1.Cytotoxicity of Human Tumor Draining Lymph Node Cells Stimulated by Cancer-Associated Apomucin in vitro

Human tumor antigen elicit a cell-mediate immune response in the host have been identified. In this study, a pancreatic cancer cell line SW1990 associated apomucin was examined for the induction of tumor draining lymph node cells(TDLN) anti-tumor immune responses.The TDLN of patients with breast cancer(4 cases), gastric cancer(3 cases), colon cancer(3 cases) were isolated, cocultured with apomucin 50 ?g/ml and low dose rhIL-2 50U/ml in vitro, the cytolytic effects were measured by MTT assay. The results showed that: the proliferated lymphocytes can lysis LS174T colon cancer cells, KATO-Ⅲ gastric cancer cells, SW1990 pancreatic cancer cells and K562 leukemia cells. The most cytolytic effects were higher than LAK. This results might provide a rational basis for future attempts to elicit and manipulate apomucin vaccine for active specific immunotherapy or adoptive immunotherapy for adenocarcinoma.

3.Clinical value of C - reactive protein, erythrocyte sedimentation rate and white blood cell count in patients with ocular syphilis

AIM:To explore the clinical value of combined detection of C-reactive protein ( CRP ) , erythrocyte sedimentation rate ( ESR) and white blood cell ( WBC) count in patients with ocular syphilis.
METHODS:Dates of CRP, ESR, WBC, TPPA and RPR of 51 ophthalmopathy patients caused by syphilis and 50 normal control from Jan. 2012 to Dec. 2015 in eye hospital were recruited and analyzed statistically.
RESULTS:The positive rates of CRP, ESR and WBC of oculopathy patients were 16%, 18% and 39%, respectively, which were higher than those in the control group. In patients group, the positive rate of ESR was higher than CRP and WBC. There were no obvious relationships between RPR titers and positive ratios of CRP, WBC and ESR.
CONCLUSION: The blood level of CRP, WBC and ESR may have certain help in estimating and monitoring condition of patients with ocular syphilis.
4.Effect of acupuncture on feeding center of hypothalamus in experimental obese rats

BACKGROUND: The therapeutic effects of acupuncture on weight loss have been verified. How does the central neural system act on weight loss with acupuncture?What relationships are there between the central regulation and peripheral action?OBJECTIVE: To study the effect of acupuncture on feeding center of hypothalamus in obese cases.DESIGN: Randomized and controlled study in which the experimental animals were taken as the subjects.SETTING: Clinical medical college of a university of traditional chinese medicine.MATERIALS: The experiment was performed in Key Laboratory of Acupuncture and Moxibustion in Nanjing University of Traditional Chinese Medicine from June 1998 to January 2000. Fifty SD male rats were employed and randomized into acupuncture group, obese group and normal group.METHODS: The experimental obese model was prepared by feeding SD rats with high-fat forage. Microelectrode record of nerve cell and cerebral stereotaxic technique were applied, by the treatment with acupuncture on experimental obese rats, the electric activity (Hz) of nerve cell was observed at unit time in hunger center of lateral hypothalamus area(LHA) and satiety center of ventral medium nucleus of hypothalamus(VMH).MAIN OUTCOME MEASURES: Mass changes at the same stage before and after acupuncture in rats of every group and discharge frequency in LHA and VMH in rats of every group.RESULTS: Acupuncture reduced remarkably the excitability in LHA, the values of which in acupuncture, obese and normal groups were(9.4 ± 3.7),(21.4 ± 6. 8), (9. 1 ± 5.2) Hz respectively( P < 0. 01 ); improved electric activity frequency in VMH, the values of which in 3 groups were (21.1 ± 4. 3), (5.0 ± 1.3), ( 14. 5 ± 2. 2)Hz, respectively( P < 0.01 ), inhibited excessive appetite of obese rats and reduced calorie intake so as to achieve weight loss.CONCLUSION: The regulation of acupuncture on central neural nuclei in obese animals is one of the important mechanisms on weight loss.
5.Impacts of abnormal lipid metabolism on prognosis and quality of life in patients with severe COPD

Objective To investigate the influence of abnormal lipid metabolism on prognosis and quality of life in patients with severe COPD. Methods 134 patients with severe COPD in our hospital from September 2013 to January 2015 were selected as research objects, and their blood lipid levels were detected. The patients were divided into the normal group (n=61) and the low level group (n=73) according to the test results of lipid levels. 1-year mortality rate was statistically analyzed in both groups , and the WHOQOL-BREF health survey profile was applied to assess quality of life before and after treatment. Results 1-year survival rate in the normal group was 90.16%(55/61), which was higher than 73.97%(54/73) in the low level group (χ2=5.741, P=0.017). As compared with the normal group, the scores on physiological function, environment, social relationship, mental state, spirit / religious / belief, and total quality of lifewere markedly lower in the low level group after treatment (P < 0.05). The correlation analysis showed that plasma levels of TC, TG, and HDL-C were negatively correlated with 1-year mortality rate in patients with severe COPD (P < 0.05) but positively related with all dimensions in quality of life; plasma HDL-C level was positively related with 1-year mortality rate (P < 0.05), but negatively related with all dimensions in quality of life in patients with severe COPD (P < 0.05). Conclusions For patients with severe COPD, abnormal lipid metabolism is closely related with prognosis and quality of life. We should pay attention to nutritional support and control of abnormal lipid metabolism in those patients , leading to improvement of prognosis and quality of life.

