1.EXPERIMENTAL STUDIES ON THE ANITUMOR EFFECT OF OYSTER EXTRACT

Balb/c mice with HAC murine hepatic cancer were fed with oyster extract at a daily dose of 0. 2 ml (20 mg) for ten successive days. We found that the tumor-caused decreased immunological indicarors including total T cell number, the ratio of helper T cells, the transformation strength induced by mitogens, and NK activity increased markedly. Compared with the control, mice fed with oyster extract showed prolonged survive and their tumor weight reduced significantly. When mice earring human colon cancer were fed with oyster extract at the same dose for 15 days, the tumor size reduced to 44. 6% of the control tumors. The possiblity that oyster extract inhibits tumor growth by enhencing host immune function, especially killer cell activity, was discussed.
2.Expression of P120 catenin mRNA in Non-Hodgkin's lymphoma cell lines.

To investigate p120 catenin mRNA expression in Non-Hodgkin's lymphoma (NHL) cell lines (U937, Raji, Jurkat and Molt4) and normal lymphocytes and explore the relationship between p120 catenin and Non-Hodgkins lymphoma, total RNA sample was extracted by using TRIzol and reversely transcripted into cDNA. Polymerase chain reaction was performed to detect mRNA expression of p120 catenin in NHL cell lines U937, Raji, Jurkat and Molt4. Normal lymphocytes were used as control. It was found expressions of p120 catenin 1A and 3A mRNA were high in above-mentioned NHL cell lines, but neither p120 catenin 1A nor 3A was found in normal lymphocytes as shown by RT-PCR. It is concluded that both P120ctn1A and P120ctn3A mRNA transcripts were found in all NHL cell lines U937, Raji, Jurkat and Molt4 but they don't exist in normal lymphocytes, suggesting p120ctn possibly is of importance in diagnosis and therapy of lymphoma.

3.Preparation of SEREX-defined tumor antigens and preliminary study of their sero-reactivity

Objective:To obtain tumor antigens for study of the sero activity in sera of normal subjects and tumor patients.Methods:Had cloned MY OVA 2,7 and 13 full length genes and expressed the fusion proteins in E.Coli.The proteins were characterized by SDS PAGE and Western Blot.Using bacterially synthesized and purified proteins,74 patients with different kinds of tumors and 13 healthy controls by dot blot were investigated.Results:Expressed fusion proteins at high level,reaching a yield at least 30% of the total bacteria protein.By using affinity chromatography and thrombin digestion,finally gained three purified proteins,which could be applied to serum screening.Dot blot results showed that auto antibodies against MY OVA 2 and MY OVA 7 were detectable in some of tumor and normal samples,while antibody against MY OVA 13 was only detected in tumor patients(5/74) and not detected in normal subjects(0/13).Conclusion:Tumor antigens prepared from genetic engineering can be adopted for effective seroscreening.
4.Hemangioblastic characteristics of human adipose tissue-derived adult stem cells in vivo

AIM: To investigate whether Flk1~+ CD31~- CD34~- cells isolated from human adult adipose tissue have characteristics of hemangioblasts in vivo. METHODS: After sublethally irradiated (300cGy) with a caesium source, the female non-obese diabetic/severe combined immunodeficient (NOD/SCID) mice were injected with human adipose tissue-derived Flk1~+ CD31~- CD34~- cells (10~5 cells per mouse) via tail vain with 0.4 mL Roswell Park Memorial Institute medium (RPMI-1640). The control mice received the same volume of RPMI-1640 medium. All mice were killed 2 months after transplantation for further study. The differentiation potential of Flk1~+ CD31~- CD34~- cells was assessed in bone marrow and gastrointestinal tract by the methods of flow cytometry, RT-PCR, FISH, and triple-color immunofluorescence. RESULTS: Flk1~+ CD31~- CD34~- human adipose tissue-derived adult stem cells differentiated into endothelial cells and hematopoietic cells at the single-cell level in vivo. CONCLUSION: Human adult adipose tissue-derived Flk1~+ CD31~- CD34~- cells bear characteristics of hemangioblast in vivo and may have potential application for the treatment of hematopoietic and vascular diseases.
5.Low-Field-MRI in Assessing the Vessels Surrounding Pancreas Involved by Pancreatic Carcinoma

Objective To study the accuracy,clinical value and limit in assessing the vessels surrounding pancreas involved by pancneutic carcinoma with low field MRI.Methods MRI scanning was performed in 55 patients with pancreatic carcinoma.We prospectively evaluated the MRI findings of vascular involvement by pancreatic carcinoma,and compared with the surgical results.Results Of total 55 cases,50(91%)cases were correctly diagnosed on MRI .MRI carries an accuracy of 86% for prediction of resectility,and accuracy of 93% for unresectility.Conclusion Low field MRI can accurately assess the peripancreative vessels ,but it has limited value in the patients with pancreatic carinomaⅠa orⅠb.
6.A simple and effective auxiliary device for detection of physiologic func-tion on isolated sciatic nerve and gastrocnemius in toads

