OBJECTIVE:To establish a method for the content determination of berberine hydrochloride in Fufang huangge ji-angtang tablet. METHODS:UPLC was performed on the column of Acquity UPLC BEH C18 with mobile phase of acetoni-trile-0.1% H3PO4(21∶79,V/V)at flow rate of 0.3 ml/min,the detection wavelength was 345 nm,column temperature was 30℃, and the volume injection was 1.0 μl. RESULTS:The linear range of berberine hydrochloride was 0.008 16-0.081 6 μg(r=0.999 9);RSDs of precision,stability and reproducibility were lower than 1%;average recovery was 97.05%-98.96%(RSD=0.66%,n=6). CONCLUSIONS: The method is simple,precise,stablity and reproducible,and can be used for the quality control of Fufang huangge jiangtang tablet.

Object To study the correlativity between the in vivo release of healthy volunteers and the in vitro dissolution of XIANGHE SUPPOSITORY(XS). Methods The electromagnetic stirring method was used to determine the in vitro dissolution of XS, and the in vivo release was got by the indirect method, assaying the content of the left XS after it had released in administering system, HPLC was used for the determination of XS with the index of variant content of berberine hydrochloride. Results A good correlativity was shown between the percentage of in vivo release and in vitro dissolution of XS. Conclusion In vitro dissolution test under the given conditions could indicate the in vivo absorption of XS.

A field study on 24 weigbt--lifters and an experiment on rats imitating kuman acute food restrictionwere done in order to find out the metabolism of water and electrolytes during the acute weight-reductionperiod and the effect of appropriate supplementation. The results showed that food restriction caused very significant loss of the water and electrolytes withsigns of muscle cramp, dehydration, ketonuria etc. The experimental results in rats were observed duringfood restriction as following: 1. Serum potham and sodium decreased and the serum chloride and magnesium increased significantly. 2. Potassium and sodium contents of cardiac muscle were significantly lower than those of the control group,but potassium content of skeletal muscles was higber. 3. Blood lactic acid (3 minutes after running on a treadmill for half an hour ) was higber than thatbefote food restriction. Supplementation of appropriate minerals is beneficial to correct the above changes and keep the athletesfit and in good form.

BACKGROUND:Hip and knee replacement is a common surgery in the clinic; the procedure is relatively complex; and the risk of surgery is relatively high, so that postoperative analgesia is not satisfactory. Perioperative pain management has been a clinical concern. To find safe and effective analgesia has become one of the important tasks of joint surgeons. OBJECTIVE:To investigate the effect of celecoxib on multi-mode analgesia after hip and knee replacement. METHODS: A total of 80 cases undergoing hip and knee replacement in the Chongqing Dongnan Hospital from September 2012 to September 2013 were enroled in this study. These patients were randomly divided into two groups. In the control group, celecoxib was not used for analgesia after replacement. In the experimental group, celecoxib was used after replacement. The pain was observed at 1-5 days after surgery in the two groups. When the analgesic pump was removed, the drug dosage and opioid analgesics use were recorded. Side effects of drug use were also recorded. RESULTS AND CONCLUSION:In terms of analgesic efficacy, the analgesic effect was better in the experimental group than in the control group (95%, 85%, P < 0.05). 95% patients in the experimental group were satisfied with the analgesia, which was significantly higher than in the control group (65%;P< 0.05). No significant difference in pain visual analogue scale score was detected between the two groups immediately, 4 and 5 days after surgery (P > 0.05). Pain visual analogue scale score was significantly lower in the experimental group than in the control group at 1, 2 and 3 days post surgery (P< 0.05). The drug dosage was significantly more in the experimental group than in the control group after surgery (P < 0.05). The frequency of opioid use in the experimental group was significantly lower than in the control group (P < 0.05). The complication rate was significantly lower in the experimental group (8%) than in the control group (18%) (P < 0.05). These findings demonstrate that the analgesic effect of celecoxib was ideal after hip and knee replacement using multi-mode analgesia, which can reduce the dose of analgesic drugs and have smal adverse reaction.

Objective To sonographically measure nuchal skin thickness(NT) and biparietal diameter/femur length (BPD/FL) for normal fetuses between 10 and 24 gestational weeks,screen abnormal cases then to culture fetal cells and chromosome analysis, diagnosis aneuploid chromosome deformed fetuses Methods Ultrosonic estimation of fetal gestation according to gestational sac or CRL or BPD/Fl and AC, measuring NT, BPD/Fl of 100 cases per gestational weeks, drawing normal curve of NT, making curve of 90 th and 10 th NT and BPD/Fl. Restropectively analysis 5 cases trisomy 21.Results The average of NT was 1 76mm between 10 and 13 gestational weeks. The average of NT was 3 52mm between14 and 24 gestational weeks. The value of 50th was 2 40～1 40 between14 and 24 gestational weeks.Conclusions If the value of NT is more than 90 percent of normal fetuses, and if BPD/FL ratio is morn than 90 percent of normal fetuses,the sensitivity of trisome 21 should increase.Combining factors as elderly and high risk parturition, the sensitivity of trisomy 21 is much more increasing.

