1.Preparation and Quality Control of Potassium Dehydroandrographolide Succinate Hollow Suppository

OBJECTIVE:To prepare Potassium dehydroandrographolide succinate(PDS)hollow suppository and establish a method for its quality control.METHODS:The PDS hollow suppository was prepared with semisynthesis fatty acid glyceride as ground substances and PEG300 or 1,2 propylene glycol as disperse medium.The content of PDS was determined by ultraviolet spectrophotometry at a detection wavelength of 251nm.RESULTS:The linear concentration range of PDS was 6.0~50.0?g?mL-1(r=0.999 9,n=5).The two kinds of PDS hollow suppository were all up to the quality standard.CONCLUSION:This method is reasonable in design and feasible in technology.
2.Estimating the copy numbers of exogenous gene in transgenic cashmere goats by real-time fluorescence quantitative PCR

The copy numbers of exogenous gene in transgenic animals is always regarded as an important information of transgenic animals.Thus,simple and sensitive methods are required for the detection of the copy numbers of exogenous gene.Three kinds of transgenic Shanbei white cashmere goats,containing Tβ4-GFP,FGF5s-GFP and VEGF164-GFP,has been obtained by using PiggyBac(PB) transposon system.Fluorescence quantitative PCR was carried out to detect the copy numbers of copGFP.Using Gluc as reference gene,the double standard curves of exogenous gene and reference gene were mapped and the genomic DNA of transgenic goats were analysized by real-time fluorescence quantitative PCR.Moreover,the copGFP/Gluc ratio in the samples was calculated as the copy numbers of copGFP.In addition,Tβ4-GFP transgenic cashmere goats were selected to detect the integration sites by using the genomic walking kit.The results showed that the standard curve equation of copGFP was y=-3.230 6x+39.216 (R2 =0.998 8) and the standard curve equation of Gluc was y=-3.564 8x+38.440 (R2 =0.996 0).The copy numbers of exogenous gene in the transgenic cashmere goats were obtained and the numbers of integration sites in the selected Tβ4-GFP transgenic goats were consistent with the copy numbers of copGFP.As a conclusion,the high throughput,fast and sensitive real-time fluorescence quantitative PCR is an efficient and convenient method for the copy number of exogenous gene in transgenic cashmere goats.
3.IL-17 contributes to autoimmune hepatitis.

The role of interleukin-17 (IL-17) in autoimmune hepatitis (AIH) was investigated. A mouse model of experimental autoimmune hepatitis was established, and the syngeneic S-100 antigen emulsified in complete Freud's adjuvant was injected intraperitoneally into adult male C57BL/6 mice. The IL-17 expression in serum and the livers of the mice models was detected by using ELISA and immunohistochemistry, respectively. IL-17 neutralizing antibody was used to study the biological effect of IL-17 in the experimental AIH. IL-17 neutralizing antibody in vivo administration alleviated the hepatic inflammation and ALT level in the AIH model. IL-17 in the peripheral blood mononuclear cells (PBMC) of AIH patients was measured by using real-time PCR method. The results showed that IL-17 level was significantly up-regulated in AIH patients and mice models. It was concluded that IL-17 contributed to the development of AIH and might be a potential therapeutic target of AIH.
4.Differential gene expression profiles in acute hepatic failure model in mice infected with MHV-3 virus intervened by anti-hepatic failure compound.

Differential gene expression profiles in Balb/cJ mouse model of acute hepatic failure infected with MHV-3 virus intervened by anti-hepatic failure compound (AHFC) and the changes of cytokines regulated by genes were investigated. The Balb/cj mice were divided into AHFC-intervened group and control group randomly. Acute hepatic failure model of Balb/cJ mice infected with MHV-3 virus was established. The survival rate in the two groups was observed. It was found that the survival rate in the AHFC-intervened group and control group was 90% and 50% respectively 48 h after intraperitoneal injection of MHV-3 (P<0.05). Before and after the experiment, the cytokines in peripheral blood of the survival mice were determined, and RNA was extracted from survival mouse liver tissue for the analysis of the differential gene expression by a 36 kb mouse oligonucleotide DNA array. In all the genes of microarray there were 332 genes expressed differently in the two groups, in which 234 genes were up-regulated and 78 genes down-regulated. Through clustering analysis, the differential expression of immune related genes, including TNF receptor superfamily, Kctd9, Bcl-2, Fgl2, IL-8, IL-6, IFN-gamma, TNF-alpha etc. might be related with the curative effectiveness of AHFC. It was suggested that AHFC can balance the immune state of mouse model of acute hepatic failure infected with MHV-3 virus mainly through regulating the expression of immune related genes, decrease the immune damage and inhibit liver cell apoptosis of mouse acute hepatic failure model obviously so as to increase the survival rate of mouse models of acute hepatic failure.