AIM: To design a simple and effective auxiliary device for detection of the compound action poten-tial (CAP) and the force on the isolated sciatic nerve and gastrocnemius in toads, and to investigate its practicability. METHODS: A simple “L” shape device (L tube), which was composed of a nerve chamber and an organ bath, was made for fixing the isolated sciatic nerve and gastrocnemius.After fixing, the sciatic CAP and the gastrocnemius force were detected by BL-420S data acquisition and analysis system.The specimens were radomly divided into control group, and the lidocaine nerve and muscle groups.The sciatic nerve or gastrocnemius of each lidocaine group was firstly treated with the corresponding concentration of lidocaine, and then washed out with Ringer’s solution, and its reversible anesthetic action on nerve conduction and muscle force was analyzed to verify the practicability of the L tube device.RESULTS: The CAP and the force of the sciatic-gastrocnemius specimens were detected concurrently by fixing the specimens in the L tube, and the liquid in the nerve chamber and organ bath was changed easily.Compared with the control, lidocaine at 0.05 and 0.2 g/L significantly increased the sciatic threshold stimulus voltage and maximal stimulus voltage (P <0.05), prolonged the absolute refractory period (P <0.01), and slowed down the conduction velocity (P <0.01).Lidocaine at 1 g/L complete-
ly blocked the sciatic conduction.The rest tension of the gastrocnemius was increased, and the maximal twitch force was decreased significantly by 1 g/L lidocaine (P <0.01), but the threshold and maximal stimulus voltage did not show statis-tic difference.The parameters of the sicatic nerve and gastrocnemius completely or partially recovered to the control level after washing out.CONCLUSION: As an auxiliary device, L tube makes the detection of CAP and force on the isolated sciatic nerve and gastrocnemius in toads more conveniently.
7.Inhibitory effects of tumor associated mitochondrial protein 12 on HepG2 cell apoptosis

AIM:To explore the inhibitory effects of tumor associated mitochondrial protein 12 (TAMP12) on tumor cell apoptosis. METHODS: (1) A retrovirus expression vector was recombinated and transfected into the packaging cell line PA317. The virus particles were obtained to infect the target cell line HepG2 low expressing of TAMP12. The expression of TAMP12 mRNA was detected by RT-PCR. The subcellular localization and quantification of TAMP12 protein labeled with double fluorescein were observed under confocal laser scanning microscope (CLSM). (2) Hoechst33258 staining and flow cytometry (FACS) were used to analysis the apoptosis of HepG2 cells treated with 5-fluorouracil (5-FU). RESULTS: (1) The CLSM observation showed that TAMP12 protein was mainly expressed in mitochondria of HepG2 cells. The expressions of TAMP12 gene and protein were stable and high in transfected HepG2 cells. (2) Upon treatment with 5-FU, the transfected HepG2 cells showed a fairly integrated nucelus while the control HepG2 cells exhibited chromatin condensation, marginalization and karyorhexix. 8.Prevalence of menopausal syndrome among postmenopausal women in Pan'an County

Objective:
To investigate the prevalence and influencing factors of menopausal syndrome among postmenopausal women in Pan'an County, Zhejiang Province, so as to provide the basis for guiding the health management of postmenopausal women.
Methods:
From May 2023 to April 2024, the postmenopausal women aged 40 to 69 years in Pan'an County were selected using the random cluster sampling method. Demographic information, lifestyle and prevalence of gynecological diseases were collected through questionnaire surveys. The prevalence of menopausal syndrome was assessed by modified Kupperman Score Scale. Factors affecting menopausal syndrome were analyzed by a multivariable logistic regression model.
Results:
A total of 816 postmenopausal women were surveyed, with an mean age of (57.63±2.92) years and a mean natural menopause age of (49.85±2.13) years. There were 574 cases with menopausal syndrome, with a prevalence of 70.34%. Flashes and sweating, insomnia and irritability were common symptoms, accounting for 62.87%, 47.43% and 41.18%, respectively. Multivariable logistic regression analysis showed that monthly personal income of ≤5 000 yuan (<3 000 yuan, OR=3.124, 95%CI: 1.829-5.335; 3 000-5 000 yuan, OR=2.399, 95%CI: 1.370-4.201) and having gynecological diseases (OR=1.970, 95%CI: 1.292-3.004) were associated with a higher risk of menopausal syndrome, while average (OR=0.141, 95%CI: 0.072-0.276) or sufficient sleep quality (OR=0.095, 95%CI: 0.049-0.185) were associated with a lower risk of menopausal syndrome.
Conclusion
The prevalence of menopausal syndrome among postmenopausal women in Pan'an County is relatively high, and is mainly influenced by personal economic status, sleep quality and the presence of gynecological diseases.
9.Research progress of parathyroid hormone replacement therapy in hypoparathyroidism

Hypoparathyroidism is a rare endocrine disease with abnormal calcium and phosphorus metabolism. It is mainly characterized by hypocalcemia,hyperphosphatemia,and with or without decreasing of the parathyroid hormone (PTH). The current standard treatment for hypoparathyroidism includes calcium and vitamin D. Calcium and vitamin D can correct hypocalcemia, but it is not easy to maintain the balance of calcium and phosphorus. PTH hormone replacement therapy has made adequate progress and may become a treatment option for patients with hypoparathyroidism. This review is trying to introduce the progress in PTH hormone replacement therapy.
