2.A study on hippocampus injury assessment with apparent diffusion effect of MRI on mouse model

Objective To determine the utility of apparent diffusion coefficient (ADC) values as an objective method of early detection of traumatic brain injury of the mice models in hippocampus injury.Methods A total of 18 healthy male adult C57/BL6 mice were randomly divided into three groups according to the degree of injury:the mild group,severe group and control group.Conventional images,diffusion-weighted images,ADC were studied.The hippocampal were selected as the interesting area.ADC values were calculated and compared with those in control subjects after 6 h,12 h and 24 h.Student t test was performed for each region to compare the injury groups and the control.Results In control group both sides hippocampal ADC values had no differences(8.65±0.26 and 8.77±0.12) × 10-4 mm2/s,P>0.05.Abnormalities were detected on ADC values,compared with conventional images in the injury groups.ADC values in mild group and severe group were significantly different from those of control group(6 h ADC values were (8.70±0.50) × 10-4 mm2/s,(6.13± 0.11) × 10-4 mm2/s,and (3.16±0.78) × 10-4 mm2/s in control,mild and severe group respectively),(12 h ADC values (8.23±0.61)× 10-4 mm2/s,(5.92 ± 0.27) × 10-4 mm2/s and (2.64±0.65) × 10-4 mm2/s) and (24 h ADC values (9.01±0.14) × 10-4 mm2/s,(5.99± 0.13)×10-4 mm2/s and (2.58±0.42)×10-4 mm2/s),P<0.05.Conclusion There are differences among the ADC values on differcnt types of hippocampal brain injury mouse-model.ADC values can identify the degree of damage on brain injury and can detect early craniocerebral injury.
3.Effect of levetiracetam on electrical status epilepticus during sleep in benign childhood epilepsy with centro-temporal spikes

Objective To investigate the effect of levetiracetam on benign childhood epilepsy with centro-temporal spikes(BECT) with early-electrical status epilepticus during sleep(ESES).Methods Since June 2010 to June 2013,35 cases of BECT with ESES were treated in our hospital.They were divided into two groups:early-ESES group and ESES group.The seizure rate and spike-wave index in different groups were observed before and after treatment.Intelligence quotient(IQ),response control quotient,attention quotient in different groups were compared before and after treatment.Results The seizure-free rate in earlyESES group was 55.00%,the total effective rate was 85.00%,EEG improvement rate was 60.00%.The seizure-free rate in ESES group was 26.67%,the total effective rate was 73.33%,EEG improvement rate was 46.67%.The seizure-free rate,total effective rate and EEG improvement rate in early-ESES group were higher than that in ESES group,but there were no statistically significant differences between two groups.The cognitive function (verbal IQ:90.29 ± 13.47 vs.83.97 ± 10.20; performance IQ:93.83 ± 11.12 vs.87.03 ± 11.15; full IQ:94.26 ± 10.96 vs.86.71 ± 11.29) and visual attention in both groups (response control quotient:100.77 ± 7.91 vs.87.40 ± 9.68 ; attention quotient:94.66 ± 7.22 vs.79.46 ± 12.52) were significantly improved after treatment(P < 0.05,respectively).Conclusion Levetiracetarn is effective on BECT with ESES and early-ESES,especially on early-ESES.Levetiracetam maybe have a preventive effect on ESES.
4.Effects of pulsed Nd:YAG laser radiation on marginal microleakage of porcelain-fused-to-metal crowns

Objective To study the effects of pulsed Nd:YAG laser radiation on the marginal microleakage of porcelain-fused-to-metal crowns. Methods Twenty in vitro premolar teeth were selected and processed by standard porcelain crown preparation. The porcelain-fused-to-metal crowns were made based on the relevant repair technical requirements. The restorations were randomly divided into control group (group A) and experimental group (group B). The restorations in the group B were treated by external Nd:YAG laser irradiation with power of 1.25 W and irradiation time of 20 s. All the restorations were fixed with composite resin cement, and then put into normal saline under 37℃for 20 d, during which 300 times thermal cycling were performed. After that, the restorations were placed into 0.5% pinkish red solution for 24 hours to dyeing, and then dissected along the longitudinal axis. The marginal microleakage of the restoration was observed by a stereomicroscopy. The data were processed by the SPSS 19.0 software. Results The degree of marginal microleakage of the restorations after Nd:YAG laser irradiation was significantly lower than that in the control group (P<0.05). Conclusions The pulsed Nd:YAG laser irradiation can effectively improve the adhesion between the resin and the hard tissue of the teeth, and reduce the marginal microleakage of the porcelain-fused-to-metal crowns.
5.Disinfection effectiveness of photodynamic therapy combined with EDTA on infected lateral canals in vitro

