3.Establishment of animal model for in vitro spermatogenesis by heterotopic grafting of neonatal mouse testis
Journal of Peking University(Health Sciences) 2003;0(06):-
Objective:To establish an in vitro model for the development of mouse spermatogenic cellsinto sperm by using the immunodefective mouse as the incubator. Methods:Tissue grafting was per-formed using testis from 1-2 days old Kun-ming mice as donor tissue and immunodefective mice as recipi-ents;the expression of TESK1 mRNA in grafts was determined by RT-PCR and the spermatogenesis fur-ther observed with histological analysis of grafts. Results:Molecular biological and histological analysesshowed that grafts post-grafting not only expressed TESK1 mRNA as in normal mouse testis, but also ex-hibited similarities in the structure of seminiferous tubules and component of spermatogenic cells,inclu-ding sperms. Conclusion:Spermatogenic cells heterotopically grafted in vitro could continuously growand complete spermatogenesis and finally develop into sperm.
4.Study on the extraction recovery of docetaxel in plasma by different solid-phase cartridges
Chongqing Medicine 2013;(22):2629-2630,2634
Objective To optimize the solid-phase extraction method by comparison of the extraction recovery of docetaxel plas-ma samples. Methods After extracted by different solid-phase cartridges with water, acetonitrile, and different content methanol e-lution,the plasma samples were analyzed on an Phenomenex LUNA C18 column(4.6 mm×250 mm,5μm)with mehhanol:acetoni-trile:water(42∶26∶32,v/v/v)as the mobile phase. From the recovery achieved, the best solid phase cartridge was found. Results This method consists of using 60%-80% methanol for the elution.Among the three kinds of solid-phase being tested,Waters Oasis HLB cartridge was found to be the best one. Conclusion The average extraction recovery of the Waters Oasis HLB cartrid-ges was between 95%-105%,and it can be used in the analysis of docetaxel in plasma samples.
6.Detection of minimal residual disease in childhood acute lymphoblastic leukemia:methodologic advances and clinical significance
International Journal of Pediatrics 2010;37(2):135-138
Multiagent chemotherapy has significantly improved survival for childhood ALL.However, the relapse rate with current therapies remains at 20% to 30%.Numerous studies have proved that the minimal residual disease(MRD)is associated with relapse rate. The major methods for the detection of MRD include polymerase chain reaction(PCR)and flow cytometry(FCM).The measurement of MRD in childhood acute lymphoblastic leukemia offers the promise of individualized,risk-stratified treatment and the prediction for post-transplant outcome.
7.Research progress on molecular targeted therapy for metastatic esophageal cancer pa-tients
Chinese Journal of Clinical Oncology 2016;43(4):170-172
Esophageal cancer is a highly aggressive malignant tumor of the digestive tract. Despite the continuous development of sur-gical techniques and multidisciplinary treatments, the prognosis and overall survival of esophageal cancer patients is still poor. In re-cent years, the development of new molecular targeted therapy has become well advanced. Drugs that target tumor-related signal pathways and block the transmission of downstream signals can inhibit the growth and metastasis of tumor cells. Currently, esopha-geal cancer-targeted drugs are rarely approved officially. HER-2 inhibitor represents these types of drugs because of its positive expres-sion of metastatic adenocarcinoma. However, studies on the preparation of new drugs are not extensive. In this paper, the research progress of a new targeted therapy for metastatic esophageal cancer is reviewed.
8.Major genetic changes of pancreatic ductal adenocarcinoma
Basic & Clinical Medicine 2006;0(01):-
Pancreatic ductal adenocarcinoma is the most common pancreatic neoplasm characterized by latent morbidit, poor prognosis, high mortality rate and limited choice of treatment.Quitealot studies focused on its pathogenesis, and showed molecular genetic alterations, which derived of genetic and environmental factors and played an important role in tumorigenesis.Recently, more and more findings laid particular emphasis on the changes of gene molecule and some were confirmed in vitro and in vivo.In this paper,we made a review and summarized the marked molecular changes and signalings of the four major pathways to understand their functions in PDAC.
9.Study on Role of Non-coding RNA in Barrett's Esophagus
Chinese Journal of Gastroenterology 2017;22(7):429-431
Non-coding RNA (ncRNA) is a class of RNA not coding the protein and plays a significant role in the process of growth and development of diseases.Barrett''s esophagus (BE) is considered as the precancerous lesion of esophageal adenocarcinoma.Several studies showed that ncRNA has the potential value in diagnosis, treatment and designing of targeting drugs. This article reviewed the role of ncRNA, especially microRNA, long non-coding RNA and circular RNA in the development and carcinogenic process of BE.
10.Clinical observation of auricular acupoint therapy for pain in early-stage extremity trauma
Journal of Acupuncture and Tuina Science 2017;15(3):219-222
Objective:To observe the efficacy of auricular acupoint sticking based on conventional treatment in treating pain in early-stage extremity trauma. Methods:A hundred eligible patients with acute soft tissue injury or acute closed fracture were randomized into an observation group and a control group by their admission sequence, 50 cases in each group. The two groups both received routine management including routine checking, external fixing, traction, raising up the affected limb, etc., as well as cold compress with Chinese medication (Xiao Zhong Zhi Tong Powder). In addition to the routine management, the control group was given oral administration of amidopyrine and phenacetin compound tablet, 1 tablet per dose, twice a day, which was then taken only when necessary or terminated after pain subsided. The observation group was given auricular acupoint sticking in addition to the routine management. The two groups were compared in terms of numerical rating scale (NRS) score, therapeutic efficacy and adverse reactions after pain was relieved. Results:After the intervention, the NRS scores dropped significantly in both groups (P<0.01); the NRS score in the observation group was significantly lower than that in the control group (P<0.05).The total effective rate in the observation group was superior to that in the control group (P<0.05).There were no severe adverse reactions in the two groups. Conclusion:Based on routine management, auricular acupoint sticking can produce a more significant efficacy in treating pain in early-stage extremity trauma compared to amidopyrine and phenacetin compound tablet; it can effectively reduce pain of the affected limb and prevent complications; it's easy-to-operate and safe; patients can learn and understand it easily; its efficacy is confirmed; it enhances the satisfaction degree of the inpatients. Therefore, this method is worth promoting in clinic.