5.Radioprotective effects of FUZHENGJIEDU on rats exposed to radon

Objective To investigate the protective effects of FUZHENGJIEDU,a kind of Chinese herb on rats exposed to radon and it's daughter.Methods Twelve male SD rats were exposed with concentration of 40 000 Bq/m3 radon and the exposed dose was 120 work level month (120 WLM),and then all the rats were randomly divided into 2 groups.One group was 120 WLM group (positive control,n=6 ),the other group was FUZHENGJIEDU treatment group (120 WLM exposed rats were given FUZHENGJIEDU 5.0 g every day,n =6),meanwhile 6 rats which lived in the normal environment ( the concentration of radon and it's daughter was lower than 50 Bq/m3 ) as control group.The levels of tumor markers induding carcinoembryonic antigen ( CEA),neuron-specific enolase (NSE),CYFRA21-1 and p53 antibody were tested by ELISA.The rate of p16 gene methylation was measured by methylation-specific PCR (MSP).Results The levels of CEA and NSE in 120 WLM group were (396.62 ± 148.74) and (9.09 ±0.90) μg/L,respectively,significantly higher than those of control group (t =2.583,2.463,P < 0.05).The levels of CEA and NSE in treatment group were (70.89 ±44.71) and (4.31 ±1.37) μg/L,respectively,remarkably lower than those of 120 WLM group (t =2.921,2.526,P <0.05 ).The concentrations of CYFRA21-1 and p53 antibody were not significantly different among the three groups (t =1.713,1.963,P > 0.05 ).The rate of p16 gene methylation in BALF cells in 120 WLM group was 16.67%,but in control group and treatment group did not arise p16 gene methylation in BALF cells.Conclusions FUZHENGJIDU would decrease the levels of CEA and NSE and the rate of p16 gene methylation,and could exert it's protective effect on rats exposed to high concentration of radon.
6.Efficacy of radiofrequency thermocoagulation guided by three-dimensional computer tomography for treatment of severe recurrent glossopharyngeal neuralgia

Objective To evaluate the efficacy of radiofrequency thermocoagulation guided by three-dimensional computer tomography(3-D CT)for the treatment of severe recurrent glossopharyngeal neuralgia(GPN). Methods Twenty-nine patients of both sexes with recurrent intractable GPN, aged 42-75 yr, with recurrent course of 1-8 yr, of Barrow Neurological Institute(BNI)Pain Scale(BNI-P) class Ⅳ or Ⅴ, were enrolled in this study. Percutaneous radiofrequency thermocoagulation was applied to the middle and lower segment of the styloid process of the glossopharyngeal nerve under 3D CT guidance. Before operation and at 72 h, 6 months and 1 year after operation, BNI-P class and BNI Numbness Scale scores were recorded, and effective treatment and operation-related complications were also recorded. Re-sults Nineteen patients underwent one operation, 8 patients underwent two operations, and 2 patients un-derwent three operations. BNI-P class was significantly lower at each time point after operation than before operation(P<0.05). The rate of effective treatment was 100% at 72 h and 6 months after operation and 97% at 1 yr after operation. Different degrees of dysesthesias at the posterior part of the tongue on the affect-ed side occurred after operation and disappeared at 1 yr after operation. Operation-related nausea and vomi-ting, facial nerve damage, accessory nerve damage and fatality were not observed. Conclusion 3D CT-guided radiofrequency thermocoagulation of the glossopharyngeal nerve provides reliable efficacy and higher safety for the patients with severe recurrent GPN.
7.Effect of Yuganlong Capsule Combined with Entecavir in Treatment of 30 Cases of Patients with Chronic Hepatitis B Cirrhosis

