1.The expression and molecular mechanisms of SH2-B in hepatocarcinoma

Objective To observe the expression and influence of SH2-B in hepatocarcinoma,and to investigate the molecular mechanisms of canceration in hepatocarcinoma.Methods By using SABC imunohis-tochemistry,the expressions of SH2-B were detected in 27 cases of hepatitis,29 cases of hepatocirrhosis and 47 cases of hepatocarcinoma.Hepatocarcinoma cell (HepG)2 with a low-expressed SH2-B was selected using immunofluorescence assay.There were 3 groups:the transfected group (transfected with pcDNA3.1 -SH2-B), the vector group (transfected with pcDNA3.1 )and the blank group (without transfection).After gene transfec-tion,SH2-B expression was detected by Western blotting;cell proliferation was measured by MTT assay;cell colony was counted by colony formation test;and cell cycle was analyzed by flowcy tometer.Results The posi-tive rate of SH2-B in hepatocarcinoma (95.7%)was significantly higher than 55.2% in hepatocirrhosis (χ2 =1 8.64,P <0.01 )and 25.9% in hepatitis (χ2 =40.01 ,P <0.01 ).After being transfected with pcDNA 3.1 -SH2-B,SH2-B expression dramatically increased in HepG2 cells.After cultured for 48 h,the average optical density value of the transfected group was 1 .1 2 ±0.1 9,obviously higher than 0.45 ±0.1 1 in the vector group (t =-31 .55,P <0.01 ),which indicated that cells proliferation was significantly enhanced after being trans-fected with SH2-B.The cell colony numbers of the transfected group was 1 66 ±1 4,significantly higher than
82 ±8 in the vector group (t =-20.33,P <0.01 )and 78 ±9 in the blank group (t =-1 9.64,P <0.01 ), which indicated that the cell colony numbers increased after being transfected with SH2-B.The S stage cells of the transfected group was (45.7 ±5.8)%,significantly higher than (1 9.4 ±4.7)% in the vector group (t =-20.33,P <0.01 )and (20.5 ±5.1 )% in the blank group (t =-34.69,P <0.01 ),which indicated that SH2-B could enhance promote cell cycle of HepG2 cells.Conclusion The expression of SH2-B in hepatocar-cinoma is high,and it may be involved in the canceration of hepatocarcinoma though promoting cell cycle,cell proliferation and cell transformation.

3.Research on correlation between E-cadherin mRNA expression and lung cancer

Objective To explore the correlation of E-cadherin(E-cad) mRNA expression with tumorigenesis and development of lung cancer.Methods The relative quantitative nested RT-PCR was used to detect the expression of E-cad mRNA in cancerous tissues,adjacent and distal tissues of tumor from 50 lung cancer patients,and non-cancerous benign lung tissues from 5 lung disorder patients.Results The means of expression rate of E-cad mRNA in cancerous tissue,adjacent and distal tissues of tumor,and non-cancerous tissues were 1.33?0.53,1.65?0.60,2.01?0.46 and 2.09?0.09 respectively.The data were evaluated by analysis of variance.There were significant difference among cancerous tissue,adjacent and distal tissue(F=18.810,P0.05).Also,there were no significant differences among squamous cell carcinoma,adenocarcinoma and adenosquamous carcinoma in the group of cancerous,adjacent and distal tissue.Conclusion The reduced expression of E-cadherin mRNA may play important role for tumorigenesis and development of lung cancer,and there was no evident relationship of E-cad mRNA with different pathological types of lung cancer.
4.Analysis on the infection and drug susceptibility of mycoplasma in 1 9 530 cases of urogenital tract infections

Objective To analyze the status of mycoplasma infection and drug resistance in the local area,and provide the ba-sis for clinical rational drug use.Methods The specimens obtained from 1 9 530 patients with urogenital tract infection were detec-ted by adopting mycoplasma culture,identification and drug sensitivity integration kit.Mycoplasma infection and drug susceptibility were analyzed.Results In the total of 1 9 530 suspected patients specimens,1 1 1 78 cases were positive with a positive rate 57.24%.The positive rate of ureaplasma urealyticum (Uu)and mycoplasma hominis (MH)was 44.63% and 0.44% respectively and the positive rate of Uu and Mh mixed infection was 12.1 7%.The positive rate of female was higher than that of male and the difference was statistically significant(P <0.05).The positive rate of mycoplasma in 2008-2014 was on the rise;The sensitive rate of mycoplasma to josamycin,doxycycline,minocycline element,clarithromycin was 88.57%,84.32%,76.09% and 71.53% respec-tively,mycoplasma was highly drug resistance to quinolone antibiotics;mixed infection resistance was higher than that of single in-fection;The number of drug resistance of Uu,MH and Uu+MH to 12 kinds of antibiotics increase.Conclusion Mycoplasma infec-tion in urogenital tract is mainly caused by Uu and Mh infection is in mixed infection way;josamycin,doxycycline is the first choice for treatment of mycoplasma in this region.Rational drug choise can be based on the drug susceptibility test.Multiple drug resist-ance of mycoplasma is serious and should be paid attention to.
5.Research of correlation between polyunsaturated fatty acids and postpartum depression

Objective To research whether the polyunsaturated fatty acids (PUFAs) level in prenatal and postpartum women af‐fect the incidence of postpartum depression .Methods 102 eligible and voluntary parturients of full‐term normal delivery were se‐lected and the serum specimens before labor and on postpartum 3 ,42 d were respectively collected for detecting serum PUFAs lev‐els .The perinatal depression related factors questionnaire and the Edinburgh Postnatal Depression Scale (EPDS) were filled in and the scores were evaluated for analyzing the incidence of postpartum depression .Results Among 102 pregnant women and parturi‐ents ,the postputum depression appeared in 28 cases with the positive rate of 27 .5% .The incidence rate of postpartum depression in the eicosapentaenoic acid (EPA) low values group and the docosahexaenoic acid (DHA) low and medium values groups was higher than that in the high values groups ;the incidence rate of postpartum depression in the arachidonic acid (AA) high value group was the higher than that in the low value group .Conclusion The lower the EPA and DHA levels ,the easier to develop depression .
6.Anti-tumor metastatic constituents from Rhodiola wallichiana.

