Objective To determine if there is an early developmental resistance to seizure-induced hi ppocampal damage. Methods Five daily pentylenetetrazol-indu ced convulsions in immature rats beginning at postnatal day P10,P60 groups.In b oth groups, the latency of seizure, the latency of Ⅳ/Ⅴ grade, the lasting time of seizure and mortality of rats after seizure were used to measure sensitivity of seizure or the resistance to brain damage. 3.Preparation of a kind of G418 resistance fibroblast feeder layers

Objective:To prepare a kind of G418 resistance fibroblast feeder layers.Methods: In order to prepare a stock primary embryonic fibroblast (EMFI) cells, the pregnant Smad3ex8+/- mice containing heterozygous neo gene were killed 14 days after coitus to get the embryo. EMFI cells were tested by PCR to identify their gene types. Only one EMFI cell which contained heterozygous neo gene was chosen and treated with mitomycin C.The mitomycin C-treated EMFI feeders were stored at -70℃. Results: The EMFI feeder layers could support the growth of embryonic stem cell line TC-1 efficiently and could live in G418 resistance medium for 1-2 weeks.
4.Pathological changes of brain at different maturational stages caused by pentylenetetrazol-induced repeated seizure in rat models.

OBJECTIVETo observe the histopathological changes in rat hippocampus at different maturational stages after repeated kindled seizures, and to explore their underlying epileptogenesis processes.
METHODSThree groups of Wistar rats (postnatal days: P10, P20, P60) were given pentylenetetrazol (PTZ) intraperitoneal injection for 5 days to induce repeated kindled seizures, and the age-matched rats in control group were injected with normal saline. The behavioral changes, the morphology and the neurons counting in hippocampus, as well as the expression of NF-kappaB were observed.
RESULTS(1) In the three groups, the latency of seizure and the latency of IV/V grade were significantly lower in the rats of group P10 and P20 [(1.2 +/- 0.6) min and (14.4 +/- 2.3) min vs. (4.7 +/- 1.6) min and (24.5 +/- 4.5) min] than group P60 [(8.6 +/- 2.0) min and (41.9 +/- 4.5) min], whereas the duration of convulsion in group P10 and P20 [(46.2 +/- 4.8) min and (29.8 +/- 5.9) min] was longer than those of group P60 [(17.1 +/- 5.0) min]. (2) The neuron counting of CA(1), CA(3) and hilar in the P10 and P20 groups showed no differences as compared to their controls, whereas adult rats (P60) had a significant neuron loss in CA(1) and CA(3) pyramidal cells, compared with the control group [(6.3 +/- 1.5)/250 microm(2), (3.6 +/- 1.4)/250 microm(2) vs. (8.2 +/- 1.9)/250 microm(2), (5.6 +/- 1.7)/250 microm(2)]. However, the dentate granule cells in immature rats (P10) with daily seizures had a significant increase as compared with the controls [(23.3 +/- 3.1)/250 microm(2) vs. (16.3 +/- 1.6)/250 microm(2)]. (3) Prominent sprouting was seen in the CA(3) stratum pyramidal layer in all experimental rats with 5 daily seizures, regardless of the age. But the degree of sprouting had significant differences among the experimental groups (P < 0.05). (4) NF-kappaB was expressed significantly in CA(3), CA(1) and dentate granule cells 24 hours after PTZ-kindling when compared with the control groups, with the spectral density decreased with age.
CONCLUSION(1) There were great differences in the vulnerability to the repeated seizure-induced brain damage at different maturational stages in rats. The immature brain appeared to be less vulnerable to the repeated seizures. (2) There was less hippocampus neuron loss and milder mossy fiber sprouting after repeated seizures in the developing rats than mature ones, which may be a pathological evidence underlying the prospect that the immature brain was more resistant to the seizure-induced neuronal injury. (3) The high expression of NF-kappaB may exert a certain biological effects in the seizure-induced neuronal injury.

5.Expression of GST-pi and MDR1 genes in operative specimens of ovarian cancer

Objectives:To study the expression of GST-pi and MDR1 genes in operative specimens of ovarian cancer,and to analyze the possible clinical role of GST-pi and MDR1. Methods:Eighteen frozen specimens of ovarian carcinoma and ten specimens of normal ovarian tissues from patients were examined for the expression of GST-pi and MDR1 genes by means of RT-PCR, and quantitative analysis was performed using β-actin as internal contrast.Results: Positive expression rate of GST-pi and MDR1 in ovarian carcinoma were 61.1% and 33.3%,respectively,and in contrast, 20% and 10% in normal ovarian tissues respectively. The level of GST-pi gene expression in ovarian carcinoma was obviously higher than that in normal ovarian tissue (P<0.05)and MDR1 gene also had high level expression in ovarian carcinoma, but had no statistical significantance. Four patients with ovarian carcinoma had GST-pi and MDR1 coexpression. Expression levels of GST-pi mRNA were lower than that of protein. Conclusions: (1) GST-pi and MDR1 had higher level expression in ovarian carcinoma than in normal ovarian tissues. (2) GST-pi and MDR1 may have same regulating factors but different mechanisms of action. (3)Processing after transcription and/or regulation of translation level may exist in GST-pi expression.
6.Cell-penetrating chimeric apoptotic peptide AVPI-LMWP/DNA co-delivery system for cancer therapy.