Objective To study the expressi on of thrombomodulin (TM) in endometrial carcinoma and explore its relation to the infiltration degree and histological grading of endometrial carcinoma. Methods The expression of TM was detected by immunohistochemistry in 50 cases of endometrial adenocarcinoma, 10 cases of endometrial complex hyper plasia and 10 cases of normal endometrium. Results The posi tive rate of TM in endometrial carcinoma was significantly higher than that in c omplex hyperplasia and normal endometrium (P 0.05). Conclusion TM can be used as a new index for judging progre ssion and prognosis of endometrial carcinoma.

We wished to sequence the full-length genomes of the DHL10M110 strain of the Akabane virus (AKV) isolated from mosquitoes in Yunnan Province, China, in 2010. We also wished to analyze the characteristics of these complete nucleotide sequences. The complete genomic sequence of the DHL10M110 strain from Yunnan Province was obtained by reverse transcription-polymerase chain reaction and direct sequencing. We found that the length of the L, M and S gene nucleotide sequences of the DHL10M110 strain were 6 869-bp, 4 309-bp and 856-bp, respectively, including the open reading frame (ORF) nucleotide sequences of 6 756-bp (L), 4 206-bp (M) and 702-bp (S), encoding 2252, 1402 and 234 amino-acid polyproteins, respectively. Phylogenetic analyses based on L-fragment ORF showed that the DHL10M110 strain had a close relationship with the OBE-1 strain of the AKV from Japan and AKVS-7/SKR/2010 strain of the AKV from South Korea. Phylogenetic analyses based on M- and S-fragment ORF showed that the DHL10M110 strain had a close relationship with the epidemic strains of the AKV from Japan, South Korea and Taiwan, but that the DHL10M110 strain had a lone evolutionary branch. In terms of nucleotide (amino acid) homology, the similarity of L-, M- and S-fragment ORFs of the DHL10M110 strain to the OBE-1 strain from Japan was 92.6% (98%), 88.5% (94%) and 96.4% (99.1%), respectively. When comparing the DHL10M110 strain with the OBE-1 strain, we noted 45, 84, and 2 different sites in the amino acids of L, M and S fragments, respectively. Homology and phylogenetic analyses also suggested that the DHL10M110 strain had a distant relationship with the epidemic strains of the AKV from Kenya and Australia. Also, we confirmed by complete genomic sequence analyses that the DHL10M110 strain was clade-Asia of the AKV. However, differences between the DHL10M110 strain compared with strains from Japan and South Korea were also noted. These results suggest that the DHL10M110 strain harbored relatively stable genetic characteristics and distinct regional features. This is the first time that full-length genomic sequences of the DHL10M110 strain of the AKV in mainland China have been obtained.

Objective To investigate the regulation of the immune function of cytokine-induced killer cells(CIK) by dendritic cell (DC) vaccine in the patients with chronic hepatitis B(CHB) in vitro.Methods CIK cells from 30 patients with CHB were cultured in vitro,and were randomly divided into two groups,DC vaccine-treated group and the control group.After 14 days of culture,the percentages of CD3 + CD4+ T,CD3 +CD8 +T and CD3 +CD56+ T cells among CIKs were analyzed by flow cytometry.The levels of IL-12,IFN-γand IL-6 in cell culture supernatant was detected by ELISA.Results The percentages of CD3 + CD4 + T,CD3 +CD8+T and CD3+ CD56+ T cells were 18.27％,64.36％ and 20.00％ in CIKs in DC vaccine group,and 17.79％ ( P ＞ 0.05 ),54.69％ ( P ＜ 0.01 ) and 13.39％ ( P ＜ 0.01 ) in the control group,respectively.The perentage of CD3 + CD4 + T cells were similar between the two groups ( P ＞ 0.05 ),but for the perentage of CD3 + CD8 +T and C D3 + CD56 + T cells were significantly different between the two groups (t =4.130 and 5.601respectively,Ps ＜ 0.01 ).The concentrations of IL- 12,IL-4 and IFN-γin supernatant were ( 177.82 ± 130.06),(31.77 ± 9.52) and (86.99 ± 56.30) ng/L in DC vaccine-treated group respectively,which were significantly different from those of (80.83 ±50.15) ng/L (t =3.811,P ＜0.01 ),(40.33 ± 19.74) ng/L( t =2.141,P ＜0.05) and (42.07 ± 19.68)ng/L(t =4.125,P ＜0.01) in the control group,respectively.Conclusion DC vaccine could enhance the killing function of CIK cells.