To study the anti-tumor metastatic constituents in Rhodiola wallichiana (HK) S H Fu var Cholaensis (Praeg) S H Fu, chemical constituents were isolated and purified by repeated column chromatography (silica gel, Toyopearl HW-40C and preparative HPLC). Their structures were elucidated on the basis of spectral data analysis. The anti-tumor metastasis assay was applied to evaluate the activities of the isolated compounds. Ten compounds (1-10) were isolated and their structures were identified by comparison of their spectral data with literature as follows: syringic acid (1), salidroside (2), tyrosol (3), scaphopetalone (4), berchemol (5), 2,6-dimethoxyacetophenone (6), rhobupcyanoside A (7), miyaginin (8), chavicol-4-O-β-D-apiofuranosyl-(1 --> 6)-O-β-D-glucopyranoside (9), eugenyol-O-β-D-apiofuranosyl-(1 --> 6)-O-β-D-glucopyranoside (10). Compounds 4-6 and 8-10, were isolated from this genus for the first time, while compound 7 was isolated from this plant for the first time. Compounds 2, 6-8 showed positive anti-tumor metastatic activities, and compounds 2 and 8 showed significant anti-tumor metastatic activities.

8.Study on material base of Carthamus tinctorius with antioxidant effect based on selective knock-out.

OBJECTIVETo establish a method for studying efficacious materials of traditional Chinese medicines from an overall perspective.
METHODCarthamus tinctorius was taken the example. Its major components were depleted by preparing liquid chromatography. Afterwards, the samples with major components depleted were evaluated for their antioxidant effect, so as to compare and analyze the major efficacious materials of C. tinctorius with antioxidant activity and the contributions.
RESULTSeven major components were depleted from C. tinctorius samples, and six of them were identified with MS data and control comparison. After all of the samples including depleted materials are compared and evaluated for their antioxidant effect, the findings showed that hydroxysafflor yellow A, anhydrosafflor yellow B and 6-hydroxykaempferol-3, 6-di-O-glucoside-7-O-glucuronide were the major efficacious materials.
CONCLUSIONThis study explored a novel and effective method for studying efficacious materials of traditional Chinese medicines. Through this method, we could explain the direct and indirect contributions of different components to the efficacy of traditional Chinese medicines, and make the efficacious material expression of traditional Chinese medicines clearer.

9.The effect of medical assessment on the rehabilitation training of autism spectrum disorder children

Objective To explore the effect of medical assessment on the rehabilitation training of children with autism spectrum disorders (ASD). Methods There were 67 children with ASD selected from special education schools .Their developmental level , ability of social life and social communication were evaluated by child health care physicians .There were 48 children classified as intervention group:individualized rehabilitation training targets were designed based on the assessment results and individualized rehabilitation training program was carried out .The other 19 children were served as controls and routine rehabilitation training was carried out .All children were reevaluated by physicians with the same methods after training for one year . Results The adaptability , language level and communication ability of children in the intervention group improved better than the control group , with statistically significant difference. Conclusion It is beneficial to develop training programs for children with ASD on the basis of medical assessment which can promote the development level of ASD children , social life ability and communication ability .
10.Analysis of Th1/Th2 response pattern for erythrodermic psoriasis.

As one of the most serious types of psoriasis, pathogenesis of erythrodermic psoriasis (EP) is unclear so far. In this study, we aimed to detect the levels of Th1/Th2 cytokine-associated transcription factors and T-lymphocyte clone in peripheral blood mononuclear cells (PBMCs) derived from EP patients, and gene expression level of T-bet/GATA-3 in skin lesion. The potential role of Th1/Th2 reaction pattern played in the pathogenesis of EP was also discussed. Serum levels of IFN-γ, IL-2, IL-4 and IL-10 were quantified by ELISA among 16 EP patients, 20 psoriasis vulgaris (PV) patients and 15 healthy controls. The expression levels of T-bet/GATA-3 in the skin lesion and PBMCs were examined by real-time qPCR. The ratio of Th1/Th2 was measured by flow cytometry. The levels of IFN-γ, IL-2, IL-4 and IL-10 were higher in EP patients than in the healthy controls. The levels of IL-4 and IL-10 were 69.44±11.45 and 12.62±4.57 pg/mL, respectively, in EP patients, significantly higher than those in PV patients and healthy controls (P<0.05). Flow cytometry revealed the levels of both Th1 and Th2 in PBMCs from EP patients were higher than those in healthy controls, and the Th1/Th2 ratio was dramatically lower than in PV patients (P<0.01). The ratios of IFN-γ/IL-4 and T-bet/GATA-3 in EP patients were both less than 1.0, suggesting a reversal when compared with the other two groups. Our study indicated that the EP patients exerted a Th1/Th2 bidirectional response pattern, and the balance of Th cell subsets inclines to Th2, which might be one of the important mechanisms of EP pathogenesis.
