1. Study on pharmacokinetics and relative bioavailability of rapid oral disintegrating tablet of dimenhydrinate

Objective: To compare the pharmacokinetics and relative bioavailability of rapid oral disintegrating tablet of dimenhydrinate (RODTD) and those of market available tablet of dimenhydrinate (DMH). Methods: Eight healthy volunteers were evenly randomized into 2 groups, one group received RODTD (25 mg) and the other received available market tablet of dimenhydrinate (25 mg). The blood levels of DMH were determined by high performance liquid chromatography (HPLC) before and after drug administration in 2 groups. Chromatography conditions were: Nova-Pak C18 as chromatographic column, methanol triethylamine buffer (1 : 1),flow rate: 1.0 ml/min, detection wavelength: 225 nm, and room temperature. The pharmacokinetics and relative bioavailability of RODTD and market available tablets were investigated. Results: The standard curve of DMH in the blank plasma was linear within the range of 5-500 ng/ml, with the regression equation being C=0.004 4 A+4.745 and R2=0.996. The limit of detection was 2 ng/ml; the average recovery rate was (90.55±4.69)% and the RSD was 0.041%. The intra-day derivations of 3 different concentrations (low, middle, and high) of plasma were 9.27%, 4.93%, and 2.95%, respectively (n=5), and the inter-day derivations were 9.97%, 3.81%, and 3.06%, respectively (n=5). Blood samples (3 ml) were subjected to HPLC assay and significant difference was found between the 2 forms of DMH. The pharmacokinetic parameters of RODTD were: AUC=(602.04±113.82) ng • h • ml-1, Cmax=(95.86±21.28) ng • h • ml-1, and TPeak=(1.8±0.32) h; the pharmacokinetic parameters of market available tablets were: AUC=(342.73±84.96) ng • h • ml-1 Cmax=(46.34± 10.32) ng • ml-1, and TPeak=(2.65±0.24) h. Statistical analysis showed there was significant difference in the relative bioavailability of 2 forms of DMH(P<0.01). The relative bioavailability of RODTD to market tablet was 175.66%. Conclusion: The developed RODTD can obviously increase the relative bioavailability of DMH.
2.Study on path of transforming traditional Chinese medicine research achievement into guideline.

At present, a number of scientific research achievements has been formed. Scientific achievement is the crystallization of great efforts from scientific workers, and it's also the valuable treasure of human civilization. Standardization is an important way to promote the international communication of Chinese medicine, and it's significant in boosting China's scientific and technological progress, improving market competitiveness and promoting international trade. Transformation of scientific research to the guideline is not only beneficial to improving the technology content of the standard, but also to the conversion from scientific research achievements into productivity. Therefore, only by absorbing the advanced scientific and technological achievements, reproducing the theory of traditional Chinese medicine (TCM) and medical technology in standard form, can make TCM keep pace with the times. This study preliminarily explores for the method to transform scientific research achievements into guideline, in order to provide reference for the future technical specifications, thus to further the development of TCM.

4.The Regulatory Functions of Aquaporin 2 by Deamino Arginine Vasopressin in Cochleas of Gui nea Pigs

Objective To investigate the expression of aquaporin 2 (AQP2) in an endolymphatic hydrops ani-mal model .Methods Forty adult guinea pigs were randomly divided into experiment group and control group (20 animals in each group ) . Guinea pigs in the experiment group were treated with deamino arginine vasopressin (DDAVP) intraperitoneally at the dosage of 4 μg · kg -1 · d-1 for 7 days ,while those in the control group were treated with physiological saline .The endolymphatic hydrops and expression of AQP2 in cochleas of all animals were evaluated by Hematoxylin and Eosin stain ,immunohistochemistry and Western blot .Results The endolymphatic hydrops in the experiment group were more severe than that in the control group(P<0 .05) .Expression of AQP2 were found in the stria vascularis and spiral ganglions in both groups with similar patterns .Both immunohistochem-istry and Western blot showed that the expressions of AQP2 in the experiment group were significantly stronger(P<0 .05 ) .Conclusion DDAVP can up-regulate the expression of AQP2 in the cochleae of guinea pigs ,thus may be closely related to the pathogenesis of endolymphatic hydrops .
5.LC-PDA-MS/MS analysis and anti-depressant study of Zhizi-Gancao-Chi decoction

