplasia verruciformis (EV) patients.
METHODSWe used immunohistochemical method to detect the expressions of CD1a and CD83 in the lesions of 10 patients with EV lesions and in the skins of 10 normal subjects.
RESULTSNo CD83 + LCs was detected in all EV patients and normal controls, but CD1a + LC was found in all cases. The quantity of CD1a + LCs in the lesions of EV patients was significantly lower than that in the normal skin (P < 0.01); furthermore, the distribution of LCs in EV lesions was uneven.
CONCLUSIONThe functions of LCs may be inhibited in EV patients.

8.Construction and immunogenicity of human papillomavirus type 6b L1 recombinant plasmid.

OBJECTIVETo construct a DNA vaccine as a prophylactic model to prevent condyloma acuminatum and detect its immunogenicity in mice.
METHODSThe major capsid protein (L1) gene of human papillomavirus (HPV) 6b was inserted into an eukaryotic expression plasmid (pcDNA3.1). The recombinant plasmid was transfected into COS-7 cells. Western blot were performed to detect whether L1 protein can be expressed in eukaryotic cells. Eighteen female BALB/c mice were tested for immunogenicity study.
RESULTSThe recombinant plasmid (pcDNA3.1-HPV6bL1) was verified as HPV6b L1 gene by sequencing. Western blot showed specific strip. Anti-L1 protein antibodies could be detected in the mice's sera inoculated with pcDNA3.1-HPV6bL1. Similarly, IL-4, IL-2, and IFN-gamma were increased in the same mice.
CONCLUSIONHPV6b L1 recombinant plasmid was constructed successfully which had immunogenicity for BALB/c mice. It provided experimental evidence for the research of DNA vaccine of condyloma acuminata.

9.Female urogenital mycoplasma infection and drug sensitivity status in Changsha.

OBJECTIVETo survey mycoplasma infection in female urogenital tract and analyze the drug sensitivity of mycoplasma in Changsha.
METHODSUreaplasma urealyticum (Uu) and Mycoplasma hominis (Mh) were detected in 6566 cases of female urogenital tract infection by means of mycoplasma culture and drug sensitivity reagent kit. Sensitivity tests for 10 antibiotics were also performed.
RESULTSA total of 2938 cases were mycoplasma-positive (positivity rate of 44.75%), including 2469 Uu-positive cases (37.6%), 52 Mh-positive cases (0.08%) and 417 cases positive for both Uu and Mh (6.35%). Josamycin, doxycycline, clarithromycin and azithromycin were more effective against Uu infection. Josamycin, doxycycline and thiamphenicol were more effective against Mh infection. Mixed infection with Uu and Mh was more resistant to most antibiotics but Josamycin and doxycycline.
CONCLUSIONThe female urogenital mycoplasma infection results mainly from Uu. Compared with simple Uu or Mh infection, mixed infection with Uu and Mh has significantly greater resistance to a wider variety of drugs. Josamycin and doxycycline are the primary choice for female urogenital mycoplasma infection in Changsha.

10.Immunogenicity of mutant and wild HPV16 DNA vaccines.

OBJECTIVETo evaluate the efficacy of mutation of human papillomavirus 16 (HPV16) E7 in two zinc-binding motifs on HPV16 E7 C terminus on antigen-specific immunity.
METHODSpcDNA3.1/16E7 and pcDNA3.1/ME7 were successfully constructed by inserting the E7 (ME7) into pcDNA3.1 BamH I, EcoR I cut sites. After intramuscular injection with pcDNA3.1, pcDNA3.1/16E7, and pcDNA3.1/16ME7 on C57BL/6 mice, splenocytes from vaccinated mice was isolated. After have been stimulated with E7-specific peptide, interferon gamma (IFN-gamma), interleukin 2 (IL-2), and cytotoxic T lymphocyte (CTL) were detected by ELISA, Eli-spot, and LDH assay respectively; splenocytes without E7 peptide stimulation were used as control group.
RESULTSSplenocytes from mice vaccinated with pcDNA/ME7, stimulated with E7 peptide, generated significantly larger number of E7-specific IL-2 compared with pcDNA3.1/16E7, pcDNA3.1, and control group. The E7-specific IL-2 generated in pcDNA-ME7 group was 5-fold of that of pcDNA3.1/16E7, and the difference was statistically significant (P < 0.05). Splenocytes from mice vaccinated with pcDNA/ME7 and stimulated with E7 peptide, generated significantly larger number of E7-specific IFN-gamma compared with other vaccines. pcDNA-ME7 generated a 2-fold increase in the number of E7-specific IFN-gamma compared with wild-type E7 and the difference was statistically significant (P < 0.05). The highest CTL activity in mice vaccinated with pcDNA/ME7 at an E:T ratio of 45:1 was achieved compared with mice vaccinated with other vaccines. The percents of specific lysis generated by pcDNA3.1/ME7, pcDNA3.1/E7, pcDNA3.1, and without vaccination were of (28.7+/-1.2) %, (55+/-2.2) %, (12.5+/-2.0) %, and (11.5+/-1.2) % respectively, and significant difference existed between the former and the latter two groups (P < 0.05). However, no significant difference was found among all the groups without specific E7 peptide stimulation (P > 0.05).
CONCLUSIONSThe mutation of zinc-binding motifs on HPV16 E7 C terminus may greatly enhance the immunogenicity.
