Objective To observe the effects of autologous myoblast transplantation on endplate regeneration and neuromuscular function restoration following direct nerve implantation (DNI) in rats so as to offer experimental basis for the use of myoblasts in neural regeneration area and further lay foundation for the research using myoblasts as transgenic carriers. Methods A total of 20 male SD rats were randomly divided into experimental group and control group,with 10 rats in each group.Models of DNI in rat gastrocnemius were established.The experimental group was injected with primarily cultured autologous myoblasts to the DNI location,while the control group was injected with isometric medium without autologous myoblasts.The effects of myoblasts on the neuromuscular function recovery following DNI were studied by detecting the tibial functional index (FTI),neuro-electrophysiology and pathohistology.Results The experimental group displayed faster tibial nerve function recovery than the control group (P ＜ 0.01 ).The peak to peak value (PPV) of gastrocnemius neuro-electrophysiology,area under the curve and regenerated endplate number of the experimental group had statistical significances as compared with the control group ( P ＜ 0.01 ). Conclusion Myoblast autotransplantation accelerates the recoveryof neuromuscular function after rat DNI and increases the number of the regenerated endplates.

Objective To observe the effect of electroacupuncture (EA) and acupuncture (A) on the proliferation of stem cells in the subependymal zone (SPZ) of the lateral ventricle and the frontal lobe cortex in hyperlipemia(HL) combined with cerebral ischemia (CI) rats. Methods A total of 72 male SD rats were randomized into control,HL,HL+ EA,CI,CI+A,HL+CI,HL+CI+EAⅠ and HL+CI+EAⅡgroups (n= 9 /group).HL model was established by feeding the animals with high fat forage for 6 weeks and CI model was established by FeCl3-induced occlusion of the unilateral middle cerebral artery. EA was applied to "Sanyinjiao" (SP 6) and "Fenglong" (ST 40) once daily for 17 days for HL+ EA group; and acupuncture to "Baihui" (GV 20) and "Shuigou" (GV 26) once daily for 7 days for CI+A group. For HL+CI+EA Ⅰ group,EA was applied to SP 6+ST 40 first before CI,once daily for 10 days,followed by EA of SP 6+ST 40 and acupuncture of GV 20+GV 26 for 7 days after CI. For HL+CI+EA Ⅱ group,no treatment was given before CI,then,acupuncture of GV 20+GV 26 and EA of SP 6+ST 40 were given once daily for 7 days after CI.The immunoactivity of Nestin and proliferation cell nuclear antigen (PCNA) of SPZ was detected by immunohistochemistry.Results In comparison with normal control group,the numbers of both Nestin and PCNA immunoreaction (IR) positive cells in the dorsolateral extension and the wall of the lateral ventricle of the brain increased significantly in CI and HL+CI groups (P0.05). Conclusion EA can upregulate Nestin and PCNA expression of the dorsolateral extension and the wall of the lateral ventricle of the brain on the ischemic side in rats with CI,and with HL+CI,which may contribute to its effects in promoting the proliferation and migration of neural stem cells in the brain.

Objective To investigate the effects of local wet hyperthermia combined with microwave ablationon tumor growth and serum levels of interferon-β(IFN-β)and interleukine-12(IL-12).Methods An experimental mouse model beating mammary carcinoma was established in Balb/c mice and then treated with microwave ablation after 14 d.Seven d after ablation,local wet hyperthermia was administered once weekly for 2 weeks.Tumor growth and lung metastasis,as well as the survival of bearing tumor mice were observed.The serum levels of IFN-β and IL-12 were detected with enzyme-linked immuno sorbent assay(ELISA). Results When local wet hyperthermia was combined with microwave ablation tumor growth and lung metastasis were significantly less than when microwave ablation or local wet hyperthermia was used alone(P＜0.05).Furthermore,the survival of bearing tumor mice in combined therapy group prolonged significantly compared with those receiving either therapy alone(P＜0.05).Serum levels of IFN-β and IL-12 were also significantly higher in combined therapy group compared with the other groups (P＜0.05). Conclusions Local wet hyperthermia combined with microwave ablation is more effective than either local wet hyperthermia or microwave ablation alone in retarding tumor growth,which may be related to the enhanced immune response.

OBJECTIVE To investigate clinical characteristics of spontaneous bacterial peritonitis(SBP) in patients with severe type of viral hepatitis.METHODS A retrospective review of 120 cases of SBP associated with severe type of viral hepatitis was performed.RESULTS Clinical manifestations were as follows:fever 85.0%,abdominal tenderness 51.6%. 63.3% of the total white blood cells count in the ascitic fluid under 0.5×109／L,85.0% of polymorphonuclear cell ratio above 0.50.The positive culture of the ascitic fluid 24.2%,whereas 42.9% of the isolated microorganisms were Escherichia coli.CONCLUSIONS Clinical manifestations in SBP may be non-specific or absent.The total white blood cells count in the ascitic fluid are often under criteria for diagnosis.E.coli was the most common culture isolate.PMN ratio is the reliable parameter for the diagnosis of SBP.

Objective To compare Western blot analysis and immunofluorescence method in Survivin expression. Methods By using Western blot analysis and immunofluorescence method, Survivin gene expressions of the peripheral blood mononuclear cells in 18 patients with leukemia and 10 healthy persons were analysed. Results Positive expression of Survivin gene was detected in 13 of 18 leukemic patients by Western blot analysis and in 11 of 18 leukemic patients by immunofluorescence method. Negative expression of Survivin gene was detected in 10 healthy persons. The results of Survivin expression were the same in 16 patients by Western blots analysis and immunofluorescence method (consistent rate is 88.89%), but the positive expression of Survivin by Western blot analysis was the negative expression by immunofluorescence method in another 2 patients. Conclusion Survivin gene expressed selectively in leukemic cells, but not in normal mononuclear cell in peripheral blood. It indicated that Survivin expression is effective monitoring index in diagnosis and prognosis of leukemia. Survivin expression by Western blot analysis was consistent with immunofluorescence method.