Objective:To investigate the disinfection effectiveness of photodynamic therapy (PDT) combined with ethylenediaminetetraacetic acid (EDTA) disodium salt solution on enterococcus faecalis in lateral root canals.Methods:Sixty-four human single root canal premolars were selected to prepare artificial root canal collaterals, and E. faecalisin root canal collateral infection models were established. The infection model was divided into PDT group ( n=16), PDT combined with EDTA group ( n=16), positive control group ( n=16) and negative control group ( n=16) according to random number table method. In the PDT group, 40 μg/ml hematoporphyrin monomethyl ether was injected into the root canal, and then the root canal was irradiated with a 45 mW laser for 90 s after 5 min incubation. In the PDT combined with EDTA group, the root canal was given 5 ml EDTA solution with 17% mass fraction for 1 min, and then treated with the method same as the PDT group. In the positive control group, the root canal were given 5 ml NaClO solution with a mass fraction of 5.25 % for 1 min. In the negative control group, the root canal were given NaCl solution with a mass concentration of 9 g/L for 1 min. Before and after the treatments, samples were taken in the lateral branches of the root canal with a K file to count plate colonies. After treatments, the roots of each group were placed in sterile brain heart infusion broth (BHI) medium for anaerobic culture for 24 h, and then sampled with a K file, and the number of root canal collaterals was detected statistically. Scanning electron microscopy was used to observe the morphology of the inner wall of lateral branches of root canals after treatments. Results:The sterilization rate of PDT combined with EDTA group was 99.56%, which was significantly higher than that of negative control group (1.98%), positive control group (85.87%) and PDT group (87.53%), the differences were statistically significant (all P<0.05). The reply experiment shows that the number of infection root canals was only 5, which was less than the negative control group (15), positive control group (12) and PDT group (11), and the differences were statistically significant (all P<0.05). Scanning electron microscopy observations showed that no obvious E. faecalis adhered to the inner wall of root canal of PDT combined with EDTA group. Conclusions:PDT combined with EDTA has a good disinfection effectiveness on E. faecalis in lateral canals, and it is expected to provide a new method for the effective killing of E. faecalis in lateral canals in clinical root canal therapy.
6.Separation of choline from Coptidis Rhizoma and impact on glucose metabolism of berberine in HepG2 cells.

OBJECTIVETo investigate action of hydrophilic constituents from Coptidis Rhizoma on glucose-lowering effect metabolism of berberine in HepG2 cells.
METHODHydrophilic fractions of Coptidis Rhizoma were prepared by high speed counter current chromatography and separated by silica gel column chromatography. MTT assay was used to monitor the proliferation of HepG2 cells, and Kit was used to test the glucose consumption in culture solution.
RESULTCholine was separated from Coptidis Rhizoma for the first time. Cell assay showed choline can significantly increase the glucose lowering effect of berberine and improve the cytotoxicity of berberine within test concentration. Compared with same dose of berberine, berberine 38 mg x L(-1) in combination with choline 100 mg x L(-1) can make glucose consumption increase by 34% and elevate cell livability up to 75% in HepG2 cells.
CONCLUSIONThe results suggest that choline had a synergistic effect on improving glucose absorption of berberine and decreasing cytotoxicity of berberine.

7.Application of cardiac magnetic resonance imaging in subclinical systemic sclerosis primary heart involvement

Objectives:Primary cardiac involvement (SSc-PHI) in systemic sclerosis is an important prognostic factor. We aimed to characterize and identify subclinical SSc-PHI using cardiovascular MRI to determine whether disease severity and serum biomarkers are associated with subclinical SSc-PHI.Methods:A total of 26 patients with SSc who had no history of cardiovascular disease or pulmonary hypertension underwent 3 T-enhanced cardiovascular MRI. Measurements included native T 1, extracellular volume, advanced gadolinium enhancement, T 2 mapping, and left ventricular volume function. Troponin T and N telencephalic natriuretic peptide precursors were also determined. Results:LGE was observed in 13 of 26 patients (50.0%), suggesting focal fibrosis, and T 2 mapping was significantly higher in the dcSSc group than in the lcSSc group ( P=0.009). Left ventricular volume and function were within the normal range in all patients, but final systolic left ventricular volume was significantly higher in dcSSc than in lcSSc ( P=0.021). The modified Rodnan skin score (mRSS) was significantly higher in patients with LGE focal fibrosis ( P=0.019). Logistic regression analysis confirmed the association between mRSS and LGE ( OR=1.224, P=0.037). In multivariate analysis, T 2 mapping was negatively correlated with disease course, and was correlated with dcSSc and fingertip ulcer ( R2=0.711, P=0.018, P=0.013, P=0.030). Troponin T was correlated with T 2 mapping ( r=0.555, P=0.049). Conclusions:Subclinical SSc-PHI is characterized by diffuse and focal myocardial fibrosis, but preserves myocardial systolic function. Subclinical SSC-Phi is associated with TNT, SSc disease severity, and complex peripheral vascular disease. These data provide information for identifying individuals at risk of SSc-PHI.