Objective To investigate the stiffness of lumbar spine after the injury caused by percutaneous diskectomy and evaluate the efficiency of percutaneous lumbar diskectomy by biomechanical study. Methods Four fresh lumbar specimens were used to analyse load-displacement curves in the intact lumbar spine and vertical disc-injured lumbar spine. 7.Early Oral-motor Exercising: An Important Aspect of Children with Cerebral Palsy

Objective To explore the effects of earlier oral-motor exercise on swallowing and feeding capability of children with cerebral palsy. Methods 36 children accepted oral-motor exercise performed by speech-language therapist once a day for 3 months. They were assessed for oral motor and feeding before and after treatment, and compared with other 36 controls matched with age and type. Results The oral motor in treatment group improved significantly (t=2.184,P<0.05) after treatment, but merely in control. The difference of scores of oral motor before and after treatment was (9.89±4.33) in treatment group, and (3.38±1.56) in control group (t=7.423, P<0.01). Conclusion Early oral-motor exercises is effective to improve oral motor function and feeding capability.

Objective To observe the effects of passive movement on the functional outcome after occlusion of the middle artery in the brain and reperfusion, and to explore the molecular mechanisms involved. Methods Cerebral infarction models were established in rats using left middle cerebral artery occlusion ( MCAO). The survivors were randomly divided into a passive movement group and a natural recovery group. There was also a sham-operated group and a normal group. Passive movement treatment (twice a day, twenty min per time) was started at different times after reperfusion. The expression of brain-derived neurotrophic factor (BDNF) and B cell lymphoma/leukemia-2 gene (Bcl-2) were determined using real-time PCRs. Results Expression of BDNF and Bcl-2 was detected a-round the infarction area in both groups. The expression of BDNF and Bcl-2 was highest in the sub-groups where passive movement was begun 24 or 48 h after the operation. Conclusions The expression of BDNF and Bcl-2 in the brain peaks when daily, moderate intensity passive movement is administered beginning 24 to 48 h after reperfusion. Passive movement might have a protective and rehabilitative effect after cerebral infarction.
9.Bioinformatic Analysis of Structural Proteins of Paramyxovirus Tianjin Strain

The amino acid sequences of the NP,P, M, F,HN and L proteins of the paramyxovirus Tianjin strain were analyzed by using the bioinformatics methods. Phylogenetic analysis based on 6 structural proteins among the Tianjin strain and 25 paramyxoviruses showed that the Tianjin strain belonged to the genus Respirovirus, in the subfamily Paramyxovirinae, and was most closely related to Sendal virus (SeV). Phylogenetic analysis with 14 known SeVs showed that Tianjin strain represented a new evolutionary lineage. Similarities comparisons indicated that Tianjin strain P protein was poorly conserved, sharing only 78.7%-91.9% amino acid identity with the known SeVs, while the L protein was the most conserved, having 96.0%-98.0% amino acid identity with the known SeVs. Alignments of amino acid sequences of 6 structural proteins clearly showed that Tianjin strain possessed many unique amino acid substitutions in their protein sequences, 15 in NP, 29 in P, 6 in M, 13 in F, 18 in HN, and 29 in L. These results revealed that Tianjin strain was most likely a new genotype of SeV. The presence of unique amino acid substitutions suggests that Tianjin strain maybe has a significant difference in biological, pathological, immunological, or epidemiological characteristics from the known SeVs.
10.Intelligence Delay in Children with Cerebral Palsy of Preschool Age

Objective To investigate the intelligence level and characters in children with cerebral palsy of 4~6 years old, and the factors related with their intelligence. Methods 215 cerebral palsy children of 4~6 years old were assessed with Wechsler Intelligence Scale for children,Gross Motor Function Classification System (GMFCS). Results 210 children finished their intelligence test, and 113 presented mental retardation. The intelligence level was higher in the children with spastic diaplegia, hemiplegia and dyskinetic type than those with quadriplegia and ataxia type. The difference was significant between VIQ and PIQ in the spastic diplegia (P<0.05). The mental retardation was more frequent in the children with GMFCS Ⅳ~Ⅴ, or with epilepsy or microcephaly than those with GMFCS Ⅰ~Ⅲ or without the complications (P<0.05). Conclusion The incidence of mental retardation in children with cerebral palsy was related with the neurotypes, motor function and complications.