Objective To observe the clinical efficacy of Yuganlong capsule combined with entecavir in treatment of liver fibrosis in patients with chronic hepatitis B cirrhosis. Methods A total of 57 cases of patients with chronic hepatitis B cirrhosis were randomly divided into treatment group(30 cases given entecavir) and control group(27 cases treated with Yuganlong capsule combined with entecavir).Fibroscan values,hepatic fibrosis indexes(HA,LN,C-Ⅳ,PC-Ⅲ),liver function index(ALT,AST,GGT,T-BiL),and ultrasonic imaging index(inner diameters of portal vein,thickness of spleen) were detected. Results Before treatment, all indicatorshad no significant differences between the two groups(P>0.05).After 96 weeks of treatment,the Fibroscan value,HA, LN,C-Ⅳ,ALT,AST and portal vein diameter were significantly reduced in the treatment group as compared with those before treatment(all P<0.05),and there were significant differences between the treatment group and the control group after the treatment (all P<0.05). Conclusion The effect of Yuganlong capsule combined with entecavir in the treatment of chronic hepatitis B liver cirrhosis is better than that of using entecavir alone to improve liver function and liver fibrosis.
8.Clinical value of peroral endoscopic cardial constriction for gastroesophageal reflux disease

Objective To study the safety and efficacy of peroral endoscopic cardial constriction (PECC) for gastroesophageal reflux disease (GERD). Methods A total of 27 patients with GERD who visited the Affiliated Hospital of Inner Mongolia Medical University from August 2016 to December 2017 and met the inclusion criteria received PECC. The 24 h esophageal pH-resistance monitoring ( DeMeester ) scores, GERD health-related quality of life ( GERD-HRQL ) scores, GERD-questionnaires ( GERD-Q ) symptom scores and clinical efficacy before and after operation were compared. Results DeMeester score at 3 months ( Z=-6. 940, P=0. 001) and 6 months ( Z=-6. 307, P=0. 001) after treatment, GERD-HRQL score at 3 months ( t=11. 499, P=0. 001) and 6 months ( t=10. 959, P=0. 001) as well as GERD-Q symptom score at 3 months ( t=30. 647, P=0. 001) and 6 months ( t=27. 217, P =0. 001) after surgery significantly decreased, compared with those before. The curative rates were 81. 5% ( 22/27) and 77. 8%(21/27) at 3 months and 6 months after operation. Conclusion PECC, a new method for the treatment of GERD with good short-term effect, safety and feasibility,can be recommended for clinical application.
9.Safety and Curative Effect of Moxa Box Moxibustion Treatment with FMEA Methods

This paper was aimed to study the safety of moxibustion with FMEA method.Failure mode and effects analysis (FMEA) were used in every aspect of the operation process of moxibustion.And the local skin temperature was measured in 80 patients treated with moxa box moxibustion.The results showed that the skin temperature reached the highest when the moxibustion was given for 15 min,which was in consistence with the patients' chief complaints and their tolerances.It indicated that moxibustion for 15 min was the best moxibustion amount.Meanwhile,inspection should be made to avoid burning.After the application of FMEA,the RPN of the inspection activities,the temperature and distance of moxibustion were significantly decreased (P < 0.05).It was concluded that the application of FMEA management mode strengthened the risk management of moxibustion treatment,standardized treatment process,provided the basis for the temperature and distance of moxibustion,and ensured the safety and efficacy of treatment.
10.The expression of FasL and ICAM-1 on the HEVs in the hypertrophic tonsils.

OBJECTIVE:
To investigate the expression of FasL and ICAM-1 on the HEVs in the hypertrophic tonsils and its clinical significance.
METHOD:
Under the light microscopy, HE and immunohistochemistry methods were used to show the morphology of HEVs, the number of T, B lymphocytes, the expression intensity of FasL and ICAM-1 in 17 patients with the idiopathic tonsillar hypertrophy (ITH) and in 20 patients with the chronic hypertrophic tonsillitis (CHT).
RESULT:
The expression intensity of ICAM-1 on the tonsillar HEVs in ITH group was significantly lower than that in CTH group (P < 0.05), but the intensity of FasL in ITH group was significantly higher than that in CHT group (P < 0.05).
CONCLUSION
FasL and ICAM-1 expression in the tonsillar HEVs contribute to the CHT and ITH. The former mainly makes IHT developed, and the latter promotes the process of the inflammation.
