1.Methodology research on determination of total arsenic in foods by inductively coupled plasma mass spectrometry based on collided reaction cell technology with microwave digestion

Objective To establish a method for determination of total arsenic in foods by inductively coupled plasma mass spectrometry based on collided reaction cell technology with microwave digestion and provide a new reliable method for accurate quantitative analysis of total arsenic in foods.Methods After microwave digestion,the samples were directly determined by inductively coupled plasma mass spectrometry based on collided reaction cell technology.Internal standard element of Yttrium(89Y) was used to correct matrix interference and signal drifting,high-performance Xs interface was used to reduce non-mass spectrum interferences,and the collided reaction cell technology (CCT mode) was used to eliminate mass spectrum interferences.Meanwhile,arsenic measurement results were compared with the arbitral method.Results The optima 1 linear range of the method standard curve was 0-40 μg/L with a correlation coefficient of 0.9998.The detection limit of the method,the lowest quantitative limit,the relative standard deviation and the recovery rate were 0.0025 mg/kg,0.082 mg/kg,1.5%-4.2% and 95.0%-105.0%,respectively.The detection results of the three standard reference materials were satisfactory.Compared with the arbitral method,there was no obvious difference (P < 0.05).Conclusions The method not only has lower detection limit,but also is simple,fast and more accurate.The results are consistent with those by arbitral method,and the new method is available for accurate quantitative analysis of total arsenic in foods.
2.Analytical quality control of determination of arsenic in water by hydride generation atomic fluorescence spectrometry

Objective To implement analytical quality control(AQC) and predict analysis error of arsenic determination in water by hydride generation atomic fluorescence spectrometry,enhancing the technical level of testing arsenic and lab quality control capability,and to ensure the accuracy and reliability of test data.Methods According to the AQC procedure of water quality analysis for evaluating precision and accuracy of trace elements,arsenic content was measured in a blank solution(3% hydrochloric acid solution),two kinds of standard solution (0.1C,0.9C arsenic standard solution,the C was the upper limit of measuring scope of the analytical method,and in this experiment the C was 20.0 μg/L),natural water sample and standard quality control sample(5.0 μg/L) with hydride generation atomic fluorescence spectrometry corresponding to GB/T 5750.5-2006.Results Of the five kinds of intra-samples and inter-assay,the difference was not significant statistically (F < F0.05,all P > 0.05).Total standard deviation (St) of every solution was less than 5% of the concentration,the confidence limit of total average rate of recovery(R/d =0.99) was in a allowable range.The proficiency of testing samples were ZWAs =1.04 and ZBAs =1.12.The blind sample testing result was satisfactory.Conclusions Arsenic in water is determined by hydride generation atomic fluorescence spectrometry,and the analytical quality control is adopted by precision-bias analysis and accuracy quality control procedure.Ensuring that the error results are in a controlled range and highly reliable analytical results are obtainable.
3.Diagnostic value of random spot urine albumin/creatinine ratio in women with hypertensive disorders complicating pregnancy

Objective To explore the correlation of random spot albuminuria to creatinine ratio (ACR) and 24 h uri?nary protein, and the diagnostic value of ACR in women with hypertensive disorders complicating pregnancy (HDCP). Meth?ods A total of 584 women with HDCP (169 with gestational hypertension, 205 with mild pre-eclampsia, 173 with severe pre-eclampsia and 37 with chronic hypertension) and 2 038 normal pregnant women were included in this study. Changes of ACR, 24-h urinary protein and other biochemical indicators were compared between groups. Bivariate correlation analysis was used to analyze the relationship between ACR, 24-h urinary protein and other biochemical indicators. Receiver operat?ing characteristic curve (ROC) was used to analyze the sensitivity, specificity and optimal cut off value of ACR for predicting HDCP. Results (1)There were significantly higher levels of ACR, 24-h urinary protein, blood urea nitrogen (BUN), serum creatinine (Cr), uric acid (UA), total cholesterol (TC) and low density lipoprotein cholesterol (LDL) and significantly lower val?ues of platelet (PLT), serum albumin and total protein in women with severe preeclampsia than those of other groups ( P <0.05). (2) There were positive correlation between ACR and 24-h urinary protein, BUN, Cr, UA, D-dimer, TC, LDL (P <0.05). (3) The optimal cut off value of ACR for predicting gestational hypertension, mild preeclampsia, severe preeclampsia (24-h urinary protein≥2 g,≥5 g) were 1.44, 10.48, 39.84, 94.91 g/mol separately. Conclusion There is positive correla?tion between ACR and 24 hour urinary protein. ACR can be used for diagnosing and estimating the severity degree of hyper?tensive disorders complicating pregnancy.
6.The damages of adrenal gland in rabbits with neisseria memingococcal septic shock

AIM: To investigate the damages of adrenal gland in rabbits infected with neisseria meningococci(MC).METHODS: 18 rabbits were injected with MC (1?10 11 organism/kg, iv, model group, n=10 ) or normal saline (control group, n=8 ).TNF-? was detected in baseline (before challenged), 60 min and 120 min after injection. 120 min after injection, TNF-? immunochemical localization of adrenal gland was detected with ABC methods by electron microscopy. RESULTS: In model group, TNF-? positive cells were located in medulla and TNF-? released into simus of medulla. The medulla damages were appeared but gland cells of cortex was normal. No damages were showed in control group. TNF-? value of plasm was elevated at 60 min and 120 min after injection compared with control group. CONCLUSION: TNF-? was released into medulla and resulted in medulla damages after MC challenging. Adrenal cortex didnt damaged in early phase of MC septic shock. [
7.The expression of Endophilin II and VEGF-C in human laryngeal cancer and connection with clinical treatment.

