2.Probability prediction in multistate survival models for patients with chronic myeloid leukaemia.

In order to find an appropriate model suitable for a multistate survival experiment, 634 patients with chronic myeloid leukaemia (CML) were selected to illustrate the method of analysis. After transplantation, there were 4 possible situations for a patient: disease free, relapse but still alive, death before relapse, and death after relapse. The last 3 events were considered as treatment failure. The results showed that the risk of death before relapse was higher than that of the relapse, especially in the first year after transplantation with competing-risk method. The result of patients with relapse time less than 12 months was much poor by the Kaplan-Meier method. And the multistate survival models were developed, which were detailed and informative based on the analysis of competing risks and Kaplan-Meier analysis. With the multistate survival models, a further analysis on conditional probability was made for patients who were disease free and still alive at month 12 after transplantation. It was concluded that it was possible for an individual patient to predict the 4 possible probabilities at any time. Also the prognoses for relapse either death or not and death either before or after relapse may be given. Furthermore, the conditional probabilities for patients who were disease free and still alive in a given time after transplantation can be predicted.

3.Assessment of statistical methods applied in the research papers of Chinese Journal of Nuclear Medicine and Molecular Imaging

Objective To assess the correctness of statistical methods applied in the research papers of Chinese Journal of Nuclear Medicine and Molecular Imaging (CJNMMI) and to provide suggestions for further improving.Methods Papers were obtained from CJNMMI by multi-stage sampling method between 2007 and 2011 according to the number of journals and articles each year.The statistical methods were reviewed in the aspects of statistical design,description,inference and expression.Results A total of 116 papers were collected.For the statistical design,99.1% (115/116) used the control group and all were applied properly.The methods of randomization and blinding were used in 16.4% (19/116) and 8.6% (10/116) of papers,respectively,with 6 and 3 papers applied incorrectly.Statistical description was used in all papers with an accurate rate over 90.0%.The index for statistical description mainly included relative number,mean and standard deviation.The frequency for the usage of statistical tables was 45.7% (53/116) and all were applied correctly.In 71.6% (83/116) of papers,statistical inference methods were used,including t test,x2 test and analysis of variance.The overall incorrect rate was 29.7% (27/91),with 27.8% (5/18),25.5% (12/47) and 24.0% (6/25) for analysis of variance,t test and x2 test,respectively.The frequency for the usage of advanced statistical inference methods was 20.5% (17/83) and all were applied correctly.For the statistical expression,98.0% of papers provided the sample size and statistics.The incorrect rate of statistical expression was 60.3% (70/116) and majority of the mistakes was the lack of basis for determining the sample size.Conclusions The overall correctness of statistical methods applied in the papers of CJNMMI was good,especially in statistical design,statistical description and advanced statistical inference methods.However,quality application skills need to be improved in areas such as methods of randomization,blinding,basic statistical inference and statistical expression.
4.LC-PDA-MS/MS analysis and anti-depressant study of Zhizi-Gancao-Chi decoction

Aim: To establish an analytical method to study the chemical components in Zhizi-Gancao-Chi(ZGC) decoction, and investigate its anti-depressant effect. Methods: An LC-PDA-MS/MS method was established and applied. The separation was performed on an ODS C_(18) column (250 mm ×4. 6 mm, 5 μm) with a gradient elu-tion. The mobile phase consisted of water/acetic acid( 1 %, v/v) and methanol. The main components were identi-fied according to the UV and MS. The anti-depressant effect was evaluated with mice tail-suspension test and forced swimming test. Results: The components of the ZGC decoction were separated isolated and ten of them were identified. These compounds are classified to iridoid glycosides, flavonoids and triterpene saponins. In addi-tion, its anti-depressant effect was observed for the first time. Conclusion: The LC-PDA-MS/MS method can ef-fectively separate and identify the components of ZGC decoction, which has significant anti-depressant effect. The relationship between the chemical component and its anti-depressant effect needs further study.
6.Experimental Research of Distribution and Migration of Oval Cells in Progressive Hepatic Injury

Objective To explore the distribution and migration of oval cells in progressive hepatic injury.Methods Sixty SD rats were divided into the control group ( n =20) and experimental group ( n =40). After the establishment of hepatic carcinoma models, C kit was continuously detected by immunohistochemistry and the liver pathologic changes were regularly observed by optical microscopy. Results The hepatic surface was smooth with eumorphism in histology in the control group. The C kit positive cells were occasionally found. In the experimental group, the oval cells with C kit positive were initially discovered in the portal regions in the second week, and these cells proliferated along the bile duct epithelia. With the hepatic injury becoming more serious, the oval cells extended into the hepatic lobular regions from the portal regions. When hepatocellular carcinoma occurred,the majority were mixed carcinomas, and the oval cells were found inside and outside the carcinoma nodes. In this period, the most of C kit positive cells still located in the portal regions. Conclusion ①The oval cells are the most sensitive cells for the hepatic injury. ②The oval cells which migrate unruly participate in the formation of hepatic pseudolobules. ③The oval cells play an important role in hepatocarcinogenesis.

9.Speckle-tracking imaging and tissue Doppler strain imaging in assessing left atrium mechanical function: A study on patients with transcatheter device closure of atrial septal defect

Objective To compare the value of speckle-tracking imaging and tissue Doppler strain imaging in evaluating left atrial (LA) mechanical contraction. Methods Forty patients after successful percutaneous atrial septal defect (ASD) closure were enrolled in this study. Two-dimensional strain imaging (2D strain echocardiography, 2DSE) and tissue Doppler imaging (TDI) were performed one weak after ASD closure. Off-line analysis was done for atrial longitudinal peak systolic strain on the interatrial septum, in correspondence of the device, and on the lateral wall of the left atrium from apical 4 chamber view with 2DSE and color-coded strain imaging, respectively. Results There was no statistical difference of the inter- and intra-observer viability as well as time consumption on the off-line analysis between 2DSE and TDI. LA lateral wall strain was much higher than that of LA septum assessed by both color-coded strain imaging (47.31%±27.25% vs 30.06%±14.29%, P<0.01) and 2DSE (43.49%±25.55% vs 12.74%±9.16%, P<0.001). LA lateral wall strain was similar with either method (P=NS). However, at the site of ASD occluder, a non-contractile element, deformation was significantly higher in color-coded strain imaging than 2DSE (P<0.001). Five patients (12.50%) presented absence of deformation on ASD occluder with 2DSE, whereas there was no patient with TDI assessment (χ~2=5.33, P=0.027). Conclusion Speckle-tracking imaging is superior to tissue Doppler strain imaging in assessing left atrium mechanical function.