To develop a cell-penetrating chimeric apoptotic peptide AVPI-LMWP/DNA co-delivery system for cancer therapy, we prepared the AVPI-LMWP/pTRAIL self-assembled complexes containing a therapeutic combination of peptide drug AVPI and DNA drug TRAIL. The chimeric apoptotic peptide AVPI-LMWP was synthesized using the standard solid-phase synthesis. The cationic AVPI-LMWP could condense pTRAIL by electrostatic interaction. The physical-chemical properties of the AVPI-LMWP/pTRAIL complexes were characterized. The cellular uptake efficiency and the inhibitory activity of the AVPI-LMWP/pTRAIL complexes on tumor cell were also performed. The results showed that the AVPI-LMWP/pTRAIL complexes were successfully prepared by co-incubation. With the increase of mass ratio (AVPI-LMWP/DNA), the particle size was decreased and the zeta potential had few change. Agarose gel electrophoresis showed that AVPI-LMWP could fully bind and condense pTRAIL at a mass ratio above 15:1. Cellular uptake efficiency was improved along with the increased ratio of W(AVPI-LMWP)/WpTRAIL. The in vitro cytotoxicity experiments demonstrated that the AVPI-LMWP/pTRAIL (W:W = 20:1) complexes was significantly more effective than the pTRAIL, AVPI-LMWP alone or LMWP/pTRAIL complexes on inhibition of HeLa cell growth. Our studies indicated that the AVPI-LMWP/pTRAIL co-delivery system could deliver plasmid into HeLa cell and induce tumor cell apoptosis efficiently, which showed its potential in cancer therapy using combination of apoptoic peptide and gene drugs.

7.Acupuncture plus bloodletting therapy for insomnia in blood stasis constitution: a clinical study

Objective: To observe the clinical efficacy of acupuncture plus bloodletting therapy in treating insomnia in people with blood stasis constitution. Methods: Seventy-two patients were randomized into a treatment group and a control group by using the random number table, 36 cases each. The treatment group received acupuncture plus bloodletting therapy, while the control group was intervened by oral administration of estazolam. One-week treatment was taken as a course, for 3 successive courses in total. A follow-up study was conducted 3 months later. The Pittsburgh sleep quality index (PSQI) was evaluated before and after the intervention, as well as in the follow-up. The clinical efficacies were also compared. Results: The total effective rate was 88.9% in the treatment group versus 83.3% in the control group, and the between-group difference was statistically significant (P<0.05). After the intervention, the PSQI scores dropped significantly in both groups (both P<0.05); the between-group difference in PSQI score was statistically significant (P<0.05). The follow-up study showed that the between-group difference in the global score of PSQI was statistically significant (P<0.05). Conclusion: Acupuncture plus bloodletting therapy can produce a more significant efficacy than oral administration of estazolam in treating insomnia in people with blood stasis constitution.
8.Relations of synovial angiogenesis and PTEN/PI3K/AKT signaling pathway in rats with adjuvant arthritis.

OBJECTIVETo observe the change of PTEN/PI3K/AKT pathway hypoxia-inducible factor (HIF-1α), vascular endothelial growth factor (VEGF) in rats with adjuvant arthritis and to explore the mechanism of neovasculization in rheumatoid arthritis.
METHODSThirty rats were randomly divided into normal control group and model control group. The model control group were established the model of adjuvant arthritis using Freund's complete adjuvant. At 19 days after modeling, the expression of microvascular density (MVD), HIF-1α, VEGF were detected by ELISA assay and PTEN, PI3K, AKT were detected by Werstern Blotting.
RESULTSCompared with the normal control group, paw swelling, arthritic index were increased, and the expression of MVD, VEGF, HIF-1α of serum, PI3K, AKT of synovial tissue were significantly increased, PTEN was significantly decreased in model control group. PI3K, HIF-1α were positively correlated with MVD; VEGF, AKT were positively correlated with paw swelling; PTEN was negatively correlated with the arthritis index; HIF-1α was positively correlated with VEGF; PI3K was positively correlated with AKT, PTEN was negatively correlated with PI3K, AKT, VEGF.
CONCLUSIONImbalance of PTEN/PI3K/AKT pathway in rats with adjuvant arthritis is one of the mechanisms of synovial neovasculization.

9.Study on chemical constituents of volatile oil and trace elements from fruits of Clausena lansium.

OBJECTIVETo study and determine the chemical constituents of the volatile oil and the trace elements in the fruits of the Clausena lansium.
METHODThe essential oils were extracted by supercritical fluid extraction (SFE) and separated on capillary columns with HP6890GC-5973MS. The components were quantitatively determined with normalization method, and were identified with GC-MS. And the trace elements were determined by ICP-MS and ICP-AES.
RESULT36 Components constituting 95% of the total essential oil were separated and identified, and 11 trace elements were identified.
CONCLUSION18 Compounds were found from the fruit of C. lansium for the first time.

10.Chemical components of essential oils from Flos chrysanthemi Indici in different areas.

OBJECTIVETo provide the foundation for reasonable utilization by analysing the essential oils of Flos chrysanthemi Indici in different areas.
METHODThe essential oils were extracted by using steam distillation and separated with GC capillary columns. The components were quantitatively determined with normalization method, and were identified with GC-MS.
RESULTS18, 17 and 20 compounds of essential oils from Guangxi, Guangdong and Hubei were identified.
CONCLUSIONThere are significant differences among the components and contents of essential oils of Flos chrysanthemi Indici from Guangxi, Guangdong and Hubei.