OBJECTIVE:
To study the expression of Endophilin II and VEGF-C in human Laryngeal cancer and its relationship with the clinical treatment effect.
METHOD:
The expression and distribution of Endophilin II and VEGF-C protein were checked in 47 patients with laryngeal carcinoma in our hospital in 2006-09-2009-10 and 23 samples of normal tissues around the cancer by immunohistochemical staining SP method. After an appropriate surgical treatment, patients accepted a 5-year followup to evaluate their recurrence situation and 5-year survival rate.
RESULT:
Immunohistochemistry showed that Endophilin II protein was strongly positive expressed in normal tissues around the cancer (positive rate 82.61%). And the expression level was dependent on the lymph metastasis, T stage and lymph invasion (P < 0.05). Patients with positive Endophilin II expression had a higher survival rate and better recurrence situation. Expression of VEGF-C was significantly higher in laryngeal carcinoma than in normal tissue around the cancer (positive rate 51.06% and 13.04%). the expression level was dependent on the lymph metastasis and lymph invasion (P < 0.05), and negatively correlated with recurrence and 5-year survival rate.
CONCLUSION
Endophilin II and VEGF-C may involve in laryngeal cancer, and the expression of Endophilin II and VEGF-C has a positive correlation with pathological features and lymph metastasis, and both are related with recurrence and 5-year survival rate. So these two proteins are likely to be a potential target for diagnosis and treatment of laryngeal cancer.

8.Comparison of Dynamics in Viremia Levels in Chickens Inoculated with Marek's Disease Virus Strains of Different Pathotypes

One-day-old unvaccinated chicks were inoculated with serotype I virulent strain GA and vaccine strain CVI988, serotype III vaccine strain HVT o f Marek's disease viruses(MDV). By using an indirect immunofluorescence assay of suspensions of peripheral blood mononuclear cells(PBMC) with monoclonal antibo dies(MAb) BA4 and BD8 specific to glycoprotein B of MDV, The dynamic course s of vir emia levels for each of 3 strains from 4 days to 45 days postinfection (PI) were determind. Viremia for virulent strain GA-infected chickens were detected from 4 days PI until days before death of infected chickens, it got its peak at abou t 14 days PI. Viremia in serotype 1 vaccine CVI988-inoculated chickens were mea sured between 4 to 22 days PI, it could be detected from 4 days PI and ended at 18 days PI, it got its peak at 8 days PI. For serotype 3 vaccine HVT-inoculated chickens, viremia could be detected beween 4 to 14 days PI, its peak happened a t about 6 days PI. Such IFA with MAbs BA4 and BD8 could be used to evaluate qu al ity of vaccination process in chicken flocks. The differential dynamic courses o f viremia for different strains may be used to diagnose virulent MDV infection i n vaccinated flocks. The viremia levels measured by IFA with the MAbs and co-cu ltivation assay for plague forming unit(PFU) were compared. The results indicate d that IFA assay of PBMC suspension was more sensitive and specific than co-cu ltivation assay, i.e. viremia levels measured by IFA were 30 to 100-fold higher than that by plague-forming assay in CEF for the same sample.
9.Safety and efficacy of primary closure in patients with normal diameters of common bile duct after laparoscopic common bile duct exploration

Objective:To study the safety and efficacy of primary closure of non-dilaed common bile ducts (CBD) after laparoscopic common bile duct exploration.Methods:A retrospective study was conducted on the clinical data of 120 patients who underwent primary closure of CBD after laparoscopic CBD exploration (LCBDE) at the Shengjing Hospital of China Medical University from April 1, 2014 to November 30, 2019. There were 44 males and 76 females, aged 22.0 to 88.0 years, (average age of 57.2 years). These patients were divided into the dilated CBD group (diameter of CBD >8 mm) and the normal CBD group (diameter of CBD ≤8 mm). The following factors, including operating time, intraoperative bleeding, postoperative hospitalization stay, abdominal drainage tube indwelling time and postoperative complications were compared.Results:The dilated CBD group consisted of 76 patients, with 25 males and 51 females, and a median age of 62.5 years. The normal CBD group consisted of 44 patients, with 19 males and 25 females, and a median age of 57.5 years. There were no significant differences in gender, age, albumin level, total bilirubin and other baseline data between groups ( P>0.05). The operation times of the two groups were: the normal CBD group [106.6(87.3, 146.3] min vs the dilated CBD group [112.0(90.5, 134.5)] min; intraoperative blood loss [the normal CBD group 20(10, 30)ml vs dilated CBD group 20(10, 20)ml]; postoperative hospital stay [the normal CBD group 7.0 (5.3, 9.0) d vs the dilated CBD group 7.0 (5.0, 7.0) d]; and postoperative abdominal drainage tube removal time [the normal CBD group 6 (4, 7) d vs the dilated CBD group 5 (4, 6)d]. The differences were all not significant ( P>0.05). There was also no significant difference in the incidences of postoperative complications (including bile leakage, biliary stricture, and stone recurrence between groups, all P>0.05). Conclusion:In patients with a normal diameter (≤8 mm) common bile duct, it was safe and efficacious to perform primary closure after laparoscopic common bile duct exploration.