Aim: To establish an analytical method to study the chemical components in Zhizi-Gancao-Chi(ZGC) decoction, and investigate its anti-depressant effect. Methods: An LC-PDA-MS/MS method was established and applied. The separation was performed on an ODS C_(18) column (250 mm ×4. 6 mm, 5 μm) with a gradient elu-tion. The mobile phase consisted of water/acetic acid( 1 %, v/v) and methanol. The main components were identi-fied according to the UV and MS. The anti-depressant effect was evaluated with mice tail-suspension test and forced swimming test. Results: The components of the ZGC decoction were separated isolated and ten of them were identified. These compounds are classified to iridoid glycosides, flavonoids and triterpene saponins. In addi-tion, its anti-depressant effect was observed for the first time. Conclusion: The LC-PDA-MS/MS method can ef-fectively separate and identify the components of ZGC decoction, which has significant anti-depressant effect. The relationship between the chemical component and its anti-depressant effect needs further study.
6.Effect of preoperative transcatheter arterial chemoembolization on Inhibitor of apoptosis protein-Survivin and Survivin mRNA of hepatocellular carcinoma.

Objective To study the effect of transcatheter arterial chemoembolizafion (TACE) on inhibitor apoptosis protein-Survivin, Survivin mRNA and proliferating cell nuclear antigen (PCNA). Methods Group A of hepatocelluar carcinoma with preoperative TACE (n = 26), group B of hepatocelluar carcinoma without any interven-tional therapy (n = 30) ,group C of Normal liver (n = 16) were divided. The expression of Survivin mRNA and the expression of Survivin and PCNA were detected. Result ①The positive Survivin mRNA expression in group A and the grey difference were lowere than that in group B (P < 0.05) ; ②The positive Survivin expression and the grey difference in group A were lower than that in group B (P < 0.05) ; ③The proliferating index (PI) of carcinoma tis-sure in group B was higher in group A and group C (P < 0.05 for each). Conclusion There is a high positive ex-pression of Survivin mRNA and Survivin, which may be decreased by TACE and then the proliferation of hepatocellu-lar carcinoma cell is surpressed.

9. Mechanisms of docetaxel resistance in triple negative breast cancer cell line CAL-51

Objective To compare sensitive difference of docetaxel between the triple negative breast cancer (TNBC) cell line CAL-51 and non TNBC line T47D and analyze mechanisms underlying docetaxel resistance in former cells. Methods Cell activity was determined by MTT method and IC50 value was calculated; Wright-Giemsa stain was used to analyze the effect of docetaxel in the morphology of CAL-51 and T47D cell lines. Flow cytometry (FCM) was performed to determine cell cycle distribution and apoptosis. Realtime fluorescence quantitative PCR was used to compare the relative gene expression levels. The anti-apoptosis protein Bcl-2 and caspase family protein expression levels were determined by Western blot. Results Wright-Giemsa stain showed significant morphology change in T47D cells by docetaxel treatment. Further flow cytometry results confirmed that docetaxel could significantly induce apoptosis in T47D cells compared to CAL-51 cells (P< 0.01). The result of realtime fluorescence quantitative PCR revealed that anti-apoptosis protein Bcl-2 was significantly higher expressed in CAL-51 cells (P<0.05). Immunoblot analysis revealed docetaxel treatment induced the instrinsic pathways in both CAL-51and T47D cells, but the activated pathway of executioner caspase was different. Conclusion Our present study shows that docetaxel induces different intrinsic apoptosis pathway in CAL-51 and T47D cell lines. Anti-apoprosis protein Bcl-2 is highly expressed, which might be the underlying mechanism of docetaxel resistance in TNBC cell line-CAL-51.
